2.Practice of designed experimental assessment in bio-analytic chemistry

By evaluation of academic designed experiment performance in Bio-Analytic Chemistry from ten different evaluating indicators including literature search, experimental design, and experimental report and so on. The quantitative analysis results indicate the average is 87.8, the standard deviation is 5.0,the difficulty index is 0.88,the dipartite degree is 0.12 and the confidence coefficient of the assess is 0.9423. The designed experimental assessment can inspire the students’experimental goaheadism, cultivate their ability to analyze and solve problems,and promote their innovative consciousness and practice skills.
3.Study on the relationship between sex hormone and lipid metabolism in overweight and obese boys aged 9 to 12 years

Objective:To explore the specific changes and correlations of sex hormones and lipid metabolism in overweight and obese boys aged 9-12 years.Methods:Seventy-two male subjects (9 to 12 years old) were divided into normal weight group ( n=42), overweight group ( n=15), and obese group ( n=15). Plasma sex hormone levels and lipid levels were detected by chemiluminescence immunoassay and ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC/QTOF/MS). All data were analyzed by multivariate statistics analysis. Results:(1) The serum concentration of sex hormone-binding globulin (SHBG) in obese group and overweight group was significantly lower than that in normal weight group, and the serum concentration of SHBG in obese group was significantly lower than that in overweight group ( P<0.05). The serum concentrations of dehydroepiandrosterone (DHEA) in overweight and obese group were significantly higher than those in normal weight group ( P<0.05). (2) There were 171 kinds of differential lipid compounds between normal weight group and overweight group, 218 species of differential lipid compounds between normal weight group and obese group, and 34 species of differential lipid compounds between overweight and obese group. Among them, 150 kinds of lipid compounds in normal weight group were significantly different from those in overweight and obese group. (3) SHGB was positively correlated with phosphatidyl cholines (PC) (20∶0/0∶0) and PC[18∶1(9E)/0∶0]( r=0.6, P<0.05), DHEA was positively correlated with diacylglycerol (DG)[17∶2(9Z, 12Z)/22∶0/0∶0]( r=0.5, P<0.05). Conclusions:9-12 years old overweight and obese boys had early growth and development, but sex hormone levels were not synchronized with growth and development. The serum DHEA level in overweight and obese boys was significantly increased, while the level of DG [17∶2 (9Z, 12Z)/22∶0/0∶0] was significantly increased, and the serum SHGB concentration was significantly decreased, while the vast majority of PC was significantly down-regulated. The mechanism of these changes needs to be further studied.

Objective To study the association of ER? expression with the expression of hormonal receptor and prognostic biomarkers.Methods The expressions of ER?,ER?,PR,Ki-67,HER-2/neu and p53 were detected in tissue samples from 109 breast cancer patients by immunohistochemistry on tissue microarray slides.The histologic grade,tumor size and lymph node metastasis status of 109 cases of breast cancer were recorded and served as comparison indexes.Results ER? was expressed in benign epithelium and stroma cells,and breast cancer cells in 59% breast cancer tissues of different histologic types.ER? was coexpressed with ER? in 45% breast cancer tissues.ER? expression was found a statistically significant association with HER-2/neu(P
5.A model-based meta-analysis to compare urate-lowering response rate of febuxostat and allopurinol in gout patient.

This study aims to compare the urate-lowering response rate of febuxostat and allopurinol in gout patient using a model-based meta-analysis. The literature search identified 22 clinical trials of gout with a total of 43 unique treatment arms that met our inclusion criteria, and a total of 6 365 gout patients were included in the study. The response rates of allopuriol and febuxostat were characterized by Tmax model and Emax model respectively, and the effect of baseline serum uric acid (sUA) and patient type on the drug effect was tested. The results showed that allopurinol can reach an average maximum response rate of 50.8% while febuxostat can reach a 100% response rate within a very short time, and the ED50 was 34.3 mg. Covariate analysis revealed that baseline sUA has a negative effect on response rate of allopurinol, and a positive effect on the predicted ED50 of febuxostat. For patients who had shown inadequate response to prior allopurinol treatment, the average response rate was about half that of the allopurinol responder patients.

After percutaneous coronary intervention (PCI) was performed,since CHD risk factors still exist, coronary restenosis rate remains high.Therefore, self-management after PCI is very important.The present article made a review on current condition and research progress of self-management in patients after PCI, aiming at providing reliable evidence for rehabilitation after PCI.
7.Reversal of Multidrug Resistance by IFN-? in KBv200 Cell Lines

Objective: To explore the effects of various concentrations IFN-? on human oral epidermic cancer cell line KB and its multidrug resistant counterpart KBv200.Methods: The toxicity of IFN-? and/or VCR was assayed by MTT method; intracellular rhodamin123 concentration and p-glycoprotein expression were measured by flow cytometry; mdr-1 mRNA was assayed by RT-PCR. Results: High-dose IFN-?(10 000 IU/ml) had significant antiproliferative effects on KB/KBv200 cell lines. Low-dose IFN-? did not have any significant effect on growth of KB/KBv200 cells, but did increase the cytotoxic effect of VCR and the accumulation of Rhodamine123 in KBv200 cell line and had no effect on parent VCR-sensitive KB cell line. IFN-? down-regulated expression of P-gp and mdr-1mRNA . Conclusions: IFN-? has significant effect on the reversal of multidrug resistance of KBv200 cell line via a mechanism of P-gp and mdr1 mRNA down-regulation.
8.The application of whole-procedure seamless nursing intervention for treatment of patients with acute coronary syndrome

Objective To investigate the feasibility and effects of whole-procedure seamless nursing intervention during regional collaborative treatment of patients with acute coronary syndrome.Methods Nursing intervention was performed on pre-hospital collaboration,transfer collaboration and catheter room collaboration during regional collaborative treatment of patients with ACS.Treatment time point,therapeutic effects and major hospitalization indicators were compared before(the control group) and after(the experimental group) implementation of nursing intervention.Results There were significant differences in mean FMC-to-B time,D-to-B time,referral time,obtaining informed consent time,mortality,LVEF and LVED between two groups(P<0.05).There were significant differences in days of hospitalization,expenditures,percentage of consumables,percentage of medication,and in-hospital mortality between two groups (P<0.05).Conclusion Whole-procedure nursing intervention can reduce time of regional collaborative treatment of patients with acute coronary syndrome,improve prognosis,decrease financial burden and increase efficiency of ACS treatment.
9.The effects of consecutive dietary nursing intervention on elderly type 2 diabetes patients

Objective To evaluate the effect of consecutive diet nursing intervention on elderly patients with type 2 diabetes. Methods 56 elderly type 2 diebetes patients were randomly selected in control group and were given conventional type 2 diabetes management. The other 56 patients in experimental group were not only given routine care, but also 3 months of consecutive diet nursing intervention. At the 1st and 3rd month, assessment of blood glucose test and quality of life were conducted by all patients conducted. T test, chi-square test were used in the statistics. Results In experimental group, the controlling effect of fasting plasma glucose and 2-hour postprandial blood glucose have been significantly improved. 3 months after the consecutive diet nursing intervention, fasting plasma glucose of control and experimental groups were (7.18±0.89) mmol/L and (6.37±0.74) mmol/L (P=0.027). After 1 months of the consecutive diet nursing intervention, 2-hour postprandial blood glucose of control group and the experimental group were (11.69 ± 1.58) mmol/ L and (9.03 ± 2.13) mmol/ L (P = 0.028) respectively. After 3 months of intervention, the number were were (12.12±2.36) mmol/L and (8.36±1.65) mmol/L respectively (P<0.01). In the experimental group, the therapeutic dimension of quality of life has been gradually decreased and the difference was statistically significant. Conclusions Consecutive diet nursing intervention can effectively improve the blood glucose control of elderly type 2 diabetes patients.
10.Bactericidal Activity of Recombinant Human Platelet Phospholipase A_2 in Vitro Against Staphylococcus Aureus and Its Impact Factors

OBJECTIVE:To evaluate the bactericidal activity of recombinant human platelet phospholipase A2(RHP-PLA2)in vitro against S.aureus and the effects of different impact factors on the bactericidal activity of it.METHODS:The bactericidal activity of RHP-PLA2 was tested by agar plating method,and the impact factors(pH,Ca2+,Mg2+,negatively charged heparin)were determined.RESULTS:The minimum bactericidal concentration(MBC)of RHP-PLA2 killing 99% S.aureus was 0.08 ng?L-1.The bactericidal rates of PLA2 at MBC were 44.4%,99.8% and 99.5% respectively in the pH of 5.0,7.4 and 9.0.1 ~5 mmol Ca2+ was suitable for the bactericidal activity of the RHP-PLA2.The bactericidal activity of RHP-PLA2 can be completely inhibited by 5.12 mmol?L-1 EGTA,10 mmol?L-1 MgCl2 or 0.30?10-4 IU?L-1 heparin.CONCLUSIONS:The bactericidal activity of RHP-PLA2 is affected by pH and Ca2+ concentration in media.The cationic property of PLA2 is the key factor influencing its bactericidal activity.
