1.The influence to atrial fibrillation occurs of long -term use of trimetazidine in treatment of dilated cardiomy-opathy

Objective To investigate the influence to atrial fibrillation occurs of long -term use of trimetazi-dine in treatment of dilated cardiomyopathy.Methods 90 cases with dilated cardiomyopathy (DCM)were averagely divided into the observation group and the control group according to a random number table,45 cases in each group. The patients of the control group were treated with conventional anti -heart failure according to their heart function, the patients of the observation group were given oral trimetazidine on the bases of conventional anti -heart failure. After follow -up six months,the left ventricular ejection fraction (LVEF),left ventricular end diastolic diameter (LVEDD),improve heart function,as well as the incidence of atrial fibrillation after treatment of two groups were compared.Results The rate of occurrence after treatment of the observation group atrial fibrillation of was 2.27%, which was significantly lower than 11.11% of the control group,the difference was statistically significant(χ2 =10.74,P <0.05).The left ventricular end diastolic diameter (LVEDD)after treatment of the observation group was (52 ±9)mm,which was significantly lower than (63 ±11)mm of the control group,the difference was statistically sig-nificant(t =6.83,P <0.05);The left ventricular ejection fraction (LVEF)of the observation group was (54 ±7)%, which was significantly higher than (41 ±6)% of the control group,the difference was statistically significant(t =8.72,P <0.05).Conclusion Dilated cardiomyopathy patients taken long -term trimetazidine,can greatly reduce the incidence of atrial fibrillation.
2.Immunotherapy experimental study of γδT cells from human peripheral blood on human liver cell nude mouse xenograft in vivo

Objective:To study the immunotherapy effects of different doses of human peripheral blood γδT cells on human hepatoma cells (SMMC-7721) xenograft model.Methods: (1)The nude mouse model of liver cancer was established by inoculated BALB/c mouse subcutaneous with human hepatoma cell line (SMMC-7721).(2)The mononuclear cells in healthy human were extracted from peripheral blood,and specific amplification γδT cells in vitro.(3) The nude mouse model divided into 5 groups by random.The positive control group was 5-Fu,negative control group was normal saline(NS).The treatment group was injected different doses of γδT cells(1×105,5×105 and 25×105)by nude mice tail vein.The positive control group injected 5-Fu by enterocoelia,negative control group injected NS by tail veins.The inhibition effect of different dose γδT cells on tumor was observed,including weight,food intake and growth conditions,etc.and the changes of tumor volume (TV),relative tumor volume (RTV)and relative tumor appreciation rate[T/C(%)] were compared with positive control group and negative control group.Results: Different dose of γδT cells had different degree of inhibition on nude mouse xenograft growth.RTV compared with saline negative control group was statistically significant (P<0.05).Compared with the positive control group of 5-Fu,the TV growth was significantly lower than the 5-Fu,degree of inhibition was similar in RTV each dose group,and all slightly higher than the 5-Fu positive control group.The each dose group of T/C (%)was slightly lower than the relative tumor proliferation rate of the control group of 5-Fu,but had no significant difference.Conclusion: The γδT cells from peripheral blood had significant inhibitory effect on nude mice transplanted liver tumor and it may be used as a new treatment for liver cancer immunotherapy provide experimental data.
3.The clinicopathological characteristics and prognosis of 30 patients with hepatoid ade-nocarcinoma of stomach

Objective:To review the clinicopathological characteristics and analyze the prognostic factors of hepatoid adenocarcinoma of stomach(HAS).Methods:From August 2012 to June 2017,30 patients with HAS were diagnosed in Daycare of Peking University Cancer Hospital.Clinicopathological data and follow-up information of these patients were retrospectively collected and analyzed.Re-sults:The median age of these 30 patients was 58 years at diagnosis,with a male-to-female ratio of 2.75:1.Twenty-nine patients were confirmed to have lymph node metastases and 7 patients had distant metastases.Ten patients died because of the cancer within the follow-up period.The 1-and 3-year survival rates were 60% and 52%,respectively.Cox multivariate regression revealed that elevated serum CA199 levels,higher lymph node staging,not having undergone radical surgery,and stronger immunohistochemical(IHC)stain-ing intensity of alpha fetoprotein(AFP)were independent poor prognostic factors.Conclusions:Elevated serum CA199 levels,lymph node staging,and IHC staining intensity of AFP are verified in this study as independent risk factors of poor outcome in HAS patients. Early detection and diagnosis of the disease may improve the clinical prognosis.Multidisciplinary team discussions are important in making therapy decisions and radical surgery should be performed whenever possible.
4. Efficacy and safety of pemetrexed combined with nedaplatin or cisplatin in the treatment of advanced non-small cell lung cancer

Objective:
To observe the clinical efficacy and adverse reactions of pemetrexed in combination with nedaplatin or cisplatin in the treatment of advanced non-small cell lung cancer(NSCLC).
Methods:
85 patients with stage Ⅲ B/IV NSCLC diagnosed by pathology or cytology were randomly divided into two groups by random double blind method.The study group(45cases) was treated with pemetrexed combined with nedaplatin, and the control group(40cases) was given pemetrexed combined with cisplatin chemotherapy.The specific method was pemetrexed 500mg/m2, the first day of intravenous infusion, cisplatin 75mg/m2 or nedaplatin 80mg/m2, divided into 3 times, the 2nd, 3rd, 4th day, intravenous infusion, every 21d for a cycle.At the end of the chemotherapy, the efficacy and adverse reaction were analyzed.The patients were followed up to record no progress survival and one-year survival rate.
Results:
Among 85 patients, there were 35 cases of partial remission(PR), 35 cases of stable(SD) and 15 cases of progression(PD), the total effective rate(ORR) was 42.2%, the disease control rate(DCR) was 82.2%.There were no statistically significant differences in ORR and DCR between the two groups(χ2=0.043, 0.001, all
5.Advances in machine learning for predicting protein functions.

Proteins play a variety of functional roles in cellular activities and are indispensable for life. Understanding the functions of proteins is crucial in many fields such as medicine and drug development. In addition, the application of enzymes in green synthesis has been of great interest, but the high cost of obtaining specific functional enzymes as well as the variety of enzyme types and functions hamper their application. At present, the specific functions of proteins are mainly determined through tedious and time-consuming experimental characterization. With the rapid development of bioinformatics and sequencing technologies, the number of protein sequences that have been sequenced is much larger than those can be annotated, thus developing efficient methods for predicting protein functions becomes crucial. With the rapid development of computer technology, data-driven machine learning methods have become a promising solution to these challenges. This review provides an overview of protein function and its annotation methods as well as the development history and operation process of machine learning. In combination with the application of machine learning in the field of enzyme function prediction, we present an outlook on the future direction of efficient artificial intelligence-assisted protein function research.
