1.Retinal toxicity of repeated intravitreal injection with bevacizumab in diabetic rats

Objective To observe the retinal toxicity of repeated intravitreal injection with bevacizumab(Avastin)in diabetic rats.Methods Forty male Sprague Dawley(SD)rats were randomly divided into normal group(Group A,10 rats)and diabetes mellitus group(30 rats).The rats in diabetes mellitus group were induced with streptozotocin injection for diabetic retinopathy model.And then randomly divided into diabetic retinopathy(DR)group(Group B,10 rats),the rats were not intervened;the left eyes of the other 20 rats were intravitreal injected with bevaeizumab 3 μ1(25 mg/m1)for 3 times as experimental group(Group C);the right eyes of the 20 rats were not intervened as experimental control group(Group D),20 days after last intravitreal injection,retinal function was measured by Flicker Electroretinogram (F-ERG);retinal vascular pattern was determined by fluorescence microscopy of ethidium bromide(EB)stained retinal flat mounts;retinal morphological changes were determined by light microscope on hematoxylin-eosin (HE) stained sections;Thy-1 and VEGF expression was measured by immunohistochemistry staining.Results F-ERG showed that-the differences of a-and b-waves-the b-wave amplitude and the Ops-wave amplitude in the implicit time between group A,B,C and D were significant (F=33.165,36.162,19.955,23.243;P=0.000);the differences of a-wave amplitude between group A,B,C and D was not significant(F=0.097,P=0v961).Retinal blood vessel pattern was normalin Group A;retinal vascular vessels were tortuous and irregularly expanded in Group B:retinal vascular vessels of Group C were regular and thinner than Group A;microaneurysm were showed in Group D.Light microscope displayed that the layers of the rat retina of Group A were regular,the retinal architectures of Group B were irregular,the retinal layers were regular in Group C,the retinal layers were irregular in Group D.Immunohistochemistry staining discovered that Thy-1 and VEGF were mainly expressed in ganglion cell layer(GCL).Conclusion Repeated intravitreal injection of bevacizumab iS toxic tO retina of diabetes mellitus rats.
2.Effects of the Therapy of Invigorating the Kidney and Qi and Dissipating Blood Stasis to Removing the Toxin on the Hepatocyte Apoptosis and Associated Genes in the Liver Fibrosis Rats

Objective To investigate the effects of the therapy of invigorating the kidney and qi and dissipating blood stasis to removing the toxin on the hepatocyte apoptosis and associated genes in the liver fibrosis rats,and to explore the treatment mechanism of liver fibrosis with TCM.Methods liver fibrosis model rats were induced by CCl4,and the Chinese herbs(Ganlike)with invigorating the kidney and qi and dissipating blood stasis to removing the toxin was given by daily gavage,after the experiment,the pathological changes of liver were observed with optical microscope.Hepatocyte apoptosis was detected with TUNEL assay,apoptosis-associated genes were detected with immunohistochemistry assay.Results The Chinese herbs Ganlike could obvously improve the pathological changes,alleviate the hepatocyte apoptosis and modulate the expression of apoptosis-associated genes.Conclusions The anti-fibrosis mechanism of Ganlike may be to regulate the hepatocyte apoptosis of liver fibrosis rats and to restrain the expressions of hepatocyte Fas/FasL,Bcl-2/Bax.
3.Testicular cancer in cryptorchids

Objective To study the diagnosis and treatment of testicular cancer in cryptorchids. Methods The data of 10 patients with cryptorchid tumor treated at our hospital between 1986 and 2003 were retrospectively analyzed.Their average age was 40 years (range,32~55 years).Of these patients,1 had the testis in the inguinal area;3,in the scrotum (with a history of inguinal cryptorchidism) and 6,in the abdomen.Cryptorchidism occurred on the left in 2 cases,on the right in 5 and bilaterally in 3.Three patients presented with stage Ⅰ A disease,5 with stage Ⅰ B,and 2 with stage Ⅱ D. Results All the 10 patients received surgical treatment including radical tumor resection,radical tumor resection with retroperitoneal lymph node dissection (RPLND),and palliative operation.Pathology showed that 9 cases had seminoma and 1 had seminoma accompanied by embryonic carcinoma.Radiotherapy was performed in 8 cases,and radiotherapy plus chemotherapy,in 2 (including 1 with embryonic carcinoma).Postoperatively,6 patients were followed up for 6 months to 14 years with a mean of 5.2 years;these patients showed no evidence of tumor recurrence and metastasis. Conclusions Cryptorchid testes have a greater risk of malignant change than do normally descended testes,and early intervention is advocated to prevent malignant canceration.Even treated with orchidopexy or orchiectomy,the patient should be followed up lifelong.Most of cryptorchid tumors are seminomas, and the ideal treatment strategies are tumor resection and radiotherapy.
4.Effects of vitrectomy on neovascular glaucoma combined with vitreous hemorrhage

Objective To explore the effects of vitrectomy on neovascular glaucoma combined with vitreous hemorrhage. Methods Seven eyes of 7 patients with neovascular glaucoma combined with vitreous hemorrhage underwent vitrectomy combined with phacoemulsification, panretinal photocoagulation, and trabeculectomy. The preoperative visual acuity ranged from light perception to 0.2, and the mean preoperative intraocular pressure was 54 mm Hg (38-64 mm Hg)(1 mm Hg=0.133 kPa). The mean follow-up was 8 months (6-15 months). Results The postoperative visual acuity ranged from light perception to 0.4, and the mean postoperative intraocular pressure was 17 mm Hg (10-30 mm Hg) which was significantly lower than preoperative one ( P

Aim To study the effect of the total extract of astragalosides (TEA) on the growth, AFP secretion and ?_GT activity of human hepatoma cells in vitro. Methods The human hepatoma HepG2 and Bel_7404 cells were cultured with TEA 5~160 mg?L-1 for 6 days. Anti_hepatoma activity was measured by MTT method. AFP was assaysed by ELISA and ?_GT was determined by enzyme_substrate method. Results The growth of the human hepatoma HepG2 and Bel_7404 cells and the secretion of AFP of HepG2 cells were reduced significantly by TEA 5~160 mg?L-1.The ?_GT activity of HepG2 cells was inhibited and its ALB content was increased by TEA 20,40 and 80 mg?L-1. The ?_GT activity of Bel_7404 cell was inhibited and its ALB content was increased by TEA 40 and 80 mg?L-1.Conclusion These results suggest that TEA has inhibitory effects on the growth, AFP secretion and ?_GT activity of human hepatoma cells.
6.Effects of total extract of astragalus(TEA) on the proliferation and the production of collagen of hepatic stellate cells

Aim To study the effects of total extract of astragalus(TEA) on activating the primary cultured hepatic stellate cells(HSC) and synthesizing collagen. Methods Livers of adult rats were perfused by pronase E and collagenase in situ, HSC and Kupffer cell (KC) were isolated by centrifugation with 18% Nycodenz. The subcultured HSC were induced by Kupffer cell conditioned medium(KCCM) and were incubated with 5~ 80 mg?L -1 TEA for 6 days. The proliferation of the cells was measured by 3H TdR and the production of collagen by 3H Proline.Results TEA (10~ 40 mg?L -1 ) could suppress the proliferation of hepatic stellate cells and TEA(40~ 80 mg?L -1 ) could suppress the production of collagen. Conlusion The suppression of hepatic stellate cell proliferation and the collagen's production by the TEA may be one of the mechanisms for depressing the hepatic fibrosis by the TEA.

8.A phantom study on the effect of target diameter and target-to-background ratio on the measurement of SUVmax in PET

Objective To investigate the effect of target diameter and target-to-background ratio (TBR) on the measurement of SUVmax [| (true SUV-SUVmax)/true SUV | × 100%,△SUVmax) in PET.Methods Six cylinders with various diameters from 5 to 29 mm were inserted in National Electrical Manufacturers Association (NEMA) NU 2-1994 phantom.The six cylinders and background of phantom were filled with18F-FDG solution.Six different TBRs (1.79,3.70,6.25,10.59,14.87 and 17.88) were obtained by changing the 18F-FDG concentration in the six cylinders.The PET data were acquired in the 2D mode,and the target's inner diameter and SUVmax were measured.The logarithmic fitting curves of △SUVmax with diameter in different TBRs using equations (y=aln(x)+b) were acquired by Microsoft excel,| a| as the influence power of diameter on △SUVmax.Statistical analysis was performed with Pearson correlation test and curve estimation utilizing SPSS 17.0.Results The Pearson correlation coefficient between △SUVmax and diameter was-0.806 (P<0.01).△SUVmax was inversely correlated with the diameter.When the diameter (the inner diameter measured on PET images) was 4.0 mm,△SUVmax could be as high as 79.73% with different TBRs.The Pearson correlation coefficient between the influence power of diameter on △SUVmax (| a |) and TBRs was 0.848 (P<0.05).When TBR was 6.25,10.59 and 14.87,the corresponding |a| was similar:38.019,39.998 and 39.362,respectively.When TBR was 17.88,| a| was the highest as 43.234.When TBR was 1.79 and 3.70,|a| was much smaller:14.141 and 23.411 respectively.Conclusions The lesion diameter is inversely correlated with △SUVmax.The influence power of diameter on △SUVmax is strongly correlated with TBR.Therefore,the effect of target diameter and TBR on the measurement of SUVmax should be taken into consideration for follow-up studies.
