1.Protective effects of allicin on acute cerebral ischemia-reperfusion injury in rats

Aim To study the protective effects of allicin on acute cerebral ischemia reperfusion injury. Methods The model of cerebral ishemia-3 h/reperfusion-24 h was induced by middle cerebral artery occlusion(MCAO) in SD rats. Allicin(10,20 mg?kg -1) was administered once daily in rats:at 0 h of reperfusion. After 24 h reperfusion, the content of malondialdehyde(MDA),the activities of superoxide dismutase(SOD),GSH and myeloperoxidase(MPO) in brain tissue,the content of NO in plasma were measured. Results Compared with vehicle group, the activity of SOD was increased from 205.87?16.52 to 259.77?19.65 and 284.54?21.79 by allicin 10,20 mg?kg -1; the MDA content was decreased from 38.69?0.10 to 28.68?1.37 and 20.97?0.44 respectively; the GSH content was increased from 28.31?0.64 to 36.19?1.12 and 47.96?0.44 respectively; the NO content was decreased from 29.7?0.49 to 27.38?1.46 and 23.00?0.71 respectively; the MPO content was decreased from 0.29?3.06?10 -2 to 0.26?2.13?10 -2 and 0.23?7.52?10 -2 respectively. Conclusion The protective effects of allicin on acute cerebral ischemia reperfusion injury may be related to increasing antioxidase activities,decreasing lipid peroxidative damage and inhibiting inflammation reaction.
2.Study on the expression of vascular endothelial growth factor in patients with adenomyosis of the uterus

Objective To investigate the role of angiogenesis and expression of vascular endothelial growth factor (VEGF) in the pathophysiology of adenomyosis of the uterus. Methods The study included 32 patients with histologically proven adenomyosis and 30 patients with asymptomatic leiomyoma of the uterus. Immunohistochemical staining was used to detect VEGF expression in different parts of the uterus. A computerized morphometric study on the VEGF expression was performed. Results The VEGF H-Score of glandular cells in the endometrium of adenomyosis uterus [proliferative phase (9.6?1.4), secretory phase (11.7?1.6)] was much higher than that of leiomyoma uterus [proliferative phase (8.3?1.7),secretory phase (10.2?1.5)] (P0.05). Background The effects of Toll-like receptor 2 (TLR2) in grafting-related immune diseases have attracted more and more attention.Blocking TLR2 signal pathway can extend the survival time of heart and kidney grafts.However, the effects of anti-TLR2 monoclonal antibody on corneal graft have not been confirmed.Objective This study was to investigate the influence of anti-TLR2 monoclonal antibody on corneal graft survival in the rats received penetrating keratoplasty (PKP).Methods Allograft corneal transplantation was performed on the right eyes of 24 SPF female Wistar rats to establish PKP models,with 12 SD rats as donors.The model eyes were randomized into the TLR2 monoclonal antibody group and the model group.Anti-TLR2 monoclonal antibody of 15 μg/30 μl was subconjunctivally injected on day 0,2,4,6 and 8 following the modeling in the TLR2 monoclonal antibody group,and equal amount of normal saline was injected in the same way in the model group.The edema,transparency and neovascularization were observed under the slit lamp microscope after surgery, and rejection index (RI) was scored based on the criteria of Holland.Corneal tissue sections of the rats were prepared for the histopathological examination on day 9 and 15 after operation.The research protocol was approved by the Southern Medical University Ethics Committee.Results Mild corneal edema was found in the two groups 1-4 days after operation.A lot of new blood vessels, edema and opacification of corneas were seen in the model group 9-14 days after operation,but in the TLR2 monoclonal antibody group,corneal opacification was found 15 days after operation.The RI scores were significantly higher in the model group than those in the TLR2 monoclonal antibody group 5,9,15 days after operation (t=4.183,4.954,13.506;all at P<0.05).The survival time in the TLR2 monoclonal antibody group was 15.5 days,with the 95% confidence interval (CI) 14.9-16.1;while that in the model group was 9.5 days,with the 95% CI 8.7-10.3, showing a significant difference between the two groups (Z =12.728,P =0.001).The corneal histopathological examination revealed that corneal stromal edema,infiltration of inflammatory cells and vascular lumen were more prominent 9 and 15 days after operation in the model group than those in the TLR2 monoclonal antibody group.Conclusions Anti-TLR2 monoclonal antibody can inhibit inflammatory response after allograft corneal transplantation and therefore extend the survival time of graft in rats.
4.Effect of abdominal surgery on enteric motor function and enteric nervous system in rat

Objective To observe the changes in enteric motor function and acetylcholine esterase(AchE)neuron and nitric oxide synthase neuron(NOS)in enteric nervous system after different abdominal operations,and to explore the mechanism of these changes.Methods Sprague Dawley rats were divided into experimental groups(laparotomy group,intestine massage group and intestinal operation group)and the control group.Rats in each group were gavaged with medicinal carbon powder,and then the transimission of carbon powder in small intestine was determined,and the spreading specimens of intestinal myenteric plexus of small intestine and large intestine were collected and stained with AchE and nicotinamide-adenine dinucleotide phosphate-D(NADPH-d)for histological study,and the density of distribution and positive stained AchE and NOS neurons were observed and compared.Results The changes in enteric motor function:the propellant rate of carbon powder was slower in enteric massage group and intestinal operation group than that in laparotomy group and control group(18.1?2.8,25.7?3.5 vs 47.4?3.6,49.8?4.5,P0.05).Compared with the control group,the number and positive expression of AchE positive neuron in intestinal myenteric plexus decreased in small intestine and large intestine in intestinal operation group and intestine massage group(small intestine 43.0?3.0,52.2?3.3 vs 65.3?3.9,P
5.The applying Diver CE thrombus-aspirating device in acute myocardial infarction

Objective To assess the effect and safety of applying Diver CE thrombus-aspirating device during direct percutaneous coronary intervention(PCI)in pations with ST-segment elevation myocardial infarction(STEMI). Methods Select 46 patients with STEMI who underwent direct PCI between July,2006 and June,2007 in Fushun Central Hospital. Dividing these patients into thrombus-aspirating group and direct PCI group,comparing the difference in chest pain relieving.magnitude of ST-segment declining in 2h,TIMI flow grade 3,the maximum value of CK and CK-MB,EF and MACE outcome(death、myocardial infarction and targert vessel revascularization).Results The percent of chest pain relieving、magnitude of ST-segment declining 、TIMI flow grade 3、the maximum value of CK and CK-MB,and EF in thrombus-aspirating group are piror to the direct PCI group(P0.05).Conclusion applying Diver CE thrombus-aspirating device during PCI can obviously reduce thrombus in coronaty artery and distal embolization,improve myocardial reperfusion more effectively.
6.Value of Doppler sonographic features in diagnosis of ovarian neoplasms

Objective To observe and measure the Doppler sonographic features of benign and malignant ovarian neoplasms,which included morphology and hemodynamics of tumor angiogenesis,and to select the most significant features. Methods Ninety-five patients with one hundred and five ovarian tumors were examined preoperatively by power Doppler sonography to observe the morphology of tumor angiogenesis. The hemodynamic features of tumor angiogenesis referred for pulsed Doppler sonography evaluation were measured,which included peak systolic velocity(V PS ),end diastolic velocity(V ED ),mean flow velocity(V M),pulsatility index(PI),resistance index(RI) and diastolic notch. To assess the variables jointly,stepwise regression analysis was used to identify the group of features that allowed the best prediction of benignity versus malignancy. Results All Doppler features significantly associated with malignancy .The ROC curve of RI revealed that the cutoff of RI( 0.51 ) with the highest accuracy had a sensitivity of 98.0 % and specificity of 96.2 %.Conclusions RI was most significantly associated with malignancy. The combination of RI and gray-scale sonography can maximize the ability to discriminate between benign and malignant ovarian tumors.
7.Inhibitory effects of allicin on HL-60 cell proliferation and its induction of apoptosis

Aim To investigate the effects of allicin on the proliferation and apoptosis of HL-60 cells,and to explore the underlying mechanism.Methods The cell viability and colony formation were detected by MTT assay.Apoptotic cells were tested by means of TUNEL labeling.RT-PCR was used to analyze the bcl-2 mRNA expressions.Results HL-60 cell growth was significantly inhibited by allicin in dose and time dependent manners.Cell colony formation obviously decreased.The typical hypo-diploid pea J apoptotic peaJ appeared in each dose group.Apoptosis occured in a dose-dependent manner.And its later stages were identified by TUNEL labeling methods respectively,and bcl-2/bax was decreased.The expression of bcl-2 mRNA was down-regulated in a time-dependent manner after treatment with allicin at different time points.Conclusion Allicin effectively inhibits growth and induces apoptosis on HL-60 cells,which may be related with the down-regulation of expressions of bcl-2.
8.Design and application of modified multifunctional dressing bag with glauber's salt for relieving swelling and pain in early patients with limb fracture

Objective:To explore the effect of modified multifunctional dressing bag with glauber's salt for relieving swelling and pain in early patients with limb fracture.Methods:A total of 111 patients with limb fracture in Tongde Hospital of Zhejiang Province were divided into two groups according to the admission time. The control group consisted of 53 patients with limbs fracture using common mirabilite bag with glauber′s salt and ice bag for external application admitted from January to August 2018, and the observation group consisted of 58 patients with limbs fracture using modified multifunctional dressing bag with glauber′s salt admitted from September 2018 to January 2020. The degree of pain, the time of appearance of dermatoglyphia, the cases of severe swelling, waiting time for operation, compliance and comfort of the patients were observed within 72 hours after injury.Results:The degree of pain, the time of appearance of dermatoglyphia, the waiting time, the cases of severe swelling for surgery in the observation group were (1.8±0.7) points, (4.5±1.2) days, (5.7±1.2) days,1 case, respectively, which were all lower than those in the control group (2.2±0.8) points, (6.0±1.5) days, (6.5±1.4) days, 19 cases, the differences between the two groups were statistically significant ( t values were -2.6,-5.6,-3.3, χ2 value was 21.8, all P<0.05). The total compliance rate in the observation group was 82.76% (48/58), higher than that in the control group 56.60% (30/53), the differences were statistically significant ( χ2 value was 9.34, P<0.05). The comfort of the observation group was 82.76% (48/58), significantly higher than that of the control group 3.77% (2/53), the difference was statistically significant ( χ2 value was 69.8, P<0.05). Conclusions:The modified multifunctional dressing bag with glauber′s salt can reduce the swelling of limbs, reduce the appearance of skin blisters, stop bleeding and pain, and shorten the waiting time for operation, at the same time, it improves the compliance and comfort of the patients.
9.Analysis on DEA-Malmquist efficiency index of medical facilities in Zhejiang province

Objective To measure the changes of total factor productivity (TFP)of Zhejiang medical facilities for decision makers to promote their service efficiency.Methods Collecting the panel data of 2005-2010(four input indexes and five output indexes)from medical facilities in Zhejiang province and measuring with Malmquist index of DEA programs of DEAP2.1.Results The average annual TFP growth rate in the period is 2.0%.A further decomposition found that the average annual growth rate of technology progress is 1.8 %,while that of technical efficiency and pure technical efficiency is only 0.2%respectively.In the meantime,no scale efficiency growth was found.Conclusion The average annual growth of total factor productivity of Zhejiang medical institutions is substantially low in the period,with technology recession found as well.To maximize productivity of the medical sector,the allocation and internal management should be strengthened to stimulate technical efficiency and scale efficiency while encouraging technology innovation.
10.UVB induces morphological changes and the expression of MMPs in human fibroblasts

Objective To observe the changes of telomere length and MMPs level in human fibroblasts induced by UVB, and to explore their roles on skin photoaging .Methods Human skin fibroblasts were extracted and cul-tured.The 5th fibroblasts were irradiated by UVB .The morphology of fibroblasts were microscoped , and the length of telomere and the mRNA expression of COL1a1 and hTERT were detected by RT-qPCR.The expression of MMP-3 and MMP-1 were detected by Western blot .Results The fibroblasts gradually became round , wrinkled and disorderly arranged after 30 mJ/cm2 UVB irradiation for 24 h.The mRNA level of COL1a1 and hTERT and the expression of MMP-3 and MMP-1 were significantly increased after UVB irradiation compared with control , and the length of telomere was shortened .Conclusions UVB may frigger the early process of photoaging by the morphologi-cal changes of human skin fibroblasts and increasing the expression of MMP-3 and MMP-1 .
