1.Anti-liver fibrosis activities of the extracellular domain of transforming growth factor beta type II receptor fusion protein in vivo

This study is performed to analyze the anti-liver fibrosis effect of the fusion protein of human serum albumin and extracellular domain of transforming growth factor beta type II receptor(eTGFBR2)in vivo to looking for the more stable anti-liver fibrosis drug. The mice model of liver fibrosis was constructed by CCl4 induction and the following groups are included in the study: the control group, CCl4 model group, the positive control group, eTGFBR2 treatment group, HSA-eTGFBR2 treatment group, and HSA group. Hematoxylin eosin staining, serum liver function index detection, and western blot are used to identify the anti-liver fibrosis activities. The results showed that: (1)CCl4 caused liver structure disorder, hepatocellular necrosis, collagen fibers proliferation, and induced liver fibrosis at last; (2)HSA-eTGFBR2 and its monomer drug improved the symptoms of liver fibrosis significantly, as well as reduced the damage of liver cells and collagen deposition, and recovered the liver basic structure to normal. Both of HSA-eTGFBR2 and its monomer drug improved liver function and reduced the expression level of liver fibrosis marker α-SMA and COL I. Moreover, the anti-liver fibrosis effect of the fusion protein is comparable to the monomer drug. In contrast, the albumin had no effect on therapeutic effect; (3)Reducing the injection frequency of HSA-eTGFBR2 achieved the comparable effects to the monomer drug with the normal injection frequency. In summary, the fusion protein HSA-eTGFBR2 has good anti-liver fibrosis effect. In addition, reducing the injection frequency of the fusion protein could also achieve the comparable treatment with the monomer drug, indicating that the fusion protein is stable and has longer half-lives and then a relatively positive application prospect in future.
2.Simultaneous HPLC Determination of Hesperidin, Magnolol, Honokiol and Liquiritin in Soft Capsule Jia-Wei Huo-Xiang Zheng-Qi

This study was aimed to develop an HPLC method for the determination of hesperidin,magnolol,honoki-oland liquiritin in Soft Capsule Jia-Wei Huo-Xiang Zheng-Qi (JWHXZQ). AKromasil C18 column (250 mmí4.6 mm, 5 μm) was used with water-methanol as mobile phasegradient elution. The flow rate was 1.0 mL·min-1, and the de-tecting wavelength was at 287 nm. The results showed that the linearity ranges ofhesperidin,magnolol,honokioland liquiritinwere 4.47-178.70 μg·mL-1, 3.42-136.96 μg·mL-1, 3.49-139.48 μg·mL-1, 3.92-157.20 μg·mL-1, respec-tively (r>0.999). The average recoveries of them were 99.48%, 99.05%, 99.57% and 99.79%, respectively. It was concluded that the method was accurate, sensitive and specific for quality control of Soft Capsule JWHXZQ.
3.Anti-liver fibrosis activities of human insulin-like growth factor-1 in vitro

This study was focus on investigating the anti-liver fibrosis effects of insulin-like growth factor-1 (IGF-1) in vitro.The effects of IGF-1 on human liver L-02 cell viability and cell cycle were observed.CC14-induced L-02 cell injury was set up to detect the anti-apoptotic activity of insulin-like growth factor-1 (IGF-1).Transforming growth factor β1 (TGF-β1) induced hepatic stellate cell line (HSC-T6) were used as a liver fibrosis model in vitro to analyze the effects of IGF-1 on the expression of liver fibrosis proteins and intracellular TGF-β1/Smad signaling pathway in HSC-T6 cells.The results showed that IGF-1 could relieve the growth inhibition effects of TGF-β1 on L-02 cells,increase the viability of L-02 cells injured by CCl4,decrease the expression of liver fibrosis proteins,and inhibit the TGF-β1/Smad signaling pathway by inhibiting the phosphorylation of Smad3.Our study suggested that IGF-1 exerted anti-liver fibrosis effects by stimulating L-02 cells proliferation,reducing cell damage and inhibiting ECM accumulation via interfering TGF-β1/Smad signaling pathway.
4.Preparation and characterization of an extracellular matrix of artificial tendon tissue from natural macromolecules.

Collagen and chitosan are well natrual polymers to be used as extracellular matrix on tissue engineering because of their biocompatibility, certain mechanical strength and biodegradability. But there are some disadvantages when they are used to construct extracellular matrix respectively. This experiment utilized their complementary performances to prepare a composite extracellular matrix of artificial tendon tissue that had adequacy mechanics strength and good biocompatibility, cell affinity, biodegradability. Collagen and chitosan were covalently crosslinked using EDC and NHS to obtain a porous scaffold material that the porous was oriented under an external force. Then RGD peptide was covalently attached to scaffold material surface to improve its affinity with cells. The microstructure of scaffold material was observed under microscope and scanning electron microscope. Simultaneously, the physical performance, hydrophilicity, ecto-degradation rate and cell compatibility of scaffold material were measured in the experiments. The results showed that this scaffold material was soft and stretchy. Its tensile strength was 15.0 MPa, corresponding shape extension was 7.33%, and its porosity was 79.4%. Its water absorption rate and water retention rate were 772% and 206% respectively. Its degradation rate in RPM 1640 culture mediun with 10% fetal bovine serum and in human serum were 4.13% and 37.2% respectively after three weeks. These degradation rates are suitable for the rehab course of injured tendon. Moreover the degradation rates can be controlled by adjusting technological conditions and degree of cross linking. Significantly higher affinity with 3T3-Ll cell was detected on the scaffold material modified by RGD peptide. The various physical performances of this complex scaffold material are appropriate for constructing extracellular matrix of artificial tendon tissue or artificial skin. Moreover, it could be used as soft tissue slurry for plastic surgery.

5.Investigation of radiation protection status in non-medical radiation institutions in Fujian Province, China

Objective To investigate the number and categorical distribution of non-medical radiation institutions in Fujian Province, China, and to explore the current situation of radiation occupational hazards prevention and control and the occupational health management of radiation workers in employing units. Methods The industrial enterprises engaged in the application of nuclear technology in our province were investigated, and different types of employing units were selected to monitor the radioactivity level in the workplace. Results A total of 377 non-medical radiation institutions were reported in the province, with a total of 5221 radiation workers (except luggage detector units); 1246 radiation workers held radiation worker certificates, accounting for 23.87%; 5103 radiation workers carried out personal dose monitoring, accounting for 97.74%; 5061 radiation workers participated in radiation protection knowledge training, accounting for 96.94%; a total of 4895 (93.76%)radiation workers received physical examination. Conclusion Health supervision authorities in Fujian Province should strengthen the supervision of non-medical radiation institutions, further improve the level of occupational health management, and protect the health rights and interests of radiation workers to the maximum extent.
6. Bioequivalence study of cetirizine hydrochloride tablets under fasting and fed conditions

AIM: To evaluate the pharmacokinetics and bioequivalence of cetirizine hydrochloride tablets under fasting and fed conditions in Chinese healthy subjects. METHODS: This was a randomized, open-label, double-sequence, two-period, crossover designed study, and healthy subjects enrolled and administrated a single dose of 10 mg test and reference cetirizine hydrochloride tablets in each period under fasting or fed condition. The plasma concentrations of cetirizine were determined by a validated LC-MS/MS method. The pharmacokinetic parameters were calculated with WinNonlin 6.3 and the bioequivalence was evaluated through SAS 9.4 software. 7.Screening of tomato cultivars in cadmium-polluted areas and study on their antioxidant capacity.

To screen the available tomato pollution-safe cultivar varieties and reduce the potential food safety risks in Cd-polluted areas, the differences of Cd accumulation in different tomato (Solanum lycopersicum) varieties in southern China were studied by soil culture and hydroponic experiments. Firstly, the high and low accumulation varieties were selected from 25 tomato varieties under 2.94 mg/kg Cd stress by soil culture test, and then the responses of high and low accumulation tomato varieties to Cd stress were determined by hydroponic experiments. The results of soil culture test show that under 2.94 mg/kg Cd stress, there were significant differences in plant height, total biomass and yield among 25 tomato cultivars, and the Cd contents of fruits of all 25 tomato cultivars exceeded the highest limit value (0.05 mg/kg) of CAC (Codex alimentarius commission). Through cluster analysis, 7, 4 and 14 varieties accumulating relatively high, medium, and low concentrations of Cd in the fruits were screened, among which the highest, the lowest, and the average Cd contents in the fruits were 3.06 mg/kg DW, 1.47 mg/kg DW, and 2.21 mg/kg DW, respectively. The results of hydroponic experiment show that under the same concentration of Cd stress, Qiantangxuri F1, a high Cd accumulating variety, absorbed Cd faster, accumulated more Cd, used shorter oxidative stress response time and had stronger tolerance to Cd than Zhefen 3053, a low Cd accumulating variety. The typical high and low Cd accumulating varieties can provide a reference for agricultural production in heavy metal polluted areas and the development of molecular-assisted breeding methods of PSC. At present, cultivating low Cd accumulating PSC varieties and dynamic monitoring of Cd contents in tomato fruits are feasible methods in medium and light Cd-polluted areas.
