Objective To investigate the distribution and expression of adiponectin receptors in different tissues of normal Wistar rats. Methods The rats were euthanized and the total RNA was extracted from equal weight tissue samples of cardiac muscle, skeletal muscle, liver, kidney, testicle and fat. RT-PCR was employed to amplify the isolated cDNA, and the expression of adiponectin receptors in different tissues was semi-quantitatively analyzed. Objective To observe the effects of dendrobium nobile Lindl(DNL) on Nuclear factor(NF)-κB and interleukin(IL)-6 expression of renal tissue in diabetic rat model .Methods The diabetes mellitus rat model was induced by streptozotocin .All rats were divided into four groups at random :the normal control group(group N) ,diabetic control group(group diabetes mellitus) ,drug group one(group DNL1) ,drug group two(group DNL2) ,and administrated for 12 weeks .The levels of the fasting plasma glucose , urea nitrogen ,serum creatinine ,urine creatinine ,24 hours urinary protein were determined .NF-κB and IL-6 protein expression levels were measured by Western-Blot .IL-6 mRNA expression levels were measured by Real-time fluorescence quantitative PCR . Results Compared with group N ,the blood sugar ,urea nitrogen ,24 hours urinary protein of group diabetes mellitus were signifi-cantly higher(P<0 .01) ,creatinine clearance was significantly reduced (P<0 .01) ,the NF-κB、IL-6 protein expression levels and IL-6 mRNA level increased significantly (all P<0 .05) .Compared with group diabetes mellitus ,the blood sugar ,urea nitrogen ,24 hours urinary protein of group DNL1 and group DNL2 were significantly decreased(P<0 .05) ,creatinine clearance increased significantly (P<0 .05) ,the NF-κB、IL-6 protein expression levels and IL-6 mRNA level decreased significantly(all P<0 .05) .Conclusion The DNL could protect the kidney of diabetic rats ,reduce the levels of NF-κB 、IL-6 protein and IL-6 mRNA expression in renal tissue of diabetic rats .
4.Changes of Nasal resistance in obstructive sleep apneasyndrome

Objective To investigate the obstructive sleep apnea syndrome ( OSAHS) associated with nasal resistance.Methods 30 cases with obstructive sleep apneahypopnea syndrome ( OSAHS ) in the OSAHS group, 30 cases with snoring but without OSAHS in the snoring group and 30 cases with normal group based on polysomnog-raphy were detected the nasal resistance.Results The index of thsnoring index[(42.7 ±27.3),(19.4 ±18.5), AHI(38.7 ±30.2),(5.9 ±1.6)and Resl+r(1.63 ±1.11)Pa· s-1· mL-1,(0.72 ±0.31)Pa· s-1· mL-1] in OSAS group and snoring group were significantly higher than those of the normal group ( t=6.451,7.444,7.698,all P<0.05);The index of lowest oxygen saturation ( SaO2 ) ,Flowl+r in OSAS group and thsnoring group were lower than that in normal group(t=7.551,7.334,all P<0.05);There were significant differences among groups of nasal resistance (t=7.451,8.144,8.608,8.904,all P<0.05);There were positive correlation between nasal resistance and snore index (r=5.658,P<0.01);The index of nasal resistance correlated with AHI index (r=0.002,P>0.05),wake times (r=0.083,P>0.05) and lowest SaO2(r=0.105,P>0.05)were not significantly correlated;The cases with OSAS cephalometric.The results suggest majority snoring symptoms significantly more at supine posi-tion in OSAS.Conclusion Nasal resistance increased,which may be associated with the occurrence in patients with OSAHS.
5.Clinical Observation of Brucea Javanica Oil Injection Combined with Oxaliplatin and Xeloda in the Treat-ment of Elderly Patients with Advanced Gastric Cancer

OBJECTIVE:To observe the clinical short-term efficacy and safety of brucea javanica oil injection combined with oxaliplatin and xeloda in the treatment of elderly patients with advanced gastric cancer. METHODS:82 elderly patients with ad-vanced gastric cancer were randomly divided into control group and observation group. Control group was treated with Oxaliplatin injection 130 mg/m2 added into 5% Glucose injection 250 ml for 2 h,iv,d1+Xeloda tablets 1 000 mg/m2,orally,twice a day, d1-14;based on the treatment of control group,observation group was additionally treated with 10% Brucea javanica oil injection 30 ml added into 0.9% Sodium chloride injection 250 ml,iv,once a day,d1-14. 21 days were as a treatment course,and it lasted 3 courses. The short-term efficacy,life quality and toxicity reactions were evaluated in 2 groups. RESULTS:The short-term efficacy in observation group was significantly higher than control group,life quality was significantly better than control group,and the in-cidences of leucopenia and liver damage were significantly lower than control group(P<0.05). CONCLUSIONS:Brucea javanica oil injection combined with oxaliplatin and xeloda has better efficacy than oxaliplatin combined with xeloda in the treatment of elder-ly patients with advanced gastric cancer,with good safety.

7.Expression and role of PN-1, thrombin and PAR-1 in rats brain edema tissue after intracerebral hemorrhage

Objective To investigate expression and role of protease catenin-1(PN-1), thrombin(thrombin), protease activated receptor-1(PAR-1) in rats brain edema tissue after intracerebral hemorrhage(ICH).Methods Adult male SD rats 80 were randomly divided into sham group,ICH group after number, 40 in each group, ICH group autologous arterial method of making a rat model of experimental ICH in the right caudate nucleus unit injection.The degree of neurological dysfunction between 2 groups was evaluated at 12 h,24 h and 120 h after post-operation.Observed the morphology of brain cells by HE staining.Changes of PN-1,thrombin, PAR-1 index in rat brain tissue at 3,6,10,12,24,48 and 120 h were detected by Western blot.Results Neurological dysfunction score ICH rats after modeling 12,48,120 h were significantly lower than the sham group(P<0.05);ICH in rats after modeling 3,6,10,12,24,48 and 120 h of brain tissue PN-1, thrombin, PAR-1 compared with sham-operated group were significantly increased (P<0.05); ICH rat brain tissue PN-1, thrombin, PAR-1 appeared in 12 h after modeling shwed a gradual downward trend (P<0.05). Conclusion ICH hematoma surrounding brain tissue in rats PN-1 has the effect of inhibiting thrombin and PAR-1 overexpression, cause nerve damage.
8.The study of thalamic 1H-MRS imaging characteristics in patients with cerebral small vessel disease and its corralation with cognition

Objective To study the thalamic metabolic alterations and its correlation with cognitive impairment in patients with cerebral small vessel disease(CSVD).Methods The cognitive function of 34 patients with CSVD and 26 matched volunteers were evaluated by Montreal cognitive assessment(MoCA), and received single voxel 1H-MRS examination to detect the content of NAA,Cho and Cr,and record the ratio of NAA/Cr and Cho/Cr on bilateral thalami.The differences of NAA/Cr, Cho/Cr on bilateral thalami between the two groups were compared, and the correlation between NAA/Cr,Cho/Cr and MoCA total score and its sub-items score in CSVD group were analyzed.Results ①The MoCA of total score as well as its sub-items such as visual space and executive ability,memory,attention and language for CSVD group were significantly lower than that for the control group(P<0.05);②NAA/Cr on bilateral thalami in CSVD group were both lower than that in the control group(left 1.57±0.18,1.68±0.17,t=2.46,P=0.02;right 1.66±0.21,1.78±0.19,t=2.23,P=0.03), the differences were statistically significant (P<0.05);there were no significantly differences in the ratio of Cho/Cr between the two groups(P>0.05);③In CSVD group, the ratio of NAA/Cr on both bilateral thalami were significantly positively correlated with MoCA score (left r=0.83,right r=0.79,P<0.05)as well as visual space and executive ability(left r=0.65,right r=0.46,P<0.05), memory(left r=0.59, right r=0.50, P<0.05), attention(left r=0.42, right r=0.52, P<0.05),language(left r=0.52, right r=0.41, P<0.05), abstraction(left r=0.47, right r=0.40, P<0.05), orientation(left r=0.48,right r=0.42, P<0.05),Cho/Cr were not significantly correlated with MoCA total score and its sub-items(P>0.05).Conclusion The thalamic neuron has been damaged and dysfunctioned in patients with CSVD,and this metabolic abnormality may be related to a wide range of cognitive impairment in CSVD patients.

Objective:To explore the characteristics of the ultraweak luminescence of human hepatocarcinoma cells(HepG2). Methods: The intensity of the ultraweak luminescence of HepG2 cells and human hepatocytes(QZG)was examined by an IFFM-D chemiluminescence instrument.The intensity of the ultraweak luminescence of HepG2 cells and human hepatocytes(QZG)were affected by different concentration of Luminol and H_(2)O_(2). Results: In 10~(-4)mol/l Luminol and 0.3% H_(2)O_(2),the intensity of the ultraweak luminescence of HepG2 cells was significantly higher than that of QZG cells(P)0.05). Conclusion:The intensity of the ultraweak luminescence of HepG2 cells is higher than that of QZG cells.The intensity of the ultraweak luminescence is affected by cell number and the concentration of Luminol and H_(2)O_(2).The ultraweak luminescence reflect functional status of tumor cells.It can become an sensitive index of physiology and metabolism of tumor cells.

Objective To observe the effect of arsenic trioxide on the apoptosis of synovium and IL-l, TNF-? in CIA rat. Methods Seventy-two rats were divided into normal control, model, and As2O3 treatment groups. The experimental models of collagen induced arthritis rats were used as the experimental moldels. The knees′ synovium, cartilage and bone tissue of the rat were taken out, then observed with light microscope and electron microscope and apoptosis were measured by TUNEL after the 15th day of treatment. Meanwhile the level of IL-1 and TNF-? were measured with ELISA kit. Results The pathological injury were improved and the apoptosis of synoviocytes were increased in the As2O3 treatment group, compared with the model group.
