1.Intervention of siRNA to the effect of gelatinase on the expression of ICAM-1 in human renal tubular epithelial cells

Objective To inhibit the expression of matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) in human kidney tubular epithelial cell(HKC) with specific small interference RNA(siRNA),then to observe whether the expression of intercellular adhesion molecule-1(ICAM-1) on HKC could be influenced.Methods HKCs were transfected with MMP-2,MMP-9 siRNA or irrespective siRNA respectively,untransfected HKC was considered as blank control.Four groups of HKCs were stimulated with 100nmol/L phorbol myristate acetate(PMA) for 18h.Total RNA or protein of cells were extracted with Trizol and RIPA cell lysate respectively,the mRNA expression levels of MMP-2,MMP-9 and ICAM-1 were detected by real time PCR,the protein expression levels of above indexes were detected by western blotting and immunofluorescence combined with confocal laser microscopy.Results The specific siRNA could inhibit the expression of MMP-2 and MMP-9 in HKCs effectively.Immunofluorescence combined with confocal assay demonstrated that while HKCs was stimulated with PMA,the expression of ICAM-1 was up-regulated,which was significantly up-regulated in the group transfected with MMP-9 siRNA(P0.05).Conclusions The inhibitory expression of MMP-9 of HKC may up-regulate the expression of ICAM-1,implying that,besides taking part in the degradation of extracellular matrix,MMP-9 may throw an influence on the proceeding of renal fibrosis by inflammatory pathway.
3.Effect of Eradication of Helicobacter Pylori on Non-alcoholic Fatty Liver Disease

Objective To observe the effect of eradication of helicobacter pylori (Hp) combined with comprehensive rehabilitation treat-ment on patients with non-alcoholic fatty liver disease (NAFLD). Methods From December, 2015 to April, 2016, 219 patients diagnosed as NAFLD with Hp infection were enrolled, in which 119 patients (observation group) were treated with anti Hp quadruple therapy. They were tested with 13C urea breath test one month after drug withdrawal. The other 100 patients were as control group. The severity of NAFLD was observed with abdominal ultrasound after six months. Results After treatment, Hp was eradicated in 107 cases (90.68%) in the observation group, one dropped out for pulmonary infection. After six months, the severity of NAFLD improved better in the patients eradicated Hp in the observation group than in the control group (χ2=8.397, P<0.05). Conclusion Eradication of Hp combined with comprehensive rehabilita-tion treatment can improve the severity of NAFLD.
4.The Correlation of ACE I/D Gene Polymorphisms with the Antihypertensive Efficacy of Irbesartan Among Hypertensive Patients

Objective To analyze the correlationship of ACE I/D gene polymerphisms with the anti-hypertensive efficacy of irbesartan among essential hypertension (EH) patients in Yunnan han people.Methods One hundred EH patients hypertension were treated with irbesartan 150mg once daily for 4 weeks,and anti-hypertensive efficacy were monitored during the treatment.Results There was a significant difference between anti-hypertensive efficacy of irbesartan among different genotype groups,DD group >ID group >Ⅱ group (P < 0.05) Conclusions There is different therapy responsiveness in diffenent genotypes.ACE I/D polymorphism may be an important hereditary factor that impacts the efficacy of irbesartan.
5.Regulatory effects of IFN-γ on Treg cells from HIV/AIDS patients receiving HAART for one year

Objective To investigate the regulatory effects of IFN-γon Treg cells from HIV/AIDS patients receiving highly active antiretroviral therapy (HAART) for one year.Methods Thirty HIV/A1DS patients whose CD4+T cells were below 350/μ1 were recruited for HAART therapy.Blood samples were collected at the time points of 0,24,48 weeks after HAART.PBMCs were isolated and randomly divided into two culture groups.One group was cultured directly in medium and another group was co-cultured with IFN-γ (40 pg/ml).The supernatants and cells were separated after 5 days of culture for analysis.The concentrations of IL-12 and CD4+CD25+Foxp3 Treg cells were measured by ELISA and flow cytometry,respectively.Results The levels of IL-12 in the supernatants from the culture without IFN-γ at time points of 0,24,48 weeks after HAART were lower than those from the co-cultured group [(37.02±12.76) vs (41.79± 15.02),t=2.336,P=0.03; (41.76±17.01) vs (47.2±14.26),t=2.702,P=0.014; (48.01± 11.84) vs (53.44± 11.30),t =3.14,P =0.003].The percentages of CD4+ CD25 + Foxp3 Treg cells in CD4+ T cells from the direct-cultured group were higher than those from the co-cultured group at the three time points [(10.41±1.10)% vs (2.40±1.11)%,t=13.89,P=0.000; (8.33±2.03)% vs (1.99± 0.86)%,t=12.93,P=0.000; (5.65±1.55)% vs (1.32±0.73)%,t=10.61,P=0.000].Moreover,the results within the same group at the time points of 0,24,48 weeks upon HAART were also significantly different.Conclusion With the interference of HAART,IL-12 levels were increased,while CD4+CD25+ Foxp3 Treg cells were decreased in patients with HIV/AIDS.IFN-γ plays an important role in this process.
6.An investigation of life value and suicide attitude of college students in the earthquake-stricken area

Objective To understand the life attitude of college students in earthquake-stricken area. Methods 166 college students were conducted by life value scale (LVS) and suicide attitude questionnaire (QSA). Results ①On the LVS, males( (1.85 ±0.57) , (1.73±0.55) ) got significantly higher scores than females ((1.66±0.43),(1.54±0.40)) on survival values-oriented and spiritual values-oriented, and lower scores on tool values-oriented ((2.59±0.46 vs (2.76±0.51) ) , there was significant difference on overcome values-oriented between different grades(F = 3.710, P = 0.013). On the QSA, males (3.01 ±0.51) got significantly lower scores than females (3.28 ±0.51) on the cognition of suicidal behavior difference (t=3.110, P=0.002). There were significant differences on attitude to relationship of suicides between different gender and areas (P<0.05) , and attitude to euthanasia between different gender, grades and subjects (P<0.05). ②Social valuesoriented can predict cognition of suicidal behavior, personal and tool values-oriented can predict attitude to relationship of suicides, the total of life value can predict cognition of suicidal behavior and attitude to relationship of suicides. Conclusion The life value can predict the suicide attitude of college students in earthquake-stricken area, thus the life education should be enhanced to improve their quality of life.
7.Association of secretory IgA with clinical pathological characteristics and complement activation in IgA nephropathy

Objective To further investigate the association among clinical pathology,complement activation and renal secretory IgA (SIgA) deposition in patients with IgA nephropathy (IgAN).Methods The activation of serum complements were detected by immunoturbidimetry and ELISA.Renal deposition of SIgA and activation of complements were detected by immunofluorescence.Then the association among clinical pathology,complement activation and renal SIgA deposition were analyzed in IgAN patients.Results In all 201 patients with IgAN,59 patients had SIgA deposition with higher incidences of mucosal infection history and hematuria (P < 0.05),lower levels of serum cystatin C,β2 microglobulin and lower tubulointerstitial lesion grades and T-grade in the Oxford classification (P < 0.05),when compared with patients without SIgA deposition.Both alternative and mannose binding lectin (MBL) pathways were activated in patients with or without SIgA deposition.Patients with MBL pathway activation had lower estimate glomerular filtration rate (P < 0.01),higher serum creatinine,higher proportion of glomerulosclerosis and S-grade in the Oxford classification,more severe tubulointerstitial lesion (P < 0.05).Conclusions Compared with patients without SIgA deposition,patients with SIgA deposition have a stronger link to mucosal immune.The deposition of SIgA is associated with different clinical and pathological manifestations;however,the complement activation is similar in both groups of patients.Patients with MBL pathway activation show more severe kidney injury.
8.Comparison of the quantitative evaluation in high-resolution CT and pulmonary function tests in chronic obstructive pulmonary disease

Objective To analyze the correlation between quantitative evaluation of high-resolution CT (HRCT) and pulmonary function tests (PFT) in chronic obstructive pulmonary disease (COPD), and evaluate the characteristics of PFT in different CT phenotypes of COPD. Methods 65 patients with COPD underwent CT chest scan and PFT. The dimensions of subsubsegmental apical bronchi of right upper lobes were measured , which included the following parameters:airway wall thickness (T), total airway diameter (D), and pulmonary artery (PA), ratio of airway wall thickness and pulmonary artery (T/PA), thickness diameter ratio (TDR), percentage of wall area (WA%) and percentage of airway luminal area (Ai%). The percentage of total lung area occupied by low attenuation area (LAA%) was measured by lung density software. The relationship between the measurements and PFT parameters were analyzed. Patients were classified into three phenotypes based on the presence of emphysema or bronchial wall thickening evaluated by HRCT. Phenotype A: without or with little emphysema (LAA% < 25%), with or without bronchial wall thickening; Phenotype E: emphysema without bronchial wall thickening (LAA%≥25%, T/PA < 30%); Phenotype M: emphysema with bronchial wall thickening (LAA%≥25%, T/PA≥30%). The characteristics of different CT phenotypes were analyzed. Results LAA% had a good correlation with PFT parameters. PA, T/PA, WA% and Ai% also had correlation with PFT parameters. There was 53 patients of Phenotype A, 12 patients of Phenotype M. PA, T/PA, LAA%and PFT were different in two groups. Conclusions LAA%and airway quantitative measurements in HRCT have certain correlation with PFT parameters. Patients with different CT phenotypes differs in PFT parameters , which can provide reference for clinical diagnosis and treatment.
9.Protein Fold Recognition With Support Vector Machines Fusion Network

One of the important approaches to structure analysis is protein fold recognition, which is oftenapplied when there is no significant sequence similarity between structurally similar proteins. A framework with athree-layer support vector machines fusion network (SFN) is presented. The framework is applied to 27-classprotein fold recognition from primary structure of proteins. SFN uses support vector machines as memberclassifiers, and adopts All-Versus-All as multi-class categorization. Six groups of features are divided into majorand minor ones by SFN, and several diversity fusion schemes are correspondingly built. The final decision is madeby dynamic selection of the results of all fusion schemes. When it is still difficult to know what kind of fusion offeature groups can achieve good prediction,SFN is a dependable solution by selecting the optimal fusion offeature groups automatically, which can ensure the best recognition. Overall recognition system achieves 61.04%fold prediction accuracy on the independent test dataset. The results and the comparison with other approachesdemonstrate the effectiveness of SFN, and thus encourage its further exploration.
10.The development of a detecting meter for pressure pain threshold.

A new instrument for measuring pressure pain strength to different bodily parts is introduced in this paper. For a new measuring appliance, it can be used conveniently for detection and evaluation of a patient's pressure pain threshold value, and for the research of bodily channels and network vessels. The system uses silicone oil as pressure conducting medium of a transducer, and five pressure detecting probes with different surface areas are designed to meet the requirements of the parts to be measured. The system has digital lock function for easy operation.
