1.Protective Effect of Yiqimingmu Oral Solution on Rats' Retina with Photochemical Damage

OBJECTIVE:To observe the protective effect of the traditional Chinese medicine Yiqimingmu oral solution on photochemical damage of retina in rats. METHODS:SD rats were divided into four groups randomly:negative control group, model group, low dose Yiqimingmu oral solution group and high dose Yiqimingmu oral solution group. All the groups except the control one were continually exposed to Lux (1 900?106.9) green fluorescent light for 24 hours to establish retinal photochemical damage model. The low dose and high dose groups were administered intragastrically with Yiqimingmu oral solution (15mL?kg-1?d-1 and 30mL?kg-1?d-1) 7 days before light expose. Normal saline solution was administered intragastrically in control and model group. At 6 hours and 6, 14 days after light expose, the content of malandialdehyde (MDA) and activity of SOD in rats' retina were measured. The histopathologic changes were observed by transmission electron microscopy and light microscopy. 2.Medication Analysis of 564 Cases with Cataract Extraction

OBJECTIVE:To get to know the preoperative and postoperative medication of patients with cataract.METHODS:The preoperative and postoperative medication of 564 cases with cataract extraction in our hospital from September 2004 to October 2004 was analyzed statistically.RESULTS:Antibacterial eye gutta was mainly used before cataract extraction while compound eye gutta containing antiseptic drugs and glucocorticosteroid were chiefly used after cataract operation.CONCLUSION:The preoperative and postoperative medication for patients with cataract in our hospital was reasonable.
3.Progress in studies of viperin, an important cellular antiviral factor.

Viperin is an endoplasmic reticulum (ER)-associated protein that has been identified as an innate antiviral protein. Viperin expression can be largely upregulated by viruses, interferons, and oligonucleotides such as poly I:C and lipopolysaccharides. Viperin inhibits viral replication by interactiing with host cell proteins and several viral proteins, and disrupting the cell membrane system. It shows a broad-spectrum of antiviral activity. Some viruses have developed activities that counteract the action of viperin during a long- term period of evolution with hosts by impairing viperin expression. In addition to its antiviral effects, viperin has several other biological functions. This article review the basic characteristics of viperin and the state of current research into its antiviral effects, demonstrating the rapid progress that has been made in this field.

4.Content Determination and Stability of Ciclosporin Ophthalmic Emulsion

OBJECTIVE:To establish the method for the content determination of Ciclosporin ophthalmic emulsion and to probe into its stability.METHODS:HPLC method was used to determine the content of ciclosporin.The separation was performed on Eclipse XDB-C8 column.The mobile phase consisted of acetonitrile-methanol-water(67.5 :5 :27.5) with the flow rate of 0.8 mL?min-1.The detection wavelength was set at 210 nm and column temperature was 55 ℃.Its stability was studied by classic isothermal accelerated test and highlight struck test.RESULTS:The linear range of ciclosporin was 5~60 mg?L-1(r=0.999 9) with average recovery of 100.39%(RSD=1.15%).The preparation was stable under heat and light,and its shelf life at 25 ℃ was predicted about 6.88 years.CONCLUSIONS:The method is accurate and feasible for the content determination of Ciclosporin ophthalmic emulsion.The preparation can be stored at room temperature.
5.Stimulation Effect of Several Bacterial Inhibitors of Eye Drops on Rabbits Eyes

OBJECTIVE: To investigate the stimulation effect of several bacterial inhibitors of eye drops on eyes.METHODS: Stimulation effect of several bacterial inhibitors on rabbits eyes were observed with the contrast on the same body.The scores were counted according to the score standard of eyes stimulation response in order to evaluate the stimulation effect of bacterial inhibitors.RESULTS: After giving single dose of inhibitors,the average scores of stimulation reaction induced by 0.002% thiomersal,0.01% benzalkonium chloride,0.01% benzalkonium bromide,0.01% chlorhexidin acetas,0.5% chlorobutanol,0.03% ethylparaben,2% boric acid were 0.20,0.32,0.36,0.60,1.20,3.88,6.32,mild stimulation reaction was induced by 2% boric acid and 0.03% ethylparaben and others inhibitors caused no stimulation response.After giving multiple doses of inhibitors,the average scores of above inhibitors were 0.49,0.79,0.84,3.83,3.91,6.51,10.84,the medium stimulation reaction was induced by 2% boric acid,mild stimulation reaction were induced by 0.03% ethylparaben,0.01% chlorhexidin acetas and 0.5% chlorobutanol;0.002% thiomersal,0.01% benzalkonium chloride,0.01% benzalkonium bromide caused no stimulation reaction.CONCLUSION: The best inhibitors of eye drops are thiomersal,benzalkonium chloride,benzalkonium bromide,followed by chlorhexidin acetas,ethylparaben,chlorobutanol.
6.Preparation and quality evaluation of ginkgolide B-loaded self microemulsifying drug delivery system.

OBJECTIVETo prepare ginkgolide B-loaded self microemulsifying drug delivery system (SMEDDS) and evaluate its quality.
METHODThe solubility of ginkgolide B in different oil, surfactant and co-surfactant were measured by HPLC-ESI-MS. The GB-SMEDDS formulation was optimized by the self emulsifying efficiency of various combinations of oil and mix-surfactant evaluated by using pseudo-temary phase diagram. The preliminary stability of GB-SEMEDDS was evaluated by the variety of loading rate of GB and dispersed medium. The morphology, the particle size and the formulation stability were evaluated after diluting by 0.1 mol x L(-1) HCl.
RESULTThe blank self microemulsified system was composed of ethyl oleate-( cremophor EL-lecithin-ethanol, 4: 1:2) (40: 60), the loading dosage was 2.5%. Little influence of GB and emulsified medium was observed on the stability of GB-SEMDDS. After diluted with 0.1 mol x L(-1) HCl, the morphology of the microemulsion was homogeneous small spherical drops observed under electro-microscope. The particle size was (41.6 +/- 1.11) nm, the self microemulsifing time was around 2 min. The formulation was stable within 8 h, without significant changes in particle size and separation of drugs.
CONCLUSIONGB-SMEDDS is easy to prepare and its quality is stable. The solubility of GB was significantly improved by SMEDDS.

7.Analysis of SLC25A13 gene mutations and prenatal diagnosis for 20 families affected with citrin deficiency.

OBJECTIVETo detect mutations of SLC25A13 gene in 20 families affected with citrin deficiency and provide prenatal diagnosis for them.
METHODSThe 20 probands and their parents were subjected to high-frequency mutation screening combined with Sanger sequencing. After confirming the genotype of each pedigree, genetic counseling and prenatal diagnosis were performed for their subsequent pregnancies.
RESULTSBiallelic pathogenic mutations of the SLC25A13 gene were identified in all probands. These included three deletions (c.851del4, c.1092_1095delT, and c.495delA), two splice-site mutations (IVS6+5G to A and IVS11+1G to A), two nonsense mutations (c.775C to T (p.Q259X) and c.72T to A (p.Y24X)), one duplication mutation (c.1638_1660dup), one insertion (IVSl6ins3kb), and one missense mutation (c.1775A to C (p.Q592P)). Among 24 fetuses undergoing prenatal diagnosis, 8 had normal genotypes, 11 were mutation carriers, while 5 harbored biallelic mutations. Those with wild type alleles or heterozygous SLC25A13 mutations were delivered. Two fetuses harboring homozygous c.851del4 mutations were also delivered. Three fetuses harboring biallelic mutations were terminated.
CONCLUSIONAnalysis of SLC25A13 gene mutations in families affected by citrin deficiency can provide evidence for molecular diagnosis and facilitate genetic counseling and prenatal diagnosis for the subsequent pregnancy, which can effectively reduce the risk of birth of further affected children.
8.Three-dimensional finite element modeling and mechanical analysis of knee joint in patients with mild knee osteoarthritis

Objective To establish a three-dimensional finite element model of knee joint in patients with mild knee osteoarthritis in the absence of CT and MRI with thin layer thickness,and to analyze the stress distributions of the tibial plateau.Methods A female patient with mild knee osteoarthritis was selected as the study object,and received serial CT scan from pelvis to distal tibia of fibula.The obtained files in DICOM format were imported into Mimics software for extracting the bony structures of knee joint through threshold segmentation,and then optimizing the extracted structures with Edit Masks,Cavity Fill and Region Grow.The generated model of bony structures was imported into 3-matic for smoothing and wrapping,and Geomagic Wrap was used for bony structure processing with mesh doctor inspection,surface defect repair and surface fitting.The cartilages,meniscus and ligaments were established by contour extension,and assembled with bony structures in SolidWorks in the way of origin coincidence.After defining material properties and contact relationships,meshing,and setting constraints and loads in ANSYS software,the stress distributions of the tibial plateau when standing with both legs were analyzed.Results A complete knee joint model including bony structure,cartilage and ligament was successfully established.When standing with two legs,the peak value of contact stress was about 1.21 MPa,and the maximum stress was 0.72 MPa at the medial middle of tibial plateau and lateral tibial plateau.The 62.7%and 37.3%of the total load were borne by the inner and outer compartments.Conclusion The model of the total knee joint is established with CT extraction and software rendering,and the finite element analysis results meet clinical expectations,indicating that the model established by the proposed method is reliable and can be used for follow-up research.
