1.THE ATTENUATING EFFECT OF SOY ISOFLAVONES EXTRACT ON BONE LOSS IN POSTMENOPAUSAL WOMEN

Objective: To investigate the effect of soy isoflavones extract on bone loss induced by estrogen deficiency. Methods: 87 postmenopausal women were randomly assigned in single blind manner, to three treatment groups with daily intake of placebo, 84 mg and 126 mg soy isoflavones respectively. Additionally a positive control group was 10 women was given daily 2.5 mg estrogen. Bone mineral density (BMD) of the lumbar spine, neck of femur and Ward`s triangle were measured at baseline and post treatment after 24 wk . Results: BMD in all sites in both isoflavones treated and estrogen treated groups did not change significantly. However, the BMD of the lumbar spine decreased in the placebo group (P

Objective To explore protein expression and significance of MAGE genes in colorectal carcinoma(CRC)tissues.Methods The expression of MAGE genes were studied by using tissue chip and immunochemistry methods in primary CRC tissue and paired adjacent tissue samples in 97 cases.Data were analyzed with x2-test by SPSS 16.0 software.Results The protein expression of MAGE-A3,MAGE-A4 and MAGE-A10 genes were 57%(56/97),63%(61/97)and 28%(27/97)respectively in 97 cases of primary adenocarcinoma.The protein expression frequency of MAGE-A3 in poor colorectal adenocarcinomas was significantly higher than in well-and moderately disfferentiated adenocarcinomas(x2 =9.133,P =0.010).MAGE-A10 in poor colorectal primary adenocarcinomas was significantly higher than in well and moderately adenocarcinomas(x2 =15.280,P =0.000); MAGE-A10 protein expression was significantly higher in stage TNM Ⅲ + Ⅳ than in stage TNM Ⅰ + Ⅱ(x2 =4.227,P=0.040); MAGE-A10 gene expression was higher in metastasis lymphoid node than in no metastasis lymphoid node(x2 =5.557,P =0.018),and the expression level was higher in primary lesion with the increasing of the numbers of lymphoid node metastasis(x2 =7.296,P =0.026).Conclusions The protein expression of MAGE genes is associated with the tumor differentiation,TNM stage and lymphoid node metastasis.MAGE-A3 and MAGE-A10 genes are the possible prognosis marker and potential target of immunotherapy of CRC.
3.The protein expression of PLAC1/CP1 genes in colorectal carcinoma and its clinical significance

Objective To explore the protein expression of PLAC1/CP1 ( cancer-placenta 1 ) and its correlation with clinicopathological characteristics in patients of colorectal carcinoma. Methods The expression of PLAC1/CP1 gene was studied by using tissue chip and immunochemistry in 125 cases CRC tissue specimens. Data were analyzed with the x2-test or Fisher's x2 test statistic by SPSS 16. 0 software.Results The protein expression of PLAC1/CP1 gene was 57.6% (72/125) in 125 cases of CRC and 56. 7% (55/97)in 97 primary adenocarcinoma cases. That was 78. 9% (15/19)in poor differentiated colorectal primary adenocarcinoma, those were significantly higher than that of 35.3% (6/17) in well and 55.7% (34/61) in moderately differentiated adenocarcinoma ( P < 0.05 ); PLAC1/CP1 protein expression was significantly higher in stage TNM Ⅲ + Ⅳ 71.2% (37/52) than in stage TNM Ⅰ + Ⅱ 40% (18/45)(P < 0.05 ); PLAC1/CP1 genes expression rate was 69.6% (32/46) in these with lymphoid node metastasis and 45.1% (23/51)in patients without lymphoid node metastasis (P < 0.05 ). The positive expression rate of PLAC1/CP1 increased in colorectal carcinoma with the increasing of the numbers of lymphoid node involved with metastasis( x2 = 13. 353, P = 0.001 ). Conclusions The protein expression of PLAC1/CP1 is associated with tumor differentiation, TNM stage and lymphoid node metastasis.
4.Differences of tacrolimus' dosage and concentration of individuals in morning periods after renal transplantation

Objective To study the influential factors of tacrolimus'dosage and concentration differences between individuals in morning periods after renal transplantation.Methods The clinical data consisted of 118 receptors in morning periods after renal transplantation,whose immune suppressions were tacrolimus,mycophenolate and hormone.At 3,7,14 and 30 d after operation,all the receptors'weight,dosage of tacrolimus,dosage of hormone,diarrhea,blood fat,liver function,renal function,albumn and erythrocrit were recorded respectively,and at the same time their concentrations of tacrolimus and genetic polymorphisms of CYP3A5,MDRl 3435,MDR1 2677 and MDRl 1236 weredetected.Multiple linear regressions were performed.Results The fitting degrees of stepwise regression equations were low.At 3,7,14 and 30 d after operation,the adjusted R2was 0.284,0.267,0.417 and 0.324,respectively.From the aspect of pharmacogenomics,the main factors rela-ted to the differences of tacrolimus'dosage and concentration included MDR1 2677,MDRl 1236 and MDR13435,which varied intensively.Age,albumn,renal function,blood fat and liver function were important factors too.Conclusions The main reasons of the differences of tacrolimus'dosage and concentration between individuals in morning periods after renal transplantation are medicines and changes of internal environment after operation.The genetic polymorphisms of MDR1,age,albumn,renal runetion.blood fat and liver function are important factors too.
5.Application of CT Scan with Iterative Reconstruction and Low Radiating Dose in Craniocerebral Trauma

PurposeTo evaluate the feasibility and clinical values of CT scan with iterative reconstruction and low radiating dose in craniocerebral trauma.Material and Methods 120 patients suffered from craniocerebral trauma were randomly assigned. All the subjects underwent CT scan using route dose of filtered back projection (FBP) and low dose of iDose (? dose and ? dose), respectively. The quartering were used to subjectively evaluate noise of imaging, skull base artifact, contrast of gyrus-white matter and lesion display in each group. Imaging noise, signal and noise rate (SNR), contrast and noise rate (CNR) of gyrus-white matter and dose length product (DLP) were compared.Results The image quality of both? iDose and ? iDose groups were lower than that of FBP group, but still met the requirement of diagnosis. The image noise of both? iDose and ? iDose groups were higher than that of FBP group (P<0.05). The SNR and CNR of both? iDose and ? iDose groups were lower than those of FBP group (P<0.05). The DLP of both? iDose and ? iDose groups were lower than that of FBP group. There was statistical difference between iDose groups and FBP group (F=2751.46,P<0.05).Conclusion Application of iDose could effectively decrease radiation dose in craniocerebral trauma. Although iDose technique has higher noise level and lower SNR and CNR, the imaging qualities and capability of displaying abnormity meet diagnosis requirement. So that iDose has clinical significance.
6.Perioperative nutrition management of liver transplantation patients

Objective:To summarize the perioperative nutrition management,improve the successful rate in liver transplantation patients and reduce the complications after operation.Methods:A retrospective analysis of the perioperative nutrition treatment was made in 71 cases with liver transplantation.The principle of enteral nutrition before operation was appropriate energy,protein and rich vitamin,and the branched-chain amino acid(BCAA) and glutamine(Gln) were supplied.Intravenous nutrition,early enteral nutrition and total enteral nutrition support were applied during the postoperation period.Results: Except 5 cases that died from respiratory failure and 2 died from hemorrhagic shock respectively,other 64 cases recovered gradually in liver function,and their nutrition indices were enhanced significantly.Conclusion:The perioperative nutrition management is very necessary for liver transplantation patients,and the appropriate nutrition support is helpful to rehabilitate the function of graft organ and the nutritional status of the recipients as possible as early.
7.EFFECT OF SOY ISOFLAVONES ON MENOPAUSAL SYMPTOMS IN EARLY POSTMENOPAUSAL CHINESE WOMEN

Objective To study the effect of soy isoflavones(SI) on menopausal symptoms,and its mechanism.Methods Ninety early postmenopausal women were randomly assigned to three treatment groups with daily dosages of 0(placebo),84,126 mg/(kg bw?d) SI(n=30).Hot flash frequency and Kuppermann score and serum 17?-estrodiol(E2),follicle-stimulating hormone(FSH) and luteinizing hormone(LH) were assessed at baseline,12 w and 24 w posttreatment.Results The percent decreases of Kuppermann scores and hot flashes frequency of the two SI treatment groups were 57.8%?37.4%and 44.3%?19.1% in low dose group and(56.7?26.7)% and(48.5?27.2)% in high dose group respectively,significantly higher than those of the placebo group(34.6?46.2)% and(27.8?15.5) %.No significant difference was observed between the two SI groups.The contents of E2,FSH and LH among all groups were insignificantly different.Conclusion A daily dosage of 84 mg soy isoflavones or above could improve menopausal symptoms especially reducing hot flashes frequency and the mechanism seems not associatedwith serum E2,FSH,and LH.
8.Effects of differential protein transgelin-2 overexpression on biological characteristics of colon cancer cell lines

Objective To study the effect of transgelin-2 expression on biological characteristics of colon cancer cells.Methods RT-PCR and Western blot were used to observe mRNA and protein expression of transgelin-2 in five colon cancer cell lines,screen for cell line with lower transgetin-2 expression;Transient transfection was performed to establish over-expression of transgelin-2 colon cancer cell line;The effect of transgelin-2 over-expression on the proliferation,apoptosis and the ability of migration and invasion of colon cancer cell were detected by CCK-8,low cytometric analysis and transwell method respectively.Results There was no statistical difference in proliferation and apoptosis in colon cancer cells with transgelin-2 over-expression compared with the controls;Enhanced ability of migration and invasion was found in colon cancer cells with transgelin-2 over-expression.After 15 hours culture in serum free medium,more transgelin-2 over-expressed colon cancer cells went through the transwell chamber bottom mnembrane than that in control group and empty vector transfected group 207 ±62 vs.114 ±29 vs.120 ±26,F =7.302,P <0.05).After 24 hours,the differences remained statistically significant (179 ± 32 vs.95 ± 33 vs.95 ± 28,F =10.960,P < 0.05).Conclusions Transgelin-2 enhances migration and invasion ability of colon cancer cells.

AIM: To investigate the role of CD134 (OX40) and NF-?B in the pathogenesis of lupus nephritis (LN). METHODS: Renal in situ CD134 and NF-?B expression were examined in 40 biopsy specimens from LN patients by immunohistochemistry and microwave-based immunohistochemistry, respectively. The relationship between expression of CD134 and NF-?B was analyzed. RESULTS: The expression of glomerular and tubular CD134 and NF-?B in LN were higher than that in normal control, especially in class Ⅳ LN, where there was intense staining of endothelial cell, distal tubules, and interstitial mononuclear cell. 10.Effect of early postoperative enteral nutritional support in abdominal surgery

Objective To evaluate the feasibility and the effect of the early postoperative enteral nutrition on improving the nutritional status, cost of nutrition and hospital stay after operation in abdominal surgery.Methods 48 patients who underwent abdominal operation were allocated into 2 groups: enteral nutrition group(EN group,23 cases) and parenteral nutrition group(PN group,25 cases). It was isonitrogenous and isocaloric intake in both groups. Each day nitrogen intake (0.18±0.02)g/kg,104.6—125.52 kJ(25—30kCal/kg). EN was given at postoperation 8 hours by naso intestinal tube which was placed 30 cm distal to ligament of Treitz or jejunal export loop. PN was provided by peripherally inserted central venous catheters(PICC). The nutritional indices were investigated. A visual analogy scale was used to estimate the infection related complication, cost of nutrition and hospital stay. These parameters were evaluated preoperatively and on the 8th day after operation.Results Early postoperative enteral nutrition showed significant nutrition effect in the patients undergoing abdominal surgery. In the EN group,the nitrogen balance became positive at the postoperative day 4,whereas the nitrogen balance still remained negative in the PN group. The EN group decreased their hospital stay and cost of nutrition compared with that of the PN group(P<0.05). Conclusion The early postoperative enteral nutrition is safe and feasible. It cannot only improve the nutritional status and intestinal function obviously, but also decreased the hospital stay and cost of nutrition compared with parenteral nutrition.
