Objective:To study the inhibition of astragaloside on the proliferation of human keloid fibroblasts. Methods: Com-pared with that of normal skin, the expression of transforming growth factor-β( TGF-β) and its transduction factors Smad in the human keloid fibroblasts was detected. The optimal concentration was screened by MTT after HFF-1 human skin fibroblast was infected with astragaloside at different concentrations. The mRNA expression of Smad2, Smad3, Smad4 and Smad7 in the fibroblasts was studied by using real-time. The protein expression of TGF-βRⅡ, Smad2, Smad3, Smad4 and Smad7 in the fibroblasts was detected by using Western blot. Results: Compared with that of normal skin tissue, the expression of Smad protein was significantly increased ( P <0. 05) in the human keloid fibroblasts, and there was no significant difference in the TGF-βRⅡ expression (P>0. 05). The optimal concentration of astragaloside was 0. 5μg·ml-1 . The expression level of Smad2 protein in the two groups was significantly increased, and the level of Smad3 expression was significantly decreased (P<0. 05). Conclusion:Astragaloside can inhibit the formation of fi-broblast possibly through Smad2 over-expression and Smad3 inhibition in the TGF-β/Smad signal transduction pathway.

Journal of Acupuncture and Tuina Science 2016;14(4):290-294
Objective:To observe the therapeutic effects of acupuncture plus cupping therapy and Western medication alone for cervical radiculopathy. Methods: A total of 139 patients with cervical radiculopathy were randomly divided based on the random digital table into an acupuncture-cupping group (70 cases) and a Western medication group (69 cases). In the acupuncture-cupping group, Fengchi (GB 20) (bilateral), Dazhui (GV 14), Jianjing (GB 21) (bilateral) and Jiaji (EX-B 2) points of the neck [bilateral, midpoint between Fengchi (GB 20) and Dazhui (GV 14)] were punctured. After the arrival of the needling sensation, bilateral Fengchi (GB 20) and Jianjing (GB 21) were linked with electric stimulation and the rest acupoints were manipulated with the moderate needling technique. In the retaining of the needles, TDP lamp radiation was used. After acupuncture, the cupping therapy was applied to the local area. The treatment was given once every other day, 20 d for a course. After one course, the therapeutic effects were observed. The Western medication group was treated by oral administration of Meloxicam (Mobic), 7.5 mg, once a day, with water and liquid after meal. The therapeutic effects were observed after 20 d. Results:The total effective rate was 92.8% in the acupuncture-cupping group and 73.9% in the Western medication group, with a statistical difference between the two groups (P<0.05). After treatment, the scores of the clinical symptoms of the two groups were obviously declined (P<0.01), and the score of the clinical symptoms of the acupuncture-cupping group was better than that of the Western medication group, with a statistical difference (P<0.05). Conclusion:Acupuncture plus cupping therapy was more effective than administration of Meloxicam tablets alone in the treatment of cervical radiculopathy.

Chinese Journal of Immunology 2016;32(10):1503-1506
Objective:To research the number and function of monocyte-macrophages in patients with chronic active hepatitis B. Methods:The 51 chronic viral hepatitis B( CHB) patients were selected randomly, which consisted of 20 cases of mild-moderate,31 cases of severe group and 13 cases of healthy controls. PBMCs were separated by percoll. Monocytes were tagged by CD14,the molecules CD80,CD86,HLA-DR and CD163 were detected by flow cytometry which expressed on the surface of PBMCs. Serum cytokine were detected for IL-10, IL-12 and IL-23 by ELISA. The distribution of CD68 was detected in the liver by immunohistochemical staining. Results:The expressions of CD80 for all chronic hepatitis B patients were lower than the controls respectively,no matter mild-moderate or even severe group. Similarly, the HBV patients expressed lower level of CD86 in the peripheral blood mononuclear cells when compared with the control group. Furthermore, there was statistically difference between the levels of CD86 in severe group compared with control group (P<0. 01). As the expression of CD80 and CD86,the levels of HLA-DR in the patents had also declined when compared with controls. While the HLA-DR levels in both the mild-moderate HBV hepatitis groups were statistically significant higher than the severe group (P<0. 01). Different from the above all,the expression of CD163 in all chronic HBV hepatitis was higher than the control group. The CD68 positive cells in chronic HBV patients were observed and infiltrated increasingly in portal area and hepatic lobules (P<0. 05). There were statistically significant differences of IL-10 levels between the mild-moderate group, severe group and the control group, respectively (P<0. 01). Conclusion:Macrophages have participated in the pathological lesions of liver in CHB patients, among peripheral blood mononuclear cells, the phenomena of imbalance between type M1/M2 and polarization to type M2 have been observed, which participated in the development of the chronicity of CHB.
