Objective To investigate the protective effect of mucopolysaccharide polysulfate cream(MPC) and micro needle array drug delivery technologarray(MNADDT) on the blood vessel of arteriovenous fistula.Methods Eighty cases maintenance hemodialysis patients with autogenous arteriovenous fistula were randomly divided into control group(with the treatment of MPC) and study group(with the treatment of MPC plus MNADDT),40 patients in each group.Both groups were studied for the duration of 12 months.The vascular complications of puncture pain sense,puncture failure times,phlebitis,hardening of the arteries,internal fistula stenosis or embolism,pseudoaneurysm,swollen hands comprehensive syndrome of two groups were observed,and the changes of arteriovenous fistula blood flow,blood vessel diameter by Doppler ultrasound were observed in the two groups for 12 months.Results (1)The incidence of puncture pain(2.71±0.11 vs.2.76±0.14,t=2.21,P<0.05),puncture failure times(38 times vs.73 times,χ2=11.425,P<0.05),phlebitis(2 cases vs.8 cases,χ2=4.341,P<0.05) and blood vessel sclerosis(2 cases vs.9 cases,χ2=5.446,P<0.05) of the treatment group were significantly lower than that of the control group during the study(P<0.05).(2)Blood flow of arteriovenous fistulain of control group before and after treatment were (859.7±148.3) ml/min and (946.5±169.2) ml/min respectively,and the difference was significant(t=2.356,P<0.05).While blood flow of arteriovenous fistulain,cephalic vein diameter,head of venous flow velocity of study group before and after treatment were (824.6±171.5) ml/min and (1 218.1±241.9) ml/min,(5.59±0.74) mm and (6.02±0.57) mm,(94.23±27.35) cm/s and (115.85±36.63) cm/s,respectively,and the differences were significant(t=8.212,2.382,2.877,P<0.05),there were no significantly changes in the above indexes in the study group after treatment(t=5.3612,2.152,2.281,P<0.05).Conclusion MPC plus MNADDT can reduce the vascular complications of arteriovenous fistula,improve blood vessel diameter and increase blood flow.

Objective To study the effect of hypoxia-ischemia (HI) on the brain-derived neurotrophic factor (BDNF) expression in the brain cortex and the hippocampus of immature rats,and to provide new therapeutic strategies for HI brain injury.Methods Three-day-old rats were divided into 2 groups.One group of rat pups were subjected to the left carotid artery ligation followed by 60 mL/L oxygen for 2.5 hours(HI-treated rats).The other group of rat pups were only subjected to the left carotid artery separation without ligation and 60 mL/L oxygen (sham-treated rats).The brain tissues were prepared at 3,7 and 14 d after treatment.Cresyl fast videt(CV) staining was used to evaluate the damage of the cortex and the hippocampus and check whether the models were successfully made.Immunostaining was used to determine the changes in BDNF positive cells in the brain cortex and the hippocampus after HI.Western blot analysis was used to evaluate the expression of BDNF protein in the brain cortex and the hippocampus after HI.Results Models were successfully made.CV staining showed that there was brain damages and area loss in the cortex and the hippocampus after HI.BDNF immunostaining showed that the number of BDNF-positive cells was significantly decreased in the cortex (t =-3.225,-2.298,all P ＜ 0.05) and the hippocampus (t =-3.751,-2.920,all P ＜ 0.05) in the damaged side of the brain compared to the contralateral side in the rats treated with HI and the sham-treated rats at 3 d after surgery,while increased at 7 d(t =3.924,2.838,all P ＜ 0.05 for cortex ; t =4.136,2.256,all P ＜0.05 for hippocampus) and 14 d (t =3.256,2.624,all P ＜ 0.05 for cortex ; t =3.051,2.719,all P ＜ 0.05 for hippocampus) after surgery.Western blot analysis showed protein expressions of BDNF:(1) Hippocampus:the protein expressions of BDNF were significantly decreased in damaged side of the brain compared to the contralateral side of rats treated with HI at 3 d(t =-3.388,P ＜ 0.05) after surgery,while increased compared to the contralateral side of rats treated with HI and the sham-treated rats at 14 d(t =4.874,4.646,all P ＜0.05) after surgery.(2)Cortex:the protein expression of BDNF was significantly decreased in damaged side of the brain compared to the contralateral side of rats treated with HI and the sham-treated rats at 3 d(t =-7.386,-3.256,all P ＜ 0.05) after surgery,compared to the sham-treated rats at 7 d(t =4.439,P ＜ 0.05) and the contralateral side of rats treated with HI and the sham-treated rats 14 d(t =24.161,3.942,all P ＜ 0.05) after surgery.Conclusions The number of BDNF-positive cells and protein expressions are decreased in the cortex and the hippocampus at the early stage of HI injury,and increased at the late stage.BDNF may play a role in the healing stage of HI brain injury.

Objective To explore the association between VEGF gene polymorphism and the changes in cardiac function after HiHiLo. Methods Sixty two men of Han nationality from northern China underwent HiHiLo for 30 days. Training programme was as follows:exposure in hypoxic environment (14.8% - 14.3%O_2) 10hrs per day, hypoxic training three times per week (15.4%～14.8%O_2), and training at sea level in the remaining time. The genotype was analyzed by PCR-RFLP. Results There is an association between VEGF gene polymorphism and the changes in left ventricular structure and function after 4-week HiHiLo. LVM, LVMI, SV and SI after HiHiLo increased significantly in men carrying CC genotype were better than those carrying CT genotype. Conclusion Men of Han nationality carrying CC genotype probably have a better left ventricular function in response to the HiHiLo than those carrying CT genotype. CC genotype can be used as a genetic marker for the better responsiveness of left ventricular function to the HiHiLo.
