2.Relationships between concentrations of free fatty acid in serum and oxidative-damage levels in placental mitochondria and preeclamupsia

Objective To investigate the relationships between concentrations of free fatty acid (FFA) in maternal serum and oxidative damage levels in placental mitochondria and preeclampsia ( PE)-Methods A total of 60 women with PE and 60 normal pregnant women as control participated in this study.All were admitted to Fujian Maternity and Child Health Hospital for delivery from August 2010 to May 2011.Patients with PE were divided into early-onset group ( n =30,presented at < 34 weeks of gestation ) and late-onset group ( n =30,presented at ≥ 34 weeks of gestation),with 30 normal pregnant women as early control group ( < 34 weeks of gestation ) and 30 as late control group ( ≥34 weeks of gestation).Improved copper agent colorimetry was used to detect FFA in maternal serum Ultraviolet colorimetry was used to detect glutathione peroxidase (GPX) and catalase (CAT) activity in maternal placenta and malondialdebyde (MDA) and permeability transiton (PT) pore in placental mitochondria.Total superoxide dismutase (SOD) assay kit-WST was used to detect SOD activity in placenta.Real-time fluorescent quantitative PCR was used to detect mitochondrial DNA (mtDNA) expression in placenta.Results ( 1 ) Maternal serum FFA was ( 1.6 ±0.5 ) mmol/L in early-onset PE group and ( 1.5 ± 0.4) mmol/L in lateonset PE group,significantly elevated as compared to ( 1.0 ± 0.5 ) mmol/L in early control group and (0.9 ±0.5) mmol/L in late control group (P < 0.05 ). However,no significant difference was found between early-onset and late-onset PE groups (P > 0.05 ).(2) The mean placental GPX,CAT and SOD activity were significantly decreased in the early-onset PE group [ (47 ±6),( 19 ±5),(62 ± 13) U/mg]and late-onset PE group [ (67 ±6),(20 ±4),(96 ± 17) U/mg] as compared to late control group [ (80 ±3),(55 ± 3 ),( 123 ± 19 ) U/mg],respectively ( P < 0.05 ).(3) The mean placental mitochondria MDA was significantly elevated in the early-onset PE group [ (115 ± 22) nmol/mg] and late-onset PE group [(90±17) nmol/mg] as compared to late control group [(52 ± 11) nmol/mg,P <0.05].The mean absorption value that present the permeability of placental mitochondria PT pore was significantly elevated in the early-onset PE group (0.086 ±0.013) and late-onset PE group (0.069 ±0.014) as compared to late control group (0.052 ± 0.0 12,P < 0.05 ).The mean placental mtDNA expression was significantly elevated in the early-onset PE group (3.0 ±0.7) and late-onset PE group (2.8 ±0.7) as compared to late control group ( 2.6 ± 0.6,P < 0.05 ).( 4 ) The mean placental mitochondria MDA concentration correlated positively with the concentrations of FFA in maternal serum in the early-onset PE group ( r =0.703,P <0.05 ) and late-onset PE group (r =0.457,P < 0.05 ),and negatively with placental antioxidant enzyme in the early-onset PE group ( r =- 0.652,- 0.787,- 0.952 ; P < 0.05 ) and late-onset PE group ( r =-0.378,-0.689,-0.854; P<0.05).Conclusions Increased FFA in maternal serum and high levels of oxidative damage in placental mitochondria may be involved in the pathogenesis of preeclampsia.Increased FFA in serum and decreased activity of antioxidant enzyme in placenta may contribute to oxidative damage levels in placental mitochondria in women with PE.
3.Analysis on blood test result of voluntary blood donors in Fuling District during the past seven years

Objective To get the information about the infection condition of blood in voluntary donors in Fuling District from 2001 to 2007, so as to take measures to improve blood quality and ensure blood safety.Methods Serum ALT was detected with Reitman-Frankel assay, and HBsAg, anti-HCV, anti-HIV and anti-TP were detected with ELISA.Results A total of 72 868 serum samples from blood donors were detected, in which 5 512 (7.56%) were positive. The positive rates of HBsAg, anti-HCV, anti-HIV and anti-TP were 3.09%, 0.48%, 0.07% and 1.27% respectively, and the abnormality rate of ALT was 2.73%. There was statistical difference of positive rate among various years (from 2001 to 2007).Conclusion The positive rate of blood in voluntary donors decreases year by year. HBsAg was the highest (1.16%), followed by ALT (2.73%), anti-TP (1.27%), anti-HCV and anti-HIV. It suggests that blood quality monitoring should be strengthed to assure blood safety.
4.Effect of peroxisome proliferator activated receptor γ agonist on the proliferation and differentiation of rat osteoblasts in vitro

Objective To study the effect of rosiglitazone (RSG), the agonist of peroxisome proliferator activated receptor γ (PPARγ), on the proliferation and differentiation of rat osteoblasts and the related mechanisms. Methods The identification of rat primary osteoblasts was performed by alkaline phosphatase (ALP) staining and mineralized nodules. The 3-(4,5)-dimethylthiahiazo (-z-y1)-3,5-di-phenytetrazoliumromide (MTT) assay and p-nitrophenyl phosphate (PNPP) assay were used to observe the effects of different concentrations of RSG on proliferation and differentiation of the osteoblasts. The reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of connective tissue growth factor (CTGF) mRNA. The effects of the different concentrations (0,1,2,5,10 and 20 μmol/L) of RSG on TGF-β1-induced CTGF mRNA expression in osteoblasts were detected. Results (1)Different concentrations of RSG could not change the proliferation of osteoblasts (P>0. 05). (2)Compared with control group, all different concentrations of RSG could suppress ALP activity in osteoblasts (P<0. 01 ). (3) RSG suppressed the osteoblats CTGF mRNA expression induced by TGF-β1 in a dose-dependent manner (P<0. 01). Conclusions In vitro, RSG can inhibit the TGF-β1 induced rat osteoblasts CTGF mRNA expression. RSG may play a potential role in preventing the differentiation of the rat osteoblasts.
5.Diagnostic significance of humoral and cellular immune function detection in patients with syphilis

Objective To investigate and analyze the diagnostic significance of humoral and cellular immune function detection in the patients with syphilis.Methods Eighty-six cases of syphilis without anti-syphilis treatment in our hospital were selected as the research subjects(syphilis),including 26 cases of primary syphilis,39 cases of secondary syphilis 21 cases of latent syphilis,and 30 healthy subjects were selected as the control group.Then the humoral immunity and cellular immunity levels were compared between the two groups.Results The levels of IgG,IgA and IgM in the syphilis group were significantly increased compared with the control group(P<0.05).The IgG,IgA and IgM levels had no statistical difference between the primary syphilis patients and control group(P>0.05),which in the patients with secondary syphilis and latent syphilis were significantly increased compared with the control group,moreover the increase in the secondary syphilis was most significant(P<0.05);the CD3+,CD4+,CD4+/CD8+ and NK levels in different stages of syphilis were significantly decreased compared with the control group(P<0.05),while the CD8+ level was significantly higher than that in the control group(P<0.05),in which the change in the secondary syphilis patients was most significant.Conclusion The humoral immune in the patients with syphilis is hyperfunction,while cellular immune is dysfunction,the patients present the overall immune function disorder,therefore the humoral and cellular immunity detection has a certain clinical value in the diagnosis of syphilis.
6.Effect of latanoprost combined with timolol treatment on visual function, intraocular pressure and ocular blood flow of open angle glaucoma

Abstract?AlM: To study the effect of latanoprost combined with timolol treatment on visual function, intraocular pressure and ocular blood flow of open angle glaucoma.?METHODS:A total of 50 cases ( 59 eyes ) with open angle glaucoma were enrolled from January 2012 to May 2014 in our hospital department of ophthalmology, and divided into observation group and control group by adopting the random number table method. Patients in observation group were treated with latanoprost combined with timolol, patients in control group were treated only with timolol. Visual function, intraocular pressure and ocular blood flow were compared.?RESULTS:1, 2, 3 and 4wk after treatment, observation group during the day intraocular pressure, night intraocular pressure were significantly lower than that in control group, vision levels (0. 27±0. 03, 0. 36±0. 06, 0. 44± 0. 06, 0. 63 ± 0. 13 ) were significantly higher than that in control group; observation group peak systolic velocity (14. 41± 1. 73) cm/s, end diastolic velocity (4. 18 ± 0. 67) cm/s were significantly higher than that in control group;vascular resistance index ( 0. 58 ± 0. 07 ) was significantly lower than that in control group.? CONCLUSlON: Latanoprost combined with timolol treatment can reduce intraocular pressure, increase blood flow of central retinal artery, reduce vascular resistance, improve visual acuity.
7.Efficacy of Foley balloon combining with uterine adhesions to prevent the recurrence of intrauterine adhesions after resection of intrauterine adhesions

Objective To observe the efficacy of Foley balloon combining with uterine adhesions to prevent the recurrence of intrauterine adhesions after resection of intrauterine adhesions.Methods Patients underwent resection of intrauterine adhesions were randomized into two groups. 30 patients in control group underwent the therapy of uterine adhesions to prevent the relapse intrauterine. On the other hand, the study group with 30 patients were placed Foley balloon for 5 ~ 7 days and combined with the therapy of uterine adhesions to intervention for 1 ~ 2 weeks. All patients were followed-up for 1 ~ 2 months, and the result of the intrauterine adhesion were assessed by hysteroscopy.Results The rate of the relapse in control group was 3.33%; the study group had the same result (the rate was 3.33%). There was no difference between these two groups.Conclusions The rate of recurrent intrauterine adhesions is no different between the control group of only use therapy of uterine adhesions and replacement of Foley balloon combining with therapy of uterine adhesions as intervention.
8.Regulation of paclitaxel on vascular endothelial related factors of serum and tissues in patients with uterine cancer

Objective To study the regulatory effect of paclitaxel injection on vascular endothelial related factors in serum of uterine cancer patients.Methods 30 cases of uterine cancer patients treated with paclitaxel (observation group)and 30 cases of uterine cancer patients treated with surgery (control group)were selected.Chemiluminescence immunoassay (CLIA)was used to detect expressions of serum tumor necrosis factor(TNF), Interleukin-8(IL-8)and tumor specific growth factor (TSGF).Results Before treatment,there was no significant difference between the serum TNF, IL-8 and TSGF in observation group and control group(P<0.05).At 3 months after treatment,the serum TNF,IL-8 and TSGFwere significantly lower than before treatment ,which was statistically significant(P<0.05).Conclusion Paclitaxel can decrease serum TNF,IL-8 and TSGF in patients with uterine cancer,which provides a new theoretical basis for its anticancer effects.
9.Characteristics and prognosis of pulmonary infection in 83 elderly patients in emergency department

Objective To study the characteristics and prognostic analysis of 83 cases of pulmonary infection in elderly patients in emergency department.Methods Thirty-eight elderly patients with pulmonary infection from January 2013 to December 2015 in our hospital were retrospectively analyzed.Results The number of neutrophils increased in 56 patients(67.46%), the neutrophil-lymphocyte ratio increased in 69 patients(83.13%), and C-reactive protein increased in 74 patients increased(89.16 %).There was significant difference between C-reactive protein detection and neutrophil count(P<0.05).61 cases(73.49%) were complicated with other diseases.While the effect of a single antibiotic treatment is less effective.Conclusion The early symptoms of pulmonary infection in the elderly are easy to ignore, and often associated with other diseases.C-reactive protein, neutrophil-lymphocyte ratio combined with lung imaging can be more accurate response to the elderly pulmonary infection.Early detection and early treatment, combined use of antibiotics can improve the prognosis.
