1.Expression of ATP synthase 6 in breast cancer cell line, T47D cells, induced by heregulin β1

Objective : To explore expression of related genes in breast cell line T47D induced by heregulin β1. Methods: Suppression subtractive hybridization was performed using cDNA sub traction kit to detect expression of heregulin-responsive genes in T47D cells. Results: ATP syn thase 6 was up-regulated by heregulin β1 in T47D cells at 1 and 6 hours. Conclusion: Heregulin β1 participates in the regulation of expression of both ATP synthase 6 and oxidative phosphoryla tion of T47D cells.
2.Preparation of lentivirus silencing SND1 and its influence on breast cancer cell proliferation and invasion

Objective: This work aimed to construct stable MCF-7 cell sublines from which staphylococcal nuclease domain con-taining 1 (SND1) expression was interfered to analyze the effect of SND1 silencing on the proliferation and metastasis of MCF-7 cells. Methods: The lentivirus that could mediate SND1 silencing was prepared. MCF-7 cells were infected with the lentiviruses to construct stable sub-cell lines. Quantitative real-time polymerase chain reaction and Western blot analysis were employed to determine SND1 ex-pression level. MTS, wound healing, and transwell assays were applied to analyze the effect of SND1 silencing on the proliferation, mi-gration, and invasion of MCF-7 cells. Results: A lentivirus expression vector that contains sequences encoding shRNAs targeting SND1 and an shRNA negative control were successfully established. The lentiviruses (LV-SH1, LV-SH2, LV-SH3, and 和 LV-NC) were then collected and packaged. Stabilized MCF-7 sublines were prepared through infection with lentiviruses. The most efficient MCF-7 stable cell subline, MCF-SH3, was selected for SND1 silencing. Compared with the control cell, the proliferation, migration, and inva-sion potential of MCF-SH3 were significantly decreased. Conclusion: SND1 could promote the proliferation, migration, and invasion of breast cancer cells. Thus, silencing SND1 expression will inhibit such proliferation, migration, and invasion. These results indicated that the unusual expression of SND1 is associated with breast cancer and may participate in cancer progression by affecting prolifera-tion, migration, and invasion.
3.Observation of the effect of Miles laparoscopic sigmoid colostomy surgery for elderly patients with colorectal cancer

Objective To investigate the effect of Miles laparoscopic sigmoid colostomy surgery for elderly patients with colorectal cancer .Methods The clinical data of 78 elderly patients with colorectal cancer underwent surgery were chosen .According to the different operation method , they were randomly divided into the observation group and control group ,39 cases in each group .The observation group was treated with laparoscopic Miles ,and the control group underwent surgery for abdominal Miles .The safety,efficacy,early complications of the two groups were analyzed and compared .Results The incidence rate of postoperative complication , excellent rate of ostomy surgery 6 months after operation in the observation group were 5.13%, 66.67%, respectively, which were better than 38.46%,20.51%in the control group (χ2 =12.70,9.69,all P<0.05).The defecate feeling of the observation group was better than that of the control group (χ2 =29.40,P<0.05).The difference of patients'quality of life and physical function between the two groups was statistically significant (t=3.30,P<0.05).Conclusion The clinical effect of laparoscopic sigmoid colostomy surgery Miles in the treatment of elderly patients with colorectal cancer is bet -ter than traditional open surgery ,it has obvious practical significance such as reducing the short and long term compli -cations and improving the quality of life of patients .
4.Characteristics of epidemic situation of HIV/AIDS in Yongzhou in 1993-2014

Objective To analyze the characteristics of epidemic situation of HIV/AIDS in Yongzhou from 1993 to 2014,and provide evidence for HIV/AIDS prevention and control strategy.Methods Descriptive epidemiological analysis was conducted on HIV/AIDS epidemic situation in Yongzhou from 1993 to 2014.Results A total of 2 917 HIV/AIDS cases were reported,the reported cases increased year by year;the ratio of male to female cases was 2.30∶1 ;52.04% were aged 20-49 years old,46.76% were aged ≥50 years old,the percentage of population ≥50 years old increased obviously in recent years;the major transmission route was sexual contact (88.48% ),gay transmission accounted for 2.64%.Conclusion The knowledge about AIDS in Yongzhou should be strengthened, intervention in high risk population should be implemented,scope of detection should be expanded,so as to prevent the transmission of AIDS effectively.
5.Pulmonary circulation effects of different ways to neutralize heparin on the congenital heart disease patients with pulmonary arteial hypertension after extracorporeal circulation surgery

Objective To investigate pulmonary circulation effects of protamine different infusion ways on the acyanotic congeni -tal heart disease(CHD) patients with pulmonary arterial hypertension(PAH) after extracorporeal circulation surgery .Methods Se-lect 80 cases of the CHD patients prepared for extracorporeal circulation surgery ,according to the different routes of administration and with or without PAH ,80 patients are divided into 4 groups (n= 20) :A1(non-PAH ,aortic root administration) ,A2(non-PAH , central venous administration) ,B1(PAH ,aortic root administration ) ,B2 (PAH ,central venous administration) .pulmonary artery pressure(PAP) ,pulmonary venous pressure (PVP) ,central venous pressure (CVP) ,peak airway pressure (Ppeak) ,plateau airway pressure(Pmean ) at the time point of preoperative(T0) ,before infusion of protamine (T1) ,1 minute after infusion of protamine (T2) ,3 min (T3) ,5 min (T4) ,10 min (T5) ,15 min (T6) and 25 min (T7) were recorded .The level of venous blood thromboxane B2(TXB2) was detected .Results After infusion protamine ,the PAP ,PVP ,CVP ,Ppeak and Pmean of group A2 and group B2 are higher compared to group A1(P< 0 .05) and group B1(P< 0 .01) respectively ;Compared with preoperation (T0) ,the TXB2 in 4 groups increased obviously after infusion protamine (T2 to T7) (P< 0 .05) ;Compared with group A1 and group B1 ,the TXB2 of group A2 and group B2 increased more significantly (P< 0 .01) .Conclusion Patients with congenital heart disease after intracardi-al surgery with extracorporeal circulation use trace pump infusion protamine through the aortic root ,which have little effect on pul-monary circulation ,can significantly reduced TXB2 release compared with central vein administration ,it can avoid a high concentra-tion of protamine directly into the pulmonary circulation that can strongly stimulate the pulmonary vascular and tracheal smooth muscle .Especially in the CHD patients with PAH .
6.The value of conventional sequence and MRA in the diagnosis of reversible cerebral vasoconstriction syndrome at 3.0T MRI

Objective To study the value of MRI sequence and MRA in the diagnosis of reversible cerebral vasoconstriction syn-drome (RCVS).Methods MRI and MRA of 5 patients with RCVS were reviewed retrospectively.Abnormal distribution,morpholo-gy,signal intensity of the affected arteries and complication of RCVS on MRI and MRA were analyzed.Results All patients showed multifocal segmental vasoconstriction and dilatation of cerebral arteries on MRA,which sometimes was described as “string and beads”.There were 4 patients with cerebral parenchyma lesions,in which posterior reversible encephalopathy syndrome was showed in one case,cortical subarachnoid hemorrhage in one case,and watershed infarcts in 3 cases.Five patients recovered after 3 months. Conclusion MRI and MRA can clearly show cerebrovascular morphology and observe parenchymal secondary lesions,which have an important guiding significance for clinical treatment and prognosis assessment.
7.Effect of protein kinase C inhibitor on the level of phosphralated extracellular regulated protein kinases in the spinal trigeminal nucleus of migraine model rats

Objective To explore the effect of protein kinase C inhibitor on the level of phosphralated extracellular regulated protein kinases in the spinal trigeminal nucleus of migraine model rats.Methods Healthy adult male SD rats were randomly divided into four groups:normal group (group C),sham operation group (group C),migraine model group(group M),and H-7group(H-7group),with 18 rats in each group.Dural blood flow and the extracellular discharge frequency in the spinal trigeminal nucleus was recorded.ERK1/2 phosphorylation was tested.Results (1) Dural blood flow:compared with group C((3.8± 1.0)%),the dural blood flow in M group ((78.0±4.2) %)increased obviously(P<0.01) ; compared with M group((78.0±4.2)%),the dural blood flow in H-7 group((-24.8±4.9) %) decreased obviously(P<0.01).(2) The percentage of extracellular discharge frequency change:two hours after treatment,the percentage of extracellula discharge frequency change in group M ((325.9 ±47.3)%)was higher than that in group C((107.3±16.4)%).The percentage of discharge frequency change in group H-7((136.0±26.5)%) was lower than that in group M((325.9±47.3)%).There was no significant difference in the percentage of discharge frequency change between group H-7((136.0±26.5) %) and group C((107.3 ± 16.4)%).(2) ERK1/2 phosphorylation:the ERK1/2 phosphorylation in group M was higher than that in group C.The ERK1/2 phosphorylation in group H-7 was lower than than that in group C and group M.Conclusion ERK1/2 is a downstream PKC signal path and PKC may have indirect activation of ERK1/2.

Objective To construct secreting type human TRAIL(shTRAIL) gene vector pcDNA3.1-HRE/Egr1-shTRAIL mediated by hypoxia/radiation double sensitive promoter,and observe the effect of hypoxia and radiation on shTRAIL.Methods HRE upper and lower strands were gotten by chemical synthesis,double strands HRE was gotten by PCR;pMD19T-Egr1 was digested by SacⅠ and Hind Ⅲ,then Egr1 was obtained,pshuttle-shTRAIL was digested by Kpn Ⅰ and BamH Ⅰ,then shTRAIL was obtained;HRE/Egr1 double sensitive promoter mediated shTRAIL expression vector pcDNA3.1-HRE/Egr1-shTRAIL was constructed by gene recombination technique,it was identified correctlly by enzyme digestion,PCR and sequencing.A549 cells were divided into normal,hypoxia(0.1%),irradiation(6 Gy) and hypoxia + irradiation groups.Results After enzyme digestion by BamH Ⅰ and Sma Ⅰ,the fragments which lengthes were 1284 bp and 4 998 bp,2 292 bp and 3 990 bp were obtained;the vector was amplified by PCR with Egr1 and shTRAIL primer,the products which lengthes were 469 bp and 820 bp were obtained;pcDNA3.1-HRE/Egr1-shTRAIL was sequenced,the result was same to designed,this demonstrated that the construction was right.The vectors were transfected into A549 cells of adenocarcinoma of lung,the expression levels of shTRAIL mRNA and protein were increased after treated with hypoxia and radiation,it had statistically significant differences compared with normal group(P
9.Analysis of PAH-DNA adducts and gene polymorphisms of CYP1A1 and GSTM1 in lymphoma patients

Objective To explore the relationship between PAH-DNA adducts and CYP1A1,GSTM1 gene polymorphisms and lymphoma.Methods PAH-DNA adducts from bone marrow of lymphoma patients and control cases were determined by competitive ELISA.The genotypes of both CYP1A1 and GSTM1 were detected by PCR-based restriction fragment length polymorphisms (PCR-RELP).Results The level of PAH-DNA adducts in lymphoma patients [(2 498±1 250) pg/ml] was significantly higher than that in control [(1 882±797) pg/ml] (t =0.006,P < 0.05).CYP1A1 mutant genotype and GSTM1 null genotype had increased risk of lymphoma,with OR being 1.36 (95 % CI 0.56-3.31,P > 0.05),4.03 (95 % CI 1.51-10.76,P < 0.05),respectively.GSTM1 null genotype individuals with PAH-DNA level higher (or equal) than 2 200 pg/ml had increased risk of lymphoma.Conclusions The content of PAH-DNA adducts and the occurrence of lymphoma may have a certain correlation.GSTM1 null genotype may be linked to lymphoma and increase the risk.
10.Study on the HPLC Fingerprint of Gynecological Lotion

Objective:To establish the HPLC fingerprint detection method for gynecological lotion. Methods:Wondasil C18 (250 mm × 4. 6 mm,5 μm)was selected as the analytical column. The mobile phase was composed of acetonitrile-0. 3% phosphric acid and 0. 3% diethylamine solution with gradient elution at a flow rate of 1. 0 ml·min-1 . The detection wavelength was set at 284 nm and the column temperature was 30℃. Ten batches of gynecological lotion were detected by the HPLC fingerprint and evaluated by Chromato-graphic Fingerprint Evaluation System of Chinese Medicine (2004 edition). Results: The separated peaks were clear and 15 common peaks were identified in the fingerprint of 10 batches of gynecological lotion. Conclusion: The HPLC fingerprint is with good repeat-ability and stability, which can provide evidence for the quality control of gynecological lotion.
