1.The influence of Xuebijing injection and low molecular weight heparin on coagulation function and prognosis in patients with severe sepsis

Objective To compare between the impact of Xuebijing injectio (a Chinese herbal medicine preparation) and that of low molecular weight heparin (LMWH) on coagulation function and prognosis in patients with severe sepsis, so as to guide the treatment of sepsis. Method A total of 89 patients with severe sepsis were randomly(random number) divided into 3 groups, namely Xuebijing group (group A),LMWH group (group B) and routine group (group C). The patients of group C received a series of remedies according to the guideline for severe sepsis. The patients of group A were treated with Xuebijing injectio in addition to routine treatment. The patients of group B were treated with LMWH along with routine treatment.Data of blood coagulation function tests and APACHE Ⅱ score of patients in three groups before and after treatment were analyzed respectively. The 28-day mortality and length of hospital stay were compared among three groups. All data were analyzed by using t-test, Analysis of Variance, q-test, x2-test and rank sum treatment, all markers of the blood coagulation function tests in group A improved significantly after treatment (P<0.01), while only some markers of the blood coagulation function tests improved in the other two groups ( P < 0. 05 or P < 0. 01 ); After treatment, the blood coagulation function in group A improved significantly more than that in other two groups ( P < 0.05 ), while there were some markers of the blood coagulawith the data before treatment, the APACHE Ⅱ score in group A and group B decreased after treatment (P<0.05). After treatment, the APACHE Ⅱ score of group A and group B decreased significantly more than that of group C ( P < 0.05). But there was no significant difference between group A and group B ( P >Xuebijing obviously improves the blood coagulation function in patients with severe sepsis, while LMWH only improves some markers of the coagulation function tests. Both Xuebijing and LMWH reduce mortality and improve prognosis of patients with sepsis.
3.Anesthetic effect of different concentrations of low-dose hypobaric ropivacaine in elderly knee or hip replacement

BACKGROUND: Unilateral spinal epidural anesthesia with ropivacaine can reduce the perioperative pain in the elderly undergoing hip or knee arthroplasty, but choosing which concentration of small dose hypobaric ropivacaine is an issue of concern.OBJECTIVE: To explore the anesthetic and analgesic effects of unilateral spinal epidural anesthesia with different concentrations of low dose hypobaric ropivacaine in senile knee or hip replacement.METHODS: Ninety patients undergoing knee or hip replacement in the Henan Province Hospital of TCM from August 2014 to December 2015 were enrolled, and randomized into 0.15%, 0.5% and 0.25% ropivacaine groups (n=30 per group), followed by spinal epidural anesthesia at L3-4 or L2-3 lumbar interspace, respectively.RESULTS AND CONCLUSION: (1) The order of the flat subsided time and motor block recovery time was as follows:0.15% ropivacaine group < 0.25% ropivacaine group < 0.5% ropivacaine group; the onset time of motor block was longest in the 0.25% ropivacaine group, followed by 0.5% ropivacaine group, and shortest in the 0.15% ropivacaine group (all P < 0.05). (2) The order of the visual analogue scale scores at 1, 5, and 10 hours after anesthesia and 24 hours postoperatively was as follows: 0.5% ropivacaine group < 0.25% ropivacaine group < 0.15% ropivacaine group (P < 0.05). (3) The change levels of heart rate and blood pressure were highest in the 0.5% ropivacaine group, followed by 0.15% ropivacaine group, and lowest in the 0.25% ropivacaine group. (4) The incidence of adverse events was highest in the 0.5% ropivacaine group, followed by 0.15% ropivacaine group, and lowest in the 0.25% ropivacaine group (P < 0.05). (5) To conclude, 3 mL of 0.25% ropivacaine exhibits desired anesthetic effect.
4.Progress in the study of allergic disease drugs targeting on IgE/FcepsilonRI signaling pathway.

Allergic diseases have become global social health problems. The binding of IgE with its high affinity receptor FcepsilonRI plays a key step in I-type allergy. Recently, more and more key molecules on the IgE/FcepsilonRI signaling transduction pathway were to be the drug candidates against allergic diseases, with in-depth study of FcepsilonRI signal pathway gradually. The main drugs include molecule antibodies, peptides, vaccines, fusion proteins, small molecules, and other drugs related to IgE/FcepsilonRI. The recent progress in the study of mechanisms of representative drugs targeting on IgE/FcepsilonRI signaling pathway was reviewed in this article.
5.Clinical effects of Yiqihuoxue decoction on remnant stomach paralysis syndrome after subtotal gastrectomy

Objective To approach effect of Yiqihuoxue decoction on remnant stomach paralysis syndrome after subtotal gastrectomy. Methods A total of 44 patient of remnant stomach paralysis syndrome were randomly recruited into a control group and a treatment group, with 22 patients in each group. The control group was treated with TPN parenteral alimentation. On this basis, the treatment group was treated with Yiqihuoxue decoction and mental intervention. The clinical effects were observed in both groups after the treatment. Results The total effective rate was 54.55% and 100% in the control group and the treatment group respectively, showing statistic difference(P=0.00625 <0.01).Conclusion It was effective to treat remnant stomach paralysis syndrome with Yiqihuoxue decoction, TPN parenteral nutrition and mental intervention.
6.A COMBINATION PROCESS OF STERILIZING,DEPLUG AND FLOOD-INCREASING PROCESS WITH SODIUM HYPOCHLORITE

A new combination process of sterilizing, deplug and waterflood-increasing with sodium hypochlorite in waterflooding horizons has been developed. Its applicability was certified by 4 treatments of injection wells. After treatments the water-intake capacity of injection wells was increased, the pressure at well heat was decreased, and the average duration of stability wster injection was 307 days.
7.Screening and characterization of aptamers of Cepsilon3-Cepsilon4 protein.

In order to obtain nucleotides aptamers bind to IgE, 80 bp nucleotides single-stranded DNA library containing 40 random nucleotides was designed and synthesized. Oligonucleotides that bind to human Cepsilon3-Cepsilon4 protein were isolated from ssDNA pools by the systematic evolution of ligands by exponential enrichment (SELEX) method using nitrocellulose filters as screening medium. Through the optimization of critical PCR and asymmetric PCR parameters including annealing temperature, cycles, and molar ratios of target protein and ssDNA etc, a suitable screening system was established. The aptamers of Cepsilon3-Cepsilon4 protein with high affinity and high specificity were identified by ELISA with biotin-streptavidin-horseradish peroxidase system, and its primary sequence and second structure were analyzed by DNAMAN package and DNA folding sever after being cloned and sequenced. Moreover, target protein was bound to one aptamer and another aptamer modified with biotion together forming a sandwich-like complex, which was captured in microwell to detect IgE concentration using the optimal combination in the sandwich method named enzyme-linked aptamers sorption assay (ELASA). The method could be used for the quantitative detection of human IgE, and whose sensitivity reached to 120 ng x mL(-1).
8.Simultaneously determination of gentianaceae and jasminoidin in Longhui Capsules

AIM:To determine gentianaceae and jasminoidin in Longhui Capsules (Radix et Rhizoma Gentia-nae, Aloe, Fructus Gardeniae, etc.) METHODS: The Waters Symmetry C_ 18 column (4.6 mm?150 mm, 5 ?m) was used with mobile phase of methanol-water (17∶83). The flow rate was 1.0 mL/min. The detecting wavelength was at 254 nm, and the column temperature was at 30 ℃. RESULTS: The methodological study showed that a good linear correlation existed in the range of 60.72-454.4 ng for gentianaceae and the range of 96.32 -722.40 ng for jasminoidin. The average recovery of gentianaceae was 99.57%(n=9) and RSD was 1.27% , jasminoidin was 99.10%(n=9) and RSD was 2.30%. CONCLUSION: The method is simple, accurate and can be used for quality control of the preparation.

10.Influence of Radix Salviae Miltiorrhizae on Methotrexate Pharmacokinetics in Rats

Objective To compare the pharmacokinetics difference of methotrexate ( MTX) alone or MTX combined with the decoction of Radix Salviae Miltiorrhizae ( RSM) in rats, and to investigate the possible impact of RSM on the efficacy and pharmacokinetics of MTX after oral administration. Methods Sprague-Dawley rats were given MTX (7 mg/kg) alone or MTX together with RSM (3.085 g/kg) respectively. At different time points, blood was sampled from orbital venous plexus and then was precipitated by perchloric acid. The serum concentration of MTX was determined by using high performance liquid chromatography ( HPLC) method. Chromatographic separation was achieved on a reversed-phase Diamonsil C18 (2) column with the mobile phase of methanol-formic acid water solution (formic acid in volume fraction of 0.1%) in gradient elution. The column temperature was 27 ℃, flow rate was 1 mL/min, detection wavelength was 302 nm, and the injection volume was 10 μL. The results were analyzed by pharmacokinetic software DAS ( 2.1.1) by non-compartment model. Results The linearity of the calibration curve for MTX was good in the range of 0.097 6 ~ 12.5 mg/L, the detection limit was 0.097 6 mg/L and the recovery was in the range of 77.5% ~ 86.6%. Compared with MTX group, the peak-arriving time of MTX-RSM group was advanced, and the clearance of MTX was delayed. Area under concentration-time curve at 0-t (AUC(0-t)) was increased by 0.609 times, and AUC(0-∞) was increased by 0.786 times. Conclusion The decoction of RSM exerts an effect on promoting the absorption of MTX, delaying the clearance of MTX and enhancing the bioavailability of MTX in vivo.
