1.The effect of tamoxifen(TAM)in the chemotherapy of hepatocellular carcinoma(HCC)and its mechanism.
Chinese Journal of Practical Internal Medicine 2006;0(S1):-
Objective To investigate The effect of TAM(2.5 uM)in the chemotherapy of HCC and its mechanism. Methods An adriamycin-resistant human HCC cell subline(Hep-3B/ADM)was established, through exposure to gradually increased concentration of ADM.Drug sensitivity was measured by MTT.Modulation of ADM-induced apop- tosis of Hep-3B/ADM cells was assessed by Flow cytometry(FCM).PKC activity was determined by applying compet- itive protein binding method. Results TAM(2.5uM )decreased the IC50 of ADM from 1.64?g/mL to 0.23?g/mL in the Hep-3B/ADM cells.TAM exhibited a 2-fold potentiation on apoptosis caused by ADM in Hep-3B/ADM cells. With TAM,PKC activity was lower than that in control groups from 30 seconds to 24 hours. But this effect of TAM could be reversed by the co-treatment of the cells with TPA.Conclusion TAM(2.5 uM)significantly en- hanced ADM-induced cytotoxicity. The mechanism is that TAM potentiates the apoptosis induced by ADM through pro- tein kinase C inhibition.

Objective To investigate how to prevent deep-vein thrombosis(DVT) after total knee replace-ment(TKR).Methods Deep vein thrombosis in 87 patients after TKR from 2004 to 2007 in the Third Hospital Affiliated to Suzhou University was retrospectively analyzed. Results Amony the 87 patients,13 were found having deep vein thrombosis,5 of 38 using low-molecular-weight beparin after TKR having deep vein thrombosis,6 of 35 cases using aspirin after TKR having deep vein thrombosis,5 of the 10 eases using mechanical preventive measures having deep vein thrombosis. The difference between low-molecular-weight heparin and aspirin group was not statistically significant. Conclusion Using low-molecular-weight heparin before TKR can prevent the occurrence of deep-vein thrombosis.Using epidural anesthesia, and a low-molee-ular-weight heparin drugs or aspirin after TKR can better prevent the formation of deep-vein thrombosis.

Objective To investigate the mechanism of acute patella bone cartilage fracture, and further study its diagnosis and treatment of clinical significance. Methods Collecting 21 cases of the acute patella bone cartilage fracture from July 2005 to December 2007.Eleven patients were male and ten were female, with the mean age of 31.6 (16~46) years. All the patients were treated using the arthroseopic microfracture technique after MR examination. Results All patients were followed up 6 to 18 months, did not show patellofemoral joint pain. Evaluated by Tagner rating, excellent outcomes were found in 17 cases, good in 4 cases, satisfactory results in 100% cases. Conclusion The arthroscopic microfracture technique in treatment of the patella cartilage fracture method is simple, easy to operate, can significantly improve knee function and pain,as well as the quality of life.

[Objective] To determine the effects against human simplex virus type I(HSV-1) and adenovirus type Ⅲ(ADV-3) anti-viral in vitro and in vivo of eye drop prepared by mixed extract of Heartleaf houttuynia and Lonicera japonica.[Methods] By using different numbers of host cells with cytopathic effect(CPE) as the observation index, models of HSV-1 infected Vero cells and ADV-3 infected HeLa cells were established to determine the anti-viral effects in vitro of HLD with different dosages.Rabbit eye conjunctivitis model infected by HSV-1 was established to observe the anti-viral effects in vivo of HLD with different dosages, and the biopsy specimens of pathogenic eye conjunctive tissue were examined by routine pathological methods. In these experiments,a commercial acilovir eye drop(ACV) was used as the control.[Results] The minimum dosages of HLD to completely inhibit the CPEs of HSV-1 and ADV-3,the host cells were 32 mg/ml and 64 mg/ml respectively. After treatment with HLD,the inflammatory reactions of rabbit eye conjunctivitis caused by HSV-1 infection were obviously weak or disappeared. The curative rates of 1,3 and 6 g/ml HLD treating experimental rabbit eye conjunctivitis were 50.0%,83.3% and 100% respectively.Furthermore, the curative effect using 6 g/ml HLD has no significant difference compared to that using ACV drop.[Conclusion] HLD has a certain anti-viral effect in vitro and a stronger anti-viral effect in vivo, indicating the potential of HLD for developing a new drug to prevent or treat viral eye conjunctivitis.

Journal of Third Military Medical University 2003;0(10):-
Objective To isolate and identify advanced oxidation protein products from human serum albumin (AOPP-HSA), expecting to search for a method of preparing highly purified and bioactive AOPP-HSA. Methods AOPP-HSA crude products were prepared in vitro by exposing HSA to HOCl. AOPP-HSA was isolated by gel chromatography and ion-exchange HPLC. Its structural features and biological activities were characterized by UV and fluorescence spectrum, SDS-PAGE, and the experiment of TNF-? release from monocytes. Results The isolated protein was purified up to 99.4% and was dityrosine-containing protein cross-linking products with molecular weight of 700?10 3. It possessed the ability of triggering the considerable release of TNF-? from monocytes. Conclusion Highly purified and bioactive AOPP-HSA can be successfully prepared by above-mentioned two-step chromatography from AOPP-HSA crude products, which builds a basis for further study of AOPP.
7.Antibody of Salmonella typhi Detected by Gel Technique
Chinese Journal of Nosocomiology 2004;0(10):-
OBJECTIVE To study the meaning of detecting the antibody of Salmonella typhi by gel technique(GEL). METHODS Minigel column technique(MGCT) and Widal reaction(WR) were used to detect the antibody among 32 suspicious typhoid and paratyphoid patients. RESULTS Among them, the titers of various kinds of antibody of S.typhi "O","H","?","?",and "?" with GEL were 1-2 orders higher than with WR.There was a significant difference in statistics. CONCLUSIONS It is convenient and fast to detected the antibody of S.typhi.And it can give important reference data for the clinic.

Objective To study the morphological changes, human bitemarks were made on the living and dead canine skin. Method The changing patterns of the human bitemarks were recorded with morphological parameters measured. The relationships among the patterns of the bitemark, tooth area, time, ocdusal force, tooth width and thickness were analyzed by multiple progressive regression. Results The multiple progressive regression equations of the morphological changes of human bitemark were established: S=-6.96-1.68?10 2T-0.11F +2.21W+3.75H(Live dog test,T

Chinese Journal of Forensic Medicine 2002;0(06):-
Objective Digital analysis of human bitemarks by blind method was assessed for its accura-cy. Method Digital analysis was used in double-blind method for identification of human bitemarks with control samples from 8 suspects. Photoshop 5.5 generates an overlay from bitemarks scanned and various coefficients were submitted for analysis by AutoCAD R14 engineering software to compare bitemarks with denture of suspects. Results Digital analysis shows that bitemarks tally with cast of denture from "suspects" in every coefficient. Conclusion Digital analysis is feasible for identification of experimental bitemarks and promises sound prospects in future forensic practice.
