1.Making energetic efforts for progress in targeted prevention and control of endemic fluorosis

Endemic fluorosis is a generalized epidemic disease with complicated types of endemic areas and difficult to prevent and control in China.After several decades of efforts made,the situation in prevention and control of endemic tluorosis in China is in the best historical period.We should take the opportunities and make energetic efforts for targeted prevention and control of endemic fluorosis in order to completely implement intensive management of endemic areas with fluorosis and ensure sustainable prevention and control of the disease,and finally achieve the goal of complete control and elimination.This article analyzes and looks ahead in some aspects of targeted prevention and control of endemic fluorosis such as ideas,goals,conditions,supporting measures and so on.
2.The effects of arsenic on histone modifications

Arsenic is one of the important metalloid elements, which is widely distributed in nature. Arsenic can cause many health hazards. However, the toxic mechanisms of arsenic have not yet been fully understood. In recent years, many studies have shown that epigenetic regulation and control mechanism is associated with arsenic toxicity. Histone modification is an important epigenetic regulation and control mechanism, changes in its patterns may be one of the mechanisms by which arsenic alters the gene expression. We reviewed the effects of arsenic on regulation of histone methylation, acetylation, phosphorylation and ubiquitination.
3.Observation on Long-term and Short-term Therapeutic Effect of Acupuncture Therapy to Chronic Mesenteric Lymphadenitis

[Objective] To assess the curative effect of acupuncture therapy to chronic mesenteric lymphadenitis. [Methods] Al 120 children patients are ran-domly divided into two groups, 60 cases each. Control group is treated with cefuroxime sodium for infections, and treatment group with acupuncture at Sifeng(EX-UE 10), Neiguan (PC06), Zhongwan(RN 12),Zusanli(ST 36) and etc. [Results] Markedly effective rate of treatment group was 91.7%,and total effective rate was 98.3%; markedly effective rate of control group was 86.7%, and total effective rate was 95%, which showed significant difference in curative effects.The two groups symptoms were decreased compared with before treatment, the difference was significant( P<0.01);after treatment, symptom scores were lower than the control group, with significant difference. The recurrence rate of treatment group after three months was 3.6%. The recurrence rate of treatment group after six months was 1.9%, evidently lower than control group ’s 57.7%,54% respectively. [Conclusion] Acupuncture therapy to chronic mesenteric lymphadenitis has unequivocal short-term effect, a stable long-term effect, and low recurrence rate,deservers clinical applications.
4.The three-year campaign for prevention and treatment of endemic diseases to help people in endemic areas get rid of poverty in an all-round way and move towards a well-off society

This paper systematically introduces the progress and main practices of the three-year campaign for prevention and treatment of endemic diseases, discusses the effectiveness of prevention and treatment of endemic diseases in improving the health level of people in endemic areas and getting rid of poverty, heading for a well-off life in an all-round way, and demonstrates the experience of prevention and treatment of endemic diseases in China and its far-reaching influence on the process of consolidating the achievements of poverty alleviation and realizing the strategy of rural revitalization.
5.Expression of cytokines in the degenerative cartilages and synovial tissues of osteoarthritis patients

BACKGROUND:The inflammation of osteoarthritis is mediated by chondrocytes and the synovial tissue-secreted cytokines. Articular cartilage and synovial tissue contains a variety of cytokines, which play important regulatory role in the repair of articular cartilage injury.
OBJECTIVE:To analyze the relationship between chondrocytes, synovial tissue-secreted cytokines and osteoarthritis, as wel as the effect of chondrocytes and synovial tissue-secreted cytokines on osteoarthritis.
METHODS:A computer-based search was conducted in Wanfang database (www.wanfangdata.com.cn), PubMed database (www.ncbi.nlm.nih.guv/pubmed) by the first author from 2005 to 2010. The key words were“osteoarthritis, degeneration, cartilage tissue, cytokines”. A total of 146 articles were obtained through computer search, then reading the titles and abstracts for initial screen, 86 articles with the unrelated research purposes were eliminated, 40 articles with the repetitive content were excluded, and final y 21 articles related to the effect of various cytokines in the degenerative cartilage and synovial tissue of the osteoarthritis patients were included for further analysis.
RESULTS AND CONCLUSION:The cytokines mainly refers to the activated immune cel s and some stromal cel-secreted non-specific regulate immune responses and inflammatory responses-mediated smal molecule proteins, including lymphokines produced by the lymphocytes, as wel as the single nuclear factor and other cytokines produced by monocytes macrophages. Synovial cel-secreted cytokines may partial y explain the pathological process of osteoarthritis and play an important role in joint inflammation. Although, more and more scholars have pay attention to the effect of synovial cel s and cartilage cel-secreted cytokines, but mainly studies the effect of exogenous cytokines on chondrocytes or synovial cel s, and the effect of endogenous cytokines in the pathogenesis of osteoarthritis has not been extensively studied.
6.Effects of human bone marrow fibroblastoid stromal cell line HFCL on the biological characteristics of migrating and homing in multiple myeloma cell line RPMI8226

Objective To investigate the effects of human bone marrow fibroblastoid stromal cell lines HFCL on the proliferation .migrating and homing of multiple myeloma cell lines RPM18226. Methods The co-culture system of RPMI8226 and HFCL was established through cell culture in vitro, growth curves were determined by trypan blue exclusion, cell cycle and expression of adhension molecule CD49d were detected by flow cytometry, and expression of CXCR4 gene was examined by RT-PCR. Results The proliferation of RPMI8226 cells in direct contact with HFCL cells group was inhibited strongerly than that in transwell group. The percentage of G1 phase cells of RPMI8226 cells in direct contact with HFCL group was higher than that of RPM 18226 in transwell group, while the percentage of S phase cells was lower. The expressions of CD49d and CXCR4 in RPMI8226 cells were down-regulated by HFCL cells, and that in transwell group was higher than that in direct contact with HFCL cells group. Conclusion Human bone marrow fibroblastoidss tromal cell HFCL can inhibit the proliferation and the expressions of CD49d and CXCR4 of multiple myeloma cell line RPMI8226, and can prevent multiple myeloma cells from migrating and homing.
7.A retrospective analysis of the assessment results of arsenide quality control in laboratories of national endemic diseases prevention and control institutions

Objective:To analyse the quality control assessment results of arsenic testing laboratories of national endemic disease prevention and control institutions from 2006 to 2020, so as to further strengthen the quality control of arsenic determination in laboratories of national endemic disease prevention and control institutions, and to ensure the accuracy and reliability of national endemic arsenic poisoning monitoring data.Methods:The water arsenic and urinary arsenic test results of arsenic quality control laboratories from 2006 to 2020 were collected and evaluated by Z-ratio scoring method(│Z│≤2 was qualified, 2 < │Z│ < 3 was basically qualified, │Z│≥3 was unqualified), and the qualification rates of water arsenic and urinary arsenic in different years and different provinces were calculated.Results:From 2006 to 2020, the number of laboratories participating in water arsenic quality control in 14 provinces has increased from 25 to 167, and the number of laboratories participating in urinary arsenic quality control increased from 22 to 90. The standard deviation of laboratory assessment samples was relatively stable, all < 0.05 mg/L. The quality control assessment results from 2006 to 2020 were calculated according to │Z│ < 3, the qualified rate of water arsenic ranged from 64.0% to 100.0%, and that in urine was 54.5% to 100.0%. The quality control assessment results from 2009 to 2020 were calculated according to │Z│≤2, the qualified rate of water arsenic was 84.6% - 100.0%, and that in urine was 83.6% - 100.0%. Among the 13 water arsenic assessment results from 2009 to 2020, according to │Z│≤2, provincial laboratories reported all qualified years for 8 times, prefecture, city and county-level laboratories reported all qualified years for 2 times; there were 8 provinces with all qualified laboratories at province level and 3 provinces with all qualified laboratories at prefecture, city and county-level. Among the 13 assessment results of urinary arsenic from 2009 to 2020, according to │Z│≤2, the provincial laboratories reported all qualified years for 6 times, and the prefecture, city and county-level laboratories all qualified for 1 times; there were 6 provinces in which all provincial laboratories were qualified, and there were 6 provinces in which all prefecture, city and county-level laboratories were qualified.Conclusions:The quality control work of laboratories is improving day by day, but there are still some problems in individual links. Laboratories at all levels should continue to strengthen the construction of laboratories and the training of relevant personnel, further improve the theoretical and technical level of inspectors, and continuously improve the detection quality, to make the analysis data more reliable and comparable, so as to ensure the quality of arsenic poisoning prevention and monitoring work.
8.Sequence analysis of 16S rDNA and pmoCAB gene cluster of trichloroethylene-degrading methanotroph.

Methanotrophs could degrade methane and various chlorinated hydrocarbons. The analysis on methane monooxygenase gene cluster sequence would help to understand its catalytic mechanism and enhance the application in pollutants biodegradation. The methanotrophs was enriched and isolated with methane as the sole carbon source in the nitrate mineral salt medium. Then, five chlorinated hydrocarbons were selected as cometabolic substrates to study the biodegradation. The phylogenetic tree of 16S rDNA using MEGE5.05 software was constructed to identify the methanotroph strain. The pmoCAB gene cluster encoding particulate methane monooxygenase (pMMO) was amplified by semi-nested PCR in segments. ExPASy was performed to analyze theoretical molecular weight of the three pMMO subunits. As a result, a strain of methanotroph was isolated. The phylogenetic analysis indicated that the strain belongs to a species of Methylocystis, and it was named as Methylocystis sp. JTC3. The degradation rate of trichloroethylene (TCE) reached 93.79% when its initial concentration was 15.64 μmol/L after 5 days. We obtained the pmoCAB gene cluster of 3 227 bp including pmoC gene of 771 bp, pmoA gene of 759 bp, pmoB gene of 1 260 bp and two noncoding sequences in the middle by semi-nested PCR, T-A cloning and sequencing. The theoretical molecular weight of their corresponding gamma, beta and alpha subunit were 29.1 kDa, 28.6 kDa and 45.6 kDa respectively analyzed using ExPASy tool. The pmoCAB gene cluster of JTC3 was highly identical with that of Methylocystis sp. strain M analyzed by Blast, and pmoA sequences is more conservative than pmoC and pmoB. Finally, Methylocystis sp. JTC3 could degrade TCE efficiently. And the detailed analysis of pmoCAB from Methylocystis sp. JTC3 laid a solid foundation to further study its active sites features and its selectivity to chlorinated hydrocarbon.

9.Polymorphism of human intestinal fatty acid binding protein gene and serum lipid levels in coronary heart disease

Objective To explore the relationship between FABP2 54 gene polymorphism and serum lipid levels in different ethnic patients with coronary heart disease.Methods We collected fasting blood from participants,then extracted DNA from whole blood,and detected lipids from serum.We performed 54A/T FABP2 genotyping for participants in the case and the control groups by polymerase chain reaction (PCR) and DNA restriction enzyme digestion techniques.Results ( 1 ) In Hans population with coronary heart disease,the allelic frequency of point in 54T gene was 0.542,and 54A gene was 0.458 ; in urban Mongolian with coronary heart disease,the allelic frequency of point in 54T gene was 0.708,and that of 54A gene was 0.292.The allelic frequency of point in 54T and 54A genes were 0.284 and 0.716 respectively in the Han population control group and 0.353 and 0.647 respectively in the normal urban Mongolian group.Compared with the control groups,mutant 54T allele frequency in the case groups of Han and urban Mongolian was significantly higher ( Han:x2 =14.967,P < 0.05 ; Mongolian:x2 =28.083,P < 0.05 ).Compared with the Han Chinese population with coronary heart disease,the mutant 54T allele frequency in urban Mongolian with coronary heart disease was significantly (x2 =7.111,P < 0.05 ).There was no significant difference in mutant 54T allele frequency between the Mongolian control group and the Han control group ( x2 =3.392,P > 0.05 ).( 2 ) Patients with coronary heart disease who carry Thr54 ( + ) had a significantly higher level of fasting plasma TG [ CC Thr54 (-):( 1.89 ± 0.57 ) mmol/L,CC Thr54 ( + ):( 3.92 ± 1.63 ) mmol/L; CM Thr54 (-):(2.23 ± 0.13 ) mmol/L,CM Thr54 ( + ):(4.03 ± 1.14) mmol/L; Hans:P =0.001,Mongolian:P =0.035 ],and LDL-C [ CC Thr54 (-):( 3.09 ± 0.92) mmol/L,CC Thr54 ( + ):(4.05 ± 1.14 ) mmol/L ; CM Thr54 (-):( 4.26 ± 0.08 ) mmol/L,CM Thr54 ( + ):( 5.10 ± 0.56 ) mmol/L; Hans:P =0.025,Mongolian:P =0.045 ] than those with Thr54 (-).Conclusion (1) Threre is FABP2 gene polymorphism in urban Mongolian and Han population.Gene mutation frequency is in accordance with Hardy-Weinberg law of genetic equilibrium,suggesting the sample group representative.(2) FABP2 polymorphism may have a certain contribution to dyslipidemia in patients with coronary heart disease.FABP2 gene polymorphism may be associated with coronary heart disease.Individuals of Mongolian with FABP2 Thr54( + ) has a higher risk of coronary heart disease.
10.Relationship between Ala 54 Thr polymorphism of intestinal fatty acid-binding protein 2 gene and plasma lipids in old population

Objective To investigate relationship between Ala 54 Thr polymorphism in the intestinal fatty acid-binding protein (I-FABP) and plasma lipids in old people.Methods We collected fasting blood samples of selected groups,then we extracted DNA from whole blood,and detected lipids from serum.The 54A/T I-FABP genotypes were analyzed in 72 Han Chinese elderly by polymerase chain reaction (PCR) and DNA restriction enzyming.Total cholesterol (TC),triglyceride (TG),high density lipoprotein (HDL-C) and low-density lipoprotein (LDL-C) were detected.Genotype subgroups were Thr54 (-) and Thr54 (+) groups (36 cases for each).Results Compared with Thr54 (-) genotype group,Thr54 (+) genotype group had higher levels of plasma TC[(4.50±0.73) mmol/L vs.(5.48±0.49)mmol/L],TG[(1.08±0.48) mmol/L vs.(2.02±0.53)mmol/L],LDL-C [(3.10±0.44) mmol/L vs.(3.50±0.66)mmol/L],and had lower level of HDL-C [(1.14±0.25) mmol/L vs.(0.96±0.23)mmol/L](t=-6.67,-7.84,-3.03,3.05,all P<0.05).
