1.Protective Effects of Verapamil on Ischemic Precondition for Resection of Liver

Objective To study the protective effect of verapamil against ischemia reperfusion injury when liver was resected.Methods The routine Pringle's maneuver hepatic vascular occlusion in 18~20min. Thirty-nine patients for resection of liver with cirrhosis were randomly divided into two groups, Verapamil group (n=20), the 20 patients with hepatectomy were given by 5mg verapamil through right gastroepiploic vein before blocking porta hepatis.Physiologic saline group (n=19), were injected by 20ml saline in the same way.The variation of postoperative liver function were measured.Liver intracellular [Ca 2+ ]i and adenosine triphosphate (ATP) after being reperfused for 1h were measured as well as the hepatic histopathologic alterations were also examined.Results The levels of intracellular [Ca 2+ ]i concentrations,alanine aminotransferase (ALT) and aspartate aminotransferase(AST)from the verapamil group increment less than those of the physiologic saline group(P
2.Effect of hepatocyte growth factor on left ventricular remodeling after acute myocardial infarction in canine

Background and objectives To investigate the effect of hepatocyte growth factor (HGF) on left ventricular (LV) remodeling after acute myocardial infarction (AMI). Methods AMI was produced by ligation of proximal left anterior descending coronary artery(LAD) in 12 mongrel canines. These animals were randomized into 2 groups. In HGF group (n=6), canines were injected with pcDNA3-HGF lml (about 300ug) at the margin of infarcted myocardium; in control group (n=6) canines were injected with equal volume of normal saline. Cardiac function and left ventricular remodeling were evaluated with echocardiography at 1, 4, 8 weeks after MI. LV myocardium specimens were obtained at 8 weeks and stained with hematoxylin and eosin for histological examination or with sirius red to assess the collagen content. Results Compared with control group, LVEF in HGF group was significantly higher at 4 weeks (49.61+6.66 vs 39.84+6.39; P<0.05) and at 8 weeks (51.57+8.53 vs 40.61+7.67; P<0.05) after AMI, while LVESV was significantly lower in HGF group than that in control group at 8 weeks after AMI (18.98+3.47 vs 25.66+5.86; P<0.05). Posterior left ventricular wall thickness decreased significantly from 1 wk to 8 wks after AMI in control group, while remained unchanged in HGF group. Compared with control group, histological examination showed more neovascularization and less scar, and sirius red staining indicated higher volume of type Ⅲ collagen (7.10±4.06% vs 3.77±1.09%; P<0.05) and lower collagen Ⅰ/Ⅲ ratio value (1.11±0.52 vs 2.94±2.48; P<0.05)in HGF group. Conclusion HGF gene transfer might improve cardiac function and LV remodeling after acute myocardial infarction by stimulating angiogenesis, reducing fibrosis, and reducing myocardial scarring.
3.Detection of chlamydia and mycoplasma in urogenital tract infection and the analysis of drug sensitivity of mycoplasma

Objective To investigate the status of Ureaplasma urealyticum (Uu) ,Mycoplasma hominis (Mh) and Chlamydia trachomatis(Ct) in patients with urogenital tract infection ,and to analyze the drug resistance of mycoplasma .Methods The myco‐plasma and its drug sensitivity of urogenital tract specimens were detected by culture‐identification‐susceptibility integration kit .Ct was detected by using immunochromatographic method .Results Among 497 cases of specimens ,there were 194 cases of simplex Uu infection (39 .0% ) ,9 cases of simplex Mh infection (1 .8% ) ,73 cases of Uu and Mh mixed infection (14 .7% ) ,and 54 cases of Ct infection (10 .9% ) .There were 38 cases of Ct and Uu mixed infection (7 .6% ) in the Ct positive specimens .The isolated myco‐plasmas were sensitive to doxycycline ,josamycin and minocycline ,but resistant to other drugs in varying degrees .Conclusion There are both simplex infection and mixed infection of chlamydia and mycoplasma in the patients with urogenital tract infection . The clinical use of antibiotics should be reasonable based on pathogen detection and drug susceptibility test results .
4.Relationship between the Expressions of Intercellular Adhesion Molecule 1,Heat Shock Protein 70 and Acute Rejection after Pancreas Allotransplantation in Miniature Swine

Objective To investigate the relationships between the expressions of intercellular adhesion molecule 1(ICAM-1),heat shock protein 70(HSP70) and acute rejection after pancreas allotransplantation in miniature swine.Methods Ultrasound guided biopsies of the pancreas and histopathologic examination were performed after the miniature swine models of pancreas allotransplantation were established.According to the histological change,the samples were divided into group Ⅰ(no rejection),group Ⅱ(mild acute rejection),group Ⅲ(moderate acute rejection)and group Ⅳ(serious acute rejection).The expressions of ICAM-1 and HSP70 were analyzed with immunohistochemistry and imaging.Results The expressions of ICAM-1 and HSP70 were increased when acute rejection occurred after pancreas allotransplantation in miniature swine.The levels of ICAM-1 and HSP70 were sequentially high when acute rejection was serious.Conclusion ICAM-1 and HSP70 were involved in pancreas allograft rejection and useful for early diagnosis of acute rejection after pancreas transplantation.
5.An Acute Rejection Monitoring Model for Pancreaticoduodenal Allotransplantation With Enteric Drainage in Piglet

Objective To establish a successful model for pancreaticoduodenal allotransplantation with enteric drainage in piglet and surveillance acute rejection by biopsy.Methods We implemented the model same as protocol of clinical pancrease transplantation.40 piglets were divided into donors and recipients randomly.20 times experimental operation were finished. The total pancreas with a short segment of duodenum was transplanted to a recipient by anastomoses of vessels to the recipient iliac vessels and small bowel segment to the jejunum with enteric drainage. During operation, mean arterial pressure(MAP), central venous pressure(CVP)were monitored. The levels of amylase activity in blood were investigated.Doppler color blood flow ultrasonography was used to evaluate blood flow of pancreatic allografts,and ultrasound-guided percutaneous pancreas allograft biopsy and histopathologic examination were performed.Results The successful rate of operation was 90%. 6.Study on Bacterial Endotoxin Test Method of Boric Acid Rinse

OBJECTIVE:To establish a method for bacterial endotoxin test of Boric acid rinse.METHODS:According to appendix volumeⅡof Chinese Pharmacopoeia(2005 edition),the tachypleus amebocyte lysate(TAL)method was applied for the pre-inference test and interfence test to investigate non-inference concentration,pH value and sensitivity of TAL.RESULTS:Boric acid did not interfere with TAL and the reaction of bacterial endotoxin at 2 times diluted concentration with pH value adjusted above 6.The sensitivity 0.25 EU?mL-1 of TAL was available.CONCLUSION:The TAL method can control the limit of bacterial endotoxins in Boric acid rinse.

In order to construct a eukaryotic co-expression plasmid containing membrane-anchored Sjcl4FABP and Sjc26GST genes and identify their expression in vitro, Sj14 and Sj26 genes were obtained by RT-PCR with total RNA of Schistosoma japonicum adult worms as the template and cloned into eukaryotic expression plasmid pVAC to construct recombinant plasmids pVAC-Sj14 and pVAC-Sj26. Then a 23 amino-acid signal peptide of human interleukin-2 (IL-2) upstream Sj14 or Sj26 gene and a membrane-anchored sequence containing 32 amino-acids of carboxyl-terminal of human placental alkaline phosphatase (PLAP) downstream were amplified by PCR as the template of plasmid pVAC-Sj14 or pVAC-Sj26 only to get two gene fragments including Sj14 gene and Sj26 gene. The two modified genes were altogether cloned into a eukaryotic co-expression plasmid pIRES, resulting in another new recombinant plasmid pIRES-Sj26-Sj14. The expression of Sj14 and Sj26 genes was detected by RT-PCR and indirect immunofluorescent assays (IFA) when the plasmid pIRES-Sj26-Sj14 was transfected into eukaryotic Hela cells. Restriction enzyme analysis, PCR and sequencing results revealed that the recombinant plasmids pVAC-Sj14, pVAC-Sj26 and plRES-Sj26-Sj14 were successfully constructed and the expression of modified Sj14 and Sj26 genes could be detected by RT-PCR and IFA. A bivalent membrane-anchored DNA vaccine encoding Sj14 and Sj26 genes was acquired and expressed proteins were proved to be mostly anchored in cellular membranes.
8.Clinical research of endoscopic great saphenous vein harvesting during coronary artery bypass grafting

Objective To summarize the clinical result of patients undergoing endoscopic vein harvest (EVH) technology to collect greater saphenous vein (GSV) in coronary artery bypass graft (CABG) operation, and to assess the operation outcome of EVH. Methods A total of 862 patients underwent primary CABG, among whom saphenous vein of 482 patients were taken using EVH, and the others by open vein harvesting (OVH) based on patients' willingness. The operation risk factors and complication were compared between the two groups. The 64 multi-slice computed tomography (64-MSCTA) was used to evaluate the vein grafts patency after surgery for 1 year. The vein patency between the two groups was compared. Results There was no significant difference in risk factors of incision complication between two groups ( P>0.05). But the incidence of various incision complication was significantly lower in EVH group (10.2%,49/482) compared with that in OVH group (35.0%,133/380) ( P<0.05). The time of harvesting and the length of conduits was similar between two groups (P>0.05). After 1 year's follow-up, the vein graft patency were 86.0%(404/470) and 87.1%(324/372) in EVH group and OVH group, and there was no significant difference ( P>0.05). Conclusions The decrease in incision complication of EVH is unquestionably superior to those of OVH, especially for those patients with risk factors of incision complication. The EVH vein graft has good patency in short time.
9.Allogeneic bone marrow mesenchymal stem cell transplantation for acute myocardial infarction in rats

BACKGROUND:A number of studies have shown that bone marrow mesenchymal stem cells can survive in the infarcted myocardium and improve cardiac function. OBJECTIVE:To investigate the effects of al ogeneic rat bone marrow mesenchymal stem cells on heart failure in acute myocardial infarction models of rats and possible mechanisms. METHODS:Rat bone marrow mesenchymal stem cells were isolated from the bone marrow of 39 male Wistar rats by density gradient centrifugation with Percol . After ligating anterior descending coronary artery, 39 female Wistar rats were randomly divided into three groups:control group (Dulbecco’s modified Eagle’s medium, n=12), mesenchyma stem cells group (n=15) and mononuclear cells group (n=12). Eight weeks later, hemodynamics and left ventricular function were measured. Histopathological and immunohistochemical examinations were performed. RESULTS AND CONCLUSION:Compared with the control group, left ventricular end diastolic pressure, left ventricular relative weight, the col agen volume fraction of type I and type III in the infarction zone of the left ventricle were al significantly decreased, in contrast to ±dp/dtmax,-dp/dtmax/left ventricular systolic pressure, body weight and vascular density in infarction zone were al significantly increased both in mesenchymal stem cells group and mononuclear cells group. There were no significant differences between two treatment groups except for interventricular septal thickness and vascular density in non-infarction zone. 5-Bromo-2'-deoxyuridine positive cells were observed in the infarction area of mesenchyma stem cells group but no positive cells in mononuclear cells group. Some bal-like cellmasses were found positively stained with desmin and cardiac troponin T. Results have suggested that embedded bone marrow mesenchymal stem cells survived in exogenous host hearts. The therapy of mononuclear cells and mesenchymal stem cells could limit the left ventricular remodeling after acute myocardial infarction and improve left ventricular function through angiogenesis inducing and col agen deposition decreasing.
10.Radiation dose contrast analysis of 320 row CT coronary artery imaging and coronary angiography*

Objective To contrast the radiation dose to patients of 320 row CT coronary artery imaging (CTCA) and coronary angiography(CAG) .Methods 90 patients random sampled with 320 row CTCA were group A .Among of them ,7 patients with cal-cification integral scanning (CI) were A1 group;43 patients with CT coronary artery imaging/cardiac function analysis scanning (CTCA/CFA) were A2 group;40 patients with Prospective scanning (Prospective CTA ) were A3 group;139 patients random sam-pled with CAG were group B .Results Radiation dose had statistically significant between Group A and group B ,A1 and B ,A2 and B ,A1 and A3 ,A2 and A3(P<0 .05) .radiation dose had no statistically significant between Group A3 and group B ,A1 and A2(P>0 .05) .Conclusion In order to reduce the radiation dose to patients ,the prospective CTCA was used in the patients need 320 row CTCA as far as possible .
