1.Effect of Aldosterone on the Proliferation and Collagen Metabolism of Cardiac Fibroblasts in Neonate Rat

OBJECTIVE: To investigate the effects of aldosterone (Ald) on the proliferation and collagen synthesis of cardiac fibroblasts (CFs) in neonate rats and to research its mechanism. METHODS: The neonatal rat cardiac fibroblasts were induced by anchorage velocity-dependent separation method. SD rats were divided into control group, Ald (1.0?10-7 mol?L-1) group, Ald combined with spirolactone group and Ald combined with losartan group. After 24 h of exposure, CFs proliferation was measured by MTT assay and cell cycle distribution was determined with flow cytometer (FCM). Spectrophotography was used to detect the content of hydroxyproline and immunofluorescence staining method was applied to determine the protein expression of proliferation cell nuclear antigen (PCNA) and fibronectin (FN). RESULTS: As compared with control group, in Ald group the activity of CFs was increased as well as its percentage in S stage. The content of hydroxyproline and the protein expression of PCNA and FN were also increased while the percentage of CFs in G0/G1 stage was decreased (P0.05). CONCLUSION: Ald-stimulated CFs proliferation and collagen synthesis were mediated by activation of AT1 receptors and Ald receptor,and Ald receptor plays a major role. A corresponding increase in the protein expression of PCNA and FN in CFs may be related to the proliferation effect.

Objective To study the identification method of Akebia trifoliata(Thunb.) Koidz. by Fourier Transform Infrared (FTIR) Spectroscopy. Methods FTIR Spectroscopy was measured of Akebia trifoliate collected from different production areas. Results At the range of 737-1032cm-1, the Spectroscopy of Akebia trifolia of different production areas showed variances in peak value of infrared absorption, peak position, peak shape and peak strength, which can be regarded as identification evidence for Akebia trifoliate. Conclusion This mehthod is rapid, reliable, simple and effective. FTIR can be used as the identification index for Akebia trifoliate.

Hypoxia-inducible factor-1 (HIF-1), as a nuclear transcription regulator of the hypoxic response, is up-regulated during hypoxia, and it regulates a series of downstream target gene expression, such as vascular endothelial growth factor, glucose transporter and erythropoietin through binding with hypoxia response element. It plays important roles in angiogenesis, anerobic metabolism, cell survival, proliferation, migration, and differentiation.This article reviews the structure, function and activity regulation of HIF-1 and its roles in acute ischemic brain injury.
7.Construction of bcr-abl gene eukaryotic expressing vector and its expression in COS-7 cells

Objective To clone and construct eukaryotic expressing vector of bcr-abl fusion gene and to express the gene in the mammal COS-7 cell lines. Methods bcr-abl fusion gene was amplified from human chronic myeloid leukemia (CML) K562 cell lines by RT-PCR and the fragment of cDNA was retrieved,purified and cloned into the pEGFP-N3 eukaryotic expressing vector. After the selection of the positive clone and by restriction enzyme analysis and DNA sequencing, the correct plasmid was transfected into COS-7 cell lines and observed the transient expression. Results A 874 bp DNA fragment was amplified by RT-PCR. The sequence analysis showed it was consistent with bcr-abl gene of GeneBank. RT-PCR, Western blotting analysis provided strong evidences that bcr-abl gene was expressed successfully in transfected COS-7 cells.Conclusion The eukaryotic expressing vector of bcr-abl fusion gene was constructed, it will lay the foundation for further study of bcr-abl gene in the diagnosis and treatment of CML.

And to reconstruct the hospital integrated operations management platform included all aspects, such as design ideas, technical architecture, software function and so on, from whole management requirement point.Results: This platform could provide the improvement solution of traceability management for medical material in hospital, and increase the efficiency and transparency of management, and improve the whole management level in hospital.Conclusion: The introduced integrated operations management platform has standardized high-value materials management, met the fine management and cost requirements, and greatly improved the management level of medical materials.
9.Changes of serum level of interleukin-6 and interleukin-10 after coronary stent implanation

Objective To investigate the changes of the serum level of IL-6 and IL-10 as well as their significance after coronary artery stent implantation. Methods The study involved 30 patients who underwent coronary artery stent implantation. Blood stamples were taken from the femoral arteries before and 24h after successful coronary stenting. Serum concentration of interleukin-6 (IL-6) and interleukin-10 (IL-10) were measured and complications as well as the occurance of restenosis were observed after the procedure. 10.Correlation between characteristics of intracranial lesions and level of cognitive function in patients with an initial diagnosis of brain metastases

Objective To investigate the correlation between the characteristics of intracranial lesions and the level of cognitive function in patients with an initial diagnosis of brain metastases.Methods A retrospective analysis was performed in 51 patients with an initial diagnosis of brain metastases who were admitted to The Second Hospital Affiliated to Suzhou University from January 2015 to April 2016.CT and (or) MRI were used to determine the characteristics of intracranial lesions and the Montreal Cognitive Assessment was used to evaluate the cognitive function of patients.Comparison between groups was made by Mann-Whitney U test.The correlation between ranked data was analyzed by Spearman rank correlation test.Results Of the 51 patients with an initial diagnosis of brain metastases,47(92%) had cognitive impairment,including mild cognitive impairment in 31(61%) and dementia in 16(31%).There was no significant difference in level of cognitive function between the patients with involvement of the left hemisphere alone and those with involvement of the right hemisphere alone (P=0.425).The patients with involvement of both hemispheres had a significantly lower level of cognitive function than those with involvement of the left hemisphere alone (P=0.042).The patients with involvement of three or more brain lobes had a significantly lower level of cognitive function than those with involvement of one or two brain lobes (P=0.015,0.024).The intracranial lesion volume and edema volume had no significant effect on the overall cognitive function of patients (P=0.077,0.178).The patients with>3 intracranial lesions had a significantly lower level of cognitive function than those with 1-3 intracranial lesions (P=0.010).Conclusions More than 90% of patients with an initial diagnosis of brain metastases have cognitive impairment.Cognitive impairment is mainly associated with lesion site,involvement of brain lobes,and number of lesions,but not with lesion volume and edema volume.
