1. Analysis of two outbreaks of respiratory infection caused by adenovirus

Objective:
To investigate the epidemiological characteristics and the causes of two outbreaks of upper respiratory tract infection in our city.
Methods:
The epidemiological investigation was carried out on the outbreaks and the pharyngeal swabs were collected to do the pathophysiologic examination by using real-time PCR. Hexon gene of adenovirus was amplified by PCR. Then the positive products were sequenced and BLAST searching was done in GenBank.
Results:
The clinical symptoms of the two cases were high fever, sore throat and cough, etc, while the main signs were pharyngeal congestion and tonsil swelling; 32 students of a class in Yunxing primary school had respiratory infection related symptoms, of which the incidence rate was 55%; while 12 of 38 students in a class of Fangcun primary school were infected. All these students were on school in the same class, and the incidence was aggregate without proliferation. The duration of the disease was about 5 days and the prognosis was good, no death occurred. The positive result of real-time PCR showed adenovirus. BLAST search analysis on hexon gene showed AdV14.
Conclusions
According to epidemiological investigation and laboratory test results, the cause of these two outbreaks of upper respiratory tract infection was adenovirus type 14.
2.A HISTOLOGICAL AND HISTOCHEMICAL STUDY OF SOME PRESERVATING METHODS IN LOCAL ISCHEMIC KIDNEY

A comparative observation about histological and histochemical study were made onthe preservating methods of local ischemic kidney of dogs.The methods of this exp-eriment were divided into five groups:A.Local hypothermia;B.Hypothermic perfu-sion solution 500ml,4℃,containing Procaine 450 mg and Heparin 100 mg,infusedinto renal artery;C.Hypothermic perfusion solution 500ml,4℃,containing 20% Ma-nniton 32ml,25% MgSO_4 0.36 ml,Heparin 100 ml,infused into renal artery;D.Localhypothermia and heparin 30 mg injected into the same vessel.E.Local room tem-perature.The specimens were taken from the kidneys of the experimental dogs atvarious period after treated with every method.The results were as follows;A,D and E group presented irreversible histological changes within 4 hoursaftertreatment.Four hours later,the reaction of SDH,ATPase,AlPase werenegative.It suggested that the kidney damage was serious,as well as expressed thatthe effect of these preservating methods for ischemic kidney are not satisfactory.In B group,the histological and histochemical alteration is very light and almostreversible,during 60 days after treatment.In C group,hypothermic perfusionsolution containing Mannitol,MgSO_4 etc either during experiment or follow observa-tion for 60 days,no histological and histochemical alterations were found.It indica-ted that this method is an ideal perservating technique for ischemic kidney.
3.Serum antibody level and diagnostic value in convalescent patients infected with novel coronavirus

Objective:To investigate the levels of serum IgM, IgG, IgA and neutralizing antibodies (NAbs) in convalescing patients infected with the novel coronavirus and their diagnostic significance.Methods:Five hundreds and eighty one infected patients and 219 non-infected patients were detected by chemiluminescence method . The positive rate of antibody and antibody level were statistically analyzed for statistical significance. The correlation between the antibodies was compared, and the ability of each antibody to identify breakthrough infection in the vaccinated population was evaluated by combining ROC curve.Results:The antibody titers of IgM, IgG, NAbs and IgA in infected patients were higher than those in uninfected individuals. The antibody positive rate and antibody level of infected persons who were vaccinated and not vaccinated, with different ages, different doses of vaccines, and different intervals of vaccination were statistically significant ( P<0.05); IgA antibody levels were positively correlated with IgG and NAbs, and NAbs is positively correlated with IgG antibody; In breakthrough infection analysis, IgG, IgA and NAbs antibodies were good at identifying infected persons in the vaccinated population. Conclusions:The positive rate of IgM antibody is low in the convalescence period, which is of little significance for monitoring the course of COVID-19 and evaluating the efficacy of vaccines. In addition, the antibody level and positive rate can be significantly improved by the booster injection of COVID-19 vaccine. Differences in IgG, IgA, and NAbs antibody levels can be used to identify breakthrough infection cases from vaccinated populations.
4.Surveillance analysis of influenza Etiological in Xuzhou from 2010 to 2015

Objective To investigate the prevalence,subtypes and infected population of influenza (flu) viruses in Xuzhou city from 2010 to 2015,providing scientific supports for influenza prevention and control.Methods Molecular detection and virus isolation were carried out with the throat swab samples collected from influenza-like illness(ILI) cases of national sentinel hospitals and the flu outbreaks.Results The influenza has obvious epidemic characteristics seasonally,two concentrated outbreaks in every December to next March and August to September.The positive rates were different among age groups,the highest was in less than ten years old group (64.36%),then 10-19 years old group (10.69%),and the lowest was in 50-59 years old group (3.77%).A total of 15 245 swab specimens were collected in which 1 459 strains were detected and identified for influenza viruses with total positive rate of 9.57%.Among these positive samples,593 samples were H3 subtype (40.64%),627 samples were B subtype (42.97%),237 samples were H1 N1 subtype (16.24%).Four hundred and fouty strains were isolated fromMDCK cells the relatively isolate rate was 30.16 % (440/1 459),fifty nine strains from embryonated chicken (13.78%).Conclusions The epidemic types of influenza in Xuzhou from 2010 to 2015 were alternation between H3 and B.There were epidemic peaks of H1N1occurred occasionally and unstably.
5.Retesting of HIV antibody positive samples and analysis of confirmatory results

Objective:To discuss the coincidence rate between the initial test and the retest result of the laboratory, and the relationship between the result of reexamination and the result of confirmation by analyzing the retesting and confirmatory result of positive samples in various medical institutions,Methods:A total of 710 HIV antibody positive samples from medical institutions within the jurisdiction of Xuzhou city from January 1 to December 31, 2018 were retrospectively collected as research materials. The source, population, reinspection result and confirmatory result were analyzed respectively.Results:The positive rate of reexamination in medical institutions, disease control institutions and blood collection and supply institutions was 62.12%(269/433), 96.53%(167/173), 14.42%(15/104), respectively. The double-reactive samples were confirmed to be positive at the time of reexamination or follow-up. The inconsistencies in the reexamination result were confirmed to be negative by the appropriate time or follow-up. The false positive rate by Bio-Rad reagents was 0.23% (1/443), the false positive rate by Alere reagents was 1.78% (8/450).Conclusions:From high to low, the retest compliance rate was in the order of disease control institutions, medical institutions, blood collection and supply institutions. The samples sent by the CDC can be tested directly without retesting. Therefore, the double-reactive samples in the reinspection should be mainly followed up. If the retest result are inconsistent and the samples are not confirmed positive or the follow-up result are positive, there is no missing test. The specificity of Bio-Rad reagents is better than that of Alere reagents.
6.Molecular epidemiological analysis of adenovirus in children with influenza-like illness in Xuzhou from 2013 to 2019

Objective:To understand the epidemiological characteristics of human adenovirus(HAdv)in Xuzhou area.Methods:A total of 7 257 swab samples from influenza-like illness children in Xuzhou children’s Hospital were collected during 2013-2019. Real-time PCR was used to detect adenovirus, and then the positive specimens were subjected to amplification for the hexon gene by polymerase chain reaction (PCR) for genotypic analysis. Adenovirus 55 was cultured by Hep-2 cells and analyzed by whole gene sequencing.Results:A total of 227 strains of adenoviruses were detected out of 7 257 samples, and the detection rate was 3.13%. Infections were mainly found in children aged 3 to 6, with a peak of detection in May and June. Typing data showed that the infection was mainly adenovirus type 3 and type 7, and the dominant type of adenovirus in different years alternated between type 3 and type 7. Complete gene sequencing showed only a few point mutations in genes of adenovirus type 55 we isolated, and no other gene mutation and recombination were found.Conclusions:The main types of adenovirus were types 3 and 7 among children with inluenza-like illness in Xuzhou from 2013 to 2019, and the infected population was mainly preschool children, and the detection peak was in spring and summer.
7. Differential expression of long non-coding RNAs in peripheral blood of type 2 diabetic patients

Objective:
To screen and verify the differential expression profiles of long non-coding RNAs(LncRNAs) in peripheral blood of patients with type 2 diabetes mellitus(T2DM), to identify the potential molecular specific markers of early T2DM.
Methods:
The blood samples of 4 type 2 diabetic patients and 4 normal control subjects were collected for microarray analysis. Then six candidate markers of LncRNAs screened from the differential expression profile were tested by qRT-PCR among the subjects (80 cases in the T2DM group and 50 cases in the control group). The possibility of these LncRNAs as molecular diagnostic markers was analyzed, and finally two of them were carried out by receiver operating characteristic (ROC) analysis.
Results:
Compared with control subjects, there were differentially expressed 133 LncRNAs in type 2 diabetic patients, among which 5 were up-regulated with the maximum up-regulated fold 3.29 and 128 were down-regulated with the maximum down-regulated fold 8.99. Six down-regulated LncRNAs were selected for validation and revealed a similar result to that of microarray.The expressions of two LncRNAs(NONHSAT160746 and NONHSAT140069) in peripheral blood of diabetic patients were significantly lower than those of control subjects (
