2.Clinical Observation of Tiao Ren Tong Du Needling for Post-stroke Insomnia

Objective To observe the clinical efficacy of Tiao Ren Tong Du (regulating the Conception Vessel and unblocking the Governor Vessel) needling method in treating post-stroke insomnia and the change of Pittsburgh Sleep Quality Index (PSQI) after treatment, and to discuss the effect of this needling method on post-stroke insomnia. Method Ninety post-stroke patients were randomized into a treatment group, a control group and a Western medicine group. For the treatment group, points from the Conception and Governor Vessels were selected as the major points; for the control group, conventional needling method was adopted; for the Western medicine group, Zopiclone was prescribed. After intervention, the clinical efficacies and PSQI scores were investigated and compared. Result After intervention, the effective rates of the three groups were equivalent (P>0.05), the recovery rate of the treatment group was significantly higher than that of the control groups (P<0.05,P<0.01), and the PSQI scores were significantly changed in all three groups (P<0.05); the one-month and 3-month follow-ups showed insignificant changes of PSQI score in the treatment group compared to that right after the intervention (P>0.05); in the Western medicine group, the one-month follow-up showed significant difference in PSQI score compared to that right after the intervention (P<0.05); in the control group, there was a significant difference in PSQI score between the one-month follow-up and 3-month follow-up results (P<0.05). Conclusion The clinical efficacies of acupuncture methods and oral administration of Zopiclone are equivalent in treating post-stroke insomnia, but Tiao Ren Tong Du needling method can produce a higher recovery rate and better long-term efficacy.
3.Transcatheter therapy for congenital heart diseases using the Amplatzer occluder: A report of 30 cases

Objective To evaluate the short-term effectiveness of Amplatzer device closure for congenital heart diseases.Methods Under X-ray fluoroscopy or/and transthoracic echocardiography(TTE),percutaneous puncture of the femoral artery or vein was conducted and the Amplatzer occluder was passed through the catheter.Effectiveness evaluation by transthoracic echocardiography,electrocardiography(ECG),and X-ray examination was applied at 24 hours,1 month,6 months,and every 1 year after the procedure.Results In 1 case of perimembranous ventricular septal defect(VSD),the detachment of the Amplatzer occluder to the arch of aorta was due to an inadequate small size.This patient was immediately operated with successful removal of the device through the femoral artery and later underwent a re-deployment of a larger-sized Amplatzer occluder closure successfully.The remaining 29 patients with atrial septal defect(ASD),patent ductus arteriosus(PDA),or perimembranous ventricular septal defect underwent a successful deployment of the Amplatzer occluder on one session without complications.The operation time was 20~90 min (38?16 min),the X-ray exposure time was 5~45 min(18?10 min),and the length of hospital stay was 3~7 days(4?2 days).Conclusions Amplatzer occluder transcatheter closure of congenital heart diseases has advantages of simplicity of deployment,good safety,and high success rate.This procedure is suitable for patients with secundum ASD,membranous VSD,and PDA.

6.Psychological stress among nurses in Department of Emergency of Beijing Jishuitan Hospital

Objective:
To investigate the psychological stress among nurses in the Department of Emergency, Beijing Jishuitan Hospital, so as to provide the evidence for formulating targeted psychological health interventions.
Methods:
All nurses from Department of Emergency, Beijing Jishuitan Hospital were enrolled, and participants' demographics, professional title, working duration, family support and participation in public health emergency responses were collected using questionnaire surveys. The development of psychological stress during the latest one week was evaluated using the Symptom Checklist-90 (SCL-90) scale, and factors affecting psychological stress were identified.
Results:
A total of 121 valid questionnaires were recovered, and the participants included 6 men (4.96%) and 115 women (95.04%) and had a mean age of (41.49±4.61) years; and mean service length of (12.14±2.61) years. There were 83 participants that were married (68.60%), 71 with a bachelor degree (58.68%), 60 with a medium-grade professional title (49.59%). The mean score of psychological stress symptoms was 213.43±18.58, which was significantly higher than national norm (124.57±14.31; t=12.194, P<0.001). The detection of psychological stress was 31.40% among the participants, and the presence of psychological stress correlated with age of 40 years and older (χ2=4.826, P=0.028), no family support (χ2=40.420, P<0.001), and no participation in public health emergency responses (χ2=33.767, P<0.001).
Conclusions
A high degree of psychological stress is detected among nurses in Department of Emergency of Beijing Jishuitan Hospital, and age, family support and emergency response capability are factors affecting psychological stress.

8.Construction and identification of a recombinant lentivirus harboring expressing Mus musculus STAT3 gene

Objective To construct recombinant lentivirus with the gene STAT3 of the Mus musculus,measure the expres-sion of STAT3,and conduct lentivirus packing and identification.Methods The mRNA of mouse myoblast was extracted and transformed into STAT3 cDNA by the special primer.then,STAT3 cDNA was amplified and reclaimed and inseted into pLVX-IRES-ZsGreen1 vector.Cleavage map and sequencing analysis were used for identification of the recombinant lentivirus vector (pLVX-IRES-ZsGreen1-STAT3).293 T cells were transfected with main vector pLVX-IRES-ZsGreen1-STAT3.and 48 h later, Western blott detected the expression of STAT3 protein.Lentiviral vectors were packaged and the titer was determined.Results The lentiviral vector plasmid pLVX-IRES-ZsGreen1-STAT3 was identified correctly by cleavage map and Co-transfection of 293 T cells with 48 h,the expression of STAT3 was significantly enhanced by western blot.And DNA sequencing analysis confirmed that STAT3 gene sequencing was exactly the same with that reported by genbank.Conclusion Lentiviral vector carrying STAT3 was successfnlly constructed and could express STAT3 with high efficiency,and can be used in further study.

BACKGROUND: Increased expression of survivin in various tumor tissues can regulate cell proliferation, division, and plays an important role in protecting cells from apoptosis.OBJECTIVE: To construct the specific micro RNA (miRNA) expression vector that can block the C_(57)BL mice survivin gene by RNA interference (RNAi) technique.DESIGN, TIME AND SETTING: The single sample observation was performed at the experimental center of Department of Neurology, The First Afliliated Hospital of Chongqing Medical University from June to November 2008.MATERIALS: Ring-shaped pcDNA~(TM)6.2-GW/EmGFPmiR and BLOCK-iT~(TM) Pol II miR RNA interfered Expression Vector Kit with EmGFP was produced by Invitrogen Company. DH5a E. coli was preserved at the laboratory. Xho I and BamH I enzyme, spectinomycin were provided by Shanghai Sangon Biological Engineering Technology & Services Co., Ltd.METHODS: According to sequence of mRNA of C_(57)BL mice survivin provided by Genebank, four pairs of specific oligonucleotide sequences were designed and synthesized by using the software. The annealed oligonucleotide fragment was sub-cloned into pcDNA~(TM)6.2-GW/EmGFPmiR expression vector by gene clone technique and transformed into DH5a E. coli, subsequently, a single colony was incubated into liquid medium containing spectionmycin. Finally the plasmid was extracted.MAIN OUTCOME MEASURES: The recombinant vector was identified by sequencing and agarose gel electrophoresis.RESULTS: The sequencing revealed that insertion element was correctly cloned into the vector without nucleotide mutation, absence or insertion abnormality. The result of double enzyme digestion demonstrated that the fragment length was coincidence with expectation.CONCLUSION: The C_(57)BL mice survivin miRNA expression vector is successfully constructed.
10.Culture of tissue-engineered human normal esophageal epithelial cells

AIM: In vitro isolation and culture of esophageal epithelial cells are basic component of tissue-engineered esophagus. This study explored the method to culture esophageal epithelial cells for research on tissue-engineered esophagus. METHODS: The experiment was performed at experimental center of Lanzhou University from May to November 2007. Normal esophagus tissues, 2.0-3.0 cm, were harvested from patient with esophagus cancer by surgery. The informed consent was obtained from the patient. Esophageal epithelial cells for tissue engineering were obtained and passaged. The cells cultured by DMEM+F12 (1∶1) after 20 minutes, 1-4 days were observed by immunohistochemistry staining and inverted phase contrast microscope. The growth curve of cells was drawn by MTT method. RESULTS: The immunohistochemistry staining results showed that 90% of the cells were positive, which indicated the cultured cells were esophageal epithelial cells. Normal cells were big and globular, floating in the culture-medium. Cells began to adhere after 20 minutes, and most cells were polygon-like or irregular globular and adherent after 1 day; the cells began to cluster after 2 days; the cells grew at peak after about 3-4 days with abundant endochylema and large and spherical nuclear. Cells growth reached the peak after about 3 days of culture and its absorbance was significantly different compared with that on the 1st, 2nd, 5th, and 6th days (P
