1.Adolescents’ Peer Attachment, Online Games Preference and Pathological Internet Use

Objective: To investigate the relationships among peer attachment, self-identity, preference of online games and pathological internet use. Methods: 404 middle school students (213 boys, 191 girls) in Beijing participated in this study, and Peer Attachment Scale, Self-identity Scale, Preference of Online Games Scale and Adolescent Pathological Internet Use Scale were used. Results: ①Peer alienation predicted PIU significantly, but peer trust and peer communication didn’t; ②The preference of online games had significant complete mediating effects on the association between peer trust and PIU, and on that between peer alienation and PIU as well; the complete mediating effect of ego identity on the relationship between peer trust, peer communication and PIU was significant. Conclusion: The study demonstrates that there are significant relations among adolescents’ peer attachment, preference of online games, ego-identity and PIU.
2.Differentiation of rabbit bone marrow mesenchymal stem cells into adipocytes in media containing dexamethasone, 3-isobutyl-1-methylxanthine, insulin and indomethacin

BACKGROUND: Adipocytes are derived from preadipocytes, which are induced by mesenchymal stem cell differentiation. This study explored the mechanism of mesenchymal stem cell differentiation into adipocytes and provided a new thinking for preventing and treating obesity.OBJECTIVE: To investigate the isolation and culture of BMSCs from rabbits in vitro, and the method of differentiating into adipocytes.DESIGN: An observational controlled in vitro experiment.SETTING: Department of Histology and Embryology, Binzhou Medical College, and Yunnan Pharmacological Laboratories of Natural Products.MATERIALS: Totally 12 healthy Japanese rabbits aged 6-8 weeks of 200 to 300g of either sex were provided by Experimental Animal Center of Kunming Medical College. Dexamethasone, 3-isobutyl-1-methylxanthine and insulin (Sigma, USA) and indomethacin (ALEXIS, USA) were used in this study.METHODS: Experiments were performed at the Yunnan Pharmacological Laboratories of Natural Products between July 2004 and January 2005. BMSCs were harvested from the femurs, tibias and humerus bones of the rabbits, and then purified and proliferated. After primary culture, the subcultured cells were cultured in media including 1μmol/L dexamethasone, 0.5mmol/L 3-isobutyl-1-methylxanthine, 10mg/L insulin and 0.2mmol/L indomethacin. The disposal was accorded with the animal ethical standards.MAIN OUTCOME MEASURES: Growth curve of the third passage of cells was evaluated by modified MTT method. Salmon pink lipid drop appeared after Oil Red O staining under a light microscope.RESULTS: After 72 hours of adipocyte induction, lipid droplet appeared in BMSCs. With the prolongation of induction time, lipid vacuoles were found and cells changed from fusiform to round or polygon. At week 3, approximately 80% BMSCs were induced into adipocytes as detected by Oil Red O.CONCLUSION: Rabbit BMSCs isolated and cultured in vitro can be induced into adipocytes in the media containing 1μmol/L dexamethasone, 0.5mmol/L 3-isobutyl-1-methylxanthine, 10mg/L insulin and 0.2mmol/L indomethacin.
3.Increased enrichment of fetal nuclear red blood cells via polyvinylidene difluoride by combined interfering membrane ionic channels

Objective The effectiveness on the enrichment of nuclear red blood cells(NRBC) in umbilical cord blood by polyvinylidene difluoride membrane was investigated with intervention on membrane ionic channels.Methods NRBC was isolated by the isolation solution with a density of 1.067 g/ml.The rates of the cell enrichment using verapamil and furosemide intervention by discontinuous density centrifugation were examined by flow cytometry.The NRBC that past through PVDF membrane with pore size of 5 ?m were checked and quantified by nested-PCR.Results The optimal centrifugation density was 1.067 g/ml.The mean purity of NRBC after centrifugation was 2.54%,yet the purity after intervention increased to 9.36%(3.7-fold increment).The NRBC passing through PVDF membrane was from 0.83% to 6.15%,with enrichment of 7.4 times.When NRBC was 120 cells/ml,the density of the nested PCR products increased by nearly 4 times after intervention(4.48 vs 17.78).Conclusions Verapamil and furosemide could affect the morphology of cells and promote the rate of NRBC enrichment.
4.Effect of silicone oil on corneal endothelium of rabbit eyes

Objective To investigate the effect of silicone oil on the corneal endothelium in experimental animal models,while the silicone oil in the anterior chamber of the experimental animals will be taken out so as to explore their corneal changes.Methods Forty healthy white rabbits were randomly divided into three groups : silicone oil in the anterior chamber group (SIG),silicone oil in and out of the anterior chamber group (SIOG) and control group(CG).The cornea of rabbit was investigated at different times by slit-lamp,corneal thickness instrument,endothelial cell count instrument,histology and electromicroscopy.The time of silicone oil in the anterior chamber without keratpathy and the pathological changes of cornea with taking out of the silicone oil were observed.Results The density of the corneal endothelium in SIOG and SIG decreased more obviously by endothelial cell count instrument from 8 weeks to 16 weeks compared with control group (P
5. Progress in aldehyde dehydrogenases and pancreatic cancer and pancreatic cancer stem cells

Pancreatic cancer is a kind of malignant tumor with high mortality rate, low diagnostic rate, and poor prognosis in the digestive system. Aldehyde dehydrogenases (ALDHs) were the intracellular nicotinamide adenine dinucleotide phosphate (NADP+)-dependent enzymes, which play an important role in the metabolism of ethanol, Vitamin A, cyclophosphamide, etc. Recent studies have found that ALDHs are involved in the regulation of many cell signaling pathways, and their expressions are related to the metastasis, prognosis, and drug-resistance of malignancies. In addition, ALDHs as common markers in normal stem cells and cancer stem cells, play an important role in the occurrence and development of pancreatic cancer and pancreatic cancer stem cells. This review discusses the expression and regulatory mechanisms of different subtypes of ALDHs family in pancreatic cancer and pancreatic cancer stem cells, and summarizes the mechanism of action of ALDHs-related anti-tumor drugs, in order to provide theoretical basis for the treatment of pancreatic cancer and the development of novel drugs.
6. Curcumin inhibits epithelial-mesenchymal transformation of human esophageal cancer KYSE70 cells

Objective: To investigate the effect of curcumin on transforming growth factor-beta 1 (TGF-β1)- induced epithelial-mesenchymal transformation (EMT) of human esophageal cancer cell line KYSE70. Methods: The proliferation of KYSE70 cells after treatment with different concentrations of curcumin for 12, 24, 48 and 72 h was detected by CCK-8 assay. After treatment with curcumin (20 μmol/L), TGF-β1 (10 ng/mL), and the combination of curcumin (20 μmol/L) and TGF-β1 (10 ng/mL), the morphology of KYSE70 cells was observed under an optical microscope, the invasion ability was measured by Transwell chamber assay, the cell cycle distribution was analyzed by FCM, and the expressions of E-cadherin, N-cadherin and vimentin mRNAs and proteins were determined by real-Time fluorescent quantitative PCR and Western blotting, respectively. Results: The proliferation of KYSE70 cells was inhibited by treatment with different concentrations of curcumin for different time (all P < 0.01). The KYSE70 cells transformed into mesenchymal cells with typical spindle-shape after treatment with TGF-β1, but this transformation could be suppressed by curcumin. The percentage of KYSE70 cells at G0/G1 phase in TGF-β1 treatment group was lower than that in the control group (KYSE70 cells without any treatment) (P = 0.021). The percentage of KYSE70 cells at S phase in the combination treatment group was higher than that in TGF-β1 treatment group (P < 0.001), and the percentage of cells at G2/M phase was lower than that in TGF-β1 treatment group (P < 0.001). The number of the invasive cells in TGF-β1 treatment group was larger than that in the control group (P < 0.001). The number of the invasive cells in curcumin treatment group was smaller than that that in the control group (P < 0.001). The number of the invasive cells in combination treatment group was smaller than that in TGF-β1 treatment group (P < 0.001). The expression levels of E-cadherin mRNA (P = 0.045) and protein (P = 0.008) in TGF-β1 treatment group were lower than those in the control group. The expression levels of N-cadherin and vimentin mRNAs (P = 0.003 and P < 0.001, respectively) and proteins (both P < 0.001) were higher than those in the control group. The expression levels of E-cadherin mRNA (P = 0.005) and protein (P = 0.006) in combination treatment group were higher than those in TGF-β1 treatment group. The expression levels of N-cadherin and vimentin mRNAs (P = 0.010 and P < 0.001, respectively) and proteins (both P < 0.001) in combination treatment group were lower than those in TGF-β1 treatment group. Conclusion: Curcumin can inhibit the proliferation and invasion of esophageal cancer KYSE70 cells, and suppress the EMT.
7.Resting-state functional connectivity in post-stroke expressive aphasia:a preliminary study

Objective To analyze the resting-state functional connectivity of the left posterior inferior frontal gyrus in normal subjects with functional MRI and to investigate its preliminary application in post-stroke aphasia.Methods Eleven male patients with post-stroke aphasia and 11 male normal subjects were recruited in this study.Resting-state fMRI data were obtained with GE 1.5 T Twin speed MR Scanner.The fMRI data were processed with SPM2, AFNI and Matlab programs.Based on the results of previous fMRI study with category member generation task , the region of interest ( ROI ) was created in the left posterior inferior frontal gyrus The time course correlations between the ROI and other voxels within whole brain were analyzed.Intra-and inter-group analysis was performed with single sample t-test and two sample t-test.Results Brain regions showed positive resting-state functional connectivity with the ROI in normal subjects including left inferior/middle frontal gyrus , left premotor cortex , left precuneus , left insula , left putamen, right inferior frontal gyrus, right cerebellum and bilateral inferior parietal lobules (voxels 33-809, t=5.80-23.55,P<0.005,uncorrected).Brain regions functionally connected with the ROI in patients with post-stroke aphasia included left inferior/middle frontal gyrus , left premotor cortex , left precuneus , left inferior parietal lobules and right inferior frontal gyrus ( voxels 34-833, t =5.08-20.34, P<0.005, uncorrected ).The left fusiform gyrus was the only region showed significantly decreased functional connectivity with the ROI in the patients group (voxels 84,t=6.25,P<0.005, uncorrected).Conclusions The neural network related to language output exists in normal subjects at the resting-state.Resting-state functional connectivity can be used to investigate the neural mechanism of development and restoration of post-stroke aphasia.
8.Functional MRI study of insula in the task-state and resting-state

Objective To study the activated status of insula during the performance of word generation task, and to explore the function of different part of insula with functional MRI (fMRI). Methods Twenty-eight subjects were recruited in this study (male 15, female 13), all of them underwent block-designed fMRI with word generation tasks and resting-state scan. SPM 5 and REST 1.3 were used to process the data. Bilateral anterior insula and posterior insula were selected as seeds to calculate the connectivity coefficiency with other voxels, and differences between the anterior and the posterior insula were compared. Results Bilateral anterior insula was significantly activated, while bilateral posterior insula was significantly deactivated. The functional connectivity areas with left anterior insula included right anterior insula, right posterior insula, supplementary motor area (SMA), left superior temporal gyrus, left middle frontal gyrus, left superior frontal gyrus, left inferior parietal lobe, middle cingulate gyrus, right striatum and right inferior frontal gyrus. The functional connectivity areas with left posterior insula included right posterior insula, left anterior insula, right superior temporal gyrus, middle cingulate gyrus, right precentral gyrus and right striatum. The functional connectivity areas with right anterior insula included SMA, left inferior frontal gyrus, right inferior parietal lobe, left inferior parietal lobe, left superior temporal gyrus, right precentral gyrus, right striatum, middle cingulate gyrus, left middle frontal gyrus, left striatum, right middle frontal gyrus, right inferior frontal gyrus and left transverse temporal gyrus. The functional connectivity areas with right posterior insula included right precentral gyrus, left superior temporal gyrus, left anterior insula, left posterior insula, right supramarginal gyrus and middle cingulate gyrus. Conclusion Anterior insula and posterior insula are functionally connected with different areas, and concerned with the language function in different manners. Left lateral anterior insula may play an important role in the language function.
9.Study on the effects of different pacing modes of cardiac resynchronization therapy on dogs with heart failure

Objective To evaluate advantages and disadvantages of different pacing modes of cardiac resynchronization therapy (CRT).Methods Twelve dogs with heart failure were performed in every dog at random,and the pacing modes employed in the test included right atrium-different sites of ventricle,and ventricular sites included right ventricular bifocal (RV-Bi),biventricular (Bi-V),left ventricular (LV).The pacing frequency was 180 times per minute,and the results were measured before pacing and after 15 minutes when the pacing became stable in Color Doppler echocardiography,including left ventricular enddiastolic diameter (LVEDd),left ventricular ejection fraction (LVEF),interventricular mechanical delay (IVMD),interventricular septum and left ventricular posterior wall motion delay (SPWMD),left ventricular 12-segment peak time standard deviation (Ts-SD).Results (1)Compared with before pacing,at the RV-Bi,Bi-V,and LV pacing modes,LVEDd,IVMD,SPWMD,and Ts-SD decreased,LVEF increased,the difference was statistically significant [(42.42 ± 3.94) mm vs (34.00 ± 4.07) mm,(34.17 ± 3.95)mm,(33.75 ±4.18)mm; (28.08 ±4.01)mm vs (13.00 ±3.64) mm,(11.95 ±2.54)mm,(12.08 ±3.51) mm; (75.00 ± 10.22)mm vs (51.75 ±9.84) mm,(20.66 ±7.41) mm,(20.75 ±7.56) mm; (25.08±4.16)mm vs (14.91 ± 3.31)mm,(7.50 ±4.24) mm,(7.41 ±3.39)mm;(32.91 ±4.46)mm vs (41.50 ±4.16)mm,(42.00 ±4.63) mm,(42.41 ±4.99)mm,P <0.05].(2)Compared with RV-Bi pacing mode,at the Bi-V,LV pacing modes,SPWMD and Ts-SD decreased,the difference was statistically significant(P < 0.05); there was no significant difference among LVEDd,IVMD,and LVEF (P >0.05).(3)There was no significant difference in LVEDd,IVMD,SPWMD,Ts-SD and LVEF between LV and Bi-V pacing (P > 0.05).Conclusions The hemodynamic effects of RV-Bi and LV pacing modes were similar to that of Bi-V pacing,and they can be used as CRT biventricular pacing alternative modes; however,the mechanisms of improving ventricular synchronization are not identical in above pacing modes.

Aim To observe the analgesic effect of oxymatrine(OMT)and its mechanism.Methods A peripheral mononeuropathy was produced in adult mice by placing loosely constrictive ligatures around the common sciatic nerve.The antinociceptive effects of the OMT were assessed in mechanical allodynia and cold allodynia tests.The CAMKII inhibitor KN-93 and AIP were adopted to investigate the influence of OMT on the analgesic effect and analyze its analgesic mecha-nisms.Western blot was used to evaluate the expres-sions of tCaMKII and pCaMKII protein.Results The intraperitoneal administration of OMT (1 60,80 mg· kg -1 )increased the paw withdrawal threshold in the
mechanical allodynia test (P <0.05 ),OMT (1 60, 80,40 mg·kg -1 ,ip)remarkably decreased the paw lifts in the cold allodynia test (P <0.05).Ith KN-93 (1 .25 μg/site),AIP (0.02 μg/site)significantly en-hanced the analgesic effect of OMT (35 mg·kg -1 ) (P <0.01 ).Protein expression of pCaMKII was de-creased by OMT(1 60 mg·kg -1 ).Conclusion OMT has significant protective effects on chronic constriction injury(CCI)in mice,and the effective mechanism of OMT inhibits the expression of CaMKII receptor.
