1.Modification of Light Environment of Training Room for Vision Rehabilitation

Light environment is an important component of vision rehabilitation training room, and play a role for the people with visual disability. Based on the relationship between the low vision and light environment, the modification of vision rehabilitation training room were discussed in term of light environment, including light for different sorts and severities of visual disability, preventing glare, color rendering and stroboscopic effect and so on, and the objects to see. The possible schemes for modification were designed.
2.Optimizing Fermentation Conditions for Expression of Esterase B1 Fusion Protein in Recombinant Bacteria

Objective In order to research the best fermentation conditions of recombinant bacteria pThioHisA-B1/DH5? which contains gene of esterase B1.Methods Recombinant bacteria pThioHisA-B1/DH5? was induced in different culture media, induction temperatures, different inductor concentrations, different induction time in order to obtain the best expression effect.The expression products of different conditions were detected by SDS-PAGE and thin-layer gel scanning analysis.Results Based on the experiments, the optimized fermentation condition were that the culture medium was TB, inducing temperature was 28 ℃ and inducing time was 6 h,the concentration of IPTG was 0.4 mmol/L.The expression quantity of target protein attained 40.3% in total bacterial protein, and the soluble fusion protein accounted for 66.2% in total target proteins at the optimum conditions.Conclusion The optimized fermentation conditions of expressing fusion protein in recombinant bacteria have been found and in the conditions, soluble esterase B1 fusion protein of high yield can be obtained.
3.Molecular Modification of a HSV-1 Protein and Its Associated Gene Transcriptional Regulation

The molecular modifications of Herpes Simplex Virus Type Ⅰ (HSV-1) proteins represented by acetylation and phosphorylation are essential to its biological functions.The cellular chromatin-remodeling/assembly is involved in HSV-1 associated gene transcriptional regulation in human cells harboring HSV-1 lytic or latent infections.Further investigation on these biological events would provide a better understanding of the mechanisms of HSV- 1 viral gene transcriptional regulation.
4.Relationship between plasma protein Z level and severity of coronary atherosclerosis in patients with coronary heart disease

Objective To explore the relationship between plasma protein Z level and severity of coronary atherosclerosis in patients with coronary heart disease,and analyze the clinical value of plasma protein Z detection.Methods Eighty-five patients who undertaken coronary arteriography were selected,and the patients were divided into coronary heart disease group (63 patients) and control group (22 patients)according to coronary arteriography results.The patients in coronary heart disease group were divided into three groups according to the Gensini score:A group (≤30 scores),B group (31-60 scores) and C group (> 60 scores).All patients' plasma was collected and stored at-80 ℃ until examined,and the plasma PZ level was detected by enzyme-linked immunosorbent assay method.Results The plasma protein Z level in coronary heart disease group was significantly lower than that in control group [(721.82 ± 289.53) μ g/L vs.(1 077.80 ± 338.12) μ g/L],and there was statistical difference (P< 0.05).The plasma protein Z level in A group,B group and C group was (856.09 ± 312.53),(665.27 ± 267.15) and (643.04 ±248.39) μg/L,respectively.The plasma protein Z level in B group and C group was significantly lower than that in A group,and there was statistical difference (P < 0.05),but there was not statistical difference between C group and B group (P > 0.05).There was negative correlation between the plasma Z level and Gensini score (r =-0.300,P =0.017).In coronary heart disease group,the plasma Z level in patients with smoking was significantly lower than that in patients without smoking [(687.83 ± 249.94) μ g/L vs.(844.29 ± 454.71) μ g/L,and there was statistical difference (P < 0.05).There was negative correlation between the plasma Z level,age and hypersensitive C reactive protein (r =-0.349,-0.339,P < 0.05).Conclusions Plasma protein Z level in patients with coronary heart disease is significantly decreased,and the plasma protein Z level has negative correlation with the severity of coronary atherosclerosis.Smoking can induce the decrease of plasma protein Z level,and the decrease of protein Z level maybe a predictor for coronary heart disease.
5.Effect of nano-sized-Fe3O4-lipoteichoic acid from Bifidobacterium on cytokine expression in human gastric cancer in nude mice

Objective To investigate the effect of lipoteichoic acid from Bifidobacterium loaded with magnetic particle nano-sized-Fe3O4(nano-sized-Fe3O4-BLTA)on several cytokines in vivo expression in beterologous graft model for human gastric cancer in nude mice.and to analyze the inhibition mechanism of nano-sized-Fe3 O4-BLTA on BGC-823 human gastric carcinoma transplanted tumor.Methods Forty male nude mice(BALB/e.nu/nu)were used for in vivo transplant tumor model.the experimental animals were randomly divided into five groups to administrated by several dosages of Hano-sized-Fe3 O4-BLTA,and were executed after 12 d.The tumors were collected and photographed.and the tumor tissues were used for differ-ent assays for measuring tumor inhibition.The peritoneal fluid was extracted to isolate the macrophages for cytokines assays.Using irmnunohistochemical staining for vascular endothelial growth factor(VEGF)and CD3 l in tumor to investigate the tumor inhibition rate.Double antibody sandwich ELISA was used to detect the level of cytokine change.Results The mice treated with low dosage(10 μg/d)of nano-sized-Fe3O4-BLTA.the growth inhibifion rate of tumor was 49%,and the levels of VEGF(0.0224±0.0763)and CD31 (57.000 4±6.790)were lower than other treated groups(P<0.01).The high dosage(100μg/d)and me-dium dosage(50μg/d)of nano-sized-Fe3 O4-BLTA groups were significant difference(P<0.01)on the content of cytokines excreted by macrophages.The level of TNF-α(39.4040 ±-1.5590)induced by the low dosage group was higher relatively(P<0.01).Conclusion Nano-sized-Fe3O4-BLTA exerts an inhibiting effect on the growth of human gastric cancer in nude mice.Using nano-sited-Fe3 O4-BLTA,LTA can get more permeability and improve the therapy effect.which will be a new drug on the stomach cancer targeted therapy.
6.Model for pancreatic cancer in rats and the count of pancreatic myofibroblast

Objective To establish a pancreatic cancer model in Sprague Dawley(SD) rats,and to study the distribution and the counts of myofibroblast(MF) in pancreatic cancer and non-cancerous pancreatic tissues.MethodsDirectly implanted DMBA into pancreas parenchyma of SD rats and established TSA intervening group and control group. The carcinogenesis of rats executed within 3~5 months were inspected by HE stain and microscopy for pathomorphological changes.Myofibroblast(MF) was stained by Heidenains method. Results (1)The prevalence of pancreatic cancer in experimental group was 48.7%(18/37), including 17 pancreatic ductal adenocarcinoma and 1 fibrosarcoma. The prevalence of pancreatic cancer in intervering group was 33.3%(12/36), including 11 pancreatic ductal adenocarcinoma and 1 fibrosarcoma. 7.IEC material development for malaria control by working with target populations

Objective To develop user-friendly information education-communication (IEC)materials and guidelines of malaria control for various target groups. Methods The participatory methods were used to make sure participation of target groups at each stages of IEC material development. Results A package consisted of interactive video, 2 posters for primary groups, sto ry booklet for students, manual for activities of "buddy system" for teachers, flipcharts for health staff at grass-root levels, guidelines on communication skill and how to use the first 5kinds of materials, was developed. Conclusion A set of user-friendly IEC materials and guidelines of malaria control has successfully been developed with sufficient participation of target groups.
8.Clinical study of post-stroke speech apraxia treated with scalp electric acupuncture under anatomic orientation and rehabilitation training.

OBJECTIVETo compare the differences in the clinical efficacy on post-stroke speech disorder between scalp electric acupuncture (EA) under anatomic orientation combined with rehabilitation training and simple rehabilitation training.
METHODSSixty patients of post-stroke speech apraxia were randomized into an observation group and a control group, 30 cases in each one. In the observation group, under anatomic orientation, the scalp EA was adopted to the dominant hemisphere Broca area on the left cerebrum. Additionally, the speech rehabilitation training was combined. In the control group, the speech rehabilitation training was simply,used. The treatment lasted for 4 weeks totally. The speech movement program module in the psychological language assessment and treatment system of Chinese aphasia was used for the evident of efficacy assessment. The scores of counting, singing scale, repeating phonetic alphabet, repeating monosyllable and repeating disyllable were observed in the patients of the two groups. The assessment was done separately on the day of grouping and 4 weeks after treatment.
RESULTSIn 4 weeks of treatment, the scores of counting, singing scale, repeating phonetic alphabet, repeating monosyllable and repeating disyllable were all improved as compared with those before treatment in the two groups (all P<0. 001). The results in the observation group were better than those in the control group (all P< 0. 05). The total effective rate was 100. 0% (30/30) in the observation group, superior apparently to 53. 3% (16/30) in the control group (P<0. 001).
CONCLUSIONThe scalp EA under anatomic orientation combined with' speech rehabilitation training obviously improves speech apraxia in stroke patients so that the speech disorder cani be relieved. The efficacy is better than that in simple rehabilitation training.

10.Clinical analysis of hemodialysis patients with pulmonary arterial hypertension

Objective To investigate the prevalence ,clinical features and prognosis of pulmonary arterial hypertension (PAH) in maintenance bemodialysis (MHD) patients for early diagnosis and treatment. Methods Complete clinical data of 184 MHD patients in Beijing Chaoyang Hospital between January 2000 and December 2007 were retrospectively analyzed. PAH diagnosis depended on echocardiography. Results Sixty-five (35.3%) patients were found having PAH, including 31 females and 34 males, with mean (56.84±14.58) years old. The mean bemadialysis duration was (29.69±21.61) months. Among 65 patients with PAH, arteriovenous fistula was used in 61 patients and central venous catheter in 4 patients. The systolic pulmonary arterial pressure (SPAP) was (44.56±8.25) mm Hg (1 mm Hg=0.133 kPa) in PAH group, which was significantly higher than (30.28±3.92) mm Hg in non-PAH group. There were significant differences of interval dialysis weight gain (IDWG), Hb, Hct, right atrial diameter, pulmonary artery diameter and right ventricular diameter between PAH group and non-PAH group (all P<0.05). Multivariate analysis revealed anemia, IDWG and right atrial diameter were independent factors (P< 0.05). There were no significant differences of age, dialysis duration, serum calcium, phosphorus, alkaline phosphatase, parathyroid hormone (PTH) between two groups. Conclusions PAH is a common complication of MHD patients. Patients with mild PAH had few clinical manifestations and good therapeutic responses, but severe PAH may be associated with poor prognos is. Doppler echocardiography should be performed periodically to detect PAH in MHD patients.
