OBJECTIVE:To establish a method for the determination of related substances in Levo-cetirizine hydrochloride chewable tablet. METHODS:RP-HPLC was performed on the column of Venusil XBP-CN with mobile phase of acetonitrile-0.05 mol/L Sodium dihydrogen phosphate solution(pH value adjusted to 3.0 by phosphoric acid)(60∶40,V/V)at a flow rate of 1.0 ml/min, the detection wavelength was 230 nm,column temperature was 25 ℃,and volume injection was 20 μl. RESULTS:Levo-cetirizine hydrochloride chewable tablet showed good separation with related substances;the linear range of levo-cetirizine hydrochloride was 1.2-2.8 μg/ml(r=0.999 9);the quantification limit and detection limit was 1.4 ng/ml and 0.3 ng/ml,respectively;RSDs of preci-sion,stability and reproducibility tests were lower than 1%;recovery was 96.3%-105.0%(RSD=1.7%,n=9);the contents of re-lated substances were no more than 0.17%. CONCLUSIONS:The method is simple,sensitive,reproducible,accurate and reli-able,and can be used for the determination of related substances in Levo-cetirizine hydrochloride chewable tablet.

Interferon-γ-inducible protein 10 (IP-10),also known as chemokine (C-X-C motif) ligand 10(CXCL10),belongs to the CXC subfamily.IP-10 exerts its function by binding to chemokine (C-X-C motif) receptor 3 or Toll-like receptor 4.IP-10 plays an important role in the occurrence and development of autoimmune diseases.The expression of IP-10 in blood and body fluid is closely associated with the activity of autoimmune disease.IP-10 is expected to become a potential new marker and possess a significant application value in early screening,condition monitoring and therapeutic evaluation of autoimmune diseases.

Adjuvant treatment, which includes chemotherapy and endocrine therapy, for early breast cancer may impair bone den-sity, resulting in bone loss. The third generation bisphosphonate-zoledronic is an anti-resorptive agent that inhibits osteoclast-mediated bone resorption. This drug can be mainly used in the treatment of hypercalcemia caused by bone metastases of the cancer. The Zome-ta-Femara Adjuvant Synergy Trial revealed that immediate zoledronic acid and endocrine therapy not only prevents bone loss, but also reduces recurrence. The Austrian Breast and Colorectal Cancer Study Group-12 (ABCSG-12) also confirmed that zoledronic acid, when combined with endocrine therapy, could reduce the risk of cancer disease progression and death. Moreover, preclinical studies and clini-cal trials have demonstrated the synergistic antitumor effects of chemotherapy and zoledronic acid. Neo-adjuvant zoledronic acid to re-duce recurrence trials showed that the addition of zoledronic acid to adjuvant chemotherapy significantly reduced the risk of disease pro-gression and death in postmenopausal women more than five years postmenopause at the beginning of the study or over 60 years of age at the baseline. The ABCSG-12 subgroup analysis based on age (≤40 years or>40 years) also showed that zoledronic acid can signifi-cantly improve the prognosis in women who were over 40 years at the study entry. These results suggest that zoledronic acid administra-tion in patients with lowered estrogen levels (naturally or as a consequence of adjuvant treatment) easily exerts anti-tumor effects. How-ever, the optimal dose and duration of zoledronic acid requires further studies. More clinical trials should be performed to provide suffi-cient evidence to support the effectiveness of zoledronic acid in the treatment of early breast cancer.

Objective To acknowledge the spouse comorbid conditions of diabetes patients by comparing the average blood sugar level of the spouse of the diabetes patients with normal Guangzhou residents.Methods To sift and analyze the spouse test results of 152 hospitalized diabetes patients from January 2012 to January 2013,by testing the blood sugar level of the experimental subjects,comparing the different blood sugar level results between when they were with empty stomach and 2 hours after oral glucose tolerance test (OGTT).Results There were 32 cases with diabetes,9 cases with impaired glucose tolerance (IGT) and 11 cases with impaired fasting glucose (IFG) in the above 152 experimental subjects.The morbidity rates were 21.05％,5.92％ and 7.24％.All the above results were higher than the Guangzhou residents survey level,there were statistical differences between them.Conclusions The high morbidity rates of diabetes and impaired glucose regulation in diabetes patients spouse are obvious.The works for diabetes sifting and analyzing together with the health education instruction need to be strengthened.

Trastuzumab is a specific inhibitor against human epidermal growth factor receptor-2 (HER-2). Trastuzumab is widely used in the neo-adjuvant treatment of HER-2 breast cancer. Large-scale randomized and controlled clinical trials have demonstrated that pathologic complete response rates (pCRs) were significantly increased with neo-adjuvant trastuzumab therapy plus chemotherapy than with regular chemotherapy. The use of trastuzumab plus chemotherapy with lapatinib supplements could further improve pCR rates. An-thracycline and non-anthracycline drugs could both be used concurrently with trastuzumab. Endocrine therapy could be used as an alter-native for estrogen receptor-positive patients. pCR is a powerful predictor of long-term outcomes in HER-2 positive patients under neo-adjuvant therapy with trastuzumab. However, patient loss of HER-2 expression with residual disease after neo-adjuvant therapy with trastuzumab is a poor prognostic factor. This study paper will provide a review of related research.

It has long been acknowledged that there is a link between inflammation and cancer, but its molecular mechanism remains unclear. A key player in inflammation is nuclear transcription factor NF-?B, that activity is triggered in response to infectious agents and proinflammatory cytokines via the I?B kinase. In parenchyma cells, inflammation through I?B kinase/NF-?B pathway suppresses apoptosis, accelerates cell cycle, then promote tumorigenesis. In mesenchyma cells inflammation through I?B kinase/NF-?B pathway produces cytokines and chemokines that may serve as tumor growth factors. To sum up, I?B kinase/NF -?B pathway represents a critical molecular link between inflammation and cancer.

Aim To examine the role of antioxidant effect of subhypotensive dose of ACE inhibitor ramipril in its anti hypertrophic effect in rat myocardium. METHODS Model of hypertension induced myocardial hypertrophy of rat was reproduced by coarctation of abdominal aorta. The effects of nitric oxide (NO) synthase inhibitor N L arginine (6 mg?kg -1 ?d -1 ) and Vitamin E(200 U?d -1 ) on cardiac hypertrophy were examined. The change of superoxide dismutase (SOD) and NO production (assayed as NO 2 -) were monitored. RESULTS The myocardial hypertrophy was effectively prevented under hypertensive condition after treatment with ramipril for 8 week, while SOD activity and NO 2 - content were significantly increased and lipid peroxidation was significantly attenuated. Vitamin E enhanced ramipril effect and N L arginine reversed its benficail effects. CONCLUSION These data showed that sub hypotensive dose of ramipril and in combination with vitamin E enhances myocardial antioxidative capability and NO release, which plays an important role in the mechanism of action involved in their antihypertensive cardiac hypertrophic effects.

Objective To investigate the protective effects of (Val8)GLP-1-Glu-PAL to dopaminergic neurons on PD mice induced by MPTP.Methods 56 male C57BL/6 mice which are 10-12 weeks old were randomly divided into Control,model (MPTP),muti-injection (Val8) and therapy (MPTP + Val8) groups by random number table method.Mice in MPTP group were received MPTP IP.treatment (30mg/kg · d),the control group were treated with 0.9％ saline with the same volume,Val8 group were injected with (Val8) GLP-1-Glu-PAL (25 nmol/kg),and MPTP+Val8 group were going to received (Val8)GLP-1-Glu-PAL 1 h after treatment of MPTP,all the groups were treated for 8 consecutive days.Behavior test were processed 2h after drug IP injection,including swimming test and rotarod test.Mice were sacrificed immediately 1h after the final trail of behavior test in 8th day,brains were withdraw for IHC assay which tested the number TH positive neurons in SNpc area.Results The classical PD behavior symptom were induced by MPTP,from 1 st to 8th,the swimming score(1.715±0.143 and dropping latency(68.048±7.823) were both decreasing compared with control.The differences were statistically significant (P＜0.05).The positive dopaminergic neurons were significantly lower in SNpc area (P＜0.01).Compared with MPTP group,(Val8) GLP-1-Glu-PAL could statistically improved the behavior deficit,the swimming score (1.120±0.143) and dropping latency(20.546±7.823) in the MPTP+Val8 group,and differences were statistically significant (P＜0.05).Meanwhile,the TH positive neurons were significantly elevated (P＜0.01).Conclusion The results suggest that (Val8)GLP-1-Glu-PAL has protective effects for MPTP induced mice PD dopaminergic neurons.

Objective To identify the clinical characteristics and prognostic factors in young women of breast cancer treated with breast conservation therapy.Methods Clinical data of 605 cases of breast cancer patients undergoing breast conserving therapy were reviewed in our hospital from Jan 1998 to July 2008.According to their age,these patients were divided into young group (age≤ 35 years,n =110 cases) and the elderly group (age ＞ 35 years,n =495 cases).Survival factors were analyzed by Cox proportional hazards regression model.Results During the median follow-up of 61 months,the 5-year local-regional relapse-free survival was 92.6％ and 96.8％,respectively (P =0.074) ; the 5-year distant metastasis-free survival and overall survival between the two groups were (89.7％ vs.95.2％,P =0.001)and (96.4％ vs.98.9％,P =0.046).Multivariate analysis revealed that lymph node metastasis and adjuvant radiotherapy were independent risk factors for local-regional relapse.Distant metastasis and overall survival were determined by lymph node metastasis,adjuvant radiotherapy and age.Conclusions Age is not an independent factors affecting tumor local recurrence in Chinese young female patients of early breast cancer receiving breast conserving surgical therapy.

Objective To investigate the diagnostic value of determination of the genotypes in codon 12 and 13 of K-ras oncogene in blood samples of patients with pancreatic carcinoma(PC).Methods Blood samples were obtained from 54 patients with pathologically confirmed PC,and 33 healthy controls.The DNAs were obtained in these samples.and then genotype of K-ras mutation was detected by using the PNA-clamping real-time quantitative PCR.Then the correlation between the K-ras genotypes of blood DNA and the clinical characteristics was analyzed.Results K-ras mutations were found in 74.1%(40/54)of patients with PC.There was no such mutation in control samples.The mutations of K-ras was associated with age,lymph node and vessel invasion.poorly differentiated tumor,CA19-9,while it was not associated with sex,tumor location,size of tumor,clinical staging and pathological type.Conclusions The one-step method was highly sensitive for detecting K-ras mutation in blood samples.Detection of circular blood cells harboring K-ras mutation suggested the tumor was highly invasive with poor prognosis.
