1.Effect of Modified Sijunzi Decoction on Humoral Immunity of Rats with Exercise-induced Exhaustion

Objective To observe the influence of modified Sijunzi Decoction on the decrease of immune function of exercise-induced exhaustion rats. Methods Thirty SD rats were given adaptive training, and then 6 dead or non-adaptive rats were dropped out. The remaining 24 SD rats were randomly divided into normal group ( N= 8) , model group ( N = 8) , and modified Sijunzi Decoction group ( MSD group, N = 8) . Normal group did not receive any medication and took diet freely. The rats of model group and MSD group were orally given distilled water or modified Sijunzi Decoction 8 g per day respectively after the modeling by exercise-induced exhaustion. After 61 days, the serum was taken from the three groups, and immunoglobulin lgA, lgG, lgM, and complement C3,C4 were detected. Results The contents of immunoglobulins IgA, IgG, IgM and complements C3, C4 in MSD group were higher than those in the model group ( P<0.01) . Conclusion The decrease of immunity occurs in the process of exercise stress, but can be delayed by modified Sijunzi Decoction..
3.Biologic treatments for hereditary diseases.

Hereditary disease, especially monogenic disease is one of the major causes for malformation and disability of children. Most hereditary diseases have no effective therapy besides clinical symptomatic treatment. Biological techniques targeting casual genes or related signaling genes, such as transgenic, RNA interfere, genome editing, have been successfully applied in treating some hereditary diseases. However, most biological, treatments were carried out in animals, further confirmation of the effectiveness and safety of these therapies, and development of more therapeutic approaches based on mechanisms are needed before clinical trials.

4.Additive effects of the variants in the peroxisome proliferator activated receptor γ2 and uncoupling protein 2 genes on type 2 diabetes

Objective To investigate the additive effects of uncoupling protein 2(UCP2) gene 3′-untranslated region(3′-UTR) 45-base pair insertion/deletion( I/D) variation and peroxisome proliferator activated receptor( PPAR)γ2 gene Pro12Ala variation on type 2 diabetes(T2DM) in Chinese Han popula-tion.Methods The UCP2 gene 3′-UTR I/D variation and PPARγ2 gene Pro12Ala variation were exam-ined by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP) in 490 type 2 dia-betes subjects and in 585 control subjects .The additive effects of the two gene mutations were analyzed . Results ⑴The frequency of PPARγ2 gene Pro12Ala variation in type 2 diabetes was not significantly dif-ferent from that in control subjects.(χ2 =0.058, P =0.809).In T2DM group, A12 allele carriers had larger waist circumference than Pro12Pro genotype carriers;Glucose stimulated insulin secretion by oral glu-cose tolerance test (OGTT) was significantly lower in carriers of the Ala12Ala or Pro12Ala genotype com-pared with Pro12Pro genotype.( Z =2.222, P =0.026; Z =2.051, P =0.040; Z =2.079, P =0.038 ) .⑵There wasn't significant difference among 3 genotypes with 3′-UTR I/D variation in UCP2 gene (χ2 =2.311 , P =0.315 ) .In control group , Glucose stimulated insulin secretion by oral glucose tolerance test ( OGTT) was significantly higher in carriers of the I/D or I/I genotype compared with D/D genotype ( Z =1.997 , P =0.046 ) .⑶The genotype carriers with Pro/Ala +Del/Del were the greatest relation to T2DM (OR=1.22, 95%CI 1.078~1.386).Conclusion Though the UCP2 gene mutation alone or the PPARγ2 gene mutation alone is not associated with T 2DM, the possible additive effects of the two micro genes increase the occurring of T 2DM.
5.Application of RBF and RSM in Optimizing the Processing Conditions of Manchurian Dutchmanspipe Stem with Alkali

Objective To optimize the processing conditions of Manchuiran Dutchmanspipe Stem with alkali. Methods The combination of radial basis function ( RBF) and response surface methodology ( RSM) was used to investigate the influence of NaHCO3 , concentration, duration and cycles of processing on the content of aristolochic acid. Results The optimal process was achieved when Manchuiran Dutchmanspipe Stem was soaked for 3 cycles in 0. 05 mol·L-1 NaHCO3 solution, for 24 hours in each cycle. The removal rate of total aristolochic acid approached to 83. 74%. Conclusion The combination of RBF and RSM provided a new method and good guidance for further toxicity attenuation for Manchuiran Dutchmanspipe Stem.

Aim To investigate the effects of isoliquiritigenin(ISL) on cognition dysfunction and brain energy metabolism following repeated cerebral ischemia-reperfusion in mice.Methods Mouse models of learning and memory deficits were made by repeated occlusion of bilateral common carotid arteries and reperfusion. Step-down test and Y-maze task were used to examine the learning and memory abilities of mice. The brain energy state was analyzed by reversed-phase high performance liquid chromatogra phy. Results ISL 10, 20 and 40 mg?kg -1 ig reduced the n umber of errors and extended the latency of memory in step-down test, decreased the number of cumulated stimulation and increased the number of right choices i n Y-maze task. ISL(10, 20, 40 mg?kg -1) increased the levels of ATP, ener gy charge (EC) and total adenine nucleotides(TAN) in a dose-dependent manner. Conclusion The results suggest that ISL improves cognition dysfun ction induced by repeated cerebral ischemia-reperfusion in mice, which may be r elated to the improvement of brain energy metabolism.
10.Dynamic changes of osteopontin in the guinea pig model of cholesterol gallstone disease

Objective To investigate the dynamic changes of osteopontin (OPN) in the guinea pig model of cholesterol gallstone disease.Methods Forty-four guinea pigs were randomly assigned to cholesterol gallstone group and control group.The animals were sequentially killed on Day 7,14,28,42,56 and 70 after operation.The expressions of OPN mRNA in gallbladder and liver tissues were detected by real-time PCR.The changes of OPN,mucin,α1-acid glycoprotein (AAP) and apolipoprotein A1 (APOA1) in bile and blood plasma were detected by ELISA kits.Results The expression of OPN mRNA in gallbladder and liver tissues increased gradually and reached the peak in the 6th week,and then decreased.The increased expression of OPN in bile in the gallstone group started from the 1st week and reached the peak in the 6th week (P < 0.05),which gradually decreased to the baseline in the 10th week (P > 0.05).The expressions of OPN in bile and blood demonstrated similar trends,while the peak time in blood samples was much earlier (4th week).The changes of APOA1 in bile and blood were similar to OPN,although there was no advanced peak value in blood.The levels of mucin and AAP in bile and blood increased after operation,and were kept at high level throughout the study.Conclusions OPN is involved in the whole process of cholesterol gallstone formation,which may be associated with other nucleation-active factors.
