1.Damage of high glucose on retinal capillary pericytes

Objective To observe the effect of high glucose on retinal capillary pericytes in vitro and research the pathogenesis of diabetic retinopathy (DR). Methods Retinas dissected from eyes of fresh killed cattle were separated and retinal capillary pericytes were cultured. The effect of high glucose on pericytes was evaluated by measurement of lactate dehydrogenase(LDH),3H-thymidine incorporation and liquid scintillation techniques.Morphological changes were observed under microscope and electric microscope.Results There was no significant difference in LDH activity in the supernatants between high glucose(10、20、40mmol*L-1) and normal glucose(5mmol*L-1)cultured for 48h or 96h. High glucose had an effective inhibition on pericytes' proliferation cultured for 48h,96h and 144h, and the inhibition rate increased when the glucose concentration rised and time passed by. The inhibition rate of pericytes was 78.09% after cultured in high glucose (40mmol*L-1) for 144h. Conclusion High glucose has an effect inhibition on pericytes' proliferation, and the inhibition rate increase when the glucose concentration rises and time passes by.

To investigate the insulin resistance and insulin secretion of the first degree relatives of T2DM with normal glucose tolerance(NGT). The levels of FIns and HOMA-IR in the groups of the parental (n=35) and the maternal (n=58) were higher than that in the group of the paternal (n=42), bwt the IAI and △I_ 30 /△G_ 30 were lower than that in the group of paternal. This suggests the first-degree relatives of type 2 diabetes have insulin resistance and impaired first phase insulin secretion in NGT.
4.The relationship of Met326Ile variant of p85? regulatory subunit of phosphatidylinositol 3-kinase(PI3-K) with type 2 diabetes

0.05),and the distribution of genotypes was in Hardy-Weinberg equilibrium.Data analysis showed Met326Ile variant did not impact on FIns,FPG,HOMA index,BMI and gene expression of PI3-K,though the expression of PI3-K gene was lower in type 2 diabetes than in control group.Conclusion The Met326Ile variant of the p85? regulatory subunit of PI3-K is likely to be functionally normal in type 2 diabetes and health people.
5.EXPERIMENT ON CULTURE, IDENTIFICATION OF NEURAL STEM CELL FROM SD RATS

To explore the feasibility of culture and identification of neural stem cells (NSCs) from ependyma, subventricular zone and cotex of fetal and adult rat respectively, and to make the basis for treatment of degenerative diseases with NSCs transplantation, all the seed cells derived from ependyma, subventricular zone and cotex were isolated from both fetal and adult SD rat, respectively. They were then cultured, induced to differentiation and subcultured continuously in a "CYTOKINE NSCs culture medium". Identification was carried out using Nestin, NSE and GFAP antibodies for differentiated NSCs, neuron and neuroglials, respectively. The seed cells from these four locations proliferated rapidly under some corresponding conditions, and formed "neurologic spheres", which consisted of many cells and expressed Nestin antigen. After continuous culture and subculture, NSCs might divide and proliferate further. Some NSCs buds developed processes and formed nerve fibers further, while the soma enlarged into the cells with "long processes", which connected or crisscrossed with each other, and were confirmed as neurons and neuroglias by immunocytochemistry. Seed cells from fetal rats might generate more NSCs than those from adult rats, and those from ependyma and subventricular zone produced more NSCs than those from cortex. There was no special morphological difference between ependyma NSCs and cortex NSCs. It is suggested that NSCs existed not only in ependyma and cotex of fetal SD rat, but also in the subventricular zone and cotex of adult SD rat. Fetal rat nerve tissue possesses much more NSCs than adult one.
6.THE ROLE OF TELOMERASE OLIGODEOXYNUCLEOTIDE IN THE INHIBITION OF GROWTH OF MALIGNANT GLIOMA CELLS

To evaluate the curative effect of antisense telomerase ODN on malignant glioma cells. The activity of telomerase in 16 fresh malignant gliomas was detected positive by PCR ELISA method. After covered with lipofectin, the 5?mol/L telomerase ODN was incubated with cultured gliomas cells. PCNA, TUNEL positive rate and the phase of cell cycle were detected by FcM per 24h. The results showed: Anti sense ODN inhibited telomerase activity after being applied for 48 hours, and inhibition was complete at 72 hours, The ODN inhibited proliferation of malignant gliomas and promoted their apoptosis markedly at 72 hours. After 96 hours, most transfected glioma cells were stopped to grow at G 2/M phase, and the number of apoptosis cells increased as time went on. The results suggested that anti sense telomerase ODN could inhibit the telomerase activity, and in turn promote aptoptosis of glioma cells and inhibit the growth of the tumor.
7.The effect of dehydroepiandrostrone to retinal capillary pericytes under high glucose

ObjectiveTo observe the effect of dehydroepiandrostrone (DHEA) on retinal capillary pericytes’ proliferation under high glucose and research the prevention and treatment of diabetic retinopathy (DR).MethodsRetinas were dissected from eyes of fresh killed cattle and retinal capillary pericytes were separated and cultured.The effect of DHEA (10、50、100、500、1 000nmol/L) on pericytes’ proliferetion was evaluated by 3H-thymidine incorporation and li-quid scintillation techniques.ResultsThere was significant difference between DHEA (50、100、500、1 000nmol/L) and control culturing for 48h.The increasing rate was 42.67% in DHEA of 500nmol/L.ConclusionDHEA could promote pericytes’ proliferation under high glucose.This provided reliable evidence for selecting effective drugs for prevention and treatment of DR clinically.

The relation of metabolic syndrome and its components with body mass index (BMI) was investigated in Zhanshan community, Qingdao. The results showed that the prevalence and relative risk of metabolic syndrome and its components were increased with BMI. Nearly one-third of individuals with overweight or obesity had metabolic syndrome.

Methods A questionnaire survey wasconducted in 66 middle-aged and aged women with UI in two communities and the results were an-alyzed. Results Most patients lacked the knowledge of UI;UI had impact on their physical andmental heahh.Must patients wished to know about the treatment knowledge,and health education of med-ical staff was the most promising way.The mastering degree of knowledge about disease was not related tothe serious degree of disease and age,but was positively related to education degree and economic income.Conclusions We should publicize the knowledge about UI and provide health education and emotionalsupport to patients.And we should emphasize the training about UI among medical staff.
