1.Determination of Related Substance in Levo-cetirizine Hydrochloride Chewable Tablet by RP-HPLC

OBJECTIVE:To establish a method for the determination of related substances in Levo-cetirizine hydrochloride chewable tablet. METHODS:RP-HPLC was performed on the column of Venusil XBP-CN with mobile phase of acetonitrile-0.05 mol/L Sodium dihydrogen phosphate solution(pH value adjusted to 3.0 by phosphoric acid)(60∶40,V/V)at a flow rate of 1.0 ml/min, the detection wavelength was 230 nm,column temperature was 25 ℃,and volume injection was 20 μl. RESULTS:Levo-cetirizine hydrochloride chewable tablet showed good separation with related substances;the linear range of levo-cetirizine hydrochloride was 1.2-2.8 μg/ml(r=0.999 9);the quantification limit and detection limit was 1.4 ng/ml and 0.3 ng/ml,respectively;RSDs of preci-sion,stability and reproducibility tests were lower than 1%;recovery was 96.3%-105.0%(RSD=1.7%,n=9);the contents of re-lated substances were no more than 0.17%. CONCLUSIONS:The method is simple,sensitive,reproducible,accurate and reli-able,and can be used for the determination of related substances in Levo-cetirizine hydrochloride chewable tablet.
2.Integrin alpha v regulates anchorage-independent growth and survival in squamous cell carcinoma cells

Purpose:During the malignant transformation, most tuomr cells will become resistant to anoikis (the apoptosis induction upon loss of anchorage), thereby gaining an important growth advantage and providing the basis for tumor spread, growth at distant places, and formation of metastasis.The present study focus on the role of soluble fibronectin, integrin and their signal transduction in the process of tumor cells anoikis apoptosis.Methods:Flow cytometry detected integrin subunits' expression on the human squamous cell carcinoma cell (HSC 3) surface. Establish anoikis induction model. The role of different integrin subunits during anoikis induction were detected by antibody blocking method, p53 and Bcl 2/Bax expression were measured during this anoikis process.Results:There is no or low expression of integrin av on the normal epithelial cells, whilst it expresses relative higher on the surface of HSC 3 cells. The treatment of soluble fibronectin decreased p53 expression, increased the ratio of Bcl 2/Bax and then protected HSC 3 cell from anoikis. Anti integrin ?v blocking antibody resumed the anoikis that was inhibited by soluble fibronectin evidently. Conclusions:We now provide the evidence that the binding of integrin ?v and soluble fibronectin protected HSC 3 cells from anoikis apoptosis, via decreasing the expression of p53, increase the ratio of Bcl 2/Bax.
3.Operating Room Management Condition and Protocol at Township Hospitals

OBJECTIVE To understand operating room management conditions and put forward improvement methods at township hospitals.METHODS An observatory study was conducted in 9 township hospitals using the evaluation criteria based on the requirements of the Guidelines for Disinfection in Hospitals.Technical Guidelines for Prevention and Control of the Hospital Infection in Operation Departments and the Guidelines for Hand Hygiene of Medical staff in Medical Institutions.RESULTS Problems were found to a different extent in all of the 9 township hospitals:faults in rules and regulations,unreasonable layout technological process,unstandardized sterilization and disinfection and short knowledge about hospital infection.CONCLUSIONS Special attention to managment of operating room,improving consciousness,strengthening medical staff study and improving system are the key issues for improving the management level of operating room at township hospitals.
4.Evaluation of anticancer efficacy of dihydromyricetin in gastric cancer cells SGC7901

Objective To investigate the anticancer efficacy of dihydromyricetin (DMY)in gastric cancer cells SGC7901. Methods The DMY was puritied using RP-HPLC was almost 98%.The cell viability was assessed by CCK-8 Assay.The cell ap-optosis was assessed by flow cytometry.Using ultrasonic extraction with neutral water and reversed-phase high-performance liquid chromatography,DMY was purified from rattan tea.Results By using RP-HPLC,the purified DMY is almost 99%.DMY inhibi-ted the growth of tumor cells in the low concentration(≤ 1 mmol)with a dose-dependent manner(P <0.05 ).Further study on DMY showed that it induced apoptosis in SGC7901 cell line with a dose-dependent manner.Conclusion DMY is a potential thera-peutic agent for gastric cancer.
5.Disinfection and Isolation in Basic Medical and Health Institutions:Current Situation Issues and Countermeasures

OBJECTIVE To develop the countermeasures how to strengthen disinfection and isolation in the basic(medical) and health institutions.METHODS Through investigation,to analyze the current situation and problem of disinfection and isolation in the basic medical and health institutions.RESULTS There were some problems of the(disinfection) and isolation in the basic medical and health institutions and some possibility for patient catching(nosocomial) infection.CONCLUSIONS It is very important for strictly observing Medical Instrument Surveillance and Management Regulations published by the State Council and Hospital Infection Management Standards,Disinfection Technology Standards and Disinfection Management Methods published by Ministry of Health,and strengthening the management of disinfection and isolation in the basic medical and health institutions.
6.Analysis and discrimination on incorrect cases in medical statistics teaching

7.The effects of paired associative stimulation on motor cortex excitability and the recovery of motor function after stroke

Objective To investigate the effects of paired associative stimulation (PAS) on the recovery of upper limb motor function in stroke patients,and to analyze the relationship between the change of motor cortex excitability in the contralesional hemisphere and the recovery of motor function in the affected upper limb.Methods Thirty hemiplegic stroke patients were divided randomly into a treatment group and a control group.Both groups were given routine rehabilitation therapy,but the treatment group also received PAS consisting of transcranial magnetic stimulation (TMS) of the intact motor cortex and electrical stimulation (ES) of the median nerve at the wrist of the intact arm with an interval of 10 ms between the TMS and ES (PAS10).The PAS10 was delivered at a frequency of 0.05 Hz and an intensity of 120% the resting motor threshold (RMT),once daily for 30 minutes,five times a week for 4 weeks.Corticospinal excitability was measured using motor evoked potentials (MEP) and the RMT.The FuglMeyer upper limb assessment (FMA),Brunnstrom staging and the modified Barthel index (MBI) were also applied before and at the end of the 4 weeks of treatment.Correlation was sought between any changes in MEP amplitude,the RMT of the contralesional hemisphere and changes in the FMA results.Results Before the intervention there were no significant differences between the two groups in terms of any of the assessments.After 4 weeks of treatment,all the assessments had changed significantly compared to those before the treatment,but there were still no significant differences between the two groups in terms of any the assessments.After 4 weeks of treatment,the differences in MEP amplitude from the contra-lesional hemisphere and the differences in FMA scores were positively and significantly correlated with a correlation coefficient of r =0.431.The lesioned hemisphere was also positively correlated with the differences in FMA scores with a significant correlation coefficient of r =0.608.Conclusion PAS10 can facilitate the recovery of upper limb motor function.The change in motor cortex excitability of the contra-lesional hemisphere significantly correlates with functional recovery in the upper limb.
8.Effects of paired associative stimulation and repetitive transcranial magnetic stimulation on the excitability of the motor cortex

Objective To compare the effects of paired associative stimulation (PAS) and repetitive transcranial magnetic stimulation (rTMS) on motor cortex excitability.Methods The baseline corticospinal excitability of the left hemispheres of 10 healthy subjects was measured in terms of resting motor threshold (RMT) and other indicators of motor evoked potentials (MEP).On the following day they received PAS composed of trascranial magnetic stimulation (TMS) to the motor cortex of the left hemisphere and electric stimulation (ES) of the median nerve contralateral to the motor cortex,with an interval of 10ms between the TMS and ES (termed PAS10).The PAS10 was delivered at a frequency of 0.05 Hz and an intensity of 120% of the RMT,for a total of 90 pulses.The MEP amplitude,MEP latency and RMT were evaluated one minute after the stimulation.After the PAS intervention,an interval of one week was allowed to eliminate any effect of PAS on motor cortex excitability.Then rTMS was delivered to the subjects' left motor cortex at the same time of day at a frequency of 1 Hz and an intensity of 120% of the RMT,for a total of 1000 pulses.MEP amplitude,MEP latency and RMT were evaluated one minute after the stimulation.The two interventions were compared in terms of MEP amplitude,MEP latency and RMT.Results The average MEP amplitude,MEP latency and RMT at baseline were (2.93 ± 0.99) mV,(20.97 ± 1.67) ms,and (46.06 ±5.32) %,respectively.One minute after PAS10,the MEP amplitude,MEP latency and RMT were (1.14 ± 0.76) mV,(21.87 ± 1.09) ms and (52.06 ±4.20) %,respectively.One minute after rTMS,the MEP amplitude and latency and the RMT were (2.24 ± 0.79) mV,(20.88 ± 1.94) ms,and (49.00 ± 4.54) %,respectively.The differences in MEP amplitude,MEP latency and RMT pre-and post-intervention were (0.69 ± 0.10) mV,(0.09 ±0.05) ms and (3.94 ± 0.93) %,respectively for rTMS.For PAS10 they were (1.83 ± 0.14) mV,(0.90 ± 0.26)ms and (6.00 ± 1.13)%,respectively.The differences in MEP amplitude decrease and MEP latency lengthening between the two stimulation protocols were significant,but the difference in RMT elevation was not.Conclusion Both PAS10 and low frequency rTMS suppressed motor cortex excitability,but the suppressive effect of PAS10 is more significant.
9.Tissue speciality of endothelium-dependent vascular relaxation mediated by EPA and its relationship with pathophysiological property

Objective:To study the tissue speciality of endothelium-dependent vascular relaxation mediated by endothelial protein activated by acetylcholine (EPA)and its relationship with atherosclerosis.Methods:The tension of the isolated artery preparations was recorded using isometric-tension method and the isolated endothelium-intact vessels derived from normal and hyperlipidemic animal artery.Results:The EPA-mediated endothelium-dependent vascular relaxation was exhibited in rabbit aorta, renal, femoral, carotid, pulmonary arteries and cat aorta, renal, femoral, mesentery, coronary arteries but with different EC50. The sensitivity of EPA to ACh in descending order: pulmonary>renal>aorta>femoral>carotid arteries in rabbits. In cats,coronary>aorta>femoral>mesentary>renal. The endothelium-dependent vascular relaxation mediated by EPA in aorta, carotid and pulmonary arteries decreased significantly in hyperlipidemia rabbits.Conclusions:EPA effect existed wildly in testing artery and was of different tissue sensitivity. The decrease in EPA potency was closely related to occurence of atheroclerosis.
10.Preparation of fluorescent polyclonal antibody against human IgG and its application in the diagnosis of nephrosis

Objective:To prepare polyclonal antibody against human immunoglobulin G(human IgG) and to use this antibody in the clinical diagnosis of nephrosis. Methods:New Zealand rabbit was immunized subcutaneously with human IgG.Enzyme linked immunosorbent assay(ELISA) was applied to reveal the titer of the prepared polyclonal antiserum against human IgG.Antiserum was purified with affinity chromatography,and the purified antibody was confirmed for its specificity by Western blot and immunohistochemical staining.The purified antbodies which have been indentified were fused with FITC(fluorescein isothiocyanate) and then analyzed by immunohistochemical staining.Finally the fused antibody was useed in the clinical diagnosis of nephrosis. Results: The titer of the obtained antiserum was up to(1∶128 000,) double agar diffusion test showed the titer of the antibody was 1∶32.By the method of affinity chromatography,we obtained purified antibody with the purity of 85%.ELISA,double agar diffusion and immunohistochemical staining tests showed that the specificity and titer of the antibody were not decreased sharply.The purified antibody fused with FITC also kept the specificity of the primitive antibody.When the FITC fused antibody was tried in nephrosis patients,it detected human IgG effectively. Conclusion:The polyclonal antibody can specifically recognize human IgG.This purified antibody fused with FITC can be used in the diagnosis of nephrosis.
