2.NSAIDs induced apoptosis of gastric cancer cell lines and the mechanism

Purpose:To investigate whether NSAIDs can induce apoptosis of gastric cancer cell lines,to observe the effect of different p53 genotype on NSAIDs induced apoptosis,to elucidate the regulation of NSAIDs on expression of apoptosis related genes. Methods:The anti-proliferative effect of NSAIDs was measured by MTT assay.Apoptosis was determined by acridine orange(AO) staining,Annexin-V/PI double staining,laser scanning cytometry(LSC) and flow cytometry (FCM).Alteration of bcl-2 and bax genes was detected by reverse transcription polymerase chain reaction (RT-PCR).Protein expression was determined by Western-blotting.Results: Indomethacin (Indo) and Aspirin (Asp) inhibited both AGS(wild-type p53)and MKN28(mutant p53) gastric cancer cell lines growth in a time/dose dependent manner.AGS cell line was more sensitive to NSAIDs,which apoptosis percentage was significantly higher than MKN28 under the same condition. The percentages of apoptosis of MKN28 were somewhat higher among NSAIDs treated groups compared with the normal control group,but these slight differences were not statistically significant. The bax mRNA kept increasing since NSAIDs treatment accompanied by a decrease of bcl-2 gene.The Bax protein increased after treatment while the Bcl-2 protein was undetectable, which tendency was more obvious during 6-24hs.Conclusion: Both Indo and Asp could induce apoptosis in gastric cancer cell lines,which adds further theoretical foundation to the anti-cancer use of NSAIDs.NSAIDs could not induce notable apoptosis of MKN28,which indicated the mutant p53 gene perhaps blocked NSAIDs induced apoptosis.One of the major pathways that mediated the anti-tumour response of NSAIDs in gastric cancer cells was through up-regulation of bax and down-regulation of bcl-2 genes and/or proteins.[
3.Screening of proteins binding with HBV X promoter by phage display technique from human liver cDNA phage library

Objective To screen the proteins binding with HBV X-promoter and to investigate their potential role in the regulation of replication and expression of HBV DNA. Methods By using HBV X biotinylated promoter DNA as the selective molecule, the T7 select human liver cDNA library was biopanned and positive clones were selected. After screening, positive plaques were performed to amplify for inserted DNA fragment and cloned into pGEM-Teasy vector. Thirty positive plaques were chosen for DNA sequencing. Results Five kinds of known and nine kinds of novel cDNA sequences were obtained. The 5 kinds of known ones are human serum albumin, NADH dehydrogenase subunit 4, prokininase, ubiquitin specific protease 10, and testis enhanced gene transcript. Conclusion The binding proteins of HBV Xp DNA were identified by phage display. The results suggest that this approach provides a new search tool for the study of replication and expression mechanism of HBV DNA.
4.Evaluation of protective effectiveness of two tuberculosis DNA vaccines in relation to pathological changes in lung, liver and spleen

Objective To evaluate the protective effectiveness of MPT64 and ESAT6 DNA vaccines against M. tuberculosis. Methods BALB/c mice were randomly divided into 14 groups and subjected to following treatments respectively, i.e. immunized with. ESAT6 (25?g)+MPT64 (25?g)(A), ESAT6(100?g)+IFN-?(100?g) (B), ESAT6 (75?g)+MPT64 (25?g)(C), ESAT6(100?g)+IL-12(100?g) (D), MPT64(100?g)+IL-12(100?g) (E), ESAT6 (25?g)+MPT64 (75?g)(F), MPT64 (100?g)(G), Pvax1 (H),ESAT6 (100?g)(I), ESAT6 (100?g)+MPT64 (100?g)(J), ESAT6 (50?g)+MPT64 (50?g)(K), MPT64(100?g)+IFN-?(100?g)(L), BCG(M ), and saline(N). Then they were infected with M. tuberculosis H37Rv via intravenous route. The pathological changes in the lung, liver, and spleen were observed after the infection. Results Eight weeks after the inoculation, there were only alveolar exudation and capillary hyperemia in the lung lesions in the mice of group N. In the mice of group M and J, main pathological changes included tuberculous granulomas consisting of numerous lymphocytes, macrophages, epithelioid cells and Langhans giant cells, and moderate hyperplasia in alveolar walls. The lung lesions of the other groups were similar, and both hyperplasia and exudate were found (A, B, C, D, E, G, H, I, K, L). No necrosis was found in all the above groups. There were hyperemia, dense lymphocytes infiltration in the portal area, and tuberculous granuloma in the liver in all the groups. No difference was found among all the groups. The pathological changes in spleen induced hyperplasia and fusion of splenic lymph nodule. The reactions in group M and J were stronger than that of the other groups. Conclusions MPT64 and ESAT6 DNA vaccines from M.tuberculosis could enhance immunity against M. tuberculosis, either they were used in combination with different dosages or with IL-12 or IFN-?. The vaccine used in group J showed the strongest effect in enhancing immunity, almost reaching that of combined use of BCG, IFN-? and IL-12.
5.A study on repetitive nerve stimulation test in myastheula gravis

Objective To study the diagnostic value of repetitive nerve stimulation test in myasthenia gravis. Method The repetitive nerve stimulation test was performed at axillary nerves, ulnar nerves and facial nerves in 36 patients with myasthenia gravis which treated in our hospital from 1996. 10 to 1999. 10. 108 nerves were tested. Result The RNS test positivity rate of amplitude decrement in 108 nerves was 60. 2% ( 65 nerves). Among them, the highest(77. 8% )positivity rate was deltoid mucle supported by axillary nerve, the lowest(41.7% ) was abductor digiti minimi mucle supported by ulnar nerve. There were 31 cases(86. 1% ) had one or more than one nerves with positive results in 36 patients. The best stimulation frequancy were 3 ~ 5 Hz. Conclusion Stimulate the axillary nerve, facial nerve and ulnar nerve in the same patient, would improve the RNS positivity rate in diagnosis of MG.
6.Comparison of Effects of Platelet Rich Fibrin and Concentrated Growth Factor on Schwann Cells

Objective To compare the effects of platelet rich fibrin(PRF)and concentrated growth factors(CGF)using Schwann cells as a pe?ripheral nerve model. Methods A total of 10 healthy volunteers aged 18 to 55 were randomly selected,and 10 mL venous blood was collected un?der aseptic conditions to prepare PRF and CGF. The cells were randomly divided into three groups,control group,PRF group and CGF group. The cell morphology was observed by inverted phase contrast microscope. The cell proliferation was analyzed by MTT assay. The secretion of nerve growth factor in supernatant was detected by enzyme?linked immunosorbent assay. Cell cycle was detected by flow cytometry. Results There was no significant difference of the morphology of cells in each group as observed under inverted phase contrast microscope. MTT results showed that the absorbance values of PRF group and CGF group were significantly higher than those of the control group(P<0.05). The secretion of nerve growth factor in the supernatant were significantly increased(P<0.05). The number of cells in S+G2M phase was significantly increased(P<0.01),but there was no significant difference between PRF group and CGF group. Conclusion Both PRF and CGF can promote proliferation of Schwann cells and increased the amount of nerve growth factor secretion ,but there is no significant difference between PRF and CGF in terms of improving cell proliferation and promoting nerve growth factor secretion.
7.The influence of whole nursing model on health-related knowledge awareness and satisfaction rate of patients with liver hydatidosis receiving pericystectomy

Objective To observe the influence of whole nursing model on health-related knowledge awareness and satisfaction rate of patients with liver hydatidosis receiving pericystectomy.Methods 180 patients in hospital receiving resection of liver hepatic cyst were randomly divided into the observation group and the control group with 90 patients in each group.The control group was given usual care,while the observation group received whole treatment model.The patients' health-related knowledge awareness and satisfaction rate were compared between two groups.Results The rate of health-related knowledge awareness and satisfaction about nursing work in the observation group preceded the control group.Conclusions This whole treatment model can boost the awareness rate of health-related knowledge and nursing satisfaction,which not only provides the care with full,comfortable and efficient services,but also advances the self-care capacity and cognition of disease of patients.
8.Correlation of tumor growth and endothelial progenitor cells entering blood induced by surgical injury

Objective To explore the correlation of tumor growth and endothelial progenitor cells (EPC) entering blood induced by surgical injury in tumor bearing nude mice. Methods Forty-two tumor bearing nude mice were randomly divided into seven groups (n=6): non-surgical injury groups (1 d and 30 d), anesthetic group, surgical injury groups (24 h, 48 h, 72 h and 30 d after surgery). Blood samples and xenograft tumor tissues were taken from anesthetic group 24 h after anaesthesia and surgical injury groups 24 h, 48 h, 72 h and 30 d after surgery. EPC levels in peripheral blood were measured by flow cytometry, serum VEGF levels were determined by ELISA, microvessel density (MVD) and expression of VEGF were detected by immunohistochemistry. Results The levels of EPC in 24 h post-surgery group, 48 h post-surgery group and 72 h post-surgery group were significantly higher than that in non-surgical injury 1 d group (P<0.05). The levels of VEGF in 24 h post-surgery group, 48 h post-surgery group, 72 h post-surgery group and anesthetic group were significantly higher than that in non-surgical injury 1 d group (P<0.05). There was no significant difference in MVD among groups (P>0.05). Pearson correlation analysis revealed that serum VEGF levels were related to EPC levels in peripheral blood (r=0.695 6, P<0.01), while EPC levels in peripheral blood were not related to MVD (r=0.221 4, P>0.05), and serum VEGF levels had no correlation with MVD (r=0.224 9, P>0.05). Conclusion Surgical injury has no obvious influence on xenograft tumor growth.
9.Application of Mobile Ultrasonoscope in Earthquake Rescue

Objective To evaluate the application of mobile ultrasonoscope in earthquake rescue. Methods According to the characteristic of environment and traumatic conditions in earthquake area, the mobile ultrasonoscope was used to diagnose for clinicians, and the advantages and disadvantages were evaluated. Results The mobile ultrasonoscope played an important role in routine diagnosis, such as organic injury and calculus, superficial organellae injury, soft tissue injury, injury of limbs and blood vessels, gynaecology and obstetrics and so on. As far as certain conditions were conferred in which the mobile ultrasonoscope was seldom used to diagnose in common practice, it played a dramatic role too in bone fracture, joint damage, craniocerebral injury and foreign objects. Conclusion In routine diagnosis or certain conditions, the mobile ultrasonoscope is very practical, especially in earthquake rescue.
10.Systematic review on the effect of flipped classroom on medical courses in undergraduate medical education

Objective:To systematically review the teaching effects of flipped classroom (FC) in undergraduate medical education in China.Methods:Such databases as CNKI, VIP Data, and WanFang, were electronically searched for literature of randomized controlled trials (RCTs) on FC versus lecture-based learning (LBL) applied in medical education in China up to March, 2018. The included literature were evaluated for the quality, and RevMan5.3 software was used for Meta-analysis.Results:A total of 26 RCTs were included in the study, including 4 249 students, of whom the FC group had 2 135 cases, while LBL group had 2 114 cases. The students in FC group got significantly higher scores than those of the students in LBL group in the theoretical scores (WMD = 7.81, 95%CI = 5.18 -10.43, P < 0.05), skills tests (WMD = 4.69, 95%CI = 0.65 -8.73, P < 0.05). Conclusion:The FC can significantly improve the teaching results in undergraduate medical education.
