1.Clinical observation on 4 cases undergone radical abdominal trachelectomy

Objective To perform radical abdominal trachelectomy on cases with cervical tumors for the removement of tumors and retain of fertility. Methods 4 cases with cervical tumors were performed radical abdominal trachelectomy in order to reserve the uterine function. Results Menstruation of all the 4 cases recovered after operation .Postoperative follow-ups revealed good results. Conclusion Radical abdominal trachelectomy can reserve the uterine function of patients with cervical tumors.However,further clinical observation needs to be performed to compare its survival rate with that of traditional modi operandi.
2.Effect of Bushen Huatan Recipe on the Akt signal pathway in polycystic ovarian syndrome model rats with insulin resistance: an experimental research.

OBJECTIVETo observe the effect of Bushen Huatan Recipe (BHR) on the Akt signal pathway in polycystic ovarian syndrome (PCOS) model rats with insulin resistance (IR).
METHODSFifty Wistar female PCOS rats were randomly and equally divided into 5 groups, i.e., the control group, the model group, the low dose BHR group (5.406 g/kg), the medium dose BHR group (10.812 g/kg), and the high dose BHR group (21.624 g/kg), 10 in each group. Levels of fasting blood glucose (FBG) and insulin were detected to calculate homeostasis model assessment of insulin resistance (HOMA-IR). The expression of insulin receptor substrate 1 (IRS-1), glycogen synthetase kinase-3beta (GSK-3beta), glucose transporter 4 (GLUT-4), and peroxisome proliferator activated receptor (PPAR-gamma) mRNA were detected by RT-PCR. The expression of insulin signal transduction molecular kinase B (Akt) was detected by Western blot.
RESULTSCompared with the control group, HOMA-IR and the mRNA expression of PPAR-gamma mRNA significantly increased, the mRNA expression of GSK-3beta, GLUT-4, and IRS-1, protein expression of Akt and p-Akt significantly decreased in the model group (P < 0.05). Compared with the model group, HOMA-IR significantly decreased, the mRNA expression of GSK-3beta, GLUT-4, IRS-1, and Akt protein significantly increased in the high dose BHR group (P < 0.05, P < 0.01). The mRNA expression of p-Akt protein increased more obviously (P < 0.05, P < 0.01). mRNA expression of GSK- 3beta and GLUT-4 significantly increased (P < 0.05), while the mRNA expression of PPAR-gamma significantly decreased in the low and middle BHR groups (P < 0.05). The expression of p-Akt significantly increased in the low dose BHR group (P < 0.05).
CONCLUSIONSIR and abnormal insulin signal pathway existed in PCOS model rats. BHR could improve IR of PCOS rats, which was correlated with regulating protein expression of insulin signal transduction molecules.

3.Effect of polyclonal antibodies against ciliary neurotrophic factor on botulinum toxin-induced axonai sprouting

Objective To study how polyclonal antibodies against ciliary neurotrophic factor (CNTF) reduce botulinum toxin-in- duced axonal sprouting and affect paralytic muscle.Design Experimental study.Participants 20 New Zealand rabbits.Methods In 10 rabbits randomly selected,left superior rectus were as control (Group 1),right superior rectus were injected with botulinum toxin A (BTXA) 2.5U (Group 3).At 14th day,bilateral superior recti were taken out under general anaesthesia.In the other 10 rabbits,left su- perior recti were injected equivalent physical saline (NS) as control (Group 2);right superior recti were injected with BTXA 2.5U,after 4 days,injected with 50?l polyclonal antibodies against CNTF at same site (Group 4).And at 14th day,bilateral superior recti were taken out for electron microscopy,dyed with acetylcholinesterase,argentums (Ag) to show nerve axonal sprout,and accounted and mea- sured with Leica microsystems.Main Outcome Measures The mean number of sprouts and the mean total length of sprouts,and the uhrastructure change of muscle by electron microscopy.Results In Group 1,the mean number of sprouts and the mean total length of sprouts were 5.75% and 10.53?m respectively;Group 2 were 6.11% and 11.16?m;Group 3 were 84.04% and 170.71?m;and Group 4 were 54.77% and 68.12?m.The differences were statistically significant (all P=0.000).Electron microscopy showed that after admin- istration BTXA alone (Group 3),muscle atrophied obviously,nerve myelin sheath increased,while the structure of nerve and muscle re- main invariable.The Group 4 showed that local myofilament disrupted and dissolved,degenerative myocytes necrotized and disintegrat- ed into fragments,which led to partial unreversible destroy.Conclusion Polyclonal anti-CNTF can reduce BTXA-induced axonal sprout- ing,lead to partial unreversible destroy of muscle,which may prolong the time of paralytic muscle resuming.It suggests that polyclonal anti-CNTF could prolong the duration of muscle paralyses induced by botulinum toxin.
4.Large-scale expansion of human umbilical cord mesenchymal stem cells in human platelet lysate as a substitute of fetal bovine serum

BACKGROUND:Classic media and fetal bovine serum are commonly used in the culture of umbilical cord mesenchymal stem cells, but the potential risk of serum culture limits its clinical application. OBJECTIVE:To use human platelet lysate alternative to fetal bovine serum for large-scale production of mesenchymal stem cells for therapeutic applications. METHODS:Human platelet lysate was prepared by repeated freezing and thawing, centrifugation, filtration, and concentration. Human umbilical cord mesenchymal stem cells were cultured in Iscove’s Modified Dulbecco’s Medium containing 5%concentrated platelet lysate (experimental group) or 10%fetal bovine serum (control group). After separation and enzymatic digestion, human umbilical cord mesenchymal stem cells were subcultured at a concentration of 3 000/cm2 up to the fifth generation. Then, cellmorphology and diameter, immune phenotype, osteogenic and adipogenic differentiation, and cloning efficiency were detected and compared between two groups. RESULTS AND CONCLUSION:Human umbilical cord mesenchymal stem cells cultivated in human platelet lysate-supplemented media showed a smal er size and more elongated morphology than those in fetal bovine serum-supplemented media. Colony forming unit-fibroblast analyses further showed no significant differences in colony efficiency. Human umbilical cord mesenchymal stem cells cultivated in human platelet lysate-supplemented media showed an increase of proliferation capacity;whereas, similar immunophenotypes remained in the two groups. In vitro assays revealed intact differentiation potential. Moreover, human umbilical cord mesenchymal stem cells cultivated in human platelet lysate-supplemented media showed a significantly higher capacity to differentiate towards osteocytes, indicating human platelet lysate is an alternative to fetal bovine serum for low-density production of mesenchymal stem cells for therapeutic applications.
5.Relationship between subjective well-being and behavioral inhibition/activation system of older adults

Objective To explore the relationship between subjective well-being (SWB) and behavioral inhibition/activation system (BIS/BAS) of older adults.Methods Using cluster sampling method and convenient random sampling method respectively for older adults in the nursing home and living home,we got 185 older subjects; then using BIS/BAS scale and SWB scale to test 185 older adults by oral questionnaire interviews face to face.Results (1) The SWB average score was 27.39±10.96,the BIS average score was 11.29±4.30,the BAS average score was 32.96±9.13.The SWB of older adults had a significant difference between older adults in the nursing (29.81±9.91) and living home (25.09± 11.47) (t=-2.69,P<0.01).The negative experience of SWB had significant differences between older adults in the nursing (4.25±3.37) and living home (4.25±3.37) (t=3.58,P<0.01),and also between males (4.25±3.37) and females (5.91±3.83) (t=-1.96,P<0.05).The BAS fun seeking had a significant gender difference (t=2.05,P<0.05),which was higher in males (12.60±3.48) than in females (11.40±3.67).(2) BAS was positively correlated with SWB (r=0.40,P<0.01),and BIS was negatively correlated with SWB (r=-0.40,P<0.01).(3) The BAS drive could forecast the 19% variance of SWB.Conclusion BIS/BAS are correlated with SWB for older adults and the BAS drive is one of factors contributing to the SWB of older adults.
6.The expression and significance of PTEN and p21ras in endometrioid endometrial adenocarcinoma

Objective To investigate the expression and significance of tumor suppressor gene PTEN and oncogenes Ki- ras in endometrioid endometrial adenocarcinoma(EEC). Methods The expression level of PTEN and p21ras were detected with the S- P immunohistochemistry method in 20 cases of normal endometrium, 20 cases of uterus atypical hyperplasia endometrium and 45 cases of EEC. The relationship between the gene expression and clinicopathological features was analyzed. Results The expression level of PTEN decreased progressively, and was 95.0 %, 40.0 %, 31.1 % respectively in normal, atypical hyperplasia endometrium and EEC. 7.Reliability and validity of the Chinese version of the Neuropsychiatric Inventory Questionnaire (NPI-Q) in patients with Aizheimer's disease

Objective: To develop the Chinese version of the Neuropsychiatric Inventory Questionnaire (NPI-Q) and evaluate its reliability and validity. Methods: Three investigators assessed 10 dementia cases together with the NPI-Q to evaluate the inter-rater reliability. Eighty-six dementia cases and 30 healthy controls were assessed with the Behavioral Pathology in Alzheimer's Disease (BEHAVE-AD), the Brief Psychiatric Rating Scale (BPRS) and NPI-Q to evaluate the criterion validity. A subgroup of 30 dementia cases was tested with the NPI-Q again 24 h later to evaluate the test-retest reliability. Results: (1) Internal consistency. Cronbach's a coefficient of the severity subscale was 0.589, mean inter-item correlation coefficient of the severity subscale was 0.107. Cronbach's α coeffi-cient of the distress subscale was 0.612, and the mean inter-item correlation coefficient of the distress subscale was 0.114. (2) Inter-rater reliability. Intraclass correlation coefficient (ICC) of the severity subscale was 0.97, and ICC of the distress subscale was 0.94. (3) Test-retest reliability. The test-retest correlation coefficient of the severi-ty subscale was 0.89, and the test-retest correlation coefficient of the distress subscale was 0.86. (4) Criterion va-lidity. The severity subscale total score was correlated significantly with BEHAVE-AD (r=0.70, P <0.001) and BPRS(r=0.40, P<0.001) total score; the distress subscale total score was correlated significantly with the global ratings in part 2 of BEHAVE-AD (r=0.76, P<0.001) . (5) The severity subscale total score in demen-tia group was significantly higher than that in control group (median: 10 vs.1.5, P <0.001). The distress sub-scale total score in dementia group was significantly higher than that in control group (median: 10 vs 0; P< 0.001) . (6) Exploratory factor analysis identified 4 common factors, including psychosis, abnormal behavior,discontrol and affective symptoms, which explained 58.3% of total variance. All of the items loaded were between 0.596 and 0.803 on their conrresponding factor. Conclusion: The reliability and validity of the Chinese version of NPI-Q meet the psychometrics properties, and deserves to conduct further study in larger samples.
8.Intervention and intervention of cisplatin combined with gemcitabine or vinorelbine in the treatment of patients with non-small cell lung cancer

Objective To investigate the effect of psychological intervention on patients with non-small cell lung cancer treated with cisplatin or gemcitabine or Changchun vinorelbine. Methods 64 cases of patients with non-small cell lung cancer after surgery, nursing for patients in the group given basic treatment, routine care control group, the observation group based on routine nursing and psychological nursing effects were compared between the two groups. Results No significant difference of anxiety and depression in the patients of the two groups before nursing, after grouping nursing, the observation group improved significantly; compared two groups of patients with quality of life score, visible two group before the intervention had no significant difference after intervention group was clearly observed after the patients in the observation group were higher than that of the control group, comparison between groups were indicates that the difference is obvious (P<0.05). Conclusion Chemotherapy in non-small cell lung cancer after surgery for patients with psychological intervention, compared to conventional nursing, can improve the psychological status of patients with better, improve the quality of life of patients, so it is worthy of reference in clinical use.
9.Manifestation and treatment of non-motor systoms of Parkinson's disease

Non-motor symptoms are an important component of the manifestations of Parkinson's disease(PD);they usually include sleep disorder,neuropsychotic symptoms,autonomic dysfunction,pain,and so on.Non-motor symptoms are multifactorial and mainly involve various non-dopaminergic neurotransmitter systems.These symptoms are not only a result of disease progression,but also can appear in the early stage of the disease,and they can have a major impact on the quality of life.The early notice of these symptoms may lead to better therapeutic strategies and subsequently to the improvement of the quality of life in PD patients.
10.HDAC1 expression and effect of curcumin on proliferation of Raji cells.

Histone deacetylase (HDAC1) has a high expression in many cancer cells and curcumin can inhibit the growth of cancer cells. This paper was designed to investigate the expression of HDAC1 of Raji cells and the effect of curcumin on their proliferation and apoptosis. Raji cells were treated with 3.125-50 micromol/L curcumin for 8-48 h and the growth inhibition rates of Raji cells were measured by MTT. The expression of HDAC1 on Raji cells were examined by mRNA, Western blot at 24 h various concertrations (1.6-50 micromol/L). Curcumin could selectively inhibit the proliferation of Raji cells in a dose- and time-dependent manner, with the inhibition rate being 52.47 %-82.18 % (P<0.01). The up-regulation of HDAC1 expression was observed within 24 h after the treatment with curcumin as shown by RT-PCR and Western blot. With the increase of concentration, the expression was down-regulated in a dose- dependent manner. It is concluded that the expression of HDAC1 plays an important role in the proliferation and apoptosis of Raji cells and curcumin can inhibit the growth of Raji cells at various concentrations and promote the apoptosis of Raji cells.
