1.Effects and mechanism of penhyclidine hydrochloride on myocardial ischemia reperfusion injury in rat

Objective To explore effects and mechanism of penhyclidine hydrochloride (PHC) on myocardial ischemia reperfusion injury in rat.Methods Sixty Sprague Dawley (SD) adult male rats were randomly divided into sham operation group(SHAM group),myocardial ischemia reperfusion injury group(MIRI group) and penhyclidine hydrochloride treatment (PHC group),and 20 rats in every group.Compared left ventricular function of 3 groups after 24 h of ischemia reperfusion.Selected 10 rats of every group and compared the myocardial infarction size after ischemia reperfusion,and the less 10 rats' the level of tumor necrosis factor-α (TNF-αt) and nuclear factor-kB(NF-kB) in myocardial tissue were measured.Results After 24 h of ischemia reperfusion,there was no significant myocardial infarction size in SHAM group,while that in MIRI group were (39.4± 3.7)%,higher than that in PHC group (20.5 ± 2.3)%,and the difference was significant (P < 0.01).The level of left ventricular end-systolic volume (LVESV) of rats in SHAM,MIRI and PHC groups were (0.138 ± 0.024),(0.457 ± 0.032) and (0.280 ± 0.027) ml respectively,left ventricular end-diastolic volume(LVEDV) were (1.012 ± 0.065),(1.395 ± 0.076) and (1.183 ± 0.102) ml respectively,and left ventricular ejection fraction (LVEF) were (85.6 ± 7.3) %,(43.2 ± 4.5) % and (64.8 ± 5.1) % respectively,and there were statistically significant differences between groups (F =54.12,32.70,35.46,P < 0.001).The level of TNF-αtand NF-kB in rats of SHAM,MIRI and PHC group were (5.4 s0.7),(34.2 ±2.9) and (21.6 ± 1.3) pg/mg respectively and (87.6 ± 7.3),(134.2 ± 9.6) and (116.3 ± 8.7) respectively,and there were statistically significant differences between groups (F =59.40,17.97,P < 0.001).There were statistically significant differences between MIRI and SHAM groups,PHC and SHAM groups in terms of above factors (P <0.05).Conclusion PHC can decrease the inflammatory reaction and protect the myocardial ischemia reperfusion injury in rat.

Objective To study the effect of RNA interference suppressing the expression of AKT2 on sensitivity of human pancreatic cancer cell line xenograft in nude mice to gemcitabine.Methods The human pancreatic cancer cell implanted tumor mode1 in the nude mice was established.By abdominal cavity administration and intratumoral injection,the mice bearing tumor were treated with gemcitabine in combination with vector of pAKT2-siRNA.Tumor growth of tumor tissues was observed,the AKT2 mRNA and protein expression levels in tumor tissues detected by RT-PCR and immunohistochemistry respectively,and tumor apoptosis measured by Tdt-mediated dUTP nick end labeling(TUNEL).Results In chemotherapy+AKT2-siRNA group the expression of AKT2 mRNA and protein was significantly lower than in control group,chemotherapy group and chemotherapy+blank plasmid group.The tumor weight and tumor volume in chemotherapy+AKT2-siRNA group were significantly reduced as compared with those in other three groups.The inhibition rate and apoptosis rate in chemotherapy+AKT2-siRNA group were significantly higher than those in other three groups.Conclusion Sensitivity of human pancreatic cancer cell line to gemcitabine could be significantly improved by RNA interference suppressing the expression of AKT2.
6.Female outlet obstruction constipation: assessment with MR defecography

Objective Using MR defecography to assess the morphological and functional anorectal anomalies related to female outlet obstruction constipation, and evaluate the joint disease of anterior and mid pelvic. Methods One hundred and seven female patients, aged 20 to 84 years ( average, 55 years), were diagnosed as outlet obstruction constipation based on clinical symptoms and signs. They all received MR defecography in our institution. The high compliance homemade balloon was inserted into rectum to simulate stool Then relevant measurements were obtained during rest, squeezing and straining, respectively. Results In all the 107 cases, 70 ( 65.4% ) presented rectocele on dynamic MRI; 28 ( 26. 2% ) presented anismus;60 (56. 1% ) presented cystocele; 59 presented vaginal or cervical prolapse(55. 1% ); and, 54 (50. 5% )presented descending perineum. In 85 females (79.4%) multiple disorders were detected, involving more than one pelvic compartment. Conclusion MR defecography allowed to accurately evaluate the morphological and functional anorectal anomalies related to female outlet obstruction constipation, and the joint disease of anterior and mid pelvic.
7.Effects of AKT2 expression on gemcitabine sensitivity in pancreatic cancer cell line Panc-1

Objective To explore the effects of AKT2 expression in pancreatic cancer cell line Panc-1 on the sensitivity towards to gemcitabine.Methods The expression vector pAKT2 shRNA was constructed and transfected into Panc-1 cells by lipofecton.The mRNA and protein expression levels of AKT2 were detected by RT-PCR and Western blot assays,respectively.The changes of gemcitabine sensitivity after shRNA were examined by the MTT assay.Results The mRNA and protein levels of AKT2 in Panc-1 cells were significantly decreased after the transfection,and the median inhibition concentration of gemcitabine against Panc-1 cells was reduced from (1.96 ± 0.22) mg/L to (0.24±0.03) mg/L.The sensitivity of Panc-1 cells to gemcitabine was increased significantly after pAKT2-shRNA transfection.Conclusion The sensitivity of Panc-1 cells to gemcitabine could be enhanced by pAKT2-shRNA.
9.Regulation of miR-1 and miR-133 a on L-type calcium channel Cavβ2 and α1C subunits in rat cardiomyocyte hypertrophy

Objective To investigate the regulation of miR-1 and miR-133 a on L-type calcium channel β2 subunit ( Cavβ2 ) and α1C subunit during rat cardiomyocyte hypertrophy .Methods Cardiomyocyte hypertrophy was in-duced by isoproterenol (ISO, 10μmol/L).The targets of miR-1 and miR-133a were predicted by online database microCosm and Targetscan , respectively .The 3′untranslated region sequences of Cavβ2 andα1C were respectively cloned into reporter vector and then transiently transfected into HEK 293 cells.The luciferase activities of samples were measured for demonstrating the expression of luciferase reporter vector .The protein expression of Cavβ2 andα1C were evaluated by Western blot .The expression levels of Cavβ2 andα1C were inhibited by RNAi to determine theeffectsofCavβ2andα1Concardiomyocytehypertrophy.Results 1)Cavβ2wasoneofpotentialtargetsof miR-1,α1C was the one of potential targets of miR-133a.2) The luciferase activities of HEK293 cells with the plasmid containing widetype Cavβ2 3′UTR sequence or α1 C significantly decreased ( P <0.05 , P <0.01 ) . 3 ) Upregulation of the miR-1 and miR-133 a by miR-1 mimic and miR-133 a mimic transfection suppressed pro-tein expression of Cavβ2 and α1C, respectively(P<0.01, P<0.05).4)Downregulation of Cavβ2 andα1C by RNAi could markedly inhibit the increase of cell surface area ( P<0.01 ) , mRNA expression of ANP andβ-MHC (P<0.05).Conclusions Cavβ2 is the target gene of miR-1 and α1C is the target gene of miR-133a.miR-1 and miR-133a can negatively regulate the expression of L-type calcium channel Cavβ2 andα1C subunit, inhibi-ting cardiomyocyte hypertrophy.

Objective To identify the charactor of wide QRS complex tachycardia( WCT)throuGh transesophaGeal atrial pacinG( TEAP ). Methods TEAP and intracadiac electrophysioloGical examination infoamation of l2 cases WCT were collected and analyzed from January to February in 20l2 of Wuhan Asia Heart Hospital. Results Comparison of TEAP and intracadiac electrophysioloGical examination showed that l0 in l2 patients were match. Conclusion TEAP is a rapid and convenient method to diaGnose most WCT.
