The initial study of the tumour DNA fingerprints using MYO minisatellite DNA probe wascarried out,and then,by means of DNA recombinant techniques,the fragment of MYO min-isatellite DNA probe obtained from plasmid pUC19-MYO was inserted into plasmid pGEM-4Z containing RNA polymerase promotor,thus a sub-clone refferred as pGEM-4Z-MYOwas constucted.That made an offer of the conditions of preparing RNA probe in order to in-cerase the sensitivities of DNA fingerprinting and laid a foundation for raised the efficiency of de-tecting the polymorphism of the minisatellite DNA.
3.Preoperative evaluation on lymphatic metastasis of gastric cancer

Evaluating lymph node metastasis of gastric cancer preoperatively can help surgeons to choose the optimal extent of surgical lymph node resection. The diagnostic accuracy of computed tomography for lymph node metastasis of gastric cancer is about 70% , which is less accurate than endoscopic ultrasonography,and more accurate than magnetic resonance and positron emission tomography. Many molecular markers can predict the lymph node metastasis. The prognostic role of gene expression profiles is better than that of a marker. Sentinel lymph node and mathematical methods (Maruyama software, artificial neural network) may be useful in predicting the lymph node metastasis. Laparoscopy and quantum dots technique are potential methods in predicting lymph node metastasis.

5.Correlation between persistent high-risk human papillomavirus infection and vaginal microecology and cervical lesions

Abstract: Objective To investigate the correlation between persistent and non-persistent HPV infection and vaginal microecology and cervical lesions, and to provide the basis for HPV prevention and treatment. Methods In this prospective study, 229 female patients with high-risk type (HR-HPV) were selected for cervical cytology and vaginal microecological examination in the gynecological outpatient department of Baise Maternal and Child Health Hospital from January 2018 to June 2021. The patients were followed up for 1 year to detect persistent HR-HPV infection. The relationship between HR-HPV persistent infection and vaginal microecology and cervical lesions was analyzed using the HPV-negative group as a control. Results Among 229 patients with HR-HPV, there were 109 patients with persistent HR-HPV infection and 120 patients with non-persistent HR-HPV infection in 1-year follow-up, and the incidence of persistent HR-HPV infection was 47.6%. In the HR-HPV persistent and non-persistent infection and HPV-negative groups, the bacterial vaginal incidence was 20.2%, 15.0% and 8.6%, respectively; vulvovaginal candidiasis was 19.3%, 13.3% and 7.9%, respectively; trichomoniasis vaginitis was 12.8%, 9.2% and 4.5%, respectively; mixed infection was 10.1%, 6.7% and 2.7%; H2O2 detection rate was 24.8%, 18.3% and 12.0%,the positive rate of pH value was 52.3%, 40.8% and 36.4%, and microecological normal detection rate was 22.9%, 32.7% and 40.2%, respectively. There were significant differences among the three groups (χ2=10.634, 10.522, 9.010, 9.374, 10.054, 8.268, P<0.01). In the HR-HPV persistent and non-persistent infection groups, the rates of atypical squamous cell detection were 12.8% and 10.0%, and 8.3% and 4.2% for low-grade squamous cell lesions, and 4.6% and 1.7% for high-grade squamous cell carcinoma, 2.8% and 0 for squamous cell carcinoma, respectively. There was no significant difference in the composition of atypical squamous cells between the two groups (χ2=4.358, P>0.05), there were significant differences in the composition of low-grade, high-grade and squamous cell carcinoma (χ2=11.472, 12.685, 11.378, P<0.01). Spearman rank correlation analysis showed that the presence or extent of HPV infection was positively correlated with bacterial vaginosis, vulvovaginal candidiasis, trichomonal vaginitis and mixed infection (P<0.05), positively correlated with H2O2, sialdase, leucocyte esterase,pH positive and positive for all four items (P<0.05), negatively correlated with microecology (P<0.01), positively correlated with low grade, high grade and squamous cell carcinoma (P<0.01), and not significantly correlated with atypical squamous cell carcinoma (P>0.05). Conclusion Persistent cervical HPV infection is an important factor of dysregulation in vaginal microecology and aggravates the degree of dysregulation in vaginal microecology, which is related to the development of cervical lesions.
6.Source Identification of Human Biological Materials and Its Prospect in Forensic Science

Source identification of human biological materials in crime scene plays an important role in reconstructing the crime process. Searching specific genetic markers to identify the source of different human biological materials is the emphasis and difficulty of the research work of legal medical experts in recent years. This paper reviews the genetic markers which are used for identifying the source of human biological materials and studied widely, such as DNA methylation, mRNA, microRNA, microflora and protein, etc. By comparing the principles and methods of source identification of human biological materials using different kinds of genetic markers, different source of human biological material owns suitable marker types and can be identified by detecting single genetic marker or combined multiple genetic markers. Though there is no uniform standard and method for identifying the source of human biological materials in forensic laboratories at present, the research and development of a series of mature and reliable methods for distinguishing different human biological materials play the role as forensic evi-dence which will be the future development direction.
7.Genetic differentiation and patterns of gene flow of seven minority populations in Xinjiang

Objective Genotype data of nine CODIS STR loci were gathered to examine the features of population differentiation and gene flow of seven Xinjiang minorities.Methods Heterozygosity,Nei's coefficient of genetic differentiation,Nei's genetic distance and Wright's F-statistics were calculated. Statistical tests using exact method were performed to measure the level of differentiation.Phylogenetic trees were constructed by Mega;AMOVA was processed by Arlequin.R-matrix model had been applied to describe the patterns of gene flow.Results It shows that average genetic heterogeneity for each population was above 0.7 with genetic differentiation coefficient below 2%.Statistical tests for population differentiation were significant for most of the loci.Phylogenetic analysis and AMOVA showed that all populations were divided into three main groups.The R-matrix analysis reflected that Uygur,Kirgiz and Ozbek had more amounts of gene flow than other populations,while the pattern of Hui was more isolated.Conclusion The seven minorities in Xinjiang are independent populations,while the level of differentiation is at average.The relationship in evolution is not far from each other,with wide gene flow.

Objective To investigate the expression and function of Fca receptor on human airway smooth muscle(ASM)cells.Methods RT-PCR and immunofluorescence technique were used to detect the expression of Fca receptor on human ASM cells.The effect of IgA on intracellular calcium concentration of human ASM cells was measured by using Fura-2/AM as a calcium indicator.Results RT-PCR and immunofluorescence staining confirmed the presence of Fca receptor on human ASM cells.Compared with control,secretory IgA(sIgA)induced a rise in intracellular calcium concentrations on human ASM cells after 90min incubation(P0.05).Conclusion Fca receptor was expressed on human ASM cells.SIgA increased the intracellular calcium concentration of human ASM cells.
9.Expression of human epidermal growth factor receptor 2 in breast cancer and its correlation with tumor necrosis factor-α

Objective To determine the expression of Human Epidermal Growth Factor Receptor 2 (HER-2) in breast cancer and its correlation with Tumor Necrosis Factor-α (TNF-α). Methods We used SABC method to detect the expression of HER-2 and TNF-α in 112 cases of primary breast cancer, and analyzed the association of HER-2 expression with clinicopathological features and TNF-α expression. Results The positive expression rate of HER-2 in breast cancer was 35.71% (40/112), of which, the expression rate was 35.05% (34/97) in 97 invasive tubic breast cancer, and 33.33% (2/6) in invasive lobular breast cancer,25.00% (1/4) in mucinous carcinoma, and 50. 00% (2/4) in intraductal carcinoma. There was no significant correlation between HER-2 and any clinicopathological features of primary breast cancer ( P > 0. 05 ). The expression of HER-2 in breast cancer had no signiciant association with TNM stage, age, age of menarche,menopause, gravidity, parity and family history of cancer ( Ps > 0. 05 ), The positive rate of TNF-α was 23.61% (17/72) when HER-2 was negative, whereas the positive rate of TNF-α was 52. 50% (21/40) when HER-2 was positive. The expression of HER-2 was signiciantly positively correlated with the expression of TNF-α (r = 0. 881, P < 0. 05 ). Conclusion HER-2 expression had a positive correlation with TNF-α in breast cancer,but the specific mechanism is still unclear.
10.Expression of tumor necrosis factor a and correlation with estrogen receptor in breast cancer

Objective To study the expression of tumor necrosis factor -α (TNF-a) and correlation with TNF-a in breast cancer. Methods We used SABC method to detect the expression of TNF-a in 112 cases of primary breast cancer, and analysis the relationship between TNF-α with histologic grades and ER. Results The positive expression rate of TNF-α in breast cancer was higher than those of normal tissues and lobular hyperplasia of the breast (X2 =8.573, P =0.014). There was no significant correlation between histologic grades and TNF-α (X2 =1.304, P =0.521). TNF-a had a positive correlation with ER in breast cancer (X2=11.949, P =0.001). Conclusion The positive expression rate of TNF-α in breast cancer was higher than those of normal tissues and lobular hyperplasia of the breast.TNF-α had a positive correlation with ER in breast cancer, but the exact mechanism is unclear.
