2.Retinitis pigmentosa-related gene researches in China

ObjectiveTo review retinitis pigmentosa (RP)-related gene researches in China. Methods RP genes and RP gene therapy as key words were searched from those online databases including China Knowledge Resource Integrated Database of China National Knowledge Infrastructure (CNKI), National Center for Biotechnology Information (NCBI), Human Gene Mutation Database (HGMD), and Human Genome Variation Society (HGVS). Related papers written by authors from China or other countries,published from 1991 to 2011 were collected for analysis. ResultsOverall 60 RP-related genes have been identified now. Chinese researchers from 29 hospitals and research institutes confirmed 17 RP-related gene changes (239 mutations, 131 of them are pathogenic) in 300 RP families and 1572 sporadic RP patients from 1991 to 2011, as reported in 66 papers. Mutations in rhodopsin (RHO) gene, NR2E3 gene and RP1 gene were found in 2.0%, 2.9% and 1.0% of Chinese RP patients respectively.The relationship between genotypes and clinical phenotypes were investigated in 15 papers, including RHO gene, RDS gene, RP1 and RPGR or RP3 gene in ADRP families. Gene therapy for RP has been investigated in 4 papers. Seven hundred and eighteen papers about RP gene-mutation screening have been published from other countries,and more than 2000 variants were identified which including 352 disease-causing mutations. Mutations of RHO gene, RDS and RP1 were found in 25%-50%, 8% and 5%-10% of ADRP patients in Caucasian populations. RPGR mutations were found in 70%-80% of XLRP patients in Caucasian populations, and 50%-60% of mutations are identified in open reading frame (ORF) 15. Totally 391 papers about gene therapy for RP have been published. ConclusionsSeventeen RP-related genes have been studied and 131pathogenic mutations were found in China. Considering Chinese population, our RP research is still behind other countries.

4.The neurodegeneration of pretectal area in the early stage of diabetes

Objective To analyze the expression of BDNF, NGF,GFAP in Wistar rat pretectel area and determine the relation between neuron damage and neurotrophin change of pretectal area and retinal neurodegeneration in the early stage of diabetes. Methods Male Wistar rats weighing about 200 g were used to create diabetic animal model by injecting STZ. Wistar rats were killed by heart perfusion with 4% FDP at 1-week, 2-week and 1-month after diabetes. The dissected pretectal area was used to determine the expression of BDNF, NGF and GFAP with immunohistochemistry. Results In pretectal area of diabetic rat, the expression of BDNF and NGF began to decrease from 1-week DM, the expression of GFAP began to increase from 2-week DM. Conclusions There are neuron damage and neurotrophin decrease in the pretectal area in the early stage of diabetes. Neurodegeneration and neurotrophin decrease in pretectal area, which is the target tissue of retina, might be one of the reasons resulting in retinal neurodegeneration at the early stage of diabetes.
5.Expression and significance of uPAR and VEGF-C in hepatocellular carcinoma

Objective To investigate the expression of uPAR and VEGF-C in hepatocellular carcinoma (HCC) and its effect on the biological behavior of HCC. Methods The expression of uPAR and VEGF-C in liver tissue specimen of 33 patients with HCC was detected by immunohistochemical staining. Results The positive expression of uPAR and VEGF-C in HCC tissues was located in cytoplasma of cancer cells. The positive rates of uPAR and VEGF-C were 69.7 %(23/33) and 78.8 %(26/33) respectively in 33 cases of HCC. The expression rates of uPAR and VEGF-C were significantly higher in HCC with metastasis than in HCC without metastasis(P<0.05). But there were not significant correlation between the expression of uPAR or VEGF-C and tumor size, tumor grade, AFP level, hepatic cirrhosis. There was a close correlation between uPAR and VEGF-C in HCC(r=0.676, P<0.01). Conclusion The expression of uPAR and VEGF-C is correlated with metastasis, uPAR and VEGF-C may be signs of aggressive phenotype for evaluating treatment and prognosis of HCC. Examination of these two proteins may be helpful to .judge biological behavior of HCC.
6.Therapeutic Observation of Acupuncture plus Chinese Herbs for Sensorineural Hearing Loss

Objective To observe the clinical efficacy of acupuncture plus Chinese herbs in treating sensorineural hearing loss.Method Eighty patients with sensorineural hearing loss were randomized into a treatment group and a control group, 40 cases in each group. The control group was intervened by conventional medication, while the treatment group by acupuncture in addition to the intervention given to the control group. The two groups were intervened once a day, 30 days as a treatment course. The serum interleukin (IL)-2, IL-6, soluble interleukin-2 receptor (sIL-2R) levels and pure tone hearing threshold value were observed before and after consecutive 2 treatment courses, and the clinical efficacies in the two groups were compared.Result The serum levels of IL-2, IL-6 and sIL-2R and pure tone hearing threshold value were significantly changed after the treatment in the two groups (P<0.05). There were significant differences in the serum IL-2, IL-6, sIL-2R levels and pure tone hearing threshold value between the two groups after the treatment (P<0.05). The total effective rate was 94.6% in the treatment group versus 84.2% in the control group, and the difference was statistically significant (P<0.05).Conclusion Acupuncture plus Chinese herbs is an effective approach in treating sensorineural hearing loss.
7.Ultrastructural Pathology of Neurons in Mouse Brain Infected with B Encephalitis Virus

The animal model for the study of B encephalitis was established by injecting Jin Wei Yan 1 strain of B encephalitis virus into the peritoneal cavity of mice. All the mice injected demonstrated the histological features of the disease. Special attention was paid to the ultrastructural changes of the granular cells, the Purkinje cells and the Golgi cells in the cerebellum, and the findings in these cells were compared with those of the nerve cells in the cerebral cortex, dience-phalon and tnesencephalonIt was found that endoplasrnic reticulum could appear in the nerves infected by B encephalitis virus. A radiating structure was-usually formed by the viral replication multivesicular bodies with microvesiculo-tubular body as its center. The morphological changes and the developmental sequence as well as the significance of this radiating structure were discussed.In the late stage of the infection, viral particles were found in the nuclei of a part of the necrotic cells. It was likely that the viral particles entered the nuclei by way of cytoplasm. Our observation confirmed the Chen Liming's hypothesis that viral particles can be formed in the perinuclear space. The way that the viral particles were evacuated from the infected nerve cells observed in this study was in agreement with that reported by Chen Liming, but most of the viral particles left the infected nerve cells through the axons.
8.Effect of Acupuncture on Anxiety and Depression Due to Vertebrobasilar Insufficient Vertigo

Objective To observe the clinical efficacy of acupuncture in treating anxiety and depression due to vertebrobasilar insufficient vertigo (VBIV).Method Eighty-two patients with anxiety and depression due to VBIV were randomized into a treatment group and a control group, 41 cases in each group. The control group was intervened by conventional medication, while the treatment group was by acupuncture in addition to the treatments given to the control group. The Hamilton Anxiety Scale (HAMA), Hamilton Depression Scale (HAMD), Self-rating Anxiety Scale (SAS), Self-rating Depression Scale (SDS), and Treatment Emergent Symptom Scale (TESS) were evaluated before and after intervention.Result The HAMA, HAMD, SAS, and SDS scores were significantly changed after intervention in both groups (P<0.05). After treatment, the HAMA score in the treatment group was significantly different from that in the control group (P<0.05). The TESS scores were significantly changed after treatment in both groups (P<0.01). After intervention, the TESS score in the treatment group was significantly different from that in the control group (P<0.01).Conclusion Acupuncture is an effective approach in treating anxiety and depression due to VBIV, and it can reduce the occurrence of adverse reactions.
9.An intervention to reduce discrimination against people living with HIV/AIDS among medical students

Objective To explore the effectiveness of short-term integrated intervention to reduce discrimination against people living with HIV/AIDS(PLWHA)among medical students.Methods A quasi-experimental survey was conducted.Two medical schools in Beijing,one as intervention group and the other as control group were selected by convenience sampling,from which 123 undergraduate students or interns of medical and nursing specialties were recruited,62 in the intervention group and 61 in the control group.The intervention group was provided with 13 hour long antidiscrimination intervention by increasing HIV/AIDS related knowledge,sympathy and skill in dealing with people living with HIV/AIDS,while the control group was deprived of any intervention.The attitude of the students in the 2 groups were assessed by questionnaire survey in 5 time points after the intervention.Results Compared with the control group,at 4 time points after the intervention students in the intervention group had less discrimination against PLWHA in general;and significantly less discrimination in dealing with them as patients(P
10.The role of TRPM7 ion channel in the proliferation and migration of oral squamous cancer cells

Objective:To investigate the role of the transient receptor potential melastatin subfamily member 7 (TRPM7) ion channel in the proliferation and migration of oral squamous cancer cells OC2.Methods:The expression of TRPM7 in OC2 cells was examined by western blotting,RT-PCR and indirect immunofluorescence assay.The influence of TRPM7 on cell proliferation and migration under the inhibition of TRPM7 ion channel with 2-APB or siRNA TRPM7 in OC2 cells was examined by MTT and Transwell respectively.The influence of PI3K/AKT signal pathway on the expression of TRPM7 in OC2 cells was tested by Western blotting.The change of positive ion current when the OC2 cells were treated with TRPM7 inhibitor and activator was explored with patch clamp technique.Results:TRPM7 overexpression in OC2 cells mainly in the cytoplasm was observed,siRNA TRPM7 downregulated the expression of TRPM7.Inhibition of TRPM7 (2-APB or siRNA TRPM7) inhibited the growth and proliferation of OC2 cells in a time-and dose-dependent manner.Blockage of TRPM7 ion channel inhibited the migration capacity of OC2 cells.Inhibition of TRPM7 downregulated the expression of phosphorylated AKT and phosphorylated ERK.Patch clamp assay showed the activation of TRPM7-like current in OC2 cells.2-APB could weaken this current while Bradykinin could enhance the current.Conclusion:TRPM7 may regulate the proliferation and migration of oral cancer cells by the regulation of PI3K/AKT and MAPK/ERK signal pathways and cell positive current.
