1.Quality Control of Dragon's Blood Spraying Film Agent

Objective To study the quality control methods for dragon's blood spraying film agent. Methods The pH value and viscosity of dragon's blood spraying film agent were detected. Drug dispersed homogeneous degree and particle sizes were determined with Nano Particle Size Analyzer and microscope. Content of Loureirin B was measured by Ultra Performance Liquid-Chromtography (UPLC). UPLC was performed on Waters C18 column (2. 1 mm×100 mm,1. 7 μm), the wavelength was 280 nm, the column temperature was 40 ℃ , and the mobile phase was 0. 1% formic acid aqueous solution and acetonitrile, and the flow rate was 0. 8 mL·min-1 . Results The pH value and viscosity of dragon's blood spraying film agent were stable, drug dispersion was homogeneous, and particle size of the drug was tiny. The concentration of Loureirin B had a good linear relationship in the range of 15. 51-77. 54 μg. Conclusion This method can be accurately controlled, has good stability and repeatability, and can fully control quality of dragon's blood spraying film agent.
3.Effects of botulinum toxin combined with treadmill training on motor and gastrocnemius function after spinal cord injury

Objective To observe the effects of botulinum toxin A(BTX-A) injection combined with treadmill training on motor and gastrocnemius function in rats after spinal cord injury (SCI),so as to develop a possible treatment.Methods A total of 48 female Sprague-Dawley rats were randomly divided into four groups(n =12 in each):a control group which received normal saline injection(group Con-NS),an exercise group with normal saline injection(group Ex-NS),the other control group with BTX-A injection(group Con-BTX)and an exercise group which also received BTX-A injection(group Ex-BTX).All rats were subjected to incomplete SCI modelling using Allen's method.Each group then had l0 members left because of death or significant weight loss.After SCI modelling,BTX-A or normal saline were injected to rats' gastrocnemius,followed by weight support treadmill training(BWSTT) on days 7 through 35 days for the two exercise groups.Motor function was evaluated using inclined plane test before and 2 days,1,2,3,4 and 5 weeks after the injury.All rats were sacrificed 35 days after the surgery.Digit Abduction Scoring(DAS) and electrophysiological testing were performed prior to sacrifice,and the general form and the wet weight of gastrocnemius were observed after resection.Results No significant differences in the inclined plane angle among 4 groups were detected before injury and 2 days afterward (P > 0.05).At the 2nd-Sth week,however,the inclined plane angles in group Ex-NS were significantly higher than those in group Con-NS (P < 0.05).Those of group Ex-NS were also significantly higher than group Ex-BTX 2-5 weeks after injury (P < 0.05).No significant differences were detected between group Con-BTX and group Ex-BTX,as well as group Con-NS and group Ex-BTX at each time point (P > 0.05).And the average DAS score in group Con-BTX and group Ex-BTX was 0,while that of Con-NS group and group Ex-NS were both 4.There were no significant differences between the two normal saline injection groups (P < 0.05),nor between the two BTX-A injection groups (P < 0.05).However,the average DAS scores in the Con-BTX and Ex-BTX groups were significantly higher than the Con-NS and Ex-NS groups (P < 0.05).No significant atrophy of gastrocnemius muscles were observed in the Con-NS group or Ex-NS group,and as would be expected,the muscles in group Ex-NS were thicker on average than those in group Con-NS.Significant atrophy was observed in group Con-BTX and group Ex-BTX.Moreover,the muscle wet weight was significantly higher in group ExNS than group Con-NS (P < 0.05).Compared with group Con-NS and group Ex-NS,the muscle wet weight was significantly lower in group Con-BTX and group Ex-BTX (P < 0.05).No significant differences in the latency of the compound muscle action potential (CAMP) were detected among 4 groups (P >0.05),but the CAMP amplitude was significantly less in the control groups than in the exercise groups.(P <0.05).Conclusions Exercise training can significantly improve motor and skeletal muscle function in SCI rats,but BTX-A injection can inhibit the improvement.
4.Clinical value of video-EEG on adrenocorticotrophic hormone therapy for infantile spasms

Objective To analyse the value of video-EEG (VEEG) in the infantile spasms (IS)children treated with adrenocorticotrophic hormone (ACTH). Methods The clinical data of 65 children with IS in our hospital from Jan 2005 to Dec 2009 were collected and the characteristics of VEEG were analyzed before and after treated with ACTH. Results All the 65 cases appeared hypsarrhythmia in the interphase before treated with ACTH. Cluster spasms and the characteristics of VEEG were recorded. After treated with ACTH,40 of 65 cases (61.5%) with epileptic seizure were controlled,in whom 27 cases (67.5% ,27/40)with hypsarrhythmia disappeared or improved obviously. The other 25 of 65 cases (38.5%) with epileptic seizure were uncontrolled,in whom only 5 cases (20. 0% ,5/25) with hypsarrhythmia disappeared. The rate of hypsarrhythmia disappeared or improved was significantly different between the epileptic seizure controlled and uncontrolled children (x2 = 13. 888, P < 0. 000). Conclusion VEEG is not only important to make a definite diagnosis for IS,but also can provide an evidence in evaluating the effect of ACTH in IS children.
5.Electroencephalography and follow-up of 128 cases of infantile convulsions with mild gastroenteritis

Objective In present study,we aimed to investigate the ictal and interictal electroencephalography (EEG) changes in patients with infantile convulsions with mild gastroenteritis,to understand the evolution of EEG and recurrence of convulsion by follow-up.Methods In this retrospective study,all patients with infantile convulsions with mild gastroenteritis visited our hospital from January 2005 to December 2009 were included,and were followed up for 3 to 7 years.All their clinical data were summarized and analyzed.Results Fist of all,we collected 128 interictal EEGs and 4 ictal (5 episodes) reports.Based on interictal EEGs,no discharge was observed in 63 subjects,and epileptic waves were discovered in other 65 patients.In most cases (48/65) epileptic waves were found to be located in the central area.For all 5 recorded ictal EEGs (2 were from same patient),epileptic waves were originated from different regions.Two months later,epileptic waves disappeared in 52/57 cases,but there were still discharge in 5 cases.Secondly,in the 103 cases who were successfully followed up,96 were seizure free,5 had recurrence during fever (3 cases) or gastroenteritis (2 cases),but remained seizure free for last 1.5 to 3.0 years.Overall,convulsion was recurred in 2 cases and finally diagnosed as epilepsy and then treated with antiepileptic drug,keeping seizure free for recent 1.5 to 2.5 years.Conclusion (1) Discharge can be found in nearly half of interictal EEGs in cases with infantile convulsion with mild gastrocnteritis,and most of them are located in central region.(2) All discharge come from local origin in ictal EEGs.(3) Seizure may recur in cases with infantile convulsion with mild gastroenteritis,especially in the presence of causative factors.Some children have risk of developing epilepsy.
6.Clinical application of vaccination with four peptides-pulsed dendritic cells in postoperative patients with malignant melanoma

Objective To investigate the safety and clinical response of dendritic cells (DC)vaccine loaded with HLA-A2-restricted peptides MAGE-3、 Tyr、 MART-1 and GP-100 against malignant melanoma. Methods Twenty three HLA-A2 positive patients with malignant melanoma were enrolled.Peripheral blood mononuclear cells were separated into adherent cells for induction of DC loaded with four peptides. DC vaccine was administered subcutaneously once a week in inguinal region. The immunological responses and clinical responses were evaluated. Results Dendritic cell vaccination were well tolerated in all patients and there was no toxicity observed. Serum levels of IL-2, IL-12 and IFN-γincreased significantly after first vaccination and fourth vaccination. The positive rate of DTH test was 50% (5/10), and 2 fold increase of CD8+ IFN-γ+ cells were observed in 6 of 10 patients. Stable disease was observed in 11 of 23 patients, one patient had a complete metastasis regression, four patients had 50% regression of metastasis,four patients had a minor response, and disease progressed in three patients. Conclusion DC vaccines loaded with peptides MAGE-3、 Tyr、 MART-1 and GP-100 can elicit non-specific and specific immune responses, leading disease control. DC vaccine is considered one of safe and effective treatments for malignant melanoma.
7.Epidemiology of uterine corpus cancer in some cancer registering areas of China from 2003-2007

Objective To analyze corpus uteri epidemiology in selected cancer registering areas of China during 2003-2007,and to provide scientific information for its prevention and control in China.Methods The incident and mortality data of corpus uteri cancer in 32 cancer registering areas of China with better quality during 2003-2007,which were selected according to the criteria of and provided by National Center for Cancer Registration,were analyzed.Results There were 8850 new cases and 1559 death cases of corpus uteri cancer,which accounted for 2.96% ( 8 850/299 306 ) of all female new cancer cases and 0.94% ( 1 559/166 305) of all female cancer death cases,respectively.Corpus uteri cancer was the 9th most common cancer for all new female cancer cases which world age adjusted incidence rates was 5.04/105,and 19th most common cancer for all female cancer death cases which world age adjusted mortality rate was 0.83/105 in 32 selected cancer registering areas of China during 2003-2007.Zhongshan city,Guangzhou city in Guangdong province and Beijing were the areas with the highest incidence rates in which were 14.51/105,8.51/105 and 6.69/105,respectively.Zhongshan city in Guangdong province,Dafeng city in Jiangsu province and Feicheng city in Shandong province were the areas with the highest mortality rates,in which were 4.03/105,3.19/10s and 1.65/105 respectively during 2003-2007.There were increasing trends for its incidence rates in above 32 areas during 2003-2007,its world age adjusted incidence rates increased from 3.94/105 in 2003 to 5.56/105 in 2007 ( P =0.026 ),while its urban world age adjusted incidence rates increased from 4.57/105 in 2003 to 6.18/105 in 2007 (P =0.038),and rural rates increased from 1.74/105 in 2003 to 3.01/105 in 2007 ( P =0.013 ),and the results showed that urban areas obviously higher than rural areas ( P < 0.01 ).Although there was a slow increasing trend for its world age adjusted mortality rates in above 32 areas during 2003-2007 which increased from 0.64/105 in 2003 to 0.87/105 in 2007 ( P =0.214 ),and from 0.66/105 in 2003 to 0.88/105 in 2007 in urban areas ( P =0.340 ),and from 0.57/105 in 2003 to 0.83/105 in 2007 in rural areas( P =0.070),while increasing trends without statistical significance.But mortality rates in urban areas were obviously higher than those of rural areas (P <0.01 ).Conclusions Although the world standardized incidence and mortality rates of corpus uteri cancer were at low level worldwide,there were increasing trends for its incidence rates during 2003-2007 in the 32 selected cancer registering areas of China.Moreover,its incidence and mortality rates were at high level worldwide in some areas such as Zhongshan city of Guangdong province and Dafeng city of Jiangsu province during the period,in which suggested that its prevention and control should be enhanced.
8.Resting-state functional connectivity in post-stroke expressive aphasia:a preliminary study

Objective To analyze the resting-state functional connectivity of the left posterior inferior frontal gyrus in normal subjects with functional MRI and to investigate its preliminary application in post-stroke aphasia.Methods Eleven male patients with post-stroke aphasia and 11 male normal subjects were recruited in this study.Resting-state fMRI data were obtained with GE 1.5 T Twin speed MR Scanner.The fMRI data were processed with SPM2, AFNI and Matlab programs.Based on the results of previous fMRI study with category member generation task , the region of interest ( ROI ) was created in the left posterior inferior frontal gyrus The time course correlations between the ROI and other voxels within whole brain were analyzed.Intra-and inter-group analysis was performed with single sample t-test and two sample t-test.Results Brain regions showed positive resting-state functional connectivity with the ROI in normal subjects including left inferior/middle frontal gyrus , left premotor cortex , left precuneus , left insula , left putamen, right inferior frontal gyrus, right cerebellum and bilateral inferior parietal lobules (voxels 33-809, t=5.80-23.55,P<0.005,uncorrected).Brain regions functionally connected with the ROI in patients with post-stroke aphasia included left inferior/middle frontal gyrus , left premotor cortex , left precuneus , left inferior parietal lobules and right inferior frontal gyrus ( voxels 34-833, t =5.08-20.34, P<0.005, uncorrected ).The left fusiform gyrus was the only region showed significantly decreased functional connectivity with the ROI in the patients group (voxels 84,t=6.25,P<0.005, uncorrected).Conclusions The neural network related to language output exists in normal subjects at the resting-state.Resting-state functional connectivity can be used to investigate the neural mechanism of development and restoration of post-stroke aphasia.
9.Screening of Film ̄forming Materials with Dragon Blood Plastics

Objective To study the film ̄forming property of film ̄forming materials in dragon blood ethanol solution, and provide the reference for screening of film ̄forming materials for dragon blood plastics. Methods PVA124,PVA1788, PVP ̄K30, EC, MC, PVB were chosen to study the film ̄forming property of film ̄forming materials based on solubility, formation time, and adhesion. Results PVA1788 and PVP ̄K30 mixed in dragon blood ethanol solution in proportion of 4:1 showed good film ̄forming property, and the membrane was smooth, uniform and easy to tear off.However, PVA124 and PVP ̄K30 mixed in proportion of 4:1 precipitated in dragon blood ethanol solution.EC, MC and PVB were not easy to tear off the membrane. Conclusion PVA1788 and PVP ̄K30 mixed in proportion of 4:1 and added with propylene glycol is the optimal choice of film ̄forming material for dragon blood spray film.
10.Functional MRI study of insula in the task-state and resting-state

Objective To study the activated status of insula during the performance of word generation task, and to explore the function of different part of insula with functional MRI (fMRI). Methods Twenty-eight subjects were recruited in this study (male 15, female 13), all of them underwent block-designed fMRI with word generation tasks and resting-state scan. SPM 5 and REST 1.3 were used to process the data. Bilateral anterior insula and posterior insula were selected as seeds to calculate the connectivity coefficiency with other voxels, and differences between the anterior and the posterior insula were compared. Results Bilateral anterior insula was significantly activated, while bilateral posterior insula was significantly deactivated. The functional connectivity areas with left anterior insula included right anterior insula, right posterior insula, supplementary motor area (SMA), left superior temporal gyrus, left middle frontal gyrus, left superior frontal gyrus, left inferior parietal lobe, middle cingulate gyrus, right striatum and right inferior frontal gyrus. The functional connectivity areas with left posterior insula included right posterior insula, left anterior insula, right superior temporal gyrus, middle cingulate gyrus, right precentral gyrus and right striatum. The functional connectivity areas with right anterior insula included SMA, left inferior frontal gyrus, right inferior parietal lobe, left inferior parietal lobe, left superior temporal gyrus, right precentral gyrus, right striatum, middle cingulate gyrus, left middle frontal gyrus, left striatum, right middle frontal gyrus, right inferior frontal gyrus and left transverse temporal gyrus. The functional connectivity areas with right posterior insula included right precentral gyrus, left superior temporal gyrus, left anterior insula, left posterior insula, right supramarginal gyrus and middle cingulate gyrus. Conclusion Anterior insula and posterior insula are functionally connected with different areas, and concerned with the language function in different manners. Left lateral anterior insula may play an important role in the language function.
