1.RELATIVE BIOAVAILABILITY OF SELENIUM IN Se-YEAST TO RATS FED ON LOW-Se DIET FROM A KESHAN DISEASE AREA

In the present study the bioavailability of selenium in Se-yeast was compared with that in sodium selenite to rats fed on low-Se diet from a keshan discaes area. The bioavailability of selenium in Se-yeast when estimated by heart, liver kidney and erythrocyte Se contents was significantly greater than that in selenite at wk 2 and 4 of supplement, indicating that selenium in Se-yeast is more cfficiently retained in tissues than selenite selenium. When the criterion used was either platelet or liver glutathione peroxidase (GSH_(?)) activity, the bioavailability was lower for selenium in Se-yeast than for in selenite at wk 2, but higher at wk 4, showing that selenium from both selenite and Se-yeast is clearly available for GSH_(?), synthesis, "although Se-yeast restores GSH_(?) activity more slowly. In addition, the effect of selenium in Se-yeast in maintaining cither Se levels in heart and erythrocyte or GSH_(?) activities in platelets and liver was superior to that of selenite after the selenium supplements were withdrawn.
2.Establishment of APP and PS-1 double gene stably transfected cell lines

AIM To study pathogenesis of Alzheimers disease and screen ?- secretase and ?- secretase inhibiting drug, PS-1(M 146L)and APP 751 gene double stably transfected CHO cell lines have been established. METHODS A? cDNA encoding human PS-1 was obtained by polymerase chain reaction from a human placental library. Mutant PS-1 (M 146L) cDNA was also generated by polymerase chain reaction from wild type PS-1 cDNA. Wild type and mutant PS1 were subcloned into CMV-based mammalian expression vectors PCI-neo,then recombined plasmid were co-transfected into APP expressing CHO cells at 1∶10 ratio using LipofectAmine. Stable expressing cell lines were screened and selected by using selection media. RESULTS Several CHO cell lines stably transfected with wide type or mutant PS-1 (M 146L) as well as APP 751 genes were established. Overexpression of PS-1 in CHO cell accumulated full length 45 kDa PS-1 protein. A? released to conditioned media were not changed in wild type PS-1 transfected APP expression CHO cells. A? 1~42 level in conditioned media of M 146L mutant PS-1 stably transfected of APP expression CHO cells were elevated about 1.6 fold. CONCLUSION Expression of M 146L mutant PS-1 in stably transfected APP expression CHO cells increased a secretion of A? 1~42. The PS-1 and APP double stably transfected CHO cell lines we generated can be used for either ?-or ?-secretase inhibitor study and related drug screening.
3.Establishment of 451 bp fragment that functionally related to transportation in hepatocellular carcinoma supressor gene-1(HCCS1)

Background and purpose:Hepatocellular carcinoma suppressor gene-1(HCCS1)is a potential hepatocellular carcinoma supressor gene,and it also plays an important role in sorting of some cytoplasmic proteins.Its carcinoma suppressor function may be related to its sorting function.So it is important to identify the minimum region of functional sequence in HCCS1 that related to the transportation.Methods:The expression vectors containing various lengths of HCCS1 gene were constructed and transfected into HeLa cells mediated by liposomes.The localizations of different HCCS1 fragments and the colocalizations of M6PR with various truncated HCCS1 were determined by laser scanning confocal microscopy,respectively.Results:10 different expression vectors containing various lengths of HCCS1 gene were successfully constructed,it had been shown that the polar localization of HCCS1 protein and the co-localizations with M6PR disappeared when HCCS1 gene was truncated to 1 120 bp from the 3'end by laser scanning confocal microscopy.Conclusions:We identified the minimum localization of the HCCS1 functional sequence that related to the transportation.
4.The anti-tumour effect of Wuxing soup and its effect on the immunity of melanoma mice

Background and purpose:Wuxing soup is popular for it's anti-cancer effect in folk medicine and deserved to be further studied by modern scientific methods.This research aimed to explore its anti-cancer effect and to study the influence on the immunity of melanoma in mice. Methods :Inhibition of different ingredients and concentrations of Wuxing soup on the growth of the mouse melanoma B16 cells was detected by MTT in vitro.Animal experiment was performed to determine its anti-cancer effect in vivo.C57/BL6 mice were randomly divided into three groups after vaccination of B16 cell,and then given intragastrically with different soups for 25 days.All mice were killed on day 26 after inoculation.The weight of tumors were recorded.The anti-tumor immune function was measured by T lymphocyte transforming assay and NK killing assay.The different effect of different ingredients was also observed. Results :Our result showed that different ingredients soup exerted different inhibition on B16 cell growth and Wuxing soup was the strongest one of all and in dose-dependent manner in vitro.The animal experiment indicated that different ingredients soup has different inhibition on melanoma,the soup-treated mouse display improved T lymphocyte transforming assay and NK killing ability.Among the different soups,Wuxing soup showed the strongest anti-cancer effect and immune enhancement. Conclusions :Wuxing soup is an effective anti-cancer agent in melanoma mice and can enhance the immunity of the mice with melanoma.
