Chinese Pharmacological Bulletin 2003;0(09):-
Aim To study the protective effect of puerarin(PUE) on cultured cerebral cells injured by anoxia-reoxygenation. Methods The anoxia-reoxygenation injury model were developed, anoxia for 60 min and reoxygenation for 30 min. the effect of PUE on cerebral ultrastructure was observed.[Ca 2+ ]_i was estimated with Adherent Cell Analysis and Sorting 570(ACAS 570) Laser Cytometer and measured with fluorescent dye Fura-2-AM, the lipid fluidity of cellular membrane was determined by fluorescence polarization technique. Results PUE improved the ultra-structure of cerebral cells, dose-dependently decreased[Ca 2+ ]_i and increased the lipid fluidity of cellular membrane. PUE markedly reduced the chromaticity value of pseudo-colour graphic model of Ca 2+ . Conclusion Puerarin has protective effect on anoxia-reoxygenation induced cerebral cell injury. The effect may be mediated by decreasing[Ca 2+ ]_i and increasing the lipid fluidity of cellular membrane.

Objective To determine the value of preoperative clinical, biochemical, cross-sec-tional imaging features and results of fine-needle aspiration for predicting malignancy in cystic neo-plasms of the pancreas (CNP). Methods The medical records of 69 patients receiving operations for CNP between 1994 and 2008 in our hospital were reviewed retrospectively. The predictive effect of va-rious preoperative factors such as sex, location, clinical manifestation, maximum diameters, tumor marker, pancreatic duct obstruction and calcification on the malignant potential of CNP was evaluated by Single and multi-factor analysis, fine needle aspiration (FNA) and intraoperative frozen-section ex-amination of the pancreatic transection margin was investigated. Results All the 69 patients were con-firmed pathologically. Of the 69 patients, 13 suffered from serous cystic neoplasms, 30 from mucinous cystic neoplasms,7 from intraductal papillary mucinous neoplasms,12 from solid pseudopapillary neo-plasms and 7 from cystic neoplasms. Forty-four lesions were diagnosed as malignant or borderline.Univariate analysis should that jaundice, raised CEA and/or CA19-9, maximum diameters and solid component of cystic neoplasmshad were of statistical significance for the risk of malignancy in CNP.The sensitivity was 34.1% (15/44), 47.7 % (21/44), 88.6%(39/44),72.7%(32/44) and specificity 96% (24/25), 84% (23/25), 68% (17/25),72% (18/25), respectively. The last three were identified as independent predictive factors for malignancy by multivariate analysis. Three cases were accurately diagnosed out of the 9 undergoing FNA preoperatively. One of 7 patients with intraductal papillary mucinous neoplasms (IPMN) undenwent total pancreatetomy for transection margin positivity. Conclusion Most malignant CNP can be accurately diagnosed preoperatively from a typical clinical, biochemical and cross-sectional imaging picture. FNA is only used in the patients who are potential candidates for nonoperative management. Margin analysis is necessary for pancreatic resection.

Journal of Applied Clinical Pediatrics 2004;0(09):-
Objective To investigate the effects of dibutylphthalate (DBP) on the proliferation and apoptosis of HL-60 leukemic cells and to study its mechanism to purge leukemic cells. Methods The effects of DBP on proliferation of HL-60 leukemic cells were measured by cell culture method. The effects on apoptosis were measured by the percentage of the apoptotic cells in morphology and of the DNA fragmentation and by DNA gel electrophoresis. The intracellular free Ca~(2+) concentration ([Ca~(2+)]i) of leukemic cells were measured by Fura-2AM method. The protein expressions of c-myc and bcl-2 genes of leukemic cells were measured by immunohistochemical assay. Results DBP could suppress the proliferation of HL-60 leukemic cells in a dose-dependent and time-dependent manner and induce them to die via apoptosis. It could elicit a intracellular Ca~(2+) redistribution and a potent extracellular calcium influx. It also down-regulated the protein expressions of c-myc and bcl-2 genes of HL-60 leukemic cells. Conclusion DBP can purge (HL-60) leukemic cells in vitro by increasing [Ca~(2+)]i of cells to initiate apoptosis and down-regulating bcl-2 and c-myc proto-gene expressions to promote cell apoptosis.
