2.Role of RIP140 and PGC-1αin angiotensin Ⅱ mediated energy metabolism in cardiomyocytes

Aim To investigate the role of transcrip-tional cofactors receptor interacting protein 140 ( RIP140 ) and peroxisome proliferator-activated recep-tor γ coactivator-1α ( PGC-1α) in the angiotensin Ⅱ( AngⅡ) mediated energy metabolism regulation in cardiomyocytes. Methods RIP140 or PGC-1α was overexpressed via adenovirus vector system. ATP con-tents were detected by luminescence detection assay system. Real-time PCR and Western blot analysis were used to measure the expressions of RIP140 and PGC-1α genes. Results After treated with 100 nmol·L-1 AngⅡfor 36h in neonatal cardiomyocytes, the content of mitochondrial ATP was reduced notably ( P <0. 01). Accordingly, the mRNA and protein levels for RIP140 were increased. However, the mRNA and pro-tein levels of PGC-1α were downregulated markedly. What’ s more,the reduction of ATP induced by AngⅡwas further aggravated by RIP 1 4 0 overexpression , but ameliorated by overexpressing PGC-1α. Conclusion The reduction of ATP mediated by AngⅡis associated with the upregulation of RIP140 , as well as the down-regulation of PGC-1α.
3.The challenges of forensic expert accepting cross-examination in trial-centered trial mode

Today, the direction of litigation reformation is changing trial pattern to be trial-centered and making the trial substantialized. Under this background, the relevant provisions have been launched from the supreme people's court and other departments to make sure that the forensic expert will appear in court and accept cross-examination, and this phenomenon should be normalized and substantiated. In order to ensure the expert opinion can be fully and effectively cross-examined, the medico-legal expert should fulfill obligations, such as respect for the court, answer questions honestly and explain expert opinions. We find that some problems show up in this process: the rate of forensic expert appearing in court is low; the legal nature of expert opinion is not clear; the requirements are not clear;the conflict between forensic expert opinion and other forms of expert evidence, including auxiliary expert, is very clearly. Based on this situation, the author give some suggestions to the forensic expert: keep on learning to improve professional skill; improve the legal knowledge; pay attention to practice and strengthen the ability of expression.
5.PREPARATION OF MONOCLONAL ANTIBODIES TO HUMAN TYPE Ⅲ COLLAGEN AND THEIR LOCALIZATION IN NORMAL TISSUES

Monoclonal antibodies, CAb-15 and CAb-16, were produced by using of spleen cells from Balb/c mice immunized with cell suspension of fresh hepatoma tissues with mouse myeloma Sp 2/0 cells. Western blotting showed that the antigen determinant of CAb-15 was situated on the ?_1 [Ⅲ] polypeptide chains and that of CAb-16 on the triple helical domain. The monoclonal antibodies were also used for immunohistochemical localization by ABC method in formalin-fixed and paraffin-embedded sections of normal tissues from liver, kidney, aorta, skin, cartilage, placenta, stomach, breast and granulation tissues. The results showed that these antibodies are useful in immunohistochemical study for collagen.
6.The clinical analysis of 27 cases of polycythemia vera complicating stroke

Objective To explore the clinical features and treatment of polycythemia vera(PV)complicating stroke.Methods Clinical data of 27 cases of PV complicating stroke were analysed retrospectively.Results(1)Among all the cases,PV complicating cerebral infarction were 22 cases(81.5%),among the 22 cases,18 cases(66.7%)of multiple cerebral infarction,2 cases(7.4%)of transient ischemic attack,2 cases(7.4%)of cerebral hemorrhage,1 case(3.7%)of subarachnoid hemorrhage(SAH).The patients's clinical manifestation,physical sign,hemogram,marrow characteristic were consistent with that of PV diagnosis criteria.(2)Imageology check show that small infarction focus were common(66.7%).Lobe of brain,basal ganglia,capsula interna were common place of cerebral infarction.The density of hemorrhagic focus was asymmetrical,there was obvious edema around the hemorrhagic focus.(3)Applied vein blood-letting and small dose chemotherapy,8 cases were recovery,18 cases were obviously improved,1 case of stroke was invalid.Conclusion The cerebral infarction are common in PV complicating stroke.Among all the PV complicating stroke cases,multiple cerebral infarction are the most common,cerebral hemorrhage and subarachnoid hemorrhage are few,vein blood-letting and small dose chemotherapy has a good curative effect for the patients.
8.Clinical investigation and drug resistance of fungal hospital-acquired infection in a certain hoapital

Objective To explore the distribution,fundamental diseases,risk factors and drug-resistance of hospital- acquired infection caused by fungi in Department of Respiratory Medicine.Methods The clinical data of 142 patients were retrospectively reviewed,and then with statistical analysis was performed.Results The main pathogen which caused fungal hospital-acquired infection was Candida albicans. The most three possible fundamental diseases were chronic obstructive pulmonary disease,bronchiectasis and lung cancer. The main risk factors were lower resistibility of patients and invasive diagnostic and therapeutic measure. The fungus resistance was gradually rising.Conclusion The hospital infection caused by fungus should be emphasized in respiratory medicine department. In order to effectively prevent the hospital infection in respiratory medicine department,we should improve the basic conditions of patients,rationally diagnose and treat,and rationally use the drug against fungi.
9.Construction of RIP140 recombinant adenovirus and its expression in neonatal rat cardiomyocytes

Aim The limited transfection efficiency for plasmid in primary neonatal rat cardiomyocytes,which are terminal differentiated cells,and long foreign DNA (the RIP140 gene sequence are as long as 3.5 kb) cause us to choose a better system to study RIP140 gene expression in primary non-replicative cells. Methods Full-length of RIP140 was cloned into pAdTracker-CMV shuttle vector,and then recombined with virus backbone pAdEasy-1 vector in BJ5183 bac-teria.Positive recombinant plasmid was confirmed by sequence analysis and restriction enzyme determina-tion,and then transfected into AD293 cells for amplifi-cation.Titers of virus particles were determined by Tis-sue Culture Infectious Dose 50 (TCID50 )method and cell vitality was analyzed by CCK-8 kit in cardiomyo-cytes.RIP140 gene was identified by Western blot. Results Sequence analysis suggested that full-length RIP140 gene was cloned correctly into AdEasyTM sys-tem.Virus titers of Ad-RIP140 and Ad-GFP were 1011.3 and 1011.7 PFU·mL-1 ,respectively.Cell vitali-ty was not affected when the Multiplicity of Infection (MOI)was lower than 200.Green fluorescent protein (GFP)and Western blot analysis showed RIP140 gene was remarkably increased in cardiomyocytes for 12h in-fection by Ad-RIP140 (P<0.05 ).Conclusion Re-combinant adenovirus containing RIP140 gene was suc-cessfully constructed and effectively expressed in car-diomyocytes.These will be helpful for further research on the function of RIP140 in cardiomyocytes.
10.On the interaction between cefonicid sodium and bovine serum albumin by spectrometry

Obiective To explore the interaction of cefonicid sodium(CS) with bovine serum albumin(BSA) by fluorescence and absorption spectroscopy. Methods The rate constant(Kq), quenching constant(Ksv), static fluo-rescence quenching association constant(KLB),binding site number(n) and binding constant(Kb) were calculated using Stern-Volmer, Lineweaver-Burk and double logarithm equations. Results CS was able to bind to BSA. The probable quenching mechanism of BSA by CS was mainly static quenching due to the formation of a CS-BSA com-plex. The results of thermodynamic parameters indicated that electrostatic force plays the main role in the binding process and the binding process was spontaneous. There was a single class of binding site for the BSA with CS. The primary binding site for CS was located at sub-domainⅡA of BSA and near by tyrosine residue. There was almost some negative cooperative effect. The results obtained from synchronous fluorescence showed that CS could change the microenvironment of Tyrand Trp residues of BSA. Conclusion The interaction between CS and BSA is dynam-ic. There is a single class of binding site for the BSA with CS. The obtained results provide references for its clini-cal application.
