1.The relationship between positive experience and perceived social support of parents with a cerebral palsy ;child

Objective To investigate the relationship between positive experience and perceived social support of the parents with a cerebral palsy child. Methods 105 parents with a cerebral palsy child who were hospitalized in the Third Affiliated Hospital of Zhengzhou University between November , 2014 and January , 2015 were conveniently selected and investigated about their positive experience and perceived social support with general information questionnaire, positive aspects of caregiving and perceived social support scale. The correlation between the two was analyzed. Results The mean score of the parent′s positive experience was (30.82 ± 5.26); the dimension of self affirmation was (16.71 ± 3.06) and the dimension of life expectation was (14.10 ± 2.85). The scores of the parents′ positive experience were significantly different among gender, education of parents and the age,gender of children. The score of the parents′ positive experience was positively correlated with the total score of perceived social support (r = 0.214, P = 0.029) and the score of other support (r = 0.227, P = 0.020). Conclusions The parents′ positive experience was medium of the high level, which is worth of more attention in order to improve it. The higher perceived social support score is , the more positive experience parents will have.
2.HEPATIC STIMULATORY SUBSTANCE (HSS) FROM THE LIVERS OF NEW-BORN CALF AND RATS

The HSS found in the cytosol of new-born calf liver was heat-stable and it promoted the DNA-synthesis of guinea-pig liver cells (P/o:5.72),HSS was also found in the cytosol of new-born rat livers,and its most effective concentration to promote incorporation of H2-TdR into DNA of guinea-pig liver cells was about 0.05mg/ml.This activity was reduced with both the increase and decrease of cytosol content,and the reason was not clear.It was proved with SDS-PAGE that the molecular weight of the active component was about 14000 and 23000 daltons.The cytosol from control rat livers showed no HSS activity and also no component measuring 23000 daltons.It needs further identification that the 23000 daltons protein is the active component in the cytosol of new-born calf and rat livers and the partially hepatectomized rat livers,but not in the cytosol of control rat livers.This finding indicateds that the livers of new-born calf and rat contain highly active HSS and are a valuable resource of HSS
3.Experimental Study of Posterior Vitreous Detachment in Rabbits Induced by Hyaluronidase and Perfluoropropane(C3F8)

Objective To evaluate the effectson experimental posterior vitreous detachment(PVD) induced by intravitrous injection of hyaluronidase and perfIuoroethane(C3F8),and whether it is safe or not.Methods Twelve rabbits(24 eyes) were divided into 3 experimental groups,the contralateral eyes in same animals served as the controls.The eyes in group A(4 eyes) and B(4 eyes) were injected with 10IU of hyaluronldase(0.2ml) and 0.2ml perfluoropropane(C3F8) respectively.The eyes in group C(4 eyes) were received vitreous injections of 10IU(0.1ml) of hyaluronldase and 0.1ml perfluoropropane(C3F8) together.All control eyes were injected with balanced salt solution(0.2ml).The ocular and fundus signs were examined for 2 following weeks and then killed for histological examination.Results Four eyes in group C showed complete separation of the vitreous cortex from the retina(PVD) by B-scan and scanning electron microscopy,and no PVD was detected in group A、B and all control eyes.No evidence of retinal toxicity was revealed by light or transmisson electron microscopy in all eyes.Conclusion Vitreous injection of hyaluronidase combined with perfluoropropane(C3F8),as a safety method,can induce posterior vitreous detachment after 2 weeks.
4.Effects of original antigenic sin on the immune response to inactivated influenza A virus subtype ;H5N1 vaccine

Objective To investigate the effects of original antigenic sin caused by previous expo-sure to influenza A virus subtype H1N1 on the immune response to inactivated H5N1 vaccine. Methods In this study, the BALB/c mice were first infected with A/PR8 (H1N1) virus or immunized with inactivated vaccine to induce immune responses against the A/PR8 virus. Then they were injected once with inactivated H5N1 vaccine at dosages of 0. 01μg, 0. 1μg and 1μg, respectively. The levels of IgG and neutralizing an-tibodies in serum samples were detected after immunization. Four weeks after immunization, the mice were challenged with a lethal dose of H5N1 virus. Some indicators including the survival rate, body weight loss and residue virus titer in lung were recorded for further evaluation. Results The pre-existing anti-A/PR8 antibodies in mice didn′t alleviate the immune responses to inactivated H5N1 vaccine. Conclusion This study indicates that the original antigenic sin associated with the previous exposure to A/PR8 virus has no significant effect on the immune efficacy of H5N1 vaccine.
5.Marking on intravenous infusion stand to increase the accuracy of head-of-bed elevation for lung cancer patients after surgery cancer patients after surgery

Objective To evaluate the accuracy of head-of-bed elevation in postoperative lung cancer patients, in order to identify the compliance of patients and nurses, and to decrease the postoperative complications and restore rehabilitation. Methods 300 postoperative lung cancer patients were divided into two groups (150 patients in experimental group and 150 patients in control group) by random digital table method. The head of the beds were elevated according to the marking sign on the intravenous infusion stand in experimental group, whereas in control group nurses elevating the bed to the height that patients felt comfortable. The compliance of patients and nurses, the indwelling time of thoracic drainage tube, postoperative complication, and the length of hospital stay were recorded and analyzed. Results The compliance of nurses and patients in the experimental group were 98.7% (148/150) and 95.3% (143/150), respectively, compared to 93.3% (140/150) and 87.3% (131/150) in the control group (X2=5.56, 6.06, P<0.05). Significant difference was found in indwelling time of thoracic drainage tube between the experimental group [(96.0±27.8) h] and the control group [(103.1±24.1) h] . The length of hospital stay in experimental group was (7.9±2.1) d, compared with (8.4±2.2) d in control group (t=-2.45,-1.99, P<0.05). There was significant difference in postoperative complications between experimental group [12.0% (18/150)] and control group [20.7% (31/150)]( X2=4.12,P<0.05). Conclusions Evaluating the head of the bed accurately by the marking sign on intravenous infusion stand stand can promote the postoperative care outcome of lung cancer patients, decrease the postoperative complications and restore rehabilitation.
6.Effects of stress on the rapid component of delayed rectifier potassium current in rat cardiomyocytes

Objective To observe the effect of stress on the rapid component of delayed rectifier potassium current (IKr) in rat cardiomyocytes. Methods Forty male SD rats were randomly divided into four groups (10 each): control group (Ctrl), exhaustive group (ES), noise group (WN) and composite group (ES+WN). Stress animal models were prepared as follows: Rats in ES group were undergoing exhaustive swimming as the stress factor, in WN group undergoing white noise and in ES+WN group undergoing exhaustive swimming + white noise as the stress factor. Langendorff device was used to reversely perfuse collagenase for isolating the rat's ventricular myocytes. The effect of stress on IKr current and gating mechanism of single ventricular myocyte was recorded by whole-cell patch clamp technique. Results Compared with the Ctrl group, the tail current density of IKr (IKr,tail) of ventricular myocytes increased significantly in ES group and WN group (P<0.01). The IKr,tail current density of the ventricular myocytes in ES+WN group was significantly higher than that in ES group and WN group (P<0.01), and the effect was voltage dependent. Gating mechanism revealed that the half inactivation voltage of IKr,tail (V1/2,inact) can be shifted to the right in ES group, WN group and ES+WN group when compared with the Ctrl group, and the recovery time constant shortened after inactivation (P<0.01). However, the steady-state activation, fast inactivation constant and voltage dependence of IKr,tail were not statistically significant in ES group, WN group and ES+WN group when compared with the Ctrl group. Conclusion Stress increases the IKr current in rat cardiomyocytes, suggesting it be one of the electrophysiological bases of stress-induced arrhythmia.
7.Immunogenicity of Hib-TT conjugate vaccine in 6-59 months-old young children and the antibody persistence in 3-5 months-old infants after primary and boosting injection with Hib-TT and Act-Hib conjugate vaccines

Objective To evaluate the immunogenicity of a Haemophilus influenzae type b capsular-tetanus toxoid(Hib-TT) conjugate vaccine produced by Lanzhou Institute of Biological products(LIBP).Methods In an open-controlled,randomized trial,the eligible and consented 6-59 months-old young children injected 2 or 1 times 1 month apart with Hib-TT conjugate vaccine,the 3-5 months-old infants received 3 injections 1 month apart for primary immunization with Hib-TT or a licensed international Hib-TT conjugate vaccine as the control vaccine,and the boosting dose of two 3-5 months-old groups was injected at the 15-17months-old.The serum anti-Hib PRP IgG GMC in both groups after primary and boosting vaccination was measured by ELISA,the percentage of geometric mean concentration (GMC) ≥ 0.15 μg/ml and ≥ 1.0μg/ml was calculated,respectively.Results The Hib-TT conjugate vaccine produced in LIBP elicited satisfactory IgG antibody response in 3-59 months-old young children,the serum IgG GMC of anti-Hib PRP were 14.52 μg/ml(95% CI:12.31-17.14)in 3-5 months-old,14.04 μg/ml(95% CI:12.40-15.90) in 6-11 months-old.the ratios of IgG antibody concentration ≥ 1.0 μg/ml were 96.90% (95% CI:92.50-99.20) in study vaccine group and 98.55% (95% CI:92.20-99.90) in the control vaccine after 3 doses,respectively.100% of the 6-11 months-old young children who injected 2 times with the Hib-TT conjugate vaccine had IgG antibody concentration ≥ 1.0 μg/ml (95% CI:95.94-100.00),91.35% (95% CI:86.13-99.48) of recipients in 12-59 months-old young children induced the IgG antibody concentration ≥ 1.0 μg/ml after a single dose.The serum IgG antibody GMC in recipients who received the study or and control vaccines increased from 6.27 μg/ml (95 % CI:5.28-7.48) and 5.57 μg/ml (95 % CI:4.45-6.97)at pre-boosting injections to 63.14 μg/ml(95% CI:52.14-76.47) and 73.48 μg/ml (95% CI:57.37-94.11) one month after boosting injection,respectively.The percentage of IgG antibody concentration ≥ 1.0 μg/ml increased from 76.35% and 79.55% of pre-boosting to 100% in the two groups after booting dose.Although the serum IgG GMC in two groups appeared to decline markedly,it remained at a relatively high levels of 25.02 μg/ml (95% CI:20.51-30.48) in the study vaccine and 23.64 μg/ml (95% CI:18.40-30.43) in the control vaccine,and all of the recipients in both groups remained 100.0% of IgG antibody concentration ≥ 1.0 μg/ml.Conclusion The study vaccine elicited a protective immune response and induced the IgG antibody concentration which indicated long-term protection of anti-Hib PRP in 3 to 59months-old infants and young children.
8.Safety and immunogenicity in 3-5 months-old infants after primary and boosting immunization with Hib PRP-TT conjugate vaccine

Objectives To evaluate the safety and immunogenicity of Haemophilus influenzae type b capsular-tetanus toxoid(Hib-TT) conjugate vaccine.Methods In an open-controlled,randomized trial,the eligible and consented infants of 3 to 5 months-old received 3 doses of Hib-TT or a licensed Hib-TT conjugate vaccine(Anerbao) as the control vaccine to evaluate safety; The serum anti-Hib PRP IgG antibody mean geometric concentration (GMC) in both groups after primary and boosting vaccination were measured by ELISA.Results No apparent difference in the frequency of total adverse reactions observed between two groups (study vaccine 23.85% vs.comparator 31.40%) (x2=0.5,P>0.05).The mild and severe fever reaction of both vaccines was 3.67% and 4.48% respectively,with no significant difference.The local reactions including erythema,swelling and induration reported was 1.22% in study vaccine group.After 3 injections,the serum anti-Hib PRP IgG antibody GMC was 6.6686 μg/ml in study group and 7.5346 μg/ml in control group,with no significant difference in both of the antibody GMC (x2 =0.147,P=0.702).After one dose boosting injection,the serum antibody GMC in study group increased from 2.6396 μg/ml of preboosting to 6.2044 μg/ml of post-boosting.Conclusion The Hib-TT conjugate vaccine is proved to be safe in 3-5 months-old infants.The primary immune schedule for 3-5 months-old infants of 3 injections 1 month apart with the Hib-TT conjugate vaccine could induce IgG antibody response against Hib PRP with the GMC of long-term protection concentration in serum.A boosting dose after primary vaccination elicited obviously immunological memory.
9.Clinical study on the changes of left ventricular function and recovery after pulmonary endarterectomy in patients with chronic thromboembolic pulmonary hypertension

Objective:To assess the configuration and systolic function of the left ventricle in patients with chronic thromboembolic pulmonary hypertension (CTEPH) by routine ultrasound, two-dimensional speckle tracking imaging and three-dimensional echocardiography, and to observe the recovery after pulmonary endarterectomy (PEA).Methods:The patients who were diagnosed with CTEPH, underwent PEA and had no left heart disease were enrolled as the CTEPH group ( n=30) in the China-Japan Friendship Hospital from November 2016 and June 2021. The right heart catheterization data before and after surgery were recorded. In the meantime, gender- and age-matched healthy individuals who sought for physical examination during the same period were included as the control group ( n=23). Echocardiography findings before and after PEA were comparatively analyzed and compared between the two groups, including left ventricular end-diastolic diameter (LVEDd), right and left ventricular cross-section ratio (RVd/LVd), left ventricular global longitudinal strain (LVGLS), left ventricular end-diastolic/systolic volume index (LVEDVi/LVESVi), left ventricular ejection fraction (LVEF) and left ventricular stroke volume (LVSV). Associations between the mean pulmonary arterial pressure (mPAP)/pulmonary vascular resistance (PVR) and left ventricular function were discussed. Results:When compared with the control group, the LVEDd, LVEDVi, LVESVi, LVSV, LVGLS and the mitral early to late diastolic flow velocity ratio (E/A) in the CTEPH group were lower (all P<0.05). There were no significant differences between the two groups regarding LVEF, cardiac output (CO), and cardiac index (CI) (all P>0.05). There were no statistical differences of the left ventricular volume and LVSV between PEA group and the control group (both P>0.05), while the LVGLS and E/A remained lower (both P<0.05). Correlation analysis showed negative associations between mPAP and LVSV as well as E/A ( r=-0.490, -0.455; both P<0.05). Conclusions:There are changes in left ventricular configuration with abnormal filling pattern and potential systolic dysfunction in CTEPH patients. The PEA surgery could lead to recovery of the left ventricular configuration and volume, but the filling pattern and LVGLS at follow-up can not recover completely.
10. Echocardiographic study of right ventricular dysfunction in patients with chronic thromboembolic pulmonary hypertension: Comparison of the right ventricular free wall longitudinal strain and conventional parameters

Objective:
To compare the value of right ventricular (RV) free wall longitudinal strain (FWLS) by speckle tracking echocardiography (STE) and conventional parameters in evaluation of RV dysfunction in chronic thromboembolic pulmonary hypertension (CTEPH).
Methods:
Sixty CTEPH patients were enrolled as group A and 45 pulmonary embolism (PE) patients with normal pulmonary pressure were enrolled as group B in this study. CTEPH patients were divided into 2 subgroups using the World Health Organization (WHO) function classification: patients with WHO Ⅰ-Ⅱ were designated as group A1 and those with WHO Ⅲ-Ⅳ were designated as group A2. Conventional RV functional parameters including tricuspid annular plane systolic excursion (TAPSE), tissue Doppler-derived tricuspid annular systolic velocity (S′), fractional area change (FAC), RV index of myocardial performance (RVIMP), and STE-derived RV FWLS were measured and compared. Clinical right heart failure (RHF) was defined as the presence of symptoms of heart failure and signs of systemic circulation congestion during hospitalization.
Results:
