1.Composing and evaluating the measurement uncertainty of two kinds of chemiluminescence detection system

Objective To compose and evaluate the measurement uncertainty of two kinds of chemiluminescence detection system using different methods.Methods The measurement uncertainty was composed by 4 different methods:(1) U 1% was composed of within-run CV(CVw %),between-run CV(CVB %)and bias (CVBias %);(2) U2% was composed of CVB % and uncertainty of calibration (CVcal %);(3) U3% was composed of CVW%,CVs% and CVcal%;(4) U4% was composed of CVW%,CVB%,CVBias% and CVcal%.The measurement uncertainty of Architect i2000SR system (Abbott,USA) and DXI800 system (Beckman,USA) was assessed.Pearson correlation analysis,Spearman correlation analysis,Paried t test and Mann-Whitney u test were performed to analyze the data.Results For Architect i2000SR system,U1%,U2%,U3% and U4% were significantly correlated (r=0.727-0.988,all P<0.05),U3% and U2% were significantly different (t =6.88,P<0.05),U4% and U1% were significantly different (t =6.21,P<0.05).For DXI800 system,U1%,U2%,U3% and U4% were also significantly correlated (r =0.608-0.975,all P<0.05),no significant difference was found between U3% and U2% (z=-1.33,P>0.05),or between U4% and U 1% (z =-1.04,P> 0.05);the expanded measurement uncertainty was correlated with CVW%,CVB%,CVBias%(rs=0.653-0.912,all P<0.05),but not with CVcal%(rs=0.548,P>0.05).Conclusions For Architect i2000SR system,the fourth method is more proper to compose the measurement uncertainty (U4%).For DXI800 system,the first method is more appropriate (U1%).According to the contribution of different components to the measurement uncertainty,the measurement quality could be improved by reducing the imprecision and bias.
2.Single nucleotide polymorphisms of TLR4 locus in Chinese Cantonese population

AIM: Toll-like receptor 4 (TLR4) was an important pathogen recognition receptor in the innate immune system. The aim of this study was to investigate the distribution of TLR4 polymorphisms in the general population of China. METHODS: Peripheral blood samples were collected from 191 unrelated healthy Chinese Cantonese individuals. The functional regions of TLR4 locus,including promoter region and all three exons with their surrounding intronic regions were amplified using polymerase chain reaction. After purified,the amplified products were directly sequenced on both strands. RESULTS: A total of eight single nucleotides polymorphisms (SNPs) were detected,five of which were novel. The most common SNP were -1607 C/T with the minor allele frequency of 0.283. Two nonsynonymous substitutions Asp299Gly and Thr399Ile,which were common in Caucasus,were not detected in Cantonese. Neutrality test revealed that TLR4 in Chinese Cantonese was not significantly deviated from the neutral model. CONCLUSION: This is the new finding on the distribution of TLR4 SNPs in the general population of China. It provides several ethnic specific SNPs for further disease association studies of TLR4 polymorphisms in Chinese populations.
3.Expressions of forkhead transcription factor O1 and Kruppel like transcription factor 9 in non-small cell lung cancer and their clinical significances

Objective To investigate the expressions of forkhead transcription factor O1 (FOXO1) and Kruppel like transcription factor 9 (KLF9) in non-small cell lung cancer (NSCLC) and their clinical significances. Methods A total of 102 patients with NSCLC who underwent surgical resection in the First Hospital of Yulin from September 2012 to June 2015 were selected as NSCLC group, and the paracancerous tissues more than 5 cm far from the edge of the cancer tissues were selected as the control group. The relative expressions of FOXO1 and KLF9 mRNA in NSCLC tissues and paracancerous tissues were detected by real-time quantitative polymerase chain reaction (qRT-PCR), and the immunohistochemistry was used to detect the expressions of FOXO1 and KLF9 in NSCLC tissues and paracancerous tissues. According to the mean mRNA expression in NSCLC tissues, FOXO1 and KLF9 were divided into high expression group and low expression group, their relationships with clinicopathological parameters were observed. The correlation of FOXO1 and KLF9 expressions was analyzed by Pearson method, and the survival of patients was analyzed by Kaplan-Meier method, logistic regression analysis was performed on the related factors affecting the prognosis of NSCLC. Results Compared with the control group, the expression level of FOXO1 in NSCLC group increased (1.28±0.47 vs. 2.82±0.15, t = 31.525, P < 0.05), while the expression level of KLF9 decreased (1.04±0.11 vs. 0.34± 0.07, t = 54.222, P < 0.05). The expressions of FOXO1 were lower in NSCLC tissues of patients with lymph node metastasis, high differentiation, and TNM stage Ⅲ-Ⅳ than those in NSCLC tissues of patients without lymph node metastasis, middle and low differentiation, and TNM stage Ⅰ-Ⅱ, and the differences were statistically significant (all P < 0.05). The expressions of KLF9 were higher in NSCLC tissues of patients with lymph node metastasis, high differentiation, and TNM stage Ⅲ-Ⅳ than those in NSCLC tissues of patients without lymph node metastasis, middle and low differentiation and TNM stage Ⅰ-Ⅱ, and the differences were statistically significant (all P < 0.05). Pearson correlation analysis showed that there was no significant correlation between FOXO1 and KLF9 expression (r = 0.154, P = 0.156). Kaplan-Meier analysis showed that the 3-year overall survival rate of FOXO1 high expression group was lower than that of FOXO1 low expression group (10.81% vs. 60.71%, χ 2 = 27.341, P < 0.01). The 3-year overall survival rate of KLF9 low expression group was lower than that of KLF9 high expression group (26.32% vs. 61.54%, χ 2 = 8.526, P = 0.003). Logistic regression analysis showed that FOXO1 was a risk factor for the prognosis of NSCLC (OR = 2.682, P = 0.003), and KLF9 was a protective factor for the prognosis of NSCLC (OR = 0.446, P = 0.012). Conclusions The high expression of FOXO1 and the low expression of KLF9 in NSCLC tissues can be used as effective indexes to predict the progression of NSCLC. The expressions of FOXO1 and KLF9 are closely related to the poor prognosis of patients.
4.Study on acupoint selection law of acupuncture for autism spectrum disorder based on data mining

Objective:To analyze the acupoint selection law of acupuncture for autism spectrum disorder (ASD) using data mining techniques.Methods:Literature related to acupuncture for ASD was retrieved from the CNKI, SinoMed, VIP, Wanfang, and PubMed databases from the establishment of the databases to April 1, 2022, and then a database of acupuncture prescriptions was established. The frequency analysis of acupoint use was performed using Microsoft Excel 2019; the Apriori algorithm was used to analyze the association law of acupoints/acupoint areas; SPSS 26.0 was used to perform intergroup cluster analysis.Results:A total of 97 relevant articles with 97 acupuncture prescriptions and 98 acupoints/acupoint areas were included. The most frequently used acupoint was Shenmen (HT 7). The acupoint area of Jin's three-needle therapy and the Governor Vessel acupoints are commonly used. The most frequently occurring part of the acupoint/acupoint area was the head, and the most commonly used specific acupoint was the rendezvous acupoint. Association rule analysis yielded 40 groups of acupoints/acupoint areas, and the most commonly used combination was Laogong (PC 8) and Shenmen (HT 7). Four categories were extracted among high-frequency acupoints/acupoint areas by cluster analysis.Conclusion:Acupuncture treatment for ASD mainly selects the head acupoints, mainly selecting the acupoint area of Jin's three-needle therapy and the Governor Vessel acupoints, and paying attention to the use of specific acupoints.
