1.Inhibitory effects of domestic incadronate on osteoclastic bone resorption in vitro

Objective To study the effects of domestic incadronate(ICD) on osteoclastic bone resorbing lacunae. Methods Rabbit's osteoclastes were coincubated with bone slices and microphotographed,microdensitometric scanned and analyzed by computer image analysis system. Results ICD (0.5,5 and 50 μmol/L) caused dose-dependent reduction in the number of osteoclastic bone resorbing lacunae. The inhibitory effect ICD (5 μmol/L) was more remarkable than that of aminopropanediphosphonate (APD). Both ICD and APD reduced the surface area of osteoclastic bone resorbing lacunae. The effect of ICD was stronger than that of APD. Conclusion ICD directly suppresses osteoclastic bone resorption and may be effective in osteoporosis.
2.Effects of total astragalosides on hepatoma cells apoptosis and wtp53 expression

AIM\ To study whether total astragalosides(TA) induces apoptosis in human hepatoma cells and its mechanism of action. METHODS\ The human hepatoma HepG2 and Bel 7404 were cultured with TA (20,40 and 80 mg?L -1 ) for 6 days and DNA fragmentation and the expression of wtp53 protein were determined by flow cytometry. RESULTS\ HepG2 and Bel 7404 cell apoptosis induced by TA(20,40 and 80 mg?L -1 ) were found by flow cytometry. The apoptotic peak of HepG2 cells increased in dose dependent manner in the range from 4 1% to 74 5%; The apoptotic peak of Bel 7404 cells also increased from 4 5% to 50 5%. The expression of wtp53 in HepG2 cells and Bel 7404 cells treated with TA (80 mg?L -1 ) were upregulated. CONCLUSION\ TA may induce apoptosis in HepG2 cells and Bel 7404 cells, its mechanism of action might be associated with upregulating the expression of wtp53.
3.Diagnostic analysis of endoscopic Lugol solution staining combined with narrow band imaging for ;esophageal cancer and precancerous lesions

Objective To investigate the diagnostic value of Lugol solution staining combined with nar-row band imaging (NBI)for esophageal cancer and precancerous lesions.Methods From December 201 3 to December 2014,1 87 patients who were with suspicious esophageal lesions and stained with Lugol solution under endoscopy were selected as control group,while from January 2015 to June 2015,124 patients who were with sus-picious esophageal lesions and examined with Lugol solution staining and NBI under endoscopy were selected as observation group.The sensitivity,specificity and accuracy between the two groups were compared and analyzed. Results One hundred and four cases (75.0%)were found among observation group,which was higher than 93 cases (55.6%)among control group (χ2 =12.07,P <0.01 ).The total detection rates of esophageal cancer and high grade intraepithelial neoplasia (HGN)were 29.0% and 17.7% in observation group respectively,which were higher than 1 8.2% and 6.4% in control group (χ2 =4.15,P =0.04;χ2 =8.54,P <0.01 ).The Kappa value in observation group was 0.55,which was higher than 0.36 in control group.The sensitivity and accuracy in observation group were 89.9% and 82.3% respectively,which were significantly higher than 71 .8% and 68.4% in control group (χ2 =9.79,P <0.01 ;χ2 =7.38,P <0.01 ).Conclusion The sensitivity and accuracy of endoscopic Lugol solution staining combined with NBI for esophageal cancer and precancerous lesions are higher than Lugol solution staining,and it is worthy to popularize the application of Lugol solution staining com-bined with NBI in diagnosing esophageal cancer and precancerous lesions.
4.Determination of 1-Deoxynojirimycin in Mulberry Resources by HPLC

OBJECTIVE:To establish a quantitative analysis method to determine the contents of 1-deoxynojirimycin in mulberry resources such as Morus alba,Excrementum bombycis,Folium mori.METHODS:1-deoxynojirimycin in mulberry resources were determined by RP-HPLC method and UV detector on Hypersil C18(250 mm?4.6 mm,5 ?m)column, using acetonitrile-0.1% glacial acetic acid(50 ∶ 50)as moble phase. The flow rate was 1.0 mL?min-1 and the detection wavelength were set at 265 nm with column temperature at 35 ℃. RESULTS:The linear range of 1-deoxynojirimycin were from 5.0?10-4 mg?mL-1 to 1.0?10-3 mg?mL-1 (r=0.999 9)with recoveries of 97.7%(RSD=2.0%,n=6).CONCLUSION:The method is simple, accurate and reproducible for the quality control of Mulberry resources.
5.Role of Helicobacter pylori Eradication in Diarrhea-predominant Irritable Bowel Syndrome

Background:Irritable bowel syndrome (IBS)is a commonly seen functional intestinal disorder. The association between Helicobacter pylori (Hp)infection and IBS is not clarified yet. Aims:To investigate the role of Hp eradication in diarrhea-predominant IBS (IBS-D)and the relationship between Hp infection and IBS-D. Methods:A total of 335 IBS-D patients and 335 health examination subjects were recruited from Jan. 2009 to Dec. 2015 at Transportation Center Hospital of Yunnan Province and People's Hospital of Xishuangbanna Dai Autonomous Prefecture. All IBS-D patients were hospitalized patients and fulfilled the Rome Ⅲ criteria. Positive for both rapid urease test and 14 C-urea breath test was defined as Hp infection. Hp-positive IBS-D patients recruited in 2005 received Hp eradication therapy and were allocated into two groups according to the results of eradication therapy. Pinaverium bromide and montmorillonite powder were given in both groups and the clinical efficacy for IBS symptoms was assessed after treatment and followed up for 6 months. Results:The Hp infection rate of IBS-D patients was significantly higher than that in healthy controls (53. 7% vs. 41. 8%,P < 0. 05). In IBS-D patients received Hp eradication therapy,51 were allocated into eradicated group and 26 into non-eradicated group. No significant differences were observed in individual symptoms including abdominal pain/discomfort,bloating and stool frequency and the overall efficacy for global symptoms (68. 6% vs. 65. 4%)between the two groups (P > 0. 05). After a 6-month follow up,the differences were still insignificant (P > 0. 05). Conclusions:Hp infection is more prevalent in IBS-D patients than in general population,however,eradication of Hp has no impact on IBS symptoms. The association between Hp infection and IBS-D needs further study.

Aim To study the protective effect of total extract of astragalus(TEA) on primary rat hepatocytes injured by CCl4 or H2O2 and its mechanism.Methods The primary hepatocytes were isolated by Ⅳ collaganase and injured by CCl4 or H2O2.The contents of malondialdehyde(MDA) and glutathion(GSH),glutathion peroxidase(GSHpx) activity of hepatocytes and the AST and /or ALT level in cultural supernatant were determined by general methods. Results (1) The elevation of MDA content of hepatocytes and AST level in supernatant of cultural hepatocytes,and the loss of GSH content and GSHpx activity induced by CCl4 were restored remarkably by TEA(5~80 mg?L-1);(2)The elevation of ALT level in the supernatant of hepatocytes and MDA content of hepatocytes, and the loss of GSH content and GSHpx activity induced by H2O2 were improved significantly by TEA(5~80 mg?L-1) treatment.Conclusion The results suggest that TEA possess direct protective action on primary hepatocyte in vitro injured by CCl4 or H2O2. These might be associated with its anti-oxidative activity.
8.CT findings of desmoplastic small round cell tumor occurred in abdomen and pelvis

Objective To investigate the CT features of desmoplastic small round cell tumor (DSRCT)occurred in abdomen and pelvis.Methods The CT images of 4 patients with pathologically proven DSRCT occurred in abdomen and pelvis were retrospectively analyzed.Results The CT features common to the 4 DSRCT in abdomen and pelvis were as follows:the lobulated soft-tissue masses were very large;the masses often involved omental and serosal surfaces of abdomen and pelvis organ and had close relationship to the surrounding organs;the originating site of the tumors were not apparent;intratumoral hemorrhage or necrosis were often present;and contrast-enhanced CT scan showed mild to medium heterogeneous enhancement.Conclusion The CT features of DSRCT in abdomen and pelvis are rather specific and can provide helpful information to the clinical diagnosis.
10.Effects of the total extract of astragalosides on the growth, AFP secretion and γ-GT activity of human hepatoma cells

AimTo study the effect of the total extract of astragalosides(TEA) on the growth, AFP secretion and γ-GT activity of human hepatoma cells invitro.Methods The human hepatoma HepG2 and Bel-7404 cells were cultured with TEA 5~160 mg · L-1 for 6 days. Anti-hepatoma activity was measured by MTT method. AFP was assaysed by ELISA and γ-GT was determined by enzyme-substrate method. Results The growth of the human hepatoma HepG2 and Bel-7404 cells and the secretion of AFP of HepG2 cells were reduced significantly by TEA 5~160 mg· L-1. The γ-GT activity of HepG2 cells was inhibited and its ALB content was increased by TEA 20, 40 and 80 mg · L-1. The γ-GT activity of Bel-7404 cell was inhibited and its ALB content was increased byTEA 40 and 80 mg · L-1 Conclusion These results suggest that TEA has inhibitory effects on the growth, AFP secretion and γ-GT activity of human hepatoma cells.
