Chinese Journal of Medical Imaging 2014;(10):781-783,785
Purpose To explore the ultrasonic features of tibial nerve (TN) in tarsal tunnel of type 2 diabetes mellitus patients with diabetic peripheral neuropathy (DPN), and to provide the basis for assessment and treatment. Materials and Methods Fifty-four patients with type 2 diabetes mellitus were divided into the DPN group including 31 patients with diabetic periphreal neuropathy and the non-DPN group including 23 patients without diabetic periphreal neuropathy, 22 cases of non diabetic patients were also enrolled as the control group, all the subjects underwent medial tibial nerve ultrasound examination, and the ultrasonic features of tibial nerve (including morphological parameters:left-right diameter, anterior-posterior diameter, thickness sectional area and color Doppler lfow signals) were compared. Results Left-right diameter and anterior-posterior diameter of DPN group were larger than those of non-DPN group and the control group (P<0.01). Ultrasonic features of tibial nerve of DPN group included nerve bundle hypoechoic, vaguedscreen clothstructure, thickening and fuzzy of neural membrane, and the unclear boundaries to adjacent tissue. There was no statistically signiifcant difference in color Doppler signal of TN (P>0.05) among the DPN group, non-DPN group and the control group. Conclusion There are some characteristics features of tibial nerve in tarsal tunnel of type 2 diabetes mellitus patients with DPN, ultrasound can be used to evaluate the morphology of TN in type 2 diabetes mellitus patients with DPN and to provide morphological basis for evaluation and treatment selection.

Objective To investigate the changes of intestinal blood flow and motility and their effects on intestinal mucosa barrier in rats with acute cerebral ischemia. Methods 64 male Wistar rats were assigned randomly into cerebral ischemia group (n=32) and control group (n=32).The two groups were divided into four subgroups according to timepoints of 6 h,12 h,24 h and 48 h after operation (8 rats for each subgroup). The intestinal mucosa blood flow, intestinal transmit index and endotoxin level in porlal vein were detected. The displacement ratio of E.coli was marked and counted with fluorescent microscope in tissue homogenate from organs. Results The intestinal blood flow in cerebral ischemia group at 6 h,12 h,24 h and 48 h after operation [(34.5±3.2)PU,(22.7±1.9)PU,(26.2±4.3) PU and (30.5± 4.1) PU] were significantly lower than control group[(46.8 ±5.4) PU,(50.1±3.6) PU,(45.4±4.1) PU and (48.7±7.3) PU] (t=2.650,2.875,2.639 and 2.507,all P<0.05).The intestinal transmit index in cerebral ischemia group at 6 h,12 h,24 h and 48h after operation[(0.59±0.07),(0.48±0.06),(0.50±0.08)and (0.57±0.04)] were decreaesd as compared with control group [(0.73±0.04),(0.75=0.02),(0.74±0.06) and (0.76±0.03)] (t=2.409,2.758,2.649 and 2.807,all P<0.05). The levels of endotoxin in cerebral ischemia group started to increase at 6 h and arrived to peak at 24 h,and the levels at four timepoints were (0.382 ± 0.014) Eu/ml, (0.466 ± 0.018) Eu/ml,(0.478 ± 0.029) Eu/ml and (0.412±0.036)Eu/ml,respectively,that were higher than those in control group[(0.102±0.007)Eu/ml,(0.114±0.021)Eu/ml,(0.112±0.018)Eu/ml and (0.108±0.011)Eu/ml] (all P<0.05).The total E.coli detection rates in multiple organs in cerebral ischemia group (11.4%,18.8 %,25.0%and 12.5%) were higher than in control group (2.1%,4.2%,2.1% and 0.0%) at four rime points (all P<0.05). The level of endotoxin was negatively related with the intestinal blood flow and intestinal transmit index( r =- 0.861 and - 0.796,both P< 0.05). Conclusions The higher intestinal mucosa blood flow and intestinal motility play important roles in the increase of intestinal mucosa permeability at early stage after acute cerebral ischemia.

Objective To investigate the role of epithelial cell apoptosis in the stress-related changes of intestinal mucosa barrier following traumatic brain injury. Methods Sixty-four health male Wistar rats were divided randomly into two groups: (1) traumatic brain injury model group (n =32) ,in which rats suffered from traumatic brain injury by Feeney's method; (2) control group ( n = 32) , rats suffered from sham operation. Each group were divided into four subgroups according 6 h, 12 h,24 h, and 48 h after operation ( n = 8, for each subgroup ). Ileum tissue were taken to observed the damage of the intestinal mucosa under microscope and electronmicroscope. The early apoptosis rate of intestinal mucosal cells were analyzed with Annexin Ⅴ-PI double stained and detected by flow cytometry. Results The intestinal mucosa were damaged and the intercellular space of intestinal mucosal were found increased in traumatic brain injury group. The early apoptosis rate of intestinal mucosal epithelial cells was significant increased in traumatic brain injury group than that in control group [6 h: (13.5 ±3.7)% vs (6.1 ± 1.7)%, P<0.05; 12 h:(66.1±6.0)% vs (5.2±1.1)%, P<0.05; 24 h:(39.8±4.8)% vs (8.4±2.6)%, P<0.05;48 h: (7.5 ±1.3)% vs (6.6 ±0.5)%]. Conclusion The increased early apoptosis rate of intestinal mucosal epithelial cells might contribute to the stress-damage of the intestinal mucosa barrier in early stage of traumatic brain injury.

Objective To study the effect of human Slitrk1 gene on proliferation and differentiation of PC12 cells. Methods The CDS sequence of Slitrk1 was amplified by PCR, and then cloned into pcDNA4 vector. The recombinant plasmid pcDNA4/Slitrk1 were transfected into rat PC12 cells by lipofectamine. The stable expression cell clones were screened by RT-PCR. MTT method was used to detect the proliferation rate of PC12 cells. The morphologic changes in PC12 cells were observed microscopically. Results The stable cell lines expressing pcDNA4 (pcDNA4/PC12) and pcDNA4/Slitrk1 (ST1/PC12) were established. Compared to pcDNA4/PC12 cells, the growth rate of ST1/PC12 cells was decreased. In addition, pcDNA4/PC12 cells tend to grow as well as the normal PC12 cells. However, most of the ST1/PC12 cells adhered to the plate with one or two neurites. Conclusion Over-expression of human Slitrk1 gene inhibited the proliferation of PC12 cells and promoted the outgrowth of neurites. It is suggested that human Slitrk1 gene may be involved in differentiation of PC12 cells.
