2.CORRELATIONS BETWEEN BLOOD PRESSURE AND BODY WEIGHT, SERUM LEPTIN IN HIGH CALORIE DIET-INDUCED OBESE RATS

Objective To examine the change of body weight (BW) and blood pressure (BP) in obese rats, clarify relationships between BP and BW and other factors. Methods Male Spraque-Dawley rats were fed either with normal diet (ND) or high calorie diet (HC) for 20 weeks. BW and BP of tail artery were observed biweekly and tetraweekly respectively; serum leptin and fasting insulin (FINS) were detected by enzyme-linked immunoadsordent assay (ELISA) and radioimmunoassay (RIA) respectively. Fasting plasma glucose (FPG) and free fatty acid(FFA) were measured by conventional means. Results BW, abdominal fat weight (AFW), ratio of abdominal fat weight to body weight (RF/W), systolic blood pressure (SBP), diastolic blood pressure (DBP), serum levels of leptin and FINS, FPG, FFA increased in the HD group after 20 weeks diet intervention (P<0.05 or P<0.01). SBP was strongly correlated with BW, leptin, FINS and FFA (P<0.05), DBP was correlated with FFA (r=0.47, P<0.05). In addition, leptin was positively correlated with BW, AFW, RF/W, FINS and FFA (P<0.05 or P<0.01). Conclusion In this study of high calorie-diet induced rats, the gain of BW is accompanied by increased BP. The obese rats have hyperleptinemia, hyperinsulinemia, hyperglycemia and dyslipidemia which may have important effects on the development of obesity-related hypertension. RF/W is the key factor in which affect serum leptin level.
3.The clinical efficacy of recombinant tissue plasminogen activator for the treatment of ischemic cerebrovascular disease caused by cerebral thrombosis

Objective To study the clinical efficacy of intra-arterial thrombolysis with recombinant tissue plasminogen activator (rt-PA) for the treatment of ischemic cerebrovascular disease caused by cerebral thrombosis. Methods A total of 245 patients accepted by our hospital during May 2013 and July 2015 were divided into the observation group (n=148) and the control group (n=97). All patients were given conventional process for controling blood pressure and blood lipids. Patients in observation group received intra-arterial thrombolysis with rt-PA, while patients in control group accepted conventional treatment. At the time of admission, the demographic characteristic, vascular influencing factors, baseline clinical findings, laboratory findings and neurological deficits were collected. The improvement of neurological function was evaluated by the modified Rankin scale 3 months after treatment. The levels of fibrinogen (FIB), D-Dimer, activated partial thromboplastin time (APTT) and thrombin time (PT) were measured before and 24 h after the treatment. Results There were no significant differences in demographic characteristic and general clinical data between the two groups ( P>0.05). The proportion of patients with improved neurological function was significantly higher in observation group than that of the control group (83.11%vs. 53.61%, P<0.05). There were no significant difference in coagulation index and fibrinolysis index before treatment between the two groups (P>0.05). Twenty-four hours after the treatment, the levels of FIB, D-Dimer, APTT and PT were significantly improved in the observation group compared with those before treatment. The level of FIB was significantly decreased, D-Dimer was significantly increased, APTT and PT were significantly prolonged in observation group compared with those of control group (P<0.05). Conclusion The rt-PA can effectively dissolve thrombosis and correct the coagulation system and fibrinolytic system.
4.Clinical studies on the combiantion of single lower limb traction and massage therapy for the lumbosacral tunnel syndrome

Objective To observe the therapeutic effect of single lower limb traction plus massage for the lumbosacral tunnel syndrome.Methods The selected 80 candidate patients were divided into the observation group with 38 patients and treatment group with 39 patients. The observation group received single lower limb traction plus massage and the treatment group received pelvic traction plus massage. Both groups therapy last 5 days, The Japanese orthopaedic association scores (JOA) was evaluated for subjective symptoms, signs, functional activity and bladder function score. The therapeutic effect was evaluated.Results The total effective rate (94.7% vs. 82.1%;χ2=12.160,P=0.007) in the observation group was significantly higher than that in the control group. O, The scores of JOA at one week after treatment (20.2 ± 2.0 vs. 18.3 ± 2.8;t=3.419, P<0.01) and two weeks after treatment (24.4 ± 1. 4vs. 22.6 ± 3.0;t=3.359,P<0.01) in the observation group were significantly higher than those in the control group.Conclusions The single lower limb traction plus massage can improve the clinical symptoms of patients with lumbosacral tunnel syndrome, and be superior to the pelvic traction plus massage.
5.Research Progress of Molecular Imprinting Cryogel

Cryogel is a kind of elastic, spongy hydrogel prepared via addition polymerization from monomers or cross-linking reaction from a polymer at a temperature lower than the freezing point of the solvent. Cryogel has been principally used in molecular imprinting cryogel based solid phase extraction in the field of analytical chemistry, which combines both the advantages of high selectivity of the molecular imprinting and the porous structure, high throughput of a cryogel. Molecular imprinting cryogel can be used as the stationary phase in a packed column, a monolith column, or the adsorbent in a separation membrane. A variety of molecules can act as template molecules, including small molecules, macromolecules, a certain fragment of a macromolecule, and even an inorganic ion. Both a simple template and a complex template can be used. When employing more than a molecule ( or an ion) as templates simultaneously, the method is termed as Mixed template. While high abundance components of a complex sample act directly as mixed template, we name the method as Pending template. Further, while an analogue, an isotopic label, or a fragment of the target substance acts as a template, the method is defined as Dummy template. However, the research on cryogel is insufficient and the application of the cryogel is narrow hitherto.
6.Advances on biological indicators of diabetic retinopathy and characteristic peculiarities

·Diabetic Retinopathy ( DR ) has gradually worsening and lingering, which is the leading cause of global young people blindness. With the progression of the disease, patients with diabetes mellitus ( DM) will have different degrees of DR. If you can not prevent and give acute early intervention, once the visual acuity decreased significantly, DR would be difficult to reverse. DR progressively has worsening, the treatment status has no optimistic. Therefore, DR in the early prevention and treatment will be indispensable. This article summarizes some of the early warning of the occurrence and development of biological markers and characteristic indicators in order to provide a basis for the early prevention of DR.
7.Thinking and practice of study on material basis of parched Chinese herbal medicines and their quality evaluation.

Though parched Chinese herbal medicines contain less effective or index components, their pharmacological actions do not reduce or even become improved to some extent. However, the current studies related to material basis could not explain the changes in property, flavour and efficacy of parched Chinese herbal medicines. Meanwhile, due to the lack of objective and specific evaluation indexes, the quality evaluation could not reflect features of parched Chinese herbal pieces. Therefore, how to break the bottleneck for the studies on parched Chinese herbal pieces, make further innovation and conduct in-depth studies on the material basis of parched Chinese herbal medicines are common problems that medical scholars are facing. According to the findings in the previous studies, the author proposed to explain the material basis of parched Chinese herbal medicines by studying Maillard reaction and establish specific quality evaluation indexes according to the features of parched Chinese herbal pieces, and conducted relevant studies.

8.Extensive Imprinting Adaptability of Polyacrylamide-based Amphoteric Cryogels Against Protein Molecules

Lysozyme, pepsin, ovalbumin, hemoglobin andγ-globumin were chosen as templates to investigate the imprinting capability of amphoteric polyacrylamide cryogels. Prepolymerizing solutions contained acrylic acid and allyl amine, as well as acrylamide and N,N'-methylenebisacrylamide as the functional monomers. As a result there were both acidic and basic functional groups in the polymers, facilitating effective interactions with likewise amphoteric proteins. The proteins differ greatly and cover wide scopes of molecular weights and isoelectric points. Regardless of the values of the molecular weights and isoelectric points, all the templates gave higher retentions on the MIP tubes than on the NIP tube. The MIP of lysozyme indicated the highest imprinting factor of 7. 0, and that of γ-globumin showed the lowest, 2. 0. The values of other proteins were intervenient. Conclusively the amphoteric polyacrylamide cryogels were suitable imprinting materials for various proteins, and could potentially be used for protein recognition, purification, and depletion.

10.Infection of tupaia hepatocytes with hepatitis C virus in vitro.

OBJECTIVETupaia belangeri (tree shrew) has a close phylogenetic relationship with primates and has been shown to be susceptible to a variety of human viruses. This study was conducted to investigate whether or not hepatitis C virus (HCV) could infect primary tupaia hepatocytes (PTHs) in vitro.
METHODSSerum-derived HCV was cultivated with PTHs, and then positive and negative strand HCV RNA in PTHs, as well as the encapsidated HCV RNA in the culture medium were detected to evaluate the infection. Virus from the culture medium of the infected PTHs was passed to naïve PTHs, and the quasispecies of HCV were compared among the inoculum and PTHs after infection and passage.
RESULTSBoth positive and negative strand HCV RNA were detected in PTHs after infection. The negative strand RNA was detectable from day 5 to day 10 after infection, while the positive strand RNA was positive up to day 14. HCV RNA, which was RNase resistant, could be detected from the culture medium of the infected PTHs from day 3 to day 14. Production of infectious virons of PTH were demonstrated by passage HCV to naïve PTHs. Compared analysis of HCV quasispecies after infection and passage showed that PTHs were selectively infected with defined HCV quasispecies, and new quasispecies emerged in PTHs after passage.
CONCLUSIONThe present study strongly indicates that PTHs could be infected by HCV and support HCV replication in vitro. Our results would be helpful for the establishment of a tupaia model of HCV infection.
