1.The effects of rosiglitazone on endothelin, nitric oxide and C-reactive protein in patients with impaired glucose tolerance

Objective To observe the changes in endothelin(ET), nitric oxide(NO) and C-reactive protein(CRP) before and after the rosiglitazone therapy in patients with impaired glucose tolerance(IGT). Methods 72 IGT patients were divided randomly into 2 groups: rosiglitazone group(n=45) and placebo group(n=27). 15 weeks later, the efficacy of rosiglitazone was compared. Results The levels of ET, CRP were decreased and NO increased significantly in rosiglitazone group compared with those before treatment (ET 54.8±10.0 pg/ml vs 72.8±24.3 pg/ml, P<0.01;CRP 4.58±0.38 mg/L vs 5.99± 0.85mg/L, P<0.01 ; NO 41.0±10.0 μmol/L. vs 31.3±9.0 μmol/L, P<0. 01)). However, the levels of ET,CRP and NO in placebo group did not change singnificantly (P>0. 05). Conclusions Rosiglitazone can singnificantly improve ET, NO, CRP level in IGT patients, which means an additional effect of rosiglitazone on the correlation of inflammation and endothelial disturbance.
2.Effects of moderate hypoxia on invasion and metastasis of colon carcinoma cell line HT-29 in vitro

Objective To investigate the correlation between moderate hypoxia and maliglant transformation of colon carcinoma cell line HT-29.Methods Hypoxic conditions were produced according to p(O2)of tumor in vivo.The heterotypic adhesiveness of HT-29 cells was detected by MTT assay.Their reaction to the hypoxic environment was determined via invasion across a Matrigel-coated Transwell filter.At 0,6,12,24 and 48 h after hypoxia,MMP-2/9 activities were assessed by gelatin zymography,and osteopontin mRNA and protein levels by RT-PCR and Western blot respectively.NF-?B/p65 levels in nucleus was measured by Western blot after extraction of nucleic protein.Results After 24-hour moderate hypoxia,the heterotypic adhesiveness and invasive capacity of HT-29 cells were significantly enchanced.MMP-2,MMP-9 activities under moderate hypoxia for 6 h showed no significant change as compared with that under normoxia,then gradually up-regulated and reached the peak at 24 h.They showed no significant difference between 24 h and 48 h after hypoxia.The expression trend of osteopontin mRNA,protein and NF-?B/p65 protein in nucleus was similar to that of MMP-2,MMP-9 under moderate hypoxia.Conclusion Moderate hypoxia could induce maliglant transformation in HT-29 cells through enhancing heterotypic adhesiveness and invasive capacity and upregulating MMP-2,MMP-9 activities.OPN-NF-?B may be an important regulatory pathway besides HIF-1 under hypoxic condition,which involves maliglant phenotype induced by moderate hypoxia.
3.Research on correlated risk factors of bile duct injury induced by laparoscopic cholecystectomy

Objective To explore the correlated risk factors of bile duct injury induced by laparoscopic cholecystectomy ,and to reduce the rate of bile duct injury companied with LC and improved patients condition after LC .Methods One thousand two hun‐dred and forty four patients who had been done the laparoscopic cholecystectomy in our hospital were searched from December 2003 to December 2013 for their case notes .The relationship between rate of bile duct injury induced by laparoscopic cholecystectomy and gender ,age ,time of LC surgery ,pathology ,gallbladder wall thickness ,liver function levels ,the existence of the gallbladder triangle anatomical anomalies and physician experience were analyzed by logistic regression method .Results Logistic regression analysis showed that patients did LC surgery at early time ,with acute cholecystitis ,with gallstones combined effusion ,with gallbladder wall thicker than 4 mm and with gallbladder triangle anatomical anomalies were likely to have bile duct injury induced by LC (P<0 .05) .And doctor′s experience on LC is another important factor as well (P<0 .05) .Conclusion According to the correlated risk factors of bile duct injury caused by laparoscopic cholecystectomy ,methods should be employed to reduce the bile duct injury .And sharing experience between doctors and learning new techniques could be helpful ,too .

7.Effect of Clinic Treatment by Tropicamide Esculin-digitalisglycosides Combination Eye Drops on Low Myopia of Children

Objective To investigate the effects of tropicamide and esculin-digitalisglycosides combination eye drops on low myopia of children (age ranging from 6 to 14). Methods Eighty children with 160 eyes of low myopia in the outpatient department of ophthalmology of our hospital were chose from July 2010 to April 2013, then the patients were equally divided into four groups:the control, esculin-digitalisglycosides eye drops,tropicamide,and tropicamide and esculin-digitalisglycosides combination groups. The naked vision and myopia correction of each group were compared before and after treatment for two weeks, and the treatment effects were compared among four groups. Results (1) The results showed there were no significant changes before and after treatment in the control, esculin-digitalisglycosides eye drops and tropicamide groups ( >0.05); (2) On the contrast, the tropicamide and esculin-digitalisglycosides combination have significantly improved naked vision and myopia correction after 2 weeks ( <0.05);(3) Prior to the treatment,there was no significant difference among four groups (<0.05);(4) After treatment,group under tropicamide and esculin- digitalisglycosides combined was statistical different from the other groups treated with other three eye drops ( <0.05);However, there was no statistical difference among groups in these three eyedrops. Conclusion Esculin-digitalisglycosides combined with tropicamide was the most effective treatment for low myopia of children.
8.Reference Gene Selection for Normalization of PCR Analysis in Chicken Embryo Fibroblast Infected with H6N1 AIV

Chicken embryo fibroblasts(CEFs)are among the most commonly used cells for the study of interactions between chicken hosts and H5N1 avian influenza virus(AIV).In this study,the expression of eleven housekeeping genes typically used for the normalization of quantitative real-time PCR(QPCR)analysis in mammals were compared in CEFs infected with H5N1 AIV to determine the most reliable reference genes in this system.CEFs cultured from 10-day-old SPF chicken embryos were infected with 100 TCID50 of H5N1 AIV and harvested at 3,12,24 and 30 hours post-infection.The expression levels of the eleven reference genes in infected and uninfected CEFs were determined by real-time PCR.Based on expression stability and expression levels,our data suggest that the ribosomal protein L4(RPL4)and tyrosine 3-monooxygenase tryptophan 5-monooxygenase activation protein zeta polypeptide(YWHAZ)are the best reference genes to use in the study of host cell response to H5N1 AIV infection.However,for the study of replication levels of H5N1 AIV in CEFs,the β-actin gene(ACTB)and the ribosomal protein L4(RPL4)gene are the best references.

Based on extensive research of anatomy and physiological function of the Eustachian tube,the authors review the literatures to determine the mechanism of patulous Eustachian tube (PET).The purpose of this review is to summarize the current knowledge on the patulous Eustachian tube,present the current diagnostic protocols for PET and determine the comparative efficacy of variable treatments for this condition.In addition,development prospects are summarized in this article.
10.Effect of Silver Needle Thermo-conduction on Cervical Spondylosis of Vertebral Artery Type

Objective To explore the effect of silver needle thermo-conduction on cervical spondylosis of vertebral artery type (CSA). Methods 82 patients with CSA were randomly divided into control group (n=41) and treatment group (n=41). The control group was treated with cervical traction combined with microwave diathermy, and the treatment group was treated with silver needle thermo-conduction combined with cervical traction. Both groups were evaluated with Evaluation Scale for Cervical Vertigo (ESCV) and transcranial Doppler before and 1 course after treatment. Results The ESCV score, the peak blood flow velocity of period of contraction of left vertebral artery, right vertebral artery and basilar artery in both groups were significantly higher after treatment than before (P<0.01), the treatment group was better than the control group after treatment (P<0.01). Conclusion Silver needle thermo-conduction combined with cervical traction therapy can accelerate the blood flow of patients with CSA and improve their clinical symptoms
