1.Comparative Research on Infection Rates and Gene Spectrum of Cervical Cells in Women Infected with HPV in Quanzhou and Wuhan Regions of China

Objective To compare the differences of infect rates and genotype distribution of 21 kinds of human papillomavir-us (HPV)in women cervical cells in Quanzhou and Wuhan regions.Methods Collected gynecological clinic of 10 068 women in cervical cell specimens from November 2012 to January 2014 in Affiliated Quanzhou First Hospital of Fujian Med-ical University and Renmin Hospital of Wuhan University,of which 6 823 cases in Quanzhou,Wuhan region 3 245 cases were detected to identify HPV genotypes by using nucleic acid hybridization technique(HybriMax).Results 2 439 in 10 068 samples of cervical cells were detected to be infected HPV with overall infection rate of 24.23%(2 439/10 068).The overall HPV infection rate in Quanzhou was 22.61% (1 543/6 823),and 27.61% (896/3 245)in Wuhan.The single HPV infection rate in Quanzhou was 17.28% (1 179/6 823),and 19.88% (645/3 245)in Wuhan.The multiple HPV infection rate in Quanzhou was 5.33% (364/6 823),and 7.73% (251/3 245)in Wuhan (The values of P<0.05).The five most common HPV genetypes in Quanzhou was HPV-16,HPV-52,HPV-58,HPV-11 and HPV-CP8304,respectively,in Wuhan was HPV-16,HPV-52,HPV-58,HPV-33 and HPV-18,respectively.Conclusion The overall,single and multiple HPV infection rates in Wuhan were all markedly higher than those in Quanzhou (the values of P<0.05).The most common HPV gene-types in two regions were different.
2.Virulence Changes between EF A7 and EF A7 cyl Mutant in the Mouse Peritonitis Models

Objective The purpose of this study was to understand the pathogenic mechanisms of enterococcal hemolysin and establish the foundation to diagnose and control relevant diseases.Methods Take 30 clean grade ICR mice were randomly divided into ten groups,each 6,1 to 4 groups according to 10-fold gradient amount per 1 ml intraperitoneal injection of 5× 107 cfu/ml~5×1010 cfu/ml of EF A7,group 5 as a control group injected 0.5 ml saline,take another 30 clean grade ICR mice,intraperitoneal injection of 5×107 cfu/ml~5×1010 cfu/ml of EF A7 cyl mutant by the same measures,compared to the original strains of mice infected with mutant strain LD50,and a week after infection mortality was observed in mice daily. Another seven groups of mice with the same bacterial concentration (5×109 cfu/ml)were injected intraperitoneally EF A7 and EF A7 cyl mutant,after infection 6 h,12 h and 24 h comparing the number of leukocytes in peripheral blood of mice,re-spectively,the concentration of TNF-αin the acute phase of cytokines.Results In the mouse peritonitis models the 50% le-thal dose (LD50)of EF A7 cyl mutant was 100 times lower than enterococcus faecalis EF A7.The differences of the survival percentage of EF A7 cyl mutant groups and EF A7 groups have significance (P<0.05).As the changes in boold leukocytes numbers and the TNF-αlevel.Conclusion Cyl gene is probably a major virulence factor of Enterococcus and plays an impor-tant role in the mouse peritonitis model.
3. Molecular epidemiological analysis on coxsackievirus B5 in certain areas of China

Objective:
To study the molecular epidemiology and genetic variation of coxsackievirus B5 (CV-B5) in certain areas in China.
Methods:
MEGA 6.0 software was used to analyze the complete VP1 region of CV-B5 isolated strains from certain areas of China by retrieving the GenBank nucleotide database. Besides, the phylogenetic tree was constructed, the homology of nucleotide and amino acids were calculated and the rate of evolution was estimated.
Results:
A total of 189 Chinese CV-B5 isolated strains were included in this study. Most of Chinese CV-B5 isolated strains belonged to genotype C, accounted for 90.5%. Compared with the genotype A, the homology of nucleotide sequences and amino acid sequences of complete VP1 region of 189 Chinese isolated strains were 79.8%-82.8% and 92.6%-97.9%, respectively; moreover, the nucleotide and the amino acid homology of 189 Chinese CV-B5 isolated strains among themselves ranged from 80.3% to 100.0% and ranged from 91.5% to 100.0%, respectively. The estimated rate of evolution of the CV-B5 was 4×10-3 substitutions/site/year.
Conclusions
The majority of CV-B5 isolated strains belonged to genotype C, and subgenotype C1 and C2 were co-circulating together in certain areas of China.
4.Expression levels of RhoA in bronchoalveolar lavage fluid of asthma patients allergic to dust mites.

OBJECTIVETo detect the expression of RhoA in the bronchoalveolar lavage fluid (BLF) from asthma patients allergic to dust mites and study its clinical significance.
METHODSBLF was collected from 47 mite-allergic asthma patients, 15 asthma patients non-allergic to dust mites and 7 healthy volunteers to detect RhoA expression by immunohistochemistry.
RESULTSThe total positivity rates of RhoA expression were 66.7% in mite-allergic asthma patients, 70.2% in asthma patients non-allergic to dust mites and 0 in healthy volunteers, showing significant difference between the 3 groups (P<0.01). No statistical difference was observed in RhoA expression levels between male and female asthma patients allergic to dust mites (P>0.05). The total positivity rate was 83.3% in asthma children allergic to dust mites, higher than that in other patients with different ages, but their RhoA expression levels showed no significant differences (P>0.05). The total positive rates were 33.3%, 57.9%, 88.2% and 100.0% in patients with clinical grades I, II, III and IV, showing significant differences (P<0.05).
CONCLUSIONSThe positivity rate of RhoA in the BLF is higher in asthma patients allergic to dust mites than in normal healthy subjects, increases with the clinical grade.

5.Study on the effects of controlling hepatitis A epidemics by building the colony immune defence.

OBJECTIVETo build the colony immune defence and to control the periodic epidemics of hepatitis A after a mass vaccination of live attenuated hepatitis A vaccine.
METHODSThrough yearly observing the correlation of the accumulative inoculation rates of live attenuated hepatitis A vaccine, the crowd immune standard and the morbidity of hepatitis A after administered live attenuated hepatitis A vaccine among susceptible population and surveilling anti-HAV IgG in the different epidemic areas.
RESULTS(1) The accumulative inoculation rates of live attenuated hepatitis A vaccine was 34.15% in 8 years from 1993 to 2000, among which they were 84.46%, 82.23% and 15.14% in the preschool children, primary and middle school student and 15 - 45 age groups respectively. The morbidity of hepatitis A decreased to 8.26/100,000 in 2000. (2) The crowd positive rates of anti-HAV IgG were 74.24% in 1998 and 83.68% by 2000. Among which they were 74.02%, 68.49%, 79.41%, 85.71% and 90.80% in 2 - 4, 6 - 8, 13 - 15, 20- and 30 - 39 age groups respectively. (3) The accumulative inoculation rates were 37.36%, 51.08% and 28.68% in the inspection areas of Tongtai, Binhai and Yandu respectively. The crowd positive rates of anti-HAV IgG in three inspect area were 85.71%, 85.94% and 78.63% respectively. It was noticed the correlation between the accumulative inoculation rates and the crowd positive rates of anti-HAV IgG was (r(city) = 0.91, F = 15.10, P < 0.03).
CONCLUSIONThe results showed that the crowd positive rates of anti-HAV IgG had increased to 85% while, the colony immune defence of hepatitis A was effectively built to break the periodic epidemics of hepatitis A. The morbidity of hepatitis A decreased to the lowest level in the history.

6.Follow-up evaluation of pulmonary lesions in patients recovering from COVID-19 with improved pulmonary ultrasonography

Objective:To evaluate the value of improved pulmonary ultrasonography in the follow-up assessment of lung damage in patients who recovered from corona virus disease 2019(COVID-19).Methods:Twenty-two patients who were cured of COVID-19 in Quanzhou First Hospital from January to May 2020 were randomly selected and divided into 7 mild cases, 12 moderate cases and 3 severe cases according to the first high-resolution CT (HRCT) at admission. Six months after recovery, modified lung ultrasonography and HRCT were used prospectively to assess the lung damage and evaluate the correlation and consistency between the two techniques.Results:①There were significant differences in lung damage between the mild group and the moderate group, severe group (all P<0.05), while there was no significant difference between the moderate group and severe group ( P>0.05). ②There was good consistency between the improved lung ultrasound examination and HRCT (Kappa=0.776, P<0.001). ③There was a positive correlation between the score of improved pulmonary ultrasound examination and HRCT Warrick score ( r=0.755, P<0.001). Conclusions:Improved pulmonary ultrasonography can be used as a priority in the evaluation of pulmonary damage follow-up in patients with COVID-19 recovery, reducing the use of CT, and providing favorable evidence for further clinical management.
7.Preliminary Result of Olfactory Ensheathing Cell Transplantation in Intractable Neuropathic Pain Following Spinal Cord Injury: 17 Cases Report

ObjectiveTo explore the feasibility and potential benefit of olfactory ensheathing cell (OEC) intraspinal transplantation in the treatment of intractable chronic neuropathic pain after spinal cord injury (SCI).Methods17 patients, 15 male and 2 female, with intractable chronic neuropathic pain after spinal cord injury was treated by OEC implant from November, 2004 to November, 2007. The age ranged from 18 to 68 (mean 40.4) years. The etiology of cord impairment included car accidents, falls, radiation damage, machine extrusion, gun-shot, and diving. The patients suffered severe persistent pain for 6 to 309 (mean 102.2) months, and the time points when cell therapy were administrated in the patients ranged from 6 to 312 (mean 105.9 months) after their injuries. Olfactory bulbs were harvested and trypsinized down to single fetal OECs. They were cultured for 12~14 days before implant. The fetal OECs were transplanted by injection into spinal cord at opposing ends of the injury site. The degree of pain was assessed and compared before operation and long-term follow-up according to the International Association of Neurorestoratology Spinal Cord Injury Functional Rating Scale (IANR-SCIFRS), i.e., 0 point means extreme pain, uncontrolled; 1 point, severe pain, narcotics required; 2 points, mild pain, ordinary pain killer effective; 3 points, no pain.ResultsThe follow-up and pain reevaluation were performed at 0.5 to 88 months with an average of 17.5 months after cell transplantation. The mean score of pain amelioration is 1.2 points.ConclusionThe OEC intraspinal transplantation appears to have a promising role in treatment of intractable chronic neuropathic pain after SCI.

Hepatotoxicity is one of the most common adverse reactions during anti -hyperlipidemia treatment .Mechanisms of anti-hyperlipidemia drug-induced hepatotoxicity are not clear yet , but most of the toxic reactions are dose-related hypersensitivity and could be released after drug withdrawal .It is accepted that clinical risk factors for the development of hepatotoxicity during anti -hyper-lipidemia treatment are high age and chronic illnesses .Treatment of anti-hyperlipidemia drug-induced hepatotoxicity has not been uni-fied and most of the treatments are non-specific and symptomatic .Researching hypotoxicity and hepatoprotection antihyperlipidemia drug, especially TCM and pharmaceutics will become a promising direction .

OBJECTIVE To study the improvement effects and its mechanism of catalpol on testicular lesions in KK-Ay spontaneous diabetic mice on the basis of glycolysis process mediated by advanced glycation end products (AGEs) and their receptors (RAGE). METHODS KK-Ay spontaneous diabetic mice fed with high-fat diet were used as diabetic model, and then randomly divided into model group, catalpol group (100 mg/kg), aminoguanidine group (AGEs inhibitor, 100 mg/kg) and FPS- ZM1 group (RAGE inhibitor, 1 mg/kg), and C57BL/6J mice fed in the same period were set as normal group, with 6 mice in each group. The catalpol group and aminoguanidine group mice were given relevant medicine intragastrically, normal group and model group mice were given constant volume of normal saline intragastrically, and FPS-ZM1 group mice were given relevant medicine 1 mL/g intraperitoneally, for consecutive 8 weeks. After the last administration, the body mass, fasting blood glucose, 24-hour food intake, water consumption, urine volume, testicular organ coefficient, and sperm motility of the mice were measured; pathological morphology and ultrastructural structure of testicular tissue were observed; the levels of reduced glutathione (GSH), superoxide dismutase (SOD), lactate dehydrogenase (LDH) and sugar metabolites in testicular tissue of mice were detected; pathway enrichment analysis was performed; the level of AGEs in serum and testicular tissue, protein expressions of RAGE, B-cell lymphoma-2 (Bcl-2) and Bcl-2-associated X protein (Bax), and mRNA expressions of key rate-limiting enzymes [hexokinase (HK), phosphofructose kinase (PFK), pyruvate kinase (PK), LDH] in testicular tissue were alldetected. RESULT S Catalpol could significantly improve the general symptoms, testicular organ coefficients and motility ofsperm in KK-Ay spontaneous diabetic mice (P<0.05 or P<0.01). The morphology and ultrastructure of spermatogenic cells in each layer of the seminiferous tubules were all improved. The levels of GSH, SOD and LDH in testicular tissue,the levels of the metabolic product glucose fructose-1,6-diphosphate, 3-phosphate glycerate, 3-phosphate glyceraldehyde, lactic acid and pyruvate, the expressions of HK, PFK, PK and LDH mRNA were all significantly increased(P<0.05 or P<0.01); the levels of AGEs in serum and testicular tissue, the expression of RAGE protein and the ratio of Bax to Bcl-2 in testicular tissue were significantly decreased(P<0.05 or P<0.01). Aminoguanidine and FPS-ZM1 could significantly improve the levels of most of above indicators in mice(P<0.05 or P<0.01). CONCLUSIONS Catalpol shows significant improvement effects on testicular lesions of KK-Ay spontaneous diabetic mice, and its mechanism of action was associated with upregulation of AGEs/RAGE signaling pathway- mediated glycolysis.
