1.The Role of IκBα in TNF-α-induced Apoptosis in Hepatic Stellate Cell Line HSC-T6

To investigate the role of NF-κB in TNF-α induced apoptosis in HSC-T6, a mutant IκBα was transfected into HSC-T6 cells by lipofectin transfection technique and its transient effect was examined 48 h after the transfection. The activation of NF-κB was detected by immune fluorescence cytochemistry and Western blotting with anti-p65 antibody. The apoptosis and the rate of inhibition by TNF-α in both transfected and untransfected HSC-T6 cells were measured respectively by FAC-Scan side scatter analysis and MTT methods. Our results showed that TNF-α could activate NF-κB in untransfected cells but not in transfected HSC-T6 cells. The percentage of apoptosis in transfected cells were significantly higher than that in the untransfected ones (P<0.01) and it was also true of the inhibition rate (P<0.01). It is concluded that the resistance of HSC-T6 towards apoptosis induced by TNF-α can be mediated by NF-κB activation. The inhibition of NF-κB activation by mutant Iκ Bα can attenuate the resistance of HSC-T6 cells and increase its sensitivity to TNF-α.
2.Research of adults frontal bones thickness and analysis its mennning

Objective To research the thickness of adults frontal bones in many parts and analyze the mean-ing. Methods We chose 130 dry skulls of the Chinese people including 70 male skulls and 60 female skulls and ver-tically devided the frontal bones according the devidement of the superciliary arch into 4 parts on average. Then we measured the thickness of the frontal squama,the frontal tuber,the forehead and temple,the superciliary arch,the or-bital surface and the frontal sinus using the ruler, vernier caliper and protractor. Results We got the thickness of the frontal squama,the frontal tuber,the forehead and temple,the superciliary arch,the orbital surface and the frontal si-nus and calculated the average datas. What's more,besides the thickness of the frontal tuber,forehead and temple,the superciliary arch had statistical significance(P < 0.05) between males and females, other parts of the frontal bones had no statistical significance(P>0. 05). Conclusion We measured the thickness of the frontal squama,the frontal tu-ber,the forehead and temple,the superciliary arch,the orbital surface and the frontal sinus and the datas will had good guiding roles to our department's work.
3.The value of rapid species identification of Mycobacterium tuberculosis by two kinds of multi-locus ;polymerase chain reaction in children

Objective To evaluate the clinical value of multi-locus polymerase chain reaction (PCR) for identifying Mycobacterium tuberculosis complex isolated in children. Methods The isolates were collected and were first determined by PNB/TCH medium. 7-point PCR sites including 16SrRNA, Rv0577, IS1561, Rv1510, Rv1970, Rv3877/8 and Rv3120, and 4-point PCR sites including ropB, RD1, RD8 (present), RD8 (deleted) were used to amplify them by PCR. Results Total of 204 isolates were collected, in which 199 were Mycobacterium tuberculosis, 3 were Mycobacterium bovis, and 2 were non-tuberculous mycobacteria by the PNB/TCH method. 4-point PCR analysis showed that 196 were Mycobacterium tuberculosis, 2 were Mycobacterium bovis, 3 were BCG species and 3 were non-tuberculous mycobacteria. 7-point PCR analysis showed that 191 were Mycobacterium tuberculosis, 2 were Mycobacterium bovis, 3 were BCG species, 4 were African Mycobacterium type I, 1 was Mycobacterium caprae, 1 was Mycobacterium microti and 2 were non-tuberculous mycobacteria. Conclusion Compared with the conventional method, the PCR identification in 4-point PCR method and 7-point PCR method could rapidly identify the BCG among the complex group in children tuberculosis. 7-point PCR method was able to identify all the subspecies of Mycobacterium, except Africa Mycobacterium. 4-point PCR method would be more rapid and easier in the identification of BCG strains.
4.Characteristics of depression in patients with Parkinson' s disease in Nanjing city

Objective To determine the frequency of depression in patients with Parkinson' s disease(PD) and healthy controls over 50 years of age and to analyze the characteristics and influencing factors of PD with depression(PDD) in Nanjing.Methods One hundred and twenty-six PD patients were diagnosed and assessed using Self-rating Depression Scale (SDS) and Hamilton Depression Scale (HAMD).The frequency,characteristics and influencing factors of depression were statistically analyzed,and the factor analysis of HAMD was carried out.Also,one hundred and twenty-four healthy subjects over the age of 50 were selected as the control group.Ressults The incidence of depression in PD group was 48.4% ( 61/126):15.1% (19/126) for mild depression,27.8% (35/126) for moderate depression,5.6% (7/126) for severe depression.The incidence of depression in the control group was 9.7% (12/124):5.7% (7/124) for mild depression,2.4% (3/124) for moderate depression,1.6% (2/124) for severe depression.There was a significant difference between these two groups( x2 =45.36,P < 0.01 ).Univariate and Logistic regression analysis revealed that a high frequency of depression occurred in patients with long PD duration,high H-Y stage and UPDRS Ⅲ.According to each factor analysis of HAMD,the scores of cognitive impairment,tardiness,anxiety and sleep disturbances of the PD patients with depressive syndromes were higher than that of the control group.Conclusion Depression is a relatively common complication of PD in Nanjing which is associated with long PD duration,severity of motor disturbances and increasing H-Y stage.
5.Current aspects in red blood cell substitutes.

Red blood cell substitutes are a group of oxygen carriers designed to temporarily replace transfused blood. Current developing products include perfluorocarbon-based and hemoglobin-based oxygen carrier. Each product is unique in its limitations and advantages. A number of products are in advanced clinical trials and nearing market. When they are available for use it is likely that development will accelerate and even better products will substantially alleviate the world-wide shortage of blood for transfusion.

6.Expression of ERK5 in multidrug-resistant hepatocellular carcinoma cell line.

OBJECTIVETo detect the expression of extracellular-signal regulated protein kinase 5 (ERK5) in multidrug-resistant hepatocellular carcinoma cell lines hepG2/ADM and BEL-7402/5-FU and in the parental cell lines (hepG2 and BEL-7402), and study the role of ERK5 in multidrug resistance of hepatocellular carcinoma.
METHODSMTT assay was used to determine the multidrug resistance (MDR) of hepG2/ADM and BEL-7402/5-FU cells. Real-time PCR and Western blotting were used to detect the expression of ERK5 at mRNA and protein levels in the 4 cell lines.
RESULTSThe resistance indices to ADM, 5-FU, and CDDP was 12.34, 5.74, and 3.81 in hepG2/ADM cells, and the resistance indices to 5-FU, VCR, OHP, MTX, and ADM was 15.32, 10.08, 5. 85, 6.74 and 3.26 in BEL-7402 cells, respectively. Compared with those in hepG2 and BEL-7402 cells, the ERK5 gene was up-regulated at both mRNA and protein levels in HepG2/ADM cells, but down-regulated at the mRNA level and up-regulated at the protein level in BEL-7402/5-FU cells.
CONCLUSIONSERK5 is related to multidrug resistance in HepG2/ADM and BEL-7402/5-FU cells, and may provide new clues for reversing multidrug resistance of hepatocellular carcinoma cells.

7.Variation of phenolic compound contents of Magnolia officinalis at different levels.

OBJECTIVETo reveal the mutative discipline of the contents of the phenols in samples from different provenances.
METHODThe contents of magnolol and honokiol in samples from the filial generation of Guanxi, Hubei and Zhejiang provinces were determined respectively by HPLC.
RESULTThere were significant variations between provenances, the contents of honokiol, magnolol and phenols were the highest in samples from Hubei province, those were the lowest in samples from Zhejiang province. There were not only significant differences in polyphenol content between provenances, but also in families in the same provenance and individuals in families.
CONCLUSIONMagnolia polyphenol content and appearance traits were significantly related. The type with the small convex tip (original form) were superior to other types of M. officinalis judging from the characteristics of tree classification. The high quality of M. officinalis should actually be attributed to their local varieties and the breeding strategy of M. officinalis should be paid full attention to the good provenances, good families and good individuals.

8.Genetic diversity and genetic relationships of spacies containing extremely aromatic compounds in leaves of Chimonanthus.

OBJECTIVESpecies containing extremely aromatic compounds in leaves of Chimonanthus was analyzed to evaluate its genetic diversity and genetic relationships.
METHODAFLP molecular marker technique was used in the study, UPGMA cluster analysis was conducted with the software of POPGENE32.
RESULTSFive hundred and fifty-nine bands were amplified by 10 pairs of primers screened, of which 226 bands were polymorphic, and the percentage of polymorphic bands was 36.8%. Observed number of alleles, effective number of alleles, Nei's genetic diversity index and Shannon's information index were 1.992 6, 1.306 5, 0.199 2 and 0.325 1, respectively. Genetic distances of the 21 populations were ranged from 0.039 2 to 0.289 4.
CONCLUSIONSpecies containing extremely aromatic compounds in leaves of Chimonanthus with low genetic diversity play an important role in enhancing the protection of species and germplasm resources. Form the molecular level, the studies demonstrated the correctness of the result by Zhang Ruohui that species containing extremely aromatic compounds in leaves of Chimonanthus were divided into Ch. salicifolius, Ch. Zhejiangensis, Ch. nitens and Ch. grammatus.

9.Advance in research of osteoblast adhesion to bioactive materials.

In the research field of bone tissue engineering, the interaction of osteoblast and substrate is pivotal and the adhesion of osteoblast to biomaterials is the basic condition. Firstly, osteoblast must adhere to biomaterials, then it can migrate, proliferate and differentiate. This paper introduces the proteins relating to the adhesion of osteoblast and the influences of relating surface character and modification of biomaterials on the adhesion ability of osteoblast. These could serve as basic data and useful reference for the development of bone tissue engineering and tissue engineering scaffold materials.

10.Comparison of bio-medical parameters in SIV infected Chinese rhesus monkeys with diverse progression correlated to the pathogenesis of simian AIDS

Objective To compare the bio-medical parameters in SIV infected Chinese rhesus monkeys with diverse disease progression,by which the pathogenesis of simian AIDS were to be investigated.Methods Sixteen Chinese rhesus monkeys were inoculated intravenously with SIVmac239 and followed-up for 18 months.Based on their progression patterns and plasma viral loads,animals were divided into 3 groups,including 1 rapid progressor( RP),13 normal progressors(NP),and 2 elite controllor(EC).Their parameters of haematology,virology,immunology and pathology were examined and compared. Results Compared with other animals,RM449(RP) showed higher viral load,unresponsive humoral immunity,and higher level of auto-antibodies against lymph node,thymus,and spleen.Additionally,its effector memory CD4 count was lower,with the transformation progress being blocked-like from naive/central memory subsets to effector memory subset,as the flow-cytometry assay showed.Notable decrease in its peripheral B cell was also observed,especially to the sub-population of tissue-like memory B cells and activated memory B cells.Pathological examination showed the depletion of lymphoid tissue,atrophy of spleen and loss of thymus.Moreover,most of these parameters of RM450 and RM453 (EC) changed opposite to that of RP.Conclusion The hallmarks of RM449 were higher viraemia and lower SIV specific IgG level,which may due to the disturbance of T cells and B cells development and differentiation.Moreover,destructions of organs of the immune system may contribute to the disturbance.Our study suggest that the change of micro-environments of thymus induced by SIV infection,which is necessary in T cell and B cell development and differentiation,may contribute at least partially to the AIDS pathogenesis.
