Chinese Journal of Tissue Engineering Research 2005;9(43):151-153
BACKGROUND: High blood lipid can cause a series of cardiovascular and cerebrovascular disease, which has been commonly accepted.OBJECTIVE: To probe into the rule of changes of blood lipid of the elders in aging process.DESIGN: Cross section survey.SETTING: Department of Nutrition, Shanghai Huadong Hospital. PARTICIPANTS: Totally 605 employees who took part in body examination and participated this experiment voluntarily from July 2000 to August 2004, were recruited, including 541 males and 64 females, 91 patients < 70 years , 514 ≥ 70 years, 271 patients having the history of coronary heart disease, 83 of malignant tumors, and 251 of other diseases.METHODS: Blood lipid level of totally 605 who accepted physical examination was detected, at the same times, the disease history and common physical examination status were inquired. Venous blood of the patients was drawn after fasting over 12 hours, separating serum on the morning of the same day. Total cholesterol and triacylglycerol of the blood were detected with enzyme assay by using Automated Biochemistry Analyzer. Total cholesterol: 3.6-6.5 mmol/L; triglyceride: 0.4-1.8 mmol/L were considered as normal level, Total cholesterol: > 5.72 mmol/L;Triglyceride: >1.72 mmol/L were considered as abnormal criteria.MAIN OUTCOME MEASURES: Comparison of the blood lipid index of the subjects of different age, disease type and gender RESULTS: Totally 605 elders were involved and all data of them was enthe blood lipid index of the subjects of different age: Peak of total cholesterol appeared at 70 to 74 years, then decreases gradually, the total cholesterol level of the subjects of 70 to 74 years was significantly higher than that at other age (P < 0.05). While Triglyceride level had the trend of going up with growing older, except that of 75-79 ages group slightly went eases: The level of total cholesterol and triglyceride of the subjects in the malignant tumor group was lower than that of other disease groups, level of triglyceride of the subjects in the coronary heart disease was higher than detection rate of blood lipid of the subjects: Detection rate of total cholesterol was 15.5%, while that of triglyceride was 35.9%. There was no significant difference of the detection rate between male and female subjects (P > 0.05).CONCLUSION: Change of blood lipid metabolism of elder with aging is related to the hormone change of different genders and disease types.

Objective To evaluate the effects of adenovirus-thediated PTEN gene transfer on growth and invasion of H22 cells in vitro. Methods H22 cells were tnmsfected with Ad-PTEN and Ad-PTENG-129R it vitro, The mRNA expressions of PTEN were detected with RT-PCR. Cell viability was determined by MTT assay. Invasion of H22 in vitro was observed using Boyden chamber. Results Gene and protein expressions of PTEN were shown to be up-regulated when H22 was transfected with Ad-PTEN. MTT assay showed that the group transfected with Ad-PTEN had much lower cell viability than other groups (P<0.05) . The number of 1422 invasion cells in the group of Ad-PTEN was (15.64±3.27) %, which was lower than the number in other groups (P<0.05). Conclusion These results imply that the number of H22 invasion cells was significantly decreased after infected with Ad-PTEN in vitro.

Journal of Pharmaceutical Analysis 2006;18(2):106-108
Distributed architectures support increased load on popular web sites by dispatching client requests transparently among multiple servers in a cluster. Packet Single-Rewriting technology and client address hashing algorithm in ONE-IP technology which can ensure application-session-keep have been analyzed, an improved request dispatching algorithm which is simple, effective and supports dynamic load balance has been proposed. In this algorithm, dispatcher evaluates which server node will process request by applying a hash function to the client IP address and comparing the result with its assigned identifier subset; it adjusts the size of the subset according to the performance and current load of each server, so as to utilize all servers' resource effectively. Simulation shows that the improved algorithm has better performance than the original one.

In this study, we aimed to investigate the influences of conditioned medium from human umbilical vein endothelial cells (HUVEC) on cancer stem cell phenotype of human hepatoma cells. HUVEC and human hepatoma cells (MHCC97H) were cultured, respectively, and then the MHCC97H cells were co-cultured with conditioned medium from HUVEC (EC-CM) with Transwell system. Anti-cancer drug sensitivity, colony-formation, migration/invasion ability, expression of cancer stem cell marker and sphere formation were performed to determine the cancer stem cell phenotype in MHCC97H cells. We found that MHCC97H cells co-cultured with EC-CM exhibited significantly higher colony-formation ability and lower sensitivity of anti-cancer drugs 5-FU and Cis. Transwell assay showed that treatment with EC-CM obviously increased migration and invasion of MHCC97H cells. Moreover, increased sphere forming capability and expression of CD133 in MHCC97H cells were observed after co-cultured with EC-CM. These results suggested that EC-CM could promote cancer stem cell phenotype of hepatoma cells.

Objective:To investigate the expression of TAZ and KLF5 and their clinical significance in hepatocellular carcinoma ( HCC).Methods:We freshly collected 76 samples of surgically resected HCC and matched normal tumor-adjacent tissues and detected TAZ and KLF5 expression in these samples using immunohistochemical staining. The clinical significance of TAZ and KLF5 protein expression were analysed. Results:The protein expression of TAZ and KLF5 in HCC tissues was significantly higher than those in matched normal tumor-adjacent tissues ( P=0.001;P=0.035 ).Clinicopathological analysis suggested that TAZ and KLF5 protein expression were associated with histopathological differentiation ( P=0.007;P=0.047 ) and TNM stage ( P=0.009;P=0.040).TAZ was positively correlated with KLF5 protein in HCC tissues (r=0.651,P=0.003).Conclusion:The high-expression of TAZ and KLF5 are correlated with poor clinicopathological characteristics, and TAZ is positively associated with KLF5 in HCC tissues, suggesting that TAZ may promote tumor progression through inhibition of KLF5 protein degradation in HCC.

Academic Journal of Second Military Medical University 2001;0(09):-
Objective:To investigate the role of SDF-1/CXCR4 in metastasis of renal cell carcinoma and to observe the intracellular location of different CXCR4 segments in renal carcinoma cells. Methods:The potential nuclear localization sequences of different CXCR4 were discovered by nuclear localization software and experiments.Full length and truncated forms of CXCR4 were fused with green fluorescent protein pEGFP-N1 and their influence on subcellular localization was examined by confocal microscopy after transfecting them into renal carcinoma cell line A498.Results:Analysis with PSORT Ⅱ Prediction revealed that the nuclear localization sequence region of CXCR4 was located between amino acids 146 and 149(RPRK).Expression products of the recombinant plasmids with SDF-1 stimulation, including EGFP-CXCR4(1-510 bp),EGFP-CXCR4(1-765 bp) and wild-type EGFP-CXCR4,were mainly located in the cell nuclei. However, expression product of EGFP-CXCR4(1-267 bp) with SDF-1 stimulation was mainly located in the renal cell cytoplasm.Expression product of wild-type EGFP-CXCR4 full length plasmid without SDF-1 stimulation was mainly located in the cell cytoplasm;these results accorded with the results of bioinformatics analysis. Conclusion:Nuclear localization sequence of CXCR4 is located in the amino acids 90 to 170,which provides a theoretical basis for further clarifying the nuclear localization sequences of CXCR4 in renal cell carcinoma cells and for finding new potential target for inhibiting the metastasis of renal cell carcinoma.
