1.The effect of Quercetin on the long-term memory and PARP-1/AIF signal path in neonatal rats with hypoxic-ischemic brain damage

Objective To explore the effect of Quercetin on the long-term memory and PARP-1/AIF signal path in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods Fifty-six 7-day-old SD rats were randomly divided into sham-operation group, HIBD group, low dose of Quercetin group (20 mg/kg), and high dose of Quercetin group (40 mg/kg), each of 14 rats. Except for sham-operation group, in the other groups HIBD model were made by right common carotid artery ligation and anoxiate. The Quercetin groups were injected with the corresponding doses of Quercetin immediately once a day continuously for 7 days after the model was made,. Sham-operation group and HIBD group were intraperitoneally injected with normal saline at the same time. Neural function was evaluated by Hanging wire test and Vertical pole test at 21 days old. The capacity of learning and memory was detected by Morris water maze at 28 days old, and then rats were killed and brains were taken. HE was used to observe the pathological changes of hippocampus. Western blot were used to detect the expression of PARP-1 and AIF in hippocampus. Results Compared with sham-operation group, the neural function and learning and memory ability decreased significantly in HIBD group. Those ability in both low dose and high dose of Quercetin groups were remarkably increased in comparison with HIBD group, and there were statistic differences (P?0.05). HE showed an neuro-structure was intact and neatly arranged in hippocampus in sham-operation group. The neurons in HIBD group were obviously decreased in number and loosely arranged while those in both low dose and high dose of Quercetin groups were more increased and neatly arranged when compared with HIBD group. The expressions of PARP-1, AIF, and Bax in rats’ hippocampus were higher in HIBD group than those in low dose or high dose of Quercetin groups which were higher than those in sham-operation group. The change of Bcl-2 was the opposite, and there were statistic differences (P?0.05). Furthermore, there was no difference between low dose of Quercetin group and high dose of Quercetin group in neural function, learning and memory ability, the expressions of PARP-1, AIF, Bcl-2, and Bax (P?>?0.05). Conclusion Quercetin could improve long-term learning memory in newborn rats with HIBD, and the mechanism may be down-regulation of PARP-1/AIF cell apoptosis signaling pathway, inhibition of neuronal apoptosis, and thus play a role in protection of brain.
2.Effects of Quercetin on nerve regeneration in neonatal rats with hypoxic-ischemic brain damage

Objective To explore the protective effects of Quercetin on hypoxic ischemic brain damage (HIBD). Methods Forty-eight 7-day-old SD rats were randomly divided into sham-operation group, HIBD group and Que treatment group, 16 rats each. HIBD group and Que treatment group were treated by ligation of right common carotid artery to make anoxia and build HIBD model; sham-operation group had the separation of the right common carotid artery only. Que treatment group were injected intraperitoneally with quercetin (40 mg/kg) once a day for 7 days immediately after modeling while sham-operation group and HIBD group received equivalent normal saline at the same time. The rats in each group were scored of neurological function at 1 h after the last administration, and the ability of spatial learning-memory was tested by Morris water maze at the age of 28 days. After performing the test above, all rats were decapitated and the brains were taken. Pathological changes of hippocampus were observed by HE staining; the expression of brain-derived neurotrophic factor (BDNF) and nerve growth associated protein (GAP-43) in hippocampus CA1 area were detected by immunohistochemistry. Results There were significant differences in neurological deficit score and learning-memory ability among the three groups (P<0.01), and neurological deficit score was the highest and the learning-memory ability was the lowest in HIBD group. Pathological examination showed that the structure of hippocampal neurons was intact in sham-operation group. It was loose and disorder, and even loss of neurons in HIBD group. Compared with the HIBD group, the loose in the structure of hippocampal was lighter, and the number of neurons was increased in the Que treatment group. There was statistical difference in the positive expression of BDNF and GAP-43 in the hippocampal CA1 area among the three groups (P<0.01), with those in HIBD group being lower than in Que treatment group and sham-operation group and those in treatment group being lower than in sham-operation group (P<0.01). Conclusions Quercetin can enhance the expression of BDNF and GAP-43 in hippocampal CA1 region, promote nerve regeneration, improve the long-term learning-memory ability of HIBD neonatal rats, and protect the brain.

Radiation-induced organ fibrosis is a common complication of tumor radiotherapy and radiation accident, which poses a serious threat to patients′life.TGF-β, CTGF, PDGF-BB and NF-κB all play important roles in this pathological process, and drugs targeting these key molecules are also being developed, some of which have shown positive therapeutic effects.This review focuses on the molecular mechanisms and therapeutic progress involved in radiation-induced organ fibrosis, intended to provide a new clue to the complex process of radiation-induced organ fibrosis, and to provide a new strategy for treatment.
5.Observation on the efficacy of 131 iodine combined with surgical treatment for differentiated thyroid cancer

Objective To discuss differentiated thyroid carcinoma (DTC)postoperative lymph node metas-tasis and clinical use and the effect of 131 I treatment.Methods to185 patients with lymph node metastasis of DTC were selected by random sampling method.The standard 131 I treatment was administrated,ultrasound,CT and Tg chan-ges and imaging characteristics of 131 I treatment were evaluated the clinical curative effect was determined.Results Imaging changes:the whole group of patients,153 patients (82.7%)were effective.153 patients with effective treat-ment,105 cases examined LMDTC completely eliminate (56.8%),48 cases examination revealed narrowing (25.9%).Mere lymph node metastasis after treatment was better than that of patients with viscera metastasis,imaging examination results were statistically significant differences (χ2 =2.57,P =0.02).After treatment,imaging examina-tion result in patients with smaller lymph node was superior to that in the patients with relatively large diameter lymph nodes (P >0.05).Tg level changes:there was a significant reduction in the Tg 97 cases of 185 patients (51.1%), and general decline in 54 patients (29.2%).Tg improvement effect of patients with mere lymphatic metastasis after treatment was better than other viscera metastasis in patients with statistically significant differences (χ2 =3.41,P =0.01).Lymph node Tg improved after treatment in patients with small diameter,which was better than that with other viscera metastasis patients with statistically significant differences(χ2 =6.34,P =0.02).Conclusion Comprehensive curative effect of 131 I treatment of lymph node metastasis postoperatively in patients with DTC is distinct,and is worth popularizing in clinical use of DTC.
6.RESEARCH ON THE RELATIONSHIP BETWEEN INSULIN RESISTANCE AND SUSCEPTIBILITY OF OBESITY

Objective To study the differences between diet induced obesity (DIO) and diet induced obesity resistance (DIO-R) during insulin resistance (IR) in rats. Method Male SD rats were fed by high-fat diet 1 (45 % fat kJ) after adaptation. At 3rd week, rats were divided into 3 groups according to their body weight: rats belonging to the middle 1/3 of their body weights were the control group, and was fed by control diet (10 % fat kJ). The heaviest 1/3 and the lightest 1/3 belong to DIO and DIO-R group, respectively, and were fed by high-fat diet 2 (60 % fat kJ). Five rats were selected randomly from each group and sacarified at 3rd, 6th, 8th, 10th and 11th week. The energy intake, body fat contents, fasting blood glucose (FBG), serum lipid, serum insulin, insulin sensitive index(ISI)and leptin of rats were monitored dynamically. Results DIO rats were significantly different from DIO-R and control rats in energy intake, body fat content, FBG, lipid, serum insulin and leptin. Conclusion Fed by the same high-fat diet, DIO rats are more sensitive than DIO-R rats to become IR.
7.Clinical study of electroacupuncture and taking oral Mecobalamin tablets in the treatment of type 2 diabetes mellitus combined with mild to moderate carpal tunnel syndrome

Objective:To observe the clinical efficacy of electroacupuncture in the treatment of type 2 diabetes mellitus combined with mild and moderate carpal tunnel syndrome.Methods:Eighty patients with type 2 diabetes mellitus combined with mild to moderate carpal tunnel syndrome were randomly divided into two groups with 40 patients in each group ( n=40). In the course of the treatment, 2 patients in each group dropped out, which means 38 patients in each group were included. All patients were instructed to actively control blood glucose The control group was given Mecobalamin tablets orally, while the treatment group was given electroacupuncture of Daling and Neiguan on the affected side based onthe control group for 6 weeks. The degree of pain was assessed by VAS, and the severity of clinical symptoms was assessed by the symptom scale of Boston carpal tunnel questionnaire (BCTQ). The sensory nerve conduction velocity (SCV), the amplitude of sensory nerve action potential (SNAP) and the distal motor latency (DML) were detected by EMG evoked potential instrument. The clinical efficacy was evaluated and adverse events were recorded. Results:The total effective rate was 81.6% (31/38) in the treatment group and 60.5% (23/38) in the control group, and the difference was statistically significant ( χ2=5.094, P=0.024). After the treatment, the scores of VAS and BCTQ in the treatment group were lower than those of the control group ( t=2.639, 2.790, P=0.010, 0.007); DML (3.62 ± 0.19 ms vs. 4.00 ± 0.17 ms, t=68.891) in the treatment group was lower than that of the control group ( P<0.01), while SCV (47.36 ± 0.18 m/s vs. 42.34 ± 0.14 m/s, t=97.163) and SNAP (14.74 ± 0.18 μV vs. 12.10 ± 0.16 μV, t=51.434) in the treatment group were higher than those of the control group ( P<0.01). During the treatment, no serious adverse events occurred in two groups. Conclusions:Electroacupuncture is safe and effective in the treatment of mild and moderate carpal tunnel syndrome with type 2 diabetes. It can reduce the degree of pain and improve the symptoms of wrist joint.

The aim of this study was to establish the methods to identify crystal form of dasatinib in tablets.X-ray powder diffraction(XRPD)and solid-state nuclear magnetic resonance(ssNMR)were used to analyze the crystal form of dasatinib in Sprycel? tablets and Yinishu? tablets.The results showed that monohydrate and anhydrate were identified in Sprycel? and Yinishu? tablets respectively;with no detectable anhydrate in Sprycel? tablets and no detectable monohydrate in Yinishu? tablets.The results of XRPD and ssNMR were consistent;and could be both applied in the crystal form identification of dasatinib in tablets.
9.Gene Cloning and Expression of PACAP and Study of Its Biological Activity

In order to study the expression and the feasibility of scaled production of neuropeptide in the routine expression system such as E.coli with the pituitary adenylate cyclase activating polypep tide(PACAP) as an example, the following experiments were carried out. First, on the basis of the reported amino acid sequence of PACAP, DNA sequence of PACAP w as deduced and six partially complementary oligonucleotide fragments were design ed. The coding region of PACAP was obtained by renaturing the DNA fragments and ligation and identified by DNA sequencing. The coding region of PACAP was cloned into plasmid pGEX-4T-3 and transformed into E.coli BL21(DE3 ). An expression strain BLPACAP was selected. SDS-PAGE analysis revealed that t he GST-PACAP fusion protein was highly expressed and accumulated to about 30% o f the total bacterial proteins. By affinity chromatography, up to 90% GST-PACAP was purified by one step from bacterial lysate. The purified protein could prom ote neurite outgrowth of PC12 cells and the survival of spinal cord neurons.
10.Analysis of the curative effect of endoscopic band ligation combined with drug injection in the treatment ;of hepatic cirrhosis complicated with upper gastrointestinal haemorrhage

Objective To explore the curative effects of endoscopic band ligation combined with drug injection in the treatment of hepatic cirrhosis complicated with upper gastrointestinal hemorrhage. Methods 76 patients with hepatic cirrhosis complicated with upper gastrointestinal hemorrhage were divided into 2 groups by random double blind method, with 38 cases in each group. The control group were treated by conservative regimen alone while the experimental group were treated by endoscopic band ligation combined with drug injection, based on the control group. The clinical curative effects, adverse reactions, hemostasis time, hospitalization time , rebleeding , haemodynamics and other indexes before and after the treatment in the 2 groups were compared. Results The total effective rate, rebleeding rate and the incidence of fever in the experimental group were 2.6% and 10.5%, respectively. Compared with those in the control group (71.1%, 21.1%, 28.9%), the differences were statistically significant (P < 0.05). The differences in volume of blood transfusion, hemostasis time , time of improvement of symptoms and hospitalization time between the experimental group and the control group were significant (P < 0.01). The blood flow of portal vein(550.2 ± 143.4)mL and splenic vein (284.3 ± 96.4)mLin the experimental group was significantly less than that in the control group [(628.1 ± 156.0)mL, (332.6 ± 100.3)mL] (P < 0.05). Condusions Endoscopic band ligation combined with drug injection in the treatment of hepatic cirrhosis complicated with upper gastrointestinal hemorrhage is effective and can significantly improve the varicose hemodynamics in patients. The rate of rebleeding is low.
