1.Pentapeptides prevent enterovirus 71 proliferation in rhabdomyosarcoma cells and mice.

Enterovirus 71 (EV71) is the main causative agent of hand, foot, and mouth disease (HFMD). This article presented the inhibitory activity of pentapeptides on the EV71 infection in rhabdomyosarcoma (RD) and suckling mice. The EV71 VP1 capsid protein expression levels and mRNA levels were analyzed by Western blotting and real-time PCR. The antiviral activity of pentapeptides in vivo was evaluated by weight changes and EV71 VP1 protein expression levels in intestines of suckling mice. Results revealed that the pentapeptide P010157 was able to inhibit EV71 replication in RD cells. After being incubated with the P010157 at a concentration of 100 microg x mL(-1) for 48 h, the level of EV71 vp1 mRNA in RD cells decreased by (92.0 +/- 6.3)%. The estimated EC50 was 2.2 microg x mL(-1). P010157 was able to inhibit EV 71-induced cytopathic effect (CPE) in RD cells. The cytotoxic activity of the compound was evaluated against RD cells by MTS assay. The results showed that P010157 had no obvious toxicity. In addition, the treated mice with P010157 did not exhibit weight loss, as was observed in untreated mice. EV71 replication reduced significantly as revealed by Western blotting. These findings suggest that P010157 could prevent EV71 proliferation in vitro and in vivo. P010157 is a novel compound for antiviral therapies against EV71, which merited further investigation.

2.MiR373 and miR542-5p regulate the replication of enterovirus 71 in rhabdomyosarcoma cells.

MicroRNAs (miRNAs) play an important role in infection and replication of virus in host cells. To identify cellular miRNAs involved in the host response to enterovirus 71 (EV71) infection, we examined miRNAs effects on the replication of EV71 in rhabdomyosarcoma cells. We constructed target gene of miRNAs screening system. 3'untranslated region (UTR) dual luciferase reporter analysis was used to identify putative miRNA targets in the EV71 virus genome. First, 13 segments of EV71 virus genomes were inserted to the pMIR vector and the luciferase expression were assayed to identify the target gene of putative miRNA. The reporter gene expression of the cells transfected with the vector containing 5'-UTR was significantly downregulated. Then we screened the miRNAs that may target to 5'-UTR using online analysis programs. Furthermore, Western blotting and real-time PCR test were performed to investigate the effect of miRNAs on viral replication. The study showed that miR373 and miR542-5p could suppress the replication of EV71 virus through binding to the 5'-UTR gene. Cellular miRNAs could regulate the replication of EV71 virus in host cells, and our paper should report the role of miR373 and miR542-5p in this regulation for the first time. Our findings supported the notion that the cellular miRNAs might be essential in the host-pathogen interactions.

Sphingolipids are essential components of cellular membrane , regulating important physiologic function of cells .Al-terations of sphingolipid metabolism are closely related to the pathogenesis of several chronic diseases .Ceramide biogenesis , the center of sphingolipid metabolism , includes de novo ceramide synthesis , ceramide formation via breakdown of complicated sphingolipids and so on . 6.Analysis of postoperative efficacy and the influencing factors of the recurrence of epilepsy in Glioma-related Epilepsy

Objective To explore the postoperative recovery of the patients with glioma-related epilepsy and the possible risk factors for recurrence of epilepsy.Methods To collect clinical data of 89 patients with glioma-related epilepsy,we recorded Engle grade score of all patients underwent resection of tumor and seizure focus in a week,1,3 and 6 month after surgery Repeated measures ANOVA was used to analysis the difference within group.Kaplan-Meier method and Cox regression analysis were used to analysis seizure recurrence risk curve and the epileptic recurrence related factors,respectively.Results The postoperative recurrence rate was 26.97% (24/89).Engle grade scores were 2.966±0.081.2.202±0.080,1.730±0.093 and 1.313±0.042 in a week,1,3 and 6 month after surgery.The difference was statistically significant (Ftime=96.076,P=0.000).Cox regression model showed that tumor accumulation site (RR =2.908,CI:1.083 ~7.806),postoperative edema formation (RR =4.769,CI:1.737 ~ 13.096),tumor relapse (RR=8.309,CI:3.379~20.432)were the important risk factors for recurrence epileptic seizures (all P< 0.05).Conclusions Surgical treatment can significantly reduce or even eliminate seizures postoperative and its long term efficacy is superior to short term efficacy.Appropriate measures should be taken to treat risk factors associated with recurrence epileptiform seizure to improve the prognosis of patients quality of life.
7.Ca_V 2.3 voltage-gated calcium channel and epilepsy

The CaV 2.3 encoded Ca2+ channel is one of the least well-known voltage-gated calcium channels in terms of physiology, pharmacology and clinical relevance. Epilepsy is a family of neurological disorders that are common and harmful to human health. More and more studies such as gene knock-out experiment have demonstrated that the channel is related to epileptic seizure, including participating to plateau potential generation, regulating intracellular Ca2+ concentration, CaV 2.3 splice variant, interacting with some muteins. In addition, some antiepileptic drugs inhibit the epileptiform discharges by targeting CaV 2.3 voltage-gated calcium channel.
8.Influencing factors of seizures in glioma patients and its relationship with recurrent epilepsy

Objective To analyze the influencing factors of epilepsy in patients with glioma,and to explore the relationship between the epileptic factors and the recurrence of epilepsy after surgery.Methods From January 2012 to December 2015,clinical data of 95 glioma patients in the Fifth Affiliated Hospital of Zhengzhou University were retrospectively analyzed,which were divided into two groups,including 54 cases with epilepsy and 41 cases without epilepsy,all patients were underwent tumor resection,to search and resect epilepsy lesions in patients with epilepsy,Follow-up using outpatient examination and telephone interview was performed to record seizure recurrence after operation,sort and analyze the clinical data of patients,to investigate the influencing factors of epileptic seizures,and the relationship between the factors associated with epileptic seizures and postoperative recurrence of epilepsy.The epileptogenic factors were analyzed using x2 test and multifactor Logistic regression model,epileptogenic factors and recurrent epilepsy after operation were analyzed usingx2 test.Results Logistic regression model showed that peritumoral edema > 2 cm(OR =6.905,95% CI:2.787-17.106),low tumor pathological grade (0R =5.032,95% CI:1.696-14.931) were independent risk factors for epileptic seizures;Follow-up situations:16 cases (13 cases of epilepsy group,3 cases of no epilepsy group) showed epileptic seizure in different time after the operation,preoperative epilepsy history,peritumoral edema were related to the recurrence of epileptic seizures in epilepsy group and the patients without epilepsy (x2 =2.940,3.049,all P < 0.05),the pathological grade was related to the recurrence of epileptic seizures in epilepsy group (x2=4.774,P < 0.05),however,it was not associated with the patients without epilepsy (x2 =0.060,P > 0.05),and there was a correlation between the preoperative history of epilepsy and the postoperative recurrent seizures (x2 =4.672,P < 0.05).Conclusions Peritumoral edema > 2 cm and low tumor pathological grade are the factors that lead to epileptic seizures and postoperation epilepsy recurrence in glioma patients.It's expected to take some positive treatment to reduce epileptic seizures,we should pay special attention to glioma patients with epilepsy.
9.Surgical treatment of congenital muscular torticollis: comparison of two operative methods

Objective To evaluate the therapeutic efficacy of congenital muscular torticollis (CMT) by two different methods. Methods A total of 86 patients with congenital muscular torticollis in recent 7 years were involved in this study, including 69 cases under 12 years old and 17 cases over 12. Two different operation methods, incision of inferior extremity of sternocleidomastoid muscle and incision of amphi-sternocleidomastoid muscle with post-operative traction, were alternatively chosen according to the ages and deformed degrees of the patients. Results 72 patients were followed-up for 1 to 7 years (averaged 4.5 years). Based on the effect of function and cosmetology in the group of 57 cases under 12 years old, there were 52 cases (91.2 %) excellent, 5 cases (8.8 %) good and no case with side-effects. Meanwhile, in the group of 15 cases over 12 years old, there were 11 cases (73.3 %) excellent, 4 cases (26.7 %) good and no case with side-effects. The effect was satisfactory. Conclusion Different patients with congenital muscular torticollis should be treated with different therapeutic methods.
10.Oxidative stress damage to mitochondria mediated the pathological processes in epilepsy

Recent studies show that epileptic seizures result in mass free radical production and induce oxidative damage because of relative deficiency in anti-oxygen system.Mitochondrium is crucial to sustaining energy metabolism,regulating cell death,synthesising neurotransmitter and oxidating fatty acid.Mitochondrium is not only the place for free radical production,but also the target of oxidative damage.Mitochondrial oxidative stress and resultant dysfunction enhance epileptic susceptibility.Seizure-induced free radical can influence energy metabolism,destroy DNA construction and induce apoptosis in neurontoxic consequences in epilepsy.
