China Pharmacist 2016;19(7):1267-1272
Objective: To investigate the effects of neferine ( Nef ) on the learning and memory function and the expression of Notch1 and SYN in hippocampus in the rats with chronic cerebral ischemia .Methods:Male SD rats (250-300g) were randomly divid-ed into the sham operation group(Sham), the model group(Mod), nimodipine (Nim) positive control group and Nef treatment group with 5 ones in each .The chronic cerebral ischemia ( CCI ) model was established using bilateral common carotid artery ligation ( 2-VO).The rats were orally administered with NS(5 mg· kg-1),Nim(1 mg· kg -1) and Nef (20 mg· kg-1),respectively for 21 days after the first day of operation .The body weight was recorded .The number of hippocampal neurons in the rats was observed by Nissel staining. The learning and memory function was evaluated by Morris water maze test .Notch1 and SYN protein expressions in hippo-campus were detected by IHC staining and Western blot .Results:Compared with the Mod group , Nef group could reverse the reduc-tion of body weight and the number of hippocampal neurons in hippocampus CA 1 region induced by 2-VO, increase the route and the time of platform finding, prolong the escape latency and decrease the number of platform cross on the 21st day, and the effects were bet-ter than those of Nim (P<0.05).Moreover, the down-regulated SYN and Notch1 protein expressions in CCI group were both increase after the Nef treatment on the 21st day. Conclusion:Nef has a protective effect on the function of learning and memory in CCI rats in-duced by 2-VO,which may be related with the strengthened Notch 1 pathway in CA1 region of hippocampus and synaptic plasticity .
5.Discussion on Chinese medicine in treating post-infectious irritable bowel syndrome
China Journal of Traditional Chinese Medicine and Pharmacy 2005;0(04):-
The pathogenesis of post- infectious irritable bowel syndrome(PI-IBS)involves abnormal intestinal immune barrier, which possibly includes the pathological aspects: Intestinal and systemic changes in T lymphocyte; the change of inflammatory cytokines; a increase in intestinal mast cells and enterochromaffin cells and so on. Treatment by western medicine based largely on symptomatic treatment. TCM thinks that the fundamental organ of the irritable bowel syndrome is the liver. Emotional factors inducing liver dysfunction lead to the symptoms of irritable bowel syndrome. The pathogenesis of IBS is, the disorder of movement of qi in liver and spleen. Stagnation of liver-QI with deficiency of the spleen or disharmony between liver and spleen is the key pathogenesis of post-infectious irritable bowel syndrome. On this basis, in various stages of the disease, there may be a different pathogenesis. As a special type of IBS, the balance of vital qi and evil factors determine the disease progression of PI-IBS, in the treatment course also should not forget dispelling pathogenic factors while strengthening body resistance, tonifying deficiency and removing dampness。
6.Blood gas analysis features of H7N9 influenza patients
Chinese Journal of Infection and Chemotherapy 2014;(3):216-218
Objective To review the blood gas analysis of H7N9 flu patients for better understanding the characteristics and prognosis of the emerging disease. Methods A retrospective review was conducted for the 15 H7N9 influenza patients, whose blood gas profile was tested with a Radiometer 80 Blood Gas Analyzerin our hospital. The profile of blood gas analysis was compared between the 12 surviving patients and the 3 deaths in terms of their outcomes. Results The 3 patients who died of H7N9 influenza averaged 83 years old.The partial pressure of oxygen was <8 kPa in 9 of the 12 surviving patients, which meet the diagnostic criteria of type I respiratory failure.Their partial pressure of oxygen all recovered to normal (11 .033 ± 2.335 kPa,reference range 10-14 kPa)on discharge. The pH value of peripheral arterial bloold averaged 7.335 (range 7.39-7.54)on adission for the 3 patients who died of H7N9 influenza. The worst average pH value was 7.233 (range 7.32-7.35), which was significantly different from the values of the surviving patients. Conclusions Elderly patients are susceptible to H7N9 influenza infection and subsequent respiratory failure.Old age was associated with higher mortality. The partial pressure of ox-ygen is not significantly associated with patient outcomes.Appropriate active treatment may recover the blood gas profile to normal regardless of the initial values. If decreasing peripheral arterial bloold pH value is highly suggestive of poor prognosis, comprehensive measures should be taken to keep acid-base balance, including assisted ventilation to correct hypoxemia.
7.The value of contrast-enhanced ultrasound guiding normal saline injection in the detection of residual common bile duct stones
Chinese Journal of Ultrasonography 2017;26(6):517-521
Objective To investigate the diagnostic value of contrast-enhanced ultrasound (CEUS) guiding normal saline (NS) injection through endoscopic nasobiliary drainage duct(ENBD) on evaluation for residual stones in common bile duct. Methods Fifty-five patients with bile duct stones were treated by endoscopic retrograde cholangio-pancreatography (ERCP) and duodenoscopic sphincterotomy incision surgery (EST) and ENBD.All patients received normal ultrasonography and CEUS guiding NS injection ultrasonography after EST.The length and width of common bile duct and the detection rate of residual stones before and after NS injection were compared. Results In the 55 patients,1 patient failed in injection of contrast agent into the ENBD.In the other 54 patients, the difference of the length and width of common blie duct before and after NS injection were statistically significant [(2.94±1.76)cm vs (6.09±1.46)cm,(0.58±0.30)cm vs (1.11±0.98)cm](all P<0.001).The full display rate of the common bile duct before and after NS injection were 13.0%(7/54) and 90.7%(49/54),respectively.Before injection, none of common bile duct stones was suspected. After injection,5 cases of common bile duct stones were suspected.Three cases were confirmed by ERCP,1 case was confirmed by operation and 1 case was false positive. Conclusions CEUS of the common bile duct through ENBD performs its patency and course. On this basis the injection of NS increases the display rates of common bile duct, thus improving the detection rate of residual stones.

Chongqing Medicine 2015;(16):2174-2176
Objective To investigate the correlation between moderate hypoxia and tumor invasiveness in colon carcinoma cell line HT‐29 and detect the expression of NF‐κB protein in nucleus and activities of MMP‐2/9 ,so as to explore its latent mecha‐nism of malignant transformation .Methods The moderate hypoxia model was established according to the pressure of O2 of tumor in vivo .After different period in moderate hypoxia ,activities of MMP‐2/9 secreted by HT‐29 cells were measured by gelatin zymog‐raphy ,NF‐κB levels in nucleus were assessed by Western blot after extraction of nucleic protein .Results MMP‐2/9 activities under moderate hypoxia gradually up‐regulated in 12 h and reached the peak at 24 h .The expression of NF‐κB protein in nucleus was simi‐lar to that of MMP‐2/9 .Conclusion Moderate hypoxia could induce malignant phenotype in HT‐29 cells through up‐regulating MMP‐2/9 activities ,in which NF‐κB may involve in this progression .
9.Screening protein interacted with enterovirus 71 3A protein by means of T7-phage display system
International Journal of Laboratory Medicine 2014;(16):2129-2131
Objective To screen and identify protein interacted with enterovirus 71 (EV71)3A protein by means of T7-phage display system. Methods The prokaryotic expression vector of 3A protein was constructed for expressing and purifying 3A pro-tein.With 3A protein as the target protein, the T7-phage display system was adopted to screen the human hepatocyte cDNA library. The screened products were performed the DNA sequence analysis and the homologous research. Results 3A protein was expressed and purified, after 4 rounds of biopanning,37 positive clones were selected. The inserted fragments were amplified by PCR using specific T7 primer. The products were sequenced,2 proteins were identified to interact with 3A protein by homologous analysis. Conclusion Trough the T7 phage display system, two proteins interacted with 3A are obtained. The possible function of 3A protein can be speculated by researching the function of these two proteins, which lays the foundation for the further studying the pathogen-ic mechanism of EV71.
10.The effects of rosiglitazone on endothelin, nitric oxide and C-reactive protein in patients with impaired glucose tolerance
Chinese Journal of Diabetes 2010;18(1):51-53
Objective To observe the changes in endothelin(ET), nitric oxide(NO) and C-reactive protein(CRP) before and after the rosiglitazone therapy in patients with impaired glucose tolerance(IGT). Methods 72 IGT patients were divided randomly into 2 groups: rosiglitazone group(n=45) and placebo group(n=27). 15 weeks later, the efficacy of rosiglitazone was compared. Results The levels of ET, CRP were decreased and NO increased significantly in rosiglitazone group compared with those before treatment (ET 54.8±10.0 pg/ml vs 72.8±24.3 pg/ml, P<0.01;CRP 4.58±0.38 mg/L vs 5.99± 0.85mg/L, P<0.01 ; NO 41.0±10.0 μmol/L. vs 31.3±9.0 μmol/L, P<0. 01)). However, the levels of ET,CRP and NO in placebo group did not change singnificantly (P>0. 05). Conclusions Rosiglitazone can singnificantly improve ET, NO, CRP level in IGT patients, which means an additional effect of rosiglitazone on the correlation of inflammation and endothelial disturbance.
