Journal of Chinese Physician 2009;11(5):619-622
Objective To investigate the effect of Helicobacter pylori infection on cell proliferation and apoptosis of gastric cancer cell line SGC 7901. Method SGC 7901 cells were incubated with 1 × 108,5 × 107,1 × 107,5 × 106 cfu / ml concentration gradient of the standard HP NCTC 11637 strains in vitro. Morphological changes of cells were observed at 24, 48, 72 hour, respectively. Cell proliferation inhibition rate were detected with MTT assay. Apoptosis were observed by flow eytometry and TUNEL analysis, mRNA expression of survivin were detected by RT-PCR, and survivin expression were determined by western blot. Results Cell proliferation inhibition rate was 54.5%, 58.9%, 67.6%, 72.9% with 1 × 108,5 × 107, 1 × 107,5 × 106 cfu / ml concentration gradient of the HP bacteria on the role of SGC 7901 cells at 72h, respectively. The rate of apoptosis after 72h detected by flow cytometry and TUNEL with HP concentration gradient of different bacteria were 42.51%, 45.67%,48.57%, 49.51% and 54.61%, 51.26%, 59.41%, 62.46.% Helieobacter pylori could significantly reduce mRNA and protein expression of survivin. These effects were strengthened as the concentration of Helicobacter pylori increased and the time extended. Conclusion H. Pylori infection may be through reducing the expression of survivin to inhibit cell proliferation and pro-mote apoptosis of SGC 7901 in vitro. Moreover, this effect was positive related with time and dose.

Objective To study the effect of metastasis-associated in colon cancer 1 ( MACC1 ) inhibition by small interference RNA( siRNA) on the formation of tumor cells that promote blood vessels, and to explore its possible molecular mechanism. Methods Synthesized MACC1 specific siRNA was transfected into human lung cancer cell lines XWLC-05. Then, the inhibitory effect of siRNA on the protein level of MET, MEK/p-MEK, ERK/p-ERK, and AKT/p-AKT was detected using Western blotting while the change in VEGF, bFGF and IL-8 protein level was detected using ELISA method. Results Western blotting results indicated that the expression level of MET, p-MEK and p-ERK was significantly decreased in the XWLC-05 cells transfected by MACC1 specific siRNA compared with control and nonsense siRNA groups, but the expression of total protein MEK and ERK did not change.Furthermore, the level of AKT and phosphorylated protein was not affected.ELISA results suggested that the secretion level of VEGF, bFGF and IL-8 was significantly suppressed at 48 h or 72 h compared with control and nonsense siRNA groups. Conclusion MACC1 may reduce the secretion levels of VEGF, bFGF and IL-8 via HGF/c-Met and MEK/ERK signal transduction pathways, and regulate angiogenesis of lung cancer.

Objective To assess the validity of a newly developed in-house ELISPOT IFN-γ release assay (IGRA) for the detection of latent tuberculosis infection among HIV infected individuals. Methods In-house ELISPOT assay were performed, together with a tuberculin skin test in 205 health controls and 110 HIV infected individuals , who had no signs of active tuberculosis at time of enrolment . Results Using the ELISPOT assay, positivity rates for the 205 health controls, 110 HIV infected individuals and 47 AIDS patients on highly active antiretrovial therapy (HAART) were 7. 3% , 24.5% , 29. 8% , respectively. These results indicated that the positive rates obtained from HIV infected individuals (include patient on HAART) was significantly higher than health controls( P < 0.001). We found no significant correlation between the CD4 cell count and positivity of ELISPOT assay (P >0.05 ). The proportion of subjects with a positive response to ELISPOT assay were higher than the proportion of tuberculin skin test(TST) responders(P<0.0001) in HIV infected individuals. Conclusion Our study indicates that IGRA using M. tuberculosis specific antigens are likely to retain their validity for the diagnosis of LTBI among HIV positive individuals.

OBJECTIVEThe objective of this study is to investigate the influence of zirconia content which is 0-30.0% weight percentage of matrix on translucency of zirconia-toughened alumina glass-infiltrated ceramics.
METHODSSeven groups were divided according to different weight percentage of zirconia (0, 2.5%, 5.0%, 7.5%, 10.0%, 20.0% and 30.0%). After sintering, infiltrating and polishing, spectral transmittance was determined with spectrophotometer under D65 standard source. Contrast ratio was also tested by whiteness colorimeter.
RESULTSWith mass fraction of zirconia increasing from 0 to 30.0%, spectral transmittance reduced from 0.406% to 0.058%, while contrast ratio value increased from 0.849 +/- 0.005 to 1.015 +/- 0.006. When zirconia content was 10.0%, contrast ratio was 0.990 +/- 0.008. When it was more than 10.0%, transmission rate of the downward trend and contrast ratio of the rising trend became flat.
CONCLUSIONZirconia content has a direct impact on translucency of zirconia-toughened alumina glass-infiltrated ceramic, which is essentially opaque when zirconia content is 10.0%. When mass fraction of zirconia is more than 10.0%, the influence of zirconia content is reduced.
