3.Role of RIP140 and PGC-1αin angiotensin Ⅱ mediated energy metabolism in cardiomyocytes

Aim To investigate the role of transcrip-tional cofactors receptor interacting protein 140 ( RIP140 ) and peroxisome proliferator-activated recep-tor γ coactivator-1α ( PGC-1α) in the angiotensin Ⅱ( AngⅡ) mediated energy metabolism regulation in cardiomyocytes. Methods RIP140 or PGC-1α was overexpressed via adenovirus vector system. ATP con-tents were detected by luminescence detection assay system. Real-time PCR and Western blot analysis were used to measure the expressions of RIP140 and PGC-1α genes. Results After treated with 100 nmol·L-1 AngⅡfor 36h in neonatal cardiomyocytes, the content of mitochondrial ATP was reduced notably ( P <0. 01). Accordingly, the mRNA and protein levels for RIP140 were increased. However, the mRNA and pro-tein levels of PGC-1α were downregulated markedly. What’ s more,the reduction of ATP induced by AngⅡwas further aggravated by RIP 1 4 0 overexpression , but ameliorated by overexpressing PGC-1α. Conclusion The reduction of ATP mediated by AngⅡis associated with the upregulation of RIP140 , as well as the down-regulation of PGC-1α.
4.PREPARATION OF MONOCLONAL ANTIBODIES TO HUMAN TYPE Ⅲ COLLAGEN AND THEIR LOCALIZATION IN NORMAL TISSUES

Monoclonal antibodies, CAb-15 and CAb-16, were produced by using of spleen cells from Balb/c mice immunized with cell suspension of fresh hepatoma tissues with mouse myeloma Sp 2/0 cells. Western blotting showed that the antigen determinant of CAb-15 was situated on the ?_1 [Ⅲ] polypeptide chains and that of CAb-16 on the triple helical domain. The monoclonal antibodies were also used for immunohistochemical localization by ABC method in formalin-fixed and paraffin-embedded sections of normal tissues from liver, kidney, aorta, skin, cartilage, placenta, stomach, breast and granulation tissues. The results showed that these antibodies are useful in immunohistochemical study for collagen.
5.The challenges of forensic expert accepting cross-examination in trial-centered trial mode

Today, the direction of litigation reformation is changing trial pattern to be trial-centered and making the trial substantialized. Under this background, the relevant provisions have been launched from the supreme people's court and other departments to make sure that the forensic expert will appear in court and accept cross-examination, and this phenomenon should be normalized and substantiated. In order to ensure the expert opinion can be fully and effectively cross-examined, the medico-legal expert should fulfill obligations, such as respect for the court, answer questions honestly and explain expert opinions. We find that some problems show up in this process: the rate of forensic expert appearing in court is low; the legal nature of expert opinion is not clear; the requirements are not clear;the conflict between forensic expert opinion and other forms of expert evidence, including auxiliary expert, is very clearly. Based on this situation, the author give some suggestions to the forensic expert: keep on learning to improve professional skill; improve the legal knowledge; pay attention to practice and strengthen the ability of expression.
6.Clinical investigation and drug resistance of fungal hospital-acquired infection in a certain hoapital

Objective To explore the distribution,fundamental diseases,risk factors and drug-resistance of hospital- acquired infection caused by fungi in Department of Respiratory Medicine.Methods The clinical data of 142 patients were retrospectively reviewed,and then with statistical analysis was performed.Results The main pathogen which caused fungal hospital-acquired infection was Candida albicans. The most three possible fundamental diseases were chronic obstructive pulmonary disease,bronchiectasis and lung cancer. The main risk factors were lower resistibility of patients and invasive diagnostic and therapeutic measure. The fungus resistance was gradually rising.Conclusion The hospital infection caused by fungus should be emphasized in respiratory medicine department. In order to effectively prevent the hospital infection in respiratory medicine department,we should improve the basic conditions of patients,rationally diagnose and treat,and rationally use the drug against fungi.
7.Viral structural proteins are associated specifically with a set of Heat shock proteins including HSP70, GRP94 and HSP27 in the brain tissues of suckling mice experimentally infected with Hantaan virus

Objective To study Heat shock proteins (HSPs) induction and their association with Hantaan virus (HTNV) structural proteins in the brain tissues of mice experimentally infected with HTNV 76~118. Methods Newborn mice less than 72 h of age were experimentally infected with HTNV by intracerebral inoculation. The brains of mice at the 8th day post infection were removed and prepared for tissue extracts. The possible interaction or association of Hantaan virus nucleocapsid protein (HTNV NP) and glycoprotein G2 (HTNV G2) with GRP94, HSP70 and HSP27 were analyzed by double specific antibo dies sandwich ELISA and immunoprecipitation methods. Results HTNV infection induced the expression a set of HSPs including GRP94 and HSP70 in the brain tissues of mice experimentally infected with HTNV. HTNV NP was associated with GRP94, HSP70 and HSP27, which led to formation of HTNV NP HSP70 GRP94 HSP27 complexes. It was also observed that HTNV G2 was associated with NP and HSP27, which led to formation of HTNV G2 NP HSP27 complexes. Conclusions HTNV infection induces expression of a set of HSPs, which were associate with viral structural proteins in the brain tissues of mice experimentally infected with HTNV. This association may demonstrate the chaperone roles of HSPs in the synthesis, transportation and maturity of viral proteins during viral replication in the host cells.
8.Advanced research on neoadjuvant therapy with trastuzumab in HER2-positive breast cancer

Trastuzumab is a specific inhibitor against human epidermal growth factor receptor-2 (HER-2). Trastuzumab is widely used in the neo-adjuvant treatment of HER-2 breast cancer. Large-scale randomized and controlled clinical trials have demonstrated that pathologic complete response rates (pCRs) were significantly increased with neo-adjuvant trastuzumab therapy plus chemotherapy than with regular chemotherapy. The use of trastuzumab plus chemotherapy with lapatinib supplements could further improve pCR rates. An-thracycline and non-anthracycline drugs could both be used concurrently with trastuzumab. Endocrine therapy could be used as an alter-native for estrogen receptor-positive patients. pCR is a powerful predictor of long-term outcomes in HER-2 positive patients under neo-adjuvant therapy with trastuzumab. However, patient loss of HER-2 expression with residual disease after neo-adjuvant therapy with trastuzumab is a poor prognostic factor. This study paper will provide a review of related research.
9.Effect of siRNA on c-myc protein in HL-60 cells

Objective To study the effect of c-myc small interference RNA(siRNA) on c-mye protein in leukemic cells, and investigate a new method of gene therapy on leukemia. Methods siRNA against c-myc mRNA were designed and synthesized. The siRNA at 100 nmol/ml density was transfected into HL-60 cells. The cells cultured at 24 h, 48 h or 72 h were collected after being transfected, and the total protein of the cells was extracted according to the instruction booklet of TRIzol reagent. The expression level of c-myc protein was examined by Western blotting. Results After being treated with siRNA of c-myc, the quantity of c-myc protein was obviously reduced. It was remarkably lower than that of the control group. Furthermore after 72 h treatment with siRNA, the expression of c-myc protein reached its lowest. Conclusion The siRNA of c-myc can suppress the expression of c-myc protein in HL-60 cells. There are sequence targeted and time-dependence effect. The siRNA of c-myc may become a new tool of gene therapy on leukemia.
10.Efficacy of skin vascular diseases treated with 595 nm pulsed dye laser

Objective To evaluate the efficacy and side-effects of 595 nm pulsed dye laser (PDL) in the treatment of cutaneous vascular diseases.Methods We analyzed 572 cases of cutaneous vascular diseases treated with 595 nm PDL retrospectively and compared the efficiency among different type of diseases.Results There were totally 243 cases of infantile hemangioma,186 cases of port wine stain,69 cases of telangiectasis,42 cases of spider angioma and 32 cases of rhinophyma.The efficiency was 93.8%,54.3%,95.7%,100% and 56.3%,respectively,with 2.27% of side-effect rate.The efficacy was correlated to age,type and location of the diseases.Conclusions 595 nm PDL is a safe option for cutaneous vascular diseases with high efficacy and low side-effect rate.
