1.The application evaluation of a rapid urine microalbumin screening test

ObjectiveTo evaluate the clinical application value of a kind of rapid urine microalbumin( mALB )screening test.Methods Six hundred and thirteen samples for urine routine detection from Affiliated Fuxing hospital,Capital Medical University were included from December 2006 to February 2007.mALB of all urine samples was detected with H-800 automated urine analyzer based on a dry chemical qualitative screening test.To validate the screening effect of the dry chemical screening test,all of the positive samples and a part of negative samples were determined by a mALB quantitative test as confirmation by lmmage-800 automated specific protein analyzer.Comparison of mALB quantitative level between screening positive group and negative group was carried out by 2 independent samples rank sum test.Comparison between enumeration data was carried out by Kappa test for consistency.Results One hundred and fourteen positive samples out of 613 were detected by screening method ( 18.6% ).Out of the 114 positive samples by screening test,seventy-one samples were confirmed positive by quantitative test (mALB > 13.3 mg/L).Out of 76 negative sample by screening test,eight samples were confirmed positive by quantitative test.The mALB level of the screening positive group and negative group was 17.70 ( 9.19 -32.90) mg/L and 4.55(2.38-7.60) mg/L,respectively,and the mALB level of the screening positive group was much higher than that of the negative group ( Z =- 8.644,P < 0.01 ).According to the quantitative test,the sensitivity and specificity of the mALB screening method were 89.9% (71/79) and 61.3% (68/111 ),respectively.ConclusionsThe mALB rapid screening test in the research has a certain screening value.But the test method should be further perfected to improve its sensitivity for its certain omission rate.

Objective: To explore behavior problems and family environment of children with video games dependence Method:34 video games dependent children and 34 normal children were assessed with EPQ for children and Achenbach Children's Behavior Checklist Parents of both groups were asked to fill up Family Environment Scale-Chinese Version Result:The P and L scores of EPQ were higher in the study group than in control Social status, school status, withdraw, anxiety/depression, social problems, delinquent problems extroversive behavior problems and the total score of CBCL were also higher in the study group Children with video games dependence had families with lower intimacy, lower expressioness, lower active-reaction orientation and organization Their families had more conflicts than families of normal children Conclusion: Video games dependent children have more behavior problems and poorer family environment
3.Research progress in role of B cell immunity in primary biliary cirrhosis

Primary biliary cirrhosis (PBC)is characterized by injury of small intrahepatic bile ducts,but its mechanism,especially the role of B cell immunity in this disease,remains unclear.The possible role of B cell immunity in the pathogenesis of PBC is reviewed from the fol-lowing aspects:distribution of B cells and plasma cells in the liver tissues of PBC patients,clearance of B cells,mechanism of abnormal in-crease in serum IgM and its sources among PBC patients,mechanism of production of antimitochondrial antibodies and their possible func-tions in bile duct injury,and typical autoantibodies in PBC.B cell immunity is considered to play a significant role in the pathogenesis of PBC,but there are still many key problems to be clarified.
4.The role of CagA on H.pylori-induced IL-8 production from gastric epithelial cells

AIM: To investigate the role of CagA and the effect of small interference RNA (siRNA) on the release of IL-8 in H. pylori-infected gastric epithelial cells. METHODS: siRNA were transferred into CagA positive strain H. pylori NCTC 11637 by using electroporation. The CagA positive strain NCTC 11637 and the CagA negative strain NCTC 11639 were co-cultured with gastric epithelial cells and the level of IL-8 in the supernatant was measure by ELISA. RT-PCR and Western blotting were performed to detect CagA expression. RESULTS: The level of IL-8 induced by CagA positive strain NCTC 11637 was higher than the level induced by CagA negative strain NCTC 11639 ( 1 200.00 ?32.51) ng/L vs (100.00?8.58) ng/L, P0.05). Meanwhile, CagA mRNA decreased significantly in siRNAⅢgroup. The levels of CagA mRNA at 6, 12, and 24 h after electroporation were 31.3% (0.270/0.861), 57.6% (0.496/0.861), and 73.9% (0.637/0.861) of the control levels, respectively. Inhibition rate by siRNAⅢ was 68.7%. The Western blotting result in siRNA Ⅲ group showed that the level of CagA protein degraded to 30.7% (0.4/1.3) at 12 h after electroporation. AIM: To study the relation ship between stress and lung injury caused by acute hemorrhagic necrotizing pancreatitis(AHNP) through AHNP model.METHODS: The AHNP model was made by using 5% sodium taurocholate retrograded injection into biliopancreatic duct in SD rats. Those rats were divided into three groups randomly, from A to C, the A group undertook sham operations, the B group was made into AHNP model, and the C group was given Metyrapone. The level of corticosteroid, CRP and amylase in serum had been observed. The lung and pancreas histological examinations were also performed.RESULTS: In C group, the level of corticosteroid, CRP and amylase were much lower than those in B group. 7.Effect of overexpression of vascular cell adhesion molecule-1 on migration of murine mesenchymal stem cells

OBJECTIVE To investigate the effect of overexpression of vascular cell adhesion molecule-1(VCAM-1)on the migration in vitro of the murine mesenchymal stem cells(MSCs)and its possible mechanism. METHODS The migration ability of normal mouse MSC (C3) ,empty vector-transfected MSC(C3+N) and VCAM-1 transfected MSC(C3+VCAM-1)was assessed by Transwell culture system in vitro after incubation for 8 and 12 h,respectively. The fetal bovine serum (FBS) was used as the chemotactic agent to induce MSC migration. The transmigrated cells were detected with methylosaniliam chloride(crystal violet)as well as DAPI staining.Furthermore,the specific chemical inhibitors of mitogen-activation protein kinase (MAPK) pathway ( SB203580,PD98059 and JNK inhibitorⅡ)were added to the Transwell system for 12 h and the alteration of the MSC migration ability was evaluated. RESULTS After incubation with FBS for 8 and 12 h,the absolute migrated cell number(7467 ± 485 and 8795 ± 255)and migration rate〔(14.9 ± 1.0)% and(17.6 ± 0.5)%〕of MSC in C3+VCAM-1 group were significantly increased compared with C3 group〔2731±562 and 4779±224, (5.5 ± 1.1)%and(9.6 ± 0.4)%〕and C3+N group〔2539 ± 321 and 5645 ± 1080,(5.1 ± 0.6)%and(11.3 ± 1.1)%〕(P<0.05,P<0.01),but there was no significant difference between C3 and C3+N groups. Moreover,the MSC migration ability of C3+VCAM-1 group was partially suppressed by addition of JNK inhibitorⅡ. The transmigrated cell number(4843 ± 167)and migration rate〔(9.7 ± 0.3)%〕were decreased compared with those of C3+VCAM-1 group without JNK inhibitorⅡ(P<0.01). SB203580 and PD98059,as specific chemical inhibitors of MAPK pathway,had no effect on MSC migration. CONCLUSION VCAM-1 can enhance mouse MSC migration in vitro and th4e mechanism may be related to JNK/MAPK pathway activation.
8.Bioinformatics analysis of dysregulated miRNAs in the tissue of children with nonsyndromic cleft lip and/or cleft palate

Objective:To screen the differentially expressed miRNAs in umbilical cord tissue of children with nonsyndromic cleft lip and/or cleft palate( NSCL/P) using miRNA microarray and comprehensive bioinformatics analysis for the prediction of related the bio-logical process and signaling pathways. Methods:Umbilical cord tissues of 4 cases of healthy newborns' and 4 lip or palate tissues of 4 cases with NSCL/P without other disease aged younger than 2 years were collected. The differentially expressed miRNAs were screened by miRNA microarray. Targets of dysrugulated miRNAs were predicted by TARGETSCAN-VERT, MIRDB and RNA22-HSA. All the gene sets were analyzed by gene ontology and pathway enrichment. Results: MiRNA microarray demonstrated that 254 miRNAs were dysregulated(181 miRNAs were up-regulated and 73 downregulated,P <0. 05). The dysregulated miRNAs targets contained 5029 genes. The dysregulated miRNAs targets were enriched in anatomical structure development,cell adhesion,cell proliferation,cell motili-ty and other biological processes. The dysregulated miRNAs targets were enriched in Wnt, mTOR, cGMP-PKG, TGFβ, PI3K-Akt and other signaling pathways. Conclusion:The target genes set of miRNAs are enriched in multiple biological processes and signaling path-ways related to NSCL/P, which indicate that genetic and environmental factors may influence the development process of NSCL/P.

Objective To explore the relationship between iodine and fluoride content in drinking water and the incidence of adult thyroid nodules in Cangzhou,Hebei.Methods According to the previous reports on iodine and fluoride levels in drinking water in Cangzhou,from November 2016 to January 2017,Cangzhou was divided into high iodine,low iodine,normal iodine and fluorine,low iodine and high fluorine,high iodine and high fluorine areas,and according to the different contents of iodine and fluorine in drinking water,high iodine and high fluorine area was further divided into high iodine and high fluorine 1 (iodine:743.30 μg/L,fluorine:4.27 mg/L),2 (iodine:119.31μg/L,fluorine:4.67 mg/L) and 3 (iodine:105.30 μg/L,fluorine:1.64 mg/L) subareas.Subjects who lived for 20 or more years and aged 30 or older,without serious disease and not taken iodized salt were selected.Palpation was used to examine the size,texture,mass,tenderness and mobility of the thyroid gland.The boundary,internal echo,blood flow and quantity of nodules were observed and recorded by color Doppler.Results The prevalence difference of thyroid nodules [36.8% (629/1 710),32.8% (636/1 938),25.1% (427/1 700)] in high iodine,low iodine and normal iodine and fluorine areas was statistically significant (x2 =55.597,P < 0.05).The prevalences of thyroid nodules in both high iodine and low iodine areas were higher than that of normal iodine and fluorine area (P< 0.016 7).The prevalence difference of thyroid nodules [43.3% (749/1 730),39.8% (712/1 790),34.9% (623/1 785)] in high iodine and high fluorine 1,2 and 3 subareas was statistically significant(x2 =26.220,P < 0.05).Compared with low iodine area,the prevalence of thyroid nodules [41.2% (735/1 785)] in low iodine and high fluorine area was increased (x2 =6.288,P < 0.05).Conclusions Both high iodine and low iodine can induce thyroid nodules.In water source areas with high iodine content,both high iodine and high fluorine are the factors inducing thyroid nodules.The prevalence of thyroid nodules in low iodine and high fluorine area is significantly higher than that of low iodine area.
