1.Effect of alcoholism history on early post-operative cognitive function in elderly patients after general an-esthesia

Objective To evaluate the effect of alcoholism history on the elderly patients’early cognitive function after lower abdominal or limbs surgery under general anesthesia through controlled clinical trials.Methods Sixty male patients aged 65-80 years of ASA grade Ⅰ or Ⅱ were selected un-dergoing lower abdominal or limbs surgery,30 cases with alcoholism history as alcohol group(group A),30 cases without alcoholism experience as control group (group C).All patients were anesthe-tized intravenously.The general data before and during operation,and the postoperative complications were recorded.The mini mental state examination(MMSE)was performed to assess the cognitive function 1d before and 1,7 d after surgery,then post-operative cognitive dysfunction(POCD)was compared between the two groups through analysis of the MMSE score.Results The MMSE score of 1,7 d after surgery in two groups was significantly lower than that of the preoperative(P < 0.05 ). Compared with the score 1 d after surgery,the score of 7 d post-operation was significantly higher in group A (P <0.05).The postoperative score in group A were significantly lower than that in group C 1,7 d after surgery (P <0.05).The incidence of POCD in group A were higher than that in group C 1,7 d after surgery (P <0.05 ).Conclusion Alcoholism history can increase the incidence of early cognitive dysfunction in elderly patients after general anesthesia.

Animal models of endometriosis with traditional Chinese medicine (TCM) symptoms, not only provided a useful experimental method for TCM syndrome differentiation of endometriosis, also verified the pathogenesis of TCM of endometriosis, which offered the theoretical support for the clinic. This article discussed the establishment, result examination, existing limit, and solutions of animal models of endometriosis with TCM symptoms.
3.The effect of adipose-derived mesenchymal stem cells on pancreatic cancer cells in vitro

Objective To explore the effects of adipose tissue-derived stem cells (ADSCs) on the proliferation and invasion of pancreatic cancer cell lines in vitro.Methods ADSCs were isolated and co-cultured with pancreatic cancer cells.The proliferation and invasion of pancreatic cancer cells was tested by CCK-8 test kit.Stromal cell derived factor (SDF-1),vascular endothelial growth factor(VEGF),matrix metalloproteinase(MMP-9)and transforming growth factor-β (TGF-β1)in the culture medium were assayed by ELISA.The expression of cytokines in pancreatic cancer cells and ADSCs were detected by qRT-PCR.CCK-8 test kit was used to measure the AMD3100 regulation for the co-culture of ADSCs and pancreatic cancer cells.Results ADSCs can promote the proliferation and invasion of pancreatic cancer cells (all P <0.05); more SDF-1,VEGF,MMP-9 and less TGF-β1 was secreted by ADSCs than by pancreatic cancer cells(SDF-1:1100±100 vs.0 vs.0,F=389.134,P<0.01;VEGF:140±4 vs.99±5 vs.93 ±4,F=174.102,P <0.05;MMP-9:61.8 ±4.2 vs.43.5 ±2.8 vs.54.5 ±3.0,F=76.279,P<0.05;TGF-β1:20.6 ±3.0 vs.35.6 ±2.6 vs.41.3 ±5.5,F =79.338,P <0.05).ADSCs promote the expression of VEGF and MMP-9 in pancreatic cancer cells(VEGF:63.7 ±5.9 vs.50.6 ±4.1,t =7.536,P<0.05; MMP-9:mRNA(55.8±3.6 vs.42.7 ±3.1,t =8.279,P<0.05).AMD3100 significantly downregulates these growth-promoting effects of ADSCs on pancreatic cancer cells(SW1990:1.539 ±0.140 vs.1.361±0.066,t=2.835,P<0.05;PANC-1:1.376±0.100 vs.1.281±0.031,t=2.860,P<0.05).Conclusions ADSCs promote the proliferation and invasion of pancreatic cancer cells.
4.Genetics of sporadic cerebral small vessel disease

Cerebral smal vessel disease (CSVD) can be divided into sporadic and hereditary CSVD. The exact pathogenesis of sporadic CSVD is unknow n. Genetic factors may also play an important role, except for environmental and vascular risk factors. As a complicated disease, sporadic CSVD has the characteristics of multigenetic susceptibility. Therefore, investigating the related genetic factors may contribute to understanding the pathogenesis of sporadic CSVD. This article review s the advances in research on the genetics of sporadic CSVD.
5.Imaging diagnosis of enlarged prostatic utricle in children

Objective To discuss the image characteristics of enlarged prostatic utricle in children.Methods 13 children with enlarged prostatic utricle were retrospectively analyzed,6 of whom checked by voiding cystic urethrography (VCUG),5 of whom by MRI,2 of whom by both VCUG and MRI.Results 8 cases with VCUG showed round-like or long circle utricle filled with contrast media located in the rear posterior urethra.Among them,3 cases showed the tubiform between utricle and posterior urethra.The other 5 cases displayed overlap between utricle and urethra,and there was no tubiform between utricle and posterior urethra.7 cases with MRI showed oblong cyst at the right rear middle of the bladder that showed low signal with T1 weighted image and high signal with T2 weighted image,crossing the center of prostate gland.Among them,5 cases showed the communication between oblong cyst and posterior urethra.Also,among them,5 cases were associated with the orchitis,epididymis phlogistic,scrotal phlogistic,and 1 case showed the right renal agenesis.Conclusion The VCUG and MRI are helpful to check enlarged prostatic utricle in children.They have different advantages in different aspect.
6.The analysis of self-care ability and cognition among disabled elderly

Objective To investigate the status of self-care ability and cognition among disabled elderly,and to analyze the relationship between self-care ability and cognition,then analyze their influencing factors.Methods A total of 813 disabled elderly accepted long-term care at community or facility were recruited by stratified sampling method and investigated by socio-demographic questionnaire,activities of daily living and short portable mental status questionnaire.Results The total scores of self-care ability of the long-term care disabled was 32.01±11.59,and the total points of cognition was 3.77±3.24.Disabled elderly with severe disorders of self-care ability had a higher obstacle rate in every cognition ability than those with mild disabilities (P<0.001).There were differences in orientation and memory function with different persistent disabled time (x2 =11.875,14.758,P<0.01).Disabled elderly with cognition obstacles have a higher obstacle rate in every self-care aspect than those with normal cognition (P<0.01).The multiple regression analysis showed that the educational level (β=-0.067),arital status (β=-0.071),disabled persistence time (β=0.121),orientation ability (β=0.403),the amount of cognition obstacles (β=0.443) were the influence factors of Basic Activities of Daily Living(BADL) among disabled elderly ; and disabled persistence time (β=0.116),orientation ability (β=0.417),recalled ability (β=0.275),memorial ability (β=0.189),calculative ability (β=0.257),the amount of cognition obstacles (β=0.358) were the important factors influence Instrumental Activities of Daily Living(IADL).Conclusion The self-care ability and cognition of the long-term care elderly were declining sharply,and influencing each other.It is advisable to offer them direct long-term care service combined with the laws and characteristics of cognition and self-care ability.
7.Effects of endometriosis on the reproductive ability of mice

Objectives The aim of the study was to establish a mouse model of endometriosis , and to observe the effects of endometriosis on the reproductive ability in the mice .Methods The mouse models of endometriosis was estab-lished by subcutaneous and intraperitoneal injection of heterogenous endometrium .The pregnancy rate and live embryo number in the experimental , sham operation and blank groups were observed and compared , and the effects of endometrio-sis on reproductive ability of the mice were evaluated .Results Two weeks after the establishment of the mouse model of endometriosis , there were lesions in the peritoneal cavity and subcutaneous tissue of the mice .Compared the pregnancy rates among the model group , sham operation group and blank group , the differences were statistically significant ( P<0.05).Compared the live embryo number in the three groups , there were also significant differences (P<0.05).Con-clusions A mouse model of endometriosis is successfully established , and it demonstrates that endometriosis may affect the reproductive ability of mice .
8.Research Progress in the Pharmacological Mechanism of Active Ingredients in Semen Ziziphi Spinosae

Semen Ziziphi spinosae ( SZS) , as a commonly used traditional Chinese medicine in clinic for insomnia therapy, is rich in pharmacological active ingredients such as saponins, flavonoids, alkaloids, oil and the other chemical compounds. The recent stud-ies indicated that some active ingredients in SZS exhibited a variety of activities including sedative hypnotics, antianxiety and anti-de-pression by regulating particular neurotransmitters such as 5-hydroxytryptamine (5-HT), gamma-amino butyric acid (GABA), norepi-nephrine, dopamine and glutamate. In accordance with the previous studies on pharmacological activities of SZS, this paper summa-rized and reviewed the applications of SZS in the treatment of central nervous system diseases, myocardial diseases and hepatic disea-ses, which might provide solid evidence for the application development of SZS.

Objectives To evaluate the effect of a Chinese traditional medicine , Huoxue Xiaoyi Fang , on the embryo development after in vitro fertilization ( IVF ) in mouse models of endometriosis .Methods To observe the quantity of oocytes including the average number of superovulation , fertility rate, 2-cell and 4-cell embryo cleavage rates at different stages in vitro fertilization, and morulation rate among the traditional Chinese medicine group (TCM group), endometriosis model group ( EM model group ) and sham-operated group .To explore the influence of endometriosis and the effect of Huoxue Xiaoyi Fang on the quantity of oocytes and embryonic development after IVF in the mouse models of endometriosis.Results All the average number of superovulation , fertility rate, 2-cell and 4-cell embryo cleavage rates in the three groups showed significant differences (P<0.05), and the average number of superovulation was in an order of sham operated group >TCM group>model group .There were no significant differences in the morulation rates in the three groups (P>0.05).There were significant differences among the IVF rates of the three groups (P<0.05), and to be in an order of sham operated group >TCM group>model group .There were significant differences in the final morulation rate among the three groups (P<0.05).Conclusions Endometriosis may influence the quality of embryonic development of EM model mice, including reduced average number of superovulation , fertility rate, and embryo cleavage rate .The Chinese traditional medicine Huoxue Xiaoyi Fang can improve the the quantity of embryonic development after IVF .
10.Establishment of a novel high throughput screening assay for identifying small molecular antagonists of human interleukin-6 receptor

Objective To establish a high throughput screening assay for identifying human small molecular antagonists targeted IL-6R.Methods The full length gene of the human IL-6R extracellular region was amplified by PCR and cloned into a eukaryotic expression vector to construct recombination expression plasmid pABHis -IL6R that was then transfected transiently into HEK293T cells to prepare recombination protein IL-6R.Western blotting assay and receptor-ligand binding experiment were used to analyze the bioactivity of IL-6R.A new screening method based on ELISA was established using the function of IL-6R binding to its ligand and the characteristics of Fc fragment binding to IgG-HRP.Then Z′-factor was calculated and a known antagonist ab 47215 was used to assess the stability and reliability of the new assay .Results Recombination plasmid pABHis-IL6R was constructed and soluble IL-6R was prepared.IL-6R reported herein could be recognized by an anti-IL-6R antibody and specifically bind to its ligand in a dose response manner .A Z′-factor of 0.53 was obtained that could serve high throughput screening assay .Ab47215 , as a known specific antagonist , was able to block rhIL-6 from binding to the receptor in a dose-dependent manner in the new screening assay , the IC50 of which was (0.55 ± 0.11)μg/ml.Conclusion An innovative and easy screening assay for identifying human IL-6R antagonists is established , which might help discover potent and specific antagonists .
